技术领域
本发明属于医用食品领域,具体涉及一种口服全营养组合物及其制备方法,主要用于为炎性肠病人及其他对营养素没有特殊要求的患者提供全面而均衡的营养支持。
背景技术
炎症性肠病(IBD)病人营养不良发生率较高,营养尤其是肠内营养是其重要的治疗措施。要素饮食和非要素饮食对炎症性肠病的疗效无明显差别,整蛋白、短肽及氨基酸单体型制剂各有其特点,应根据病情选择应用。肠内营养治疗炎症性肠病的机制是多方面的,目前营养支持治疗在大多数IBD病人中还是一种“辅助支持治疗”而非针对疾病本身的一种基础治疗,随着临床营养学的发展和对炎症性肠病认识的深入,肠内营养支持将会成为炎症性肠病的基本治疗措施。
根据病人病情选用饮食营养、肠内或肠外营养支持、或营养补充剂等方法,使病人获得适宜的营养,是减少并发症和患者死亡的有效办法。从现有的炎性肠病肠内营养食品的品种和数量来看,有全营养型的、组件型的等几种类型,品种和数量少,且多靠进口。因此,我国炎性肠病营养食品有着很大的市场潜力。
目前,市场上可供选择的适合炎性肠病人食用的肠内营养食品还比较缺乏,开发一种稳定的、便于携带和使用,且制备方法相对简单的口服炎性肠病人用全营养配方组合物,用来为炎性肠病人及其他对营养素没有特殊要求的患者提供全面而均衡的营养支持尤为重要。

本发明主要涉及一种口服炎性肠病人用全营养配方组合物及其制备方法,具体如下:
一种口服全营养组合物,其特征在于所述组合物含有非脂溶性成分,脂溶性成分和纯化水,其中每1000mL所述组合物含有的非脂溶性成分为蛋白质30-80g、碳水化合物150-200g、钠0.5-2g、氯0.4-2.0g、镁0.01-0.6g、磷0.5-1.5g、钾1-4g、钙0.3-2g、铁0.005-0.05g、锌0.001-0.05g、铜0-0.01g、锰0-0.01g、硒0.01-0.5mg、碘0-1.5mg、柠檬酸0-15g、水溶性维生素0.1-2g、甜味剂0-0.5g、稳定剂2-15g;含有的脂溶性成分为脂肪10-30g、脂溶性维生素0.02-1g、乳化剂2-18g。
进一步地,所述蛋白质选自耐酸水解蛋白、中性水解蛋白或其混合;碳水化合物选自麦芽糊精、果糖、淀粉或其混合;甜味剂选自三氯蔗糖、甜菊糖苷、赤藓糖醇、木糖醇或其混合;稳定剂为食用胶;水溶性维生素选自泛酸、生物素、维生素K1、叶酸、维生素C、维生素B1、维生素B2、烟酸、维生素B6、维生素B12或其混合;脂肪选自葵花籽油、大豆油、红花籽油、中链甘油三酯或其混合,优选葵花籽油与中链甘油三酯混合;脂溶性维生素选自维生素E、维生素A、维生素D或其混合;乳化剂为变性淀粉。
一种口服全营养组合物,其特征在于所述组合物含有脂溶性成分,非脂溶性成分和纯化水,其中每1000mL上述组合物含有的非脂溶性成分为水解蛋白30-80g、麦芽糊精150-200g、磷酸氢二钾3-8g、柠檬酸钠2-7g、乳酸钙0-5g、氯化镁0.3-5g、焦磷酸铁20-80mg、硫酸锌20-100mg、***钠200-750μg、碘化钾0-2mg、硫酸锰0-5mg、硫酸铜5-20mg、泛酸0-30mg、生物素0-20mg、叶酸0-20mg、维生素K1 0-2mg、维生素C 100-300mg、维生素B1 1-5mg、维生素B2 1-5mg、烟酸10-20mg、维生素B6 1-6mg、维生素B12 0.001-0.3mg、柠檬酸0-15g、甜味剂0-0.5g、稳定剂2-15g;脂溶性成分为葵花籽油5-15g、中链甘油三酯5-15、维生素E 20-800mg、维生素A 0.1-4mg、维生素D 20-60μg、乳化剂2-18g。

进一步地,如上所述组合物中焦磷酸铁为乳化焦磷酸铁。
