Abstract
本发明公开了一种保加利亚乳杆菌参与酸奶后酸化基因及其应用。本发明通过设计引物LDB_RS05285‑F/R(Ldb1239‑F/R),经PCR扩增后与表达载体进行连接,构建重组质粒,将重组质粒转化进入乳酸菌中,构建重组菌并测定重组菌和对照菌在酸、酸冷处理下存活率,同时将LDB_RS05285基因过表达菌株和对照菌株以相同的菌量分别接种到灭活后的酸奶中,测定酸奶贮藏期的活菌数和pH值的测定,建立了该基因的克隆、表达与功能验证的方法。该基因过量表达可以有效减弱酸奶储存过程中的后酸化问题,提高酸奶储存的稳定性,延长酸奶保质期。为解决酸奶后酸化问题提供了新的思路。

本发明属于生物工程技术领域,具体涉及一种保加利亚乳杆菌后酸化相关 基因LDB_RS05285的克隆及其在酸奶中的应用。
背景技术
酸奶是以鲜牛奶为原料,向其中添加发酵剂和其它配料,经发酵后,再冷 却灌装的一种乳制品。牛奶经乳酸菌发酵后,赋予了牛奶不曾有的功效,如缓 解产生乳糖不耐症。酸奶中的益生菌还在改善人体胃肠道功能、抗肿瘤以及延 缓衰老、促进人体吸收等方面起到重要的保健作用。研究表明,产品质量高的 发酵乳在食用时pH值应为4.2~4.3,然而在发酵乳储存的过程中,产品的pH 值可继续下降,这样就出现发酵乳酸味太重,上边有少量乳清析出,导致酸奶 的感官质量下降等现象,这是由于发酵乳发生后酸化造成的。所谓的发酵乳后 酸化是指酸奶在发酵结束后,储存、运输、销售、食用前这一过程中,菌体仍 在生长繁殖,以致发酵乳由于酸度太高以及感官品质下降而无法被消费者接受 的现象,这种现象被称为酸奶的后酸化。
德式乳杆菌保加利亚亚种(Lactobacillus delbrueckii subsp.bulgaricus)又称 保加利亚乳杆菌,因发酵性能优良被广泛应用到发酵乳制品行业,其可将大分 子的营养物质发酵成小分子的营养成分并且赋予产品特殊的风味,例如可以发 酵乳糖生成易于人体吸收的葡萄糖和半乳糖,在一定程度上可以缓解乳糖不耐 症。此外,L.bulgaricus还具有调节胃肠道、抑制有害菌的生长、增强免疫力 等。然而,L.bulgaricus在发酵工业生产中存在制约行业发展的关键问题,即 后酸化。近年来研究表明,在酸奶的发酵中,控制保加利亚乳杆菌的生长是解 决后酸化的关键,学者们通过巴氏杀菌,菌株诱变和补充抗菌物质等方法来控 制保加利亚乳杆菌的生长和产酸。这些方法都都可以缓解酸乳后酸化的进程,然而效果没有达到绝对抑制,我们前期模拟保加利亚乳杆菌ATCC 11842在酸 奶后酸化过程中所受的环境胁迫条件,通过转录组组学测定了在这一过程中的 基因表达量变化,分析结果表明保加利亚乳杆菌ATCC 11842通过一系列的适 应性调节来应对低pH环境的胁迫作用,主要包括:提高糖酵解途径相关酶的 表达量,提供给细胞ATP;调整丙酮酸代谢产物的去向,使丙酮酸代谢更多的 转向脂肪酸的生物合成,然而这种代谢机制在冷胁迫作用下退出了酸响应机制; 降解错误折叠蛋白,同时提高蛋白质翻译的精确性;DNA的损伤修复等。在 这些途径中我们发现了许多参与到酸奶后酸化过程的基因,通过调控后酸化关 键基因来缓解酸乳的后酸化程度是一个新的思路,还未有相关的报道。
发明内容
本发明的目的在于提供一种来源于保加利亚乳杆菌ATCC 11842的后酸化 相关基因。
本发明的目的还在于提供上述后酸化相关基因在抑制保加利亚乳杆菌酸奶 后酸化过程中的应用。
一种保加利亚乳杆菌后酸化蛋白,是具有下述氨基酸残基序列之一的蛋白:
1)自序列表中SEQ ID NO:1所示的氨基酸残基序列;
2)将自序列表中的SEQ ID NO:1所示的氨基酸残基序列经过一个或几个 氨基酸残基的取代和/或缺失和/或添加且具有抑制后酸化活性的蛋白质。
所述保加利亚乳杆菌后酸化蛋白的基因序列如序列表SEQ ID NO:2和SEQ ID NO:3所示。
一种含上述保加利亚乳杆菌后酸化基因的载体。
含有上述基因载体的工程菌。
扩增权上述的保加利亚乳杆菌后酸化基因中任一片段的引物。
上述保加利亚乳杆菌后酸化蛋白在延缓酸奶后酸化的过程中的应用。
本发明保加利亚乳杆菌后酸化关键基因LDB_RS05285,采用如下方法获得:
(1)将保加利亚乳杆菌ATCC 11842接种与MRS培养基置于42℃培养箱, 培养至对数末期,提取细菌基因组;
(2)设计LDB_RS05285特异引物扩增目的基因,加尾反应,连接pMD19-T 载体,筛选阳性克隆,提取质粒进行酶切验证然后进行测序;
(3)对该基因所编码的蛋白三维结构进行了预测;
(4)运用RT-qPCR技术检测目的基因在酸奶后酸化过程中的表达量;
(5)构建基因过表达载体,通过电转化的方法将重组载体导入宿主菌,筛 选阳性克隆子;
(6)运用RT-qPCR技术检测目的基因在重组菌的表达量;
(7)验证该基因在酸奶后酸化过程中的应用。
本发明的有益效果:本发明克隆的LDB_RS05285基因,通过在乳酸菌中过 量表达合成铜离子转运蛋白以减弱酸奶储存过程中的后酸化问题,提高酸奶储 存的稳定性,延长酸奶保质期,为弱后酸菌株的研发提供良好的保证。

利用高保真酶从提取的乳酸菌基因组(图1中左半图)中分别进行PCR扩 增,95℃预变性5min,95℃变性30s,52℃退火30s,72℃延伸2.5min,共35 个循环,循环结束后72℃延伸10min(PCR扩增结果见图1右半图)。扩增完成 后利用普通Taq酶进行加“A”反应(72℃,10min),反应结束后进行纯化,连 接克隆载体pMD19-T,构建pMD19-LDB_RS05285质粒并转化大肠杆菌DH5a感受 态细胞,筛选阳性克隆菌株,进行测序。
使用Swiss-Model工具对该基因编码的氨基酸序列进行蛋白质的三级结 构预测。
实施例2 LDB_RS05285基因在脱脂乳中发酵不同时期的表达量分析
将保加利亚乳杆菌ATCC 11842接种到脱脂乳培养基中,根据菌株在脱脂乳 培养基中的生长曲线和pH值变化,我们选取发酵初期、发酵对数中后期、发酵 终点、发酵结束后4℃冰箱冷藏2h、7d、14d,6个不同时间节点的发酵脱 脂乳。提取6个时期的总RNA,进行目的基因的相对表达量的测定,通过分析 基因在酸奶贮藏过程中表达量变化表明该基因参与到酸奶的后酸化过程。
实施例3重组表达菌株的构建
确定测序结果准确无误后,提取质粒pMD19-LDB_RS05285和pMG36e质粒 同时用用PstI和SacI对两个质粒分别进行双酶切,回收酶切产物后用T4 DNA Ligase进行连接,再次转入大肠杆菌DH5a感受态细胞,筛选阳性克隆子,进 行测序比对。测序结果正确后,摇菌提取质粒pMG36e-LDB_RS05285,利用电转 仪将重组质粒转入乳酸乳球菌ML23感受态细胞中,30℃培养,挑选转化子进行 PCR验证(图2),电泳检测片段大小正确为重组菌ML23-36e-LDB_RS05285。
实施例4 qRT-PCR检测基因LDB_RS05285在乳酸乳球菌中的表达
试验采用qRT-PCR技术对目基因LDB_RS05285在重组菌株和对照菌内的表 达量进行分析,因L.lactis ML23菌株内也存在与目的基因功能相同且两者的 同源性较高的序列所以在对照菌株内也检测到了目的基因的表达。结果如图 3-5所示,目的基因在重组菌株(ML23-36e-LDB_RS05285)和对照菌株 (ML23-36e)内都有表达且重组菌株的表达量显著高于对照菌株(P<0.05)。
实施例5重组菌株对酸奶后酸化过程的影响
将活化好的保加利亚乳杆菌ATCC 11842接种到脱脂乳培养基中,37℃培养 11h至脱脂乳凝固然后进行巴氏杀菌,对酸奶中的乳酸菌进行灭活,冷却至室 温,将事先培养好的LDB_RS05285基因过表达菌株和对照菌株以相同的菌量分 别接种到灭活后的酸奶中,放在4摄氏度冰箱进行冷藏0d、1d、2d、3d、4d、 5d、6d、7d。按时取样进行活菌数和pH值的测定。结果表明:随着贮藏时间的 延长,活菌数逐渐减少,0-3d下降明显,4-7d趋于平缓(图6)。在贮藏后期, 目的基因过表达的菌株的菌落总数为对照组的92.3%(P>0.05)。在酸奶的贮藏期对照菌株的pH值明显低于目的基因过表达菌株(P<0.05)(图7)。结果表明, LDB_RS05285基因的过量表达一定程度上减弱酸奶储存过程中的后酸化问题, 提高酸奶储存的稳定性,延长酸奶保质期。
