豆科植物主要抗霉菌感染的防御性反应是在感染部位的异黄酮(isoflavonoid)植物抗毒素的快速累积,异黄酮植物抗毒素是一种抗菌素。(R.A.Dixon;Biol.Rev.;1986,61,239-291)。这里,查尔酮合成酶和苯丙氨酸解氨酶的基因是与植物抗毒素生物合成有关的酶。(G.J.Loake;A.D.Choudhary;M.J.Harrison;M.Mavandad;C.J.Lamb;R.A.Dixon;The Plant Cell;1991,3,829-840),肉桂酸是L-苯丙氨酸与苯丙氨酸解氨酶反应时产生苯基丙酸的中间体,当用低浓度的肉桂酸处理时,发现上述酶被刺激,但当用高浓度的肉桂酸处理时,发现上述酶被抑制。(G.J.Loake;A.D.Choudhary;M.J.Harrison;M.Mavandad;C.JLamb;R.A.Dixon;The Plant Cell;1991,3,829-840)。另外,肉桂酸是产生于上述过程中的一种产物,而且已知它总是存在于豆科植物中。
在检测公开于韩国专利No.93-1374的N-肉桂酰苯胺对豆芽的消毒效果的过程中,发现肉桂酰苯胺转化成肉桂酸。基于这些材料,制备了五种肉桂酸类化学物质如肉桂酸,肉桂酸钠盐,肉桂酸钾盐,肉桂酸甲酯和肉桂酸乙酯,并测定这些化学物质的消毒效果,用上述化学物质培育豆芽,然后比较和检测它们的消毒效果。在此实验中使用商业上可得的肉桂酸或根据已知方法可制备的肉桂酸(Adams;“Organic Reaction”Coll Vol 1,p.25l,John Wiley and Sons,New York,1942)。所使用的肉桂酸盐是通过肉桂酸与碳酸钠或碳酸钾反应制备的。至于实验中使用的肉桂酸甲酯和肉桂酸乙酯的制备,是通过肉桂酸与醇在酸性催化剂存在下反应制备的。
上述肉桂酸类是韩国健康和社会事务部公布的列于允许使用的食品添加剂通知单中的安全化学物质,而且特别是,肉桂酸是在豆科植物中生物合成的天然物质。吸收到豆芽中的肉桂酸酯会分解并最终转化为肉桂酸,肉桂酸盐易溶于水并因此容易处理。到目前为止,这些化学物质只是被化学合成或从生物化学的角度研究了它们对酶的反应活性,但它们从没有被用作消毒剂。
为测定肉桂酸类的消毒效果,实验在三个因素存在下进行。下面表1列出了1kg干豆在最初浸泡在水中时吸收化学物质的浓度达到0.75-15ppm所需的各个化学物质的用量。豆子吸收的化学物质的浓度0.75-15ppm在豆芽生长过程中会逐渐降低,直到在豆芽培育的最后阶段达到。0.1-2.0ppm的肉桂酸浓度。这里,尽管起始吸收物质分别是不同的,但最后所剩的物质都是肉桂酸。

由于肉桂酸存在顺反异构体,它的加速植物生长的植物生长素作用是内部竞争性抑制的,因此可在豆芽生长过程中引起生长抑制作用(Morris,K.A.;Arthur,E.D.Phytochemistry 1984,23,2136),但在上述浓度的化学物质时,未发现生长抑制作用。
当肉桂酸类的浓度增加时,出现生长抑制作用,而且由于豆芽的培育者愿意使用大量化学物质而不是特定量,由于生长抑制引起的影响就应被考虑。
为了解决上述问题,使用了不同的生长控制剂,其结果是,N-苄基腺嘌呤可作为添加物。即,使用这种化学物质,不会产生细根,而得到平满的软的根(韩国专利公开81-1405,81-1406,93-1374)。
本发明的发明者测定了五种肉桂酸类化学物质具有消毒效果同时无生长抑制作用发生的肉桂酸类浓度范围。肉桂酸类在豆子浸泡在水中的初始阶段的合适用量列于实施例3和实施例4的表3和表4中,研究结果发现,不管肉桂酸类化学物质的类型如何,优选的化学物质吸收浓度为0.75~15ppm/重量单位豆子。以肉桂酸为例,当1kg干豆子吸收3mg化学物质时,消毒作用很弱,但是当吸收100mg化学物质时,生长抑制作用变强,当吸收20mg化学物质时,由于严重的生长抑制已证明是不可用的。
第二,本发明者测定了与有消毒效果且无生长抑制的N-苄基腺嘌呤混合的肉桂酸的用量范围。N-苄基腺嘌呤的用量是4~11mg(韩国专利公开93-1374),且相应的肉桂酸类的浓度范围列于表2。以肉桂酸为例,即使使用3mg肉桂酸,也显示出消毒效果。最佳的浓度范围是5~30mg肉桂酸和4~11mg苄基腺嘌呤/1kg干豆,但即使使用150mg肉桂酸,也没有引起生长抑制。