同时,本发明还提供了如上所述组合物的制备方法,包含以下步骤:
步骤1:配料
a)按配方分别称量非脂溶性成分,待用;
b)按配方分别称量脂溶性成分,待用;
步骤2:溶解
c)将称得的脂溶性成分加热搅拌至全溶,得到油相溶液;
d)将称得的非脂溶性成分蛋白质及碳水化合物、稳定剂干混,用配制总量60%的水50-80℃水温剪切搅拌至全溶;
e)向步骤d)溶液中加入称得的除水溶性维生素外的剩余非脂溶性成分,搅匀;
步骤3:以高剪切转速向步骤e)溶液中缓慢加入步骤c)溶液,继续循环剪切30min,剪切结束前5min加入用水溶解的水溶性维生素,剪切均匀;
步骤4:磷酸氢二钾调节产品pH为6.0-6.9或柠檬酸调节产品pH为3.7-4.7,定容至配制总体积,制得初乳;
步骤5:将步骤4制得的初乳自循环;定容至总体积,冷却到20℃以下;
步骤6:灌装,灭菌,温度≥100℃,灭菌时间≥8分钟,或UHT 130~135℃灭菌10~30秒,灌装。
另外,本发明还提供了如上所述的组合物用于为炎性肠病人及其他对营养素没有特殊要求的患者提供全面而均衡的营养支持的应用。
本发明在研发过程中发现,各成分如乳化剂等的选择对产品稳定性影响很大,乳化剂选择不当则会产生大量沉淀和或浮油,出现分层现象,严重则导致产品变性凝固。
1)乳化剂的选择:
结果显示使用变性淀粉作为乳化剂时产品状态更稳定,加速(温度40℃,湿度:70%)放6个月产品呈均一液体,底部有少量沉淀,但摇晃即消失。
2)钙的选择
钙的来源对产品的稳定性影响也很大,除部分来源于蛋白之外,必要时还需添加其他钙源,本发明对钙源氯化钙、碳酸钙、磷酸氢钙、乳酸钙进行制样筛选:
结果显示筛选出的乳酸钙和磷酸氢钙效果优于其他两种,使用其制备样品,加速(温度40℃,湿度:70%)放置6个月,结果显示选择乳酸钙的产品状态均一,包装瓶底部仅有少量沉淀,稍一晃动即消失,比加入磷酸氢钙的产品更稳定,因此选择乳酸钙作为钙源。
3)本发明在研发期发现,产品灭菌后包装瓶底部有少量黑色碳粉状沉淀物,影响产品质量,经对矿物盐筛查,发现该沉淀物是由铁盐造成的,利用包埋的乳化焦磷酸铁代替焦磷酸铁纯粉,制备的样品灭菌后则可避免黑色碳粉状沉淀物的产生,因此最终选用经包埋的乳化焦磷酸铁。
本发明是一款短肽型预消化配方组合物,是一款根据炎性肠病人营养代谢特点设计的全营养炎性肠病人食用的肠内营养混悬剂(TPF-DM),以水解蛋白为蛋白源,无需消化,直接吸收利用,直接营养肠粘膜,快速维护肠屏障,高效补充蛋白质,在对炎性肠病人的治疗中提供营养支持的同时能更好减少并发症的发生;另外,本发明针对病人对pH不同需求,设计了中性及偏酸性两种产品,其中酸性产品更有利于病人消化吸收,但有些病人由于体质偏酸或胃酸过多则不适合饮用酸性的产品,因此可根据病人的需求选择适合的产品。
本发明中组合物中各成分含量符合规定,通过实验研究选用适合的乳化剂、钙源及稳定剂,同时根据病人需求设计酸性及中性两种产品,具有稳定性高,便于携带和使用等优点;另外其制备方法简单,成本低,更便于工业化大生产。
具体实施方式
下面将通过实施例对本发明作进一步的描述,这些描述并不是对本发明内容作进一步的限定。本领域的技术人员对发明内容所作的等同替换或改进,仍属于本发明的保护范围之内。
实施例1:
制备工艺:
步骤1:配料
a)按配方分别称量非脂溶性成分中性水解蛋白、麦芽糊精、磷酸氢二钾、柠檬酸钠、乳酸钙、氯化镁、乳化焦磷酸铁、硫酸锌、***酸钠、碘化钾、硫酸锰、硫酸铜、泛酸、生物素、叶酸、维生素K1、维生素C、维生素B1、维生素B2、烟酸、维生素B6、维生素B12、三氯蔗糖、食用胶,待用;
b)按配方分别称量脂溶性成分葵花籽油、中链甘油三酯、变性淀粉,待用;
步骤2:溶解
c)将称得的脂溶性成分葵花籽油、中链甘油三酯、维生素E、维生素A、维生素D、变性淀粉加热搅拌至全溶,得到油相溶液;
d)将称得的非脂溶性成分蛋白质及碳水化合物、食用胶干混,用配制总量60%的水50-80℃水温剪切搅拌至全溶;
e)向步骤d)溶液中加入称得的除水溶性维生素外的剩余非脂溶性成分,搅匀;
步骤3:以高剪切转速向步骤e)溶液中缓慢加入步骤c)溶液,继续循环剪切30min,剪切结束前5min加入用水溶解的水溶性维生素,剪切均匀;
步骤4:磷酸氢二钾调节产品pH为6.0-6.9,定容至配制总体积,制得初乳;
步骤5:将步骤4制得的初乳自循环;定容至总体积,冷却到20℃以下;
步骤6:灌装,灭菌,温度≥100℃,灭菌时间≥8分钟,或UHT 130~135℃灭菌10~30秒,灌装。

Claims (10)
1.一种口服全营养组合物,其特征在于所述组合物含有非脂溶性成分,脂溶性成分和纯化水,其中每1000mL所述组合物含有的非脂溶性成分为蛋白质30-80g、碳水化合物150-200g、钠0.5-2g、氯0.4-2.0g、镁0.01-0.6g、磷0.5-1.5g、钾1-4g、钙0.3-2g、铁0.005-0.05g、锌0.001-0.05g、铜0-0.01g、锰0-0.01g、硒0.01-0.5mg、碘0-1.5mg、柠檬酸0-15g、水溶性维生素0.1-2g、甜味剂0-0.5g、稳定剂2-15g;含有的脂溶性成分为脂肪10-30g、脂溶性维生素0.02-1g、乳化剂2-18g。
2.根据权利要求1所述的组合物,其特征在于所述蛋白质选自耐酸水解蛋白、中性水解蛋白或其混合;碳水化合物选自麦芽糊精、果糖、淀粉或其混合;甜味剂选自三氯蔗糖、甜菊糖苷、赤藓糖醇、木糖醇或其混合;稳定剂为食用胶;水溶性维生素选自泛酸、生物素、维生素K1、叶酸、维生素C、维生素B1、维生素B2、烟酸、维生素B6、维生素B12或其混合;脂肪选自葵花籽油、大豆油、红花籽油、中链甘油三酯或其混合,优选葵花籽油与中链甘油三酯混合;脂溶性维生素选自维生素E、维生素A、维生素D或其混合;乳化剂为变性淀粉。
3.一种口服全营养组合物,其特征在于所述组合物含有脂溶性成分,非脂溶性成分和纯化水,其中每1000mL所述组合物含有的非脂溶性成分为中性水解蛋白或耐酸水解蛋白30-80g、麦芽糊精150-200g、磷酸氢二钾3-8g、柠檬酸钠2-7g、乳酸钙0-5g、氯化镁0.3-5g、焦磷酸铁20-80mg、硫酸锌20-100mg、***钠200-750μg、碘化钾0-2mg、硫酸锰0-5mg、硫酸铜5-20mg、泛酸0-30mg、生物素0-20mg、叶酸0-20mg、维生素K1 0-2mg、维生素C 100-300mg、维生素B11-5mg、维生素B21-5mg、烟酸10-20mg、维生素B61-6mg、维生素B120.001-0.3mg、柠檬酸0-15g、甜味剂0-0.5g、稳定剂2-15g;脂溶性成分为葵花籽油5-15g、中链甘油三酯5-15、维生素E 20-800mg、维生素A 0.1-4mg、维生素D 20-60μg、乳化剂2-18g。
4.根据权利要求3所述的组合物,其特征在于每1000mL所述组合物含有的非脂溶性成分为中性水解蛋白40g、麦芽糊精186g、磷酸氢二钾4.5g、柠檬酸钠3.622g、乳酸钙3.0g、氯化镁2.3g、焦磷酸铁50mg、硫酸锌60mg、***钠400μg、碘化钾1.3mg、硫酸锰2.4mg、硫酸铜10mg、泛酸20mg、生物素10mg、叶酸10mg、维生素K1 0.5mg、维生素C 240mg、维生素B12mg、维生素B22mg、烟酸18mg、维生素B6 1.6mg、维生素B120.0018mg、甜味剂0.2g、稳定剂8g;含有的脂溶性成分为葵花籽油8g、中链甘油三酯8g、维生素E 40mg、维生素A 2mg、维生素D 40μg、乳化剂8g。