至于化学物质的制备方法,当只使用肉桂酸类时,将3~60mg肉桂酸(1kg干豆所需的用量)与表面活性剂如羧甲基纤维素钠(以下称NaCMC)混合制备水溶液,并将肉桂酸溶于含Na2CO3或K2CO3水溶液中。另一方面,如实施例中那样,当合适量的肉桂酸酯与表面活性剂如NaCMC混合后,所述化学物质很容易溶于水。当肉桂酸类与苄基腺嘌呤混合时,将肉桂酸类和苄基腺嘌呤与NaCMC混合制成水溶液,当只用肉桂酸类时,将肉桂酸和苄基腺嘌呤溶于乙醇作为处理化学物质。
豆芽作为作物可全年生产,并依据温度,水温或其他所需条件培育到适宜的尺寸,一般在晚春干燥时培育易出现一些问题。因此,实验重点在五月初到六月中进行。用three-kwan容器(即3×3.75kg=11.25kg)培育豆芽。在塑料容器的底部钻许多孔以便水可自由地慢慢流。下面将详述培育豆芽的每一步。
在水中浸泡豆子:将1.5Kg干豆子装入上述容器,每三小时向其中加入足够的水。然后将豆子放置24小时使其重量加倍。如上所述制备的化学物质在此过程中用3L水稀释。尽管水的量很少,但水慢慢通过容器底部的孔使得水再次喷淋到豆子上而达到最大程度的吸收效果。
将豆子放入另一容器培育:将经水浸泡的豆子转移到另一容器用于培育豆芽。这时,将按上述过程制备的化学物质如上用水稀释喷淋到豆子上。
培育豆芽:为了培育豆芽,首先用厚布将光线挡住以避免光线进来。豆芽生长直至生长到约11cm,且从开始的1.5Kg干豆生长至重量达到约9.0Kg豆芽。这六至七天,可根据相应的培育实验目的在适宜的时间向1.5Kg干豆子上喷淋所需的水,然后比较和检查结果。
下面将详述依据本发明的培育方法的一些实施例。
实施例1(化学物质的制备)
例1. 14.8mg肉桂酸和45mgNaCMC溶于1ml水中。所得化学物质用于1Kg干豆。
例2. 14.8mg肉桂酸和10.6mgNa2CO3溶于1ml水中。所得化学物质用于1Kg干豆。
例3. 14.8mg肉桂酸和13.8mgK2CO3溶于1ml水中。所得化学物质用于1Kg干豆。
例4. 16.2mg肉桂酸甲酯和45mgNaCMC溶于1ml水中。所得化学物质用于1Kg干豆。
例5. 17.6mg肉桂酸乙酯和45mgNaCMC溶于1ml水中。所得化学物质用于1Kg干豆。
例6. 14.8mg肉桂酸和5mg苄基腺嘌呤溶于1ml水中。所得化学物质用于1Kg干豆。
例7. 17.0mg肉桂酸钠盐,5mg苄基腺嘌呤和45mgNaCMC溶于1ml水中。所得化学物质用于1Kg干豆。
例8. 18.6mg肉桂酸钾盐,5mg苄基腺嘌呤和45mgNaCMC溶于1ml水中。所得化学物质用于1Kg干豆。
例9. 16.2mg肉桂酸甲酯,5mg苄基腺嘌呤和45mgNaCMC溶于1ml水中。所得化学物质用于1Kg干豆。
例10. 17.6mg肉桂酸乙酯,5mg苄基腺嘌呤和45mgNaCMC溶于1ml水中。所得化学物质用于1Kg干豆。
实施例2
将豆子浸泡在水中:将1.5Kg干豆子装入容器,每三小时向其中加入足够的水。然后将豆子放置24小时使其重量加倍。然后使用将上述化学物质用3L水稀释,喷淋到豆子上。
在另一容器中培育豆芽:将上述经水浸泡的豆子转移到另一容器用于培育豆芽。然后用厚布将光线挡住以避免光线进来。豆芽生长六至七天,直至生长到约11cm,且从开始的1.5Kg干豆生长至重量达到约9.0Kg豆芽。
使用实施例中例1-5制备的不同化学物质配方应用到1Kg干豆上,并进行实施例2的过程,检测化学物质的消毒效果*1,结果列于表3。

Claims (4)
2.一种培养豆芽和绿豆芽的方法,包括如下步骤:
(i)在水中浸泡豆子或绿豆;
(ii)将浸泡过的豆子或绿豆放入用于培育豆芽的另一容器中;和
(iii)进行豆芽的培育;
其特征在于,在步骤(ii)中,用其中溶解有作为消毒剂的式(1)所示化合物的水喷淋豆子或绿豆,其中式(1)所示化合物的用量为2.5-200mg/kg干豆子:
R=H,Na,K,CH3,C2H5
3.根据权利要求1或2所述的方法,其特征在于式(1)所示化合物的用量为3-100mg/kg干豆子。
4.根据权利要求1或2所述的方法,其特征在于除了使用式(1)所示化合物作为消毒剂外,每1千克干豆子还使用4-11mg的苄基腺嘌呤作为消毒剂。