5.根据权利要求3所述的组合物,其特征在于每1000mL所述组合物含有的非脂溶性成分为中性水解蛋白40g、麦芽糊精186g、磷酸氢二钾4.5g、柠檬酸钠3.622g、乳酸钙3.0g、氯化镁0.34g、焦磷酸铁50mg、硫酸锌60mg、***钠400μg、维生素C 240mg、维生素B12mg、维生素B22mg、烟酸18mg、维生素B6 1.6mg、维生素B120.0018mg、甜味剂0.2g、稳定剂8g;脂溶性成分为葵花籽油8g,中链甘油三酯8g、维生素E40mg、维生素A 1mg、维生素D 40μg、乳化剂8g。
6.根据权利要求3所述的组合物,其特征在于每1000mL所述组合物含有的非脂溶性成分为耐酸水解蛋白40g、麦芽糊精186g、磷酸氢二钾0.83g、柠檬酸钠2.85g、乳酸钙3.0g、氯化镁2.3g、焦磷酸铁50mg、硫酸锌60mg、***钠400μg、碘化钾1.3mg、硫酸锰2.4mg、硫酸铜10mg、泛酸20mg、生物素10mg、叶酸10mg、维生素K1 0.5mg、维生素C 240mg、维生素B12mg、维生素B22mg、烟酸18mg、维生素B6 16mg、维生素B120.0018mg,柠檬酸13g、甜味剂0.2g、稳定剂8g;含有的脂溶性成分为葵花籽油8g、中链甘油三酯8g、维生素E 40mg、维生素A 2mg、维生素D 40μg、乳化剂8g。
7.根据权利要求3所述的组合物,其特征在于每1000mL所述组合物含有的非脂溶性成分为耐酸水解蛋白40g、麦芽糊精186g、磷酸氢二钾0.83g、柠檬酸钠2.85g、乳酸钙3.0g、氯化镁0.34g、焦磷酸铁50mg、硫酸锌60mg、***钠400μg、维生素C 240mg、维生素B12mg、维生素B22mg、烟酸18mg、维生素B6 1.6mg、维生素B120.0018mg、柠檬酸13g、甜味剂0.2g、稳定剂8g;脂溶性成分为葵花籽油8g、中链甘油三酯8g、维生素E 40mg、维生素A 1mg、维生素D40μg、乳化剂8g。
8.根据权利要求3-7任一项所述的组合物,其特征在于所述焦磷酸铁为乳化焦磷酸铁。
9.一种权利要求1-8任一项所述组合物的制备方法,其特征在于包含以下步骤:
步骤1:配料
a)按配方分别称量非脂溶性成分,待用;
b)按配方分别称量脂溶性成分,待用;
步骤2:溶解
c)将称得的脂溶性成分加热搅拌至全溶,得到油相溶液;
d)将称得的非脂溶性成分蛋白质及碳水化合物、稳定剂干混,用配制总量60%的水50-80℃水温剪切搅拌至全溶;
e)向步骤d)溶液中加入称得的除水溶性维生素外的剩余非脂溶性成分,搅匀;
步骤3:以高剪切转速向步骤e)溶液中缓慢加入步骤c)溶液,继续循环剪切30min,剪切结束前5min加入用水溶解的水溶性维生素,剪切均匀;
步骤4:磷酸氢二钾调节产品pH为6.0-6.9或柠檬酸调节产品pH为3.7-4.7,定容至配制总体积,制得初乳;
步骤5:将步骤4制得的初乳自循环;定容至总体积,冷却到20℃以下;
步骤6:灌装,灭菌,温度≥100℃,灭菌时间≥8分钟,或UHT 130~135℃灭菌10~30秒,灌装。
10.权利要求1-8任一项所述的组合物用于为炎性肠病人及其他对营养素没有特殊要求的患者提供全面而均衡的营养支持的应用。
