Abstract

本发明涉及植物组培快繁技术领域,具体公开了一种昙花脱毒种苗的工厂化快繁方法,包括如下步骤:(1)脱毒种苗增殖培养,(2)脱毒种苗生根培养,(3)炼苗驯化,(4)移栽。本发明首次建立了昙花脱毒快繁的方法,不仅能在短时间内扩繁出大量优良昙花脱毒种苗,而且还避免了昙花扦插繁殖所带来的品种退化问题,而且操作简单、成本低、繁殖效率高、易于工厂化推广,具有较高的生产实用价值。

技术领域
本发明涉及一种昙花脱毒种苗的工厂化快繁方法,具体涉及一种能够在较短时间内繁殖出大量昙花脱毒种苗的扩繁方法,属于园林科学技术领域。
背景技术
昙花(学名:Epiphyllum oxypetalum (DC.)Haw),别名琼花、月下美人,属仙人掌科、昙花属灌木状木质植物。主茎呈细圆柱形,分枝***呈扁平叶状,花多为白色、大型、漏斗状、有芳香。原产墨西哥及中、南美洲的热带森林中,性喜温暖湿润和半阴环境,现在世界各地区广泛栽培,在中国各省区也常见栽培。昙花一般常在夏、秋晚间开放,持续时间较短,一般为2~3h,所以有“昙花一现”之说。昙花株型挺立,叶大肥厚鲜绿,花大美丽而不失优雅,因开花时间短,难以欣赏到,尤显珍贵,故受很多百姓和花农的喜爱。
昙花不仅可供观赏,而且花、茎叶均可入药,其茎叶具有软便去毒,清热疗喘的功效,主治大肠热症、便秘便血、肿疮、肺炎、痰中有血丝、哮喘等症。花朵具有的很强的治疗功效,可以治疗心悸、肺痨、失眠、肺热咳嗽等症,同时可以使高血压和脂肪过高的症状得到缓解。此外昙花还具有改善环境的作用,昙花可以增加室内的负离子含量,让室内的空气清新怡人。因此昙花栽培在观赏、医药、环保等领域有着广泛的应用前景和很高的经济价值。
目前市场上的昙花栽种方式主要为扦插繁殖,这种方法属于常规无性繁殖方法,其弊端是繁殖系数低,无法满足产业化生产的要求,并且随着繁殖代数的增加,病毒在植株体内积累,使种质严重退化,产量和品质大幅下降,商品价值也大大降低。植物病毒分布广、繁殖快、防治难,有植物“癌症”之称。已知的对昙花危害较大的病毒有仙人掌X病毒、黄瓜花叶病毒(CMV)、烟草花叶病毒(TMV)等,感染病毒的植株通常表现为花叶***或局部坏死,基部变褐或全株萎蔫、矮化、畸形或***斑驳等症状。
通过植物组培脱毒技术来生产脱毒组培苗可以实现防治目的, 可以消除危害植物的病毒、植物菌原体、类细菌对植株的危害,使植株原来的一些优良性状重新表现出来,同时由于消除了病毒的影响,植物的生长性能大大增强,能有效提高植物生长指标和经济指标。植物组织培养脱毒方法有茎尖培养脱毒、热处理脱毒、冷处理脱毒、化学药剂处理脱毒、花药培养脱毒、愈伤组织脱毒、珠心胚培养脱毒、茎尖微体嫁接脱毒等,其中由于茎尖培养脱毒效果好, 是目前植物无病毒苗培育应用最广泛、最重要的一个途径。
利用组织培养、离体快繁等技术可以解决昙花病毒病积累和快速推广的问题。目前国内对昙花的研究和应用仍处于起步阶段,对于昙花的组培脱毒快繁方法在文献上也未见报道,因此严重影响了昙花种苗工厂化、规模化生产。本发明旨在建立一种简单经济的昙花脱毒种苗的工厂化快繁方法,对促进昙花脱毒种苗工厂化、规模化生产,对昙花优良品种的推广具有广阔的市场前景。
发明内容
为了解决目前缺乏昙花脱毒苗大规模快速繁殖方法的问题,本发明提供了一种昙花脱毒种苗的工厂化快繁方法,通过建立健全了培养程序、培养条件和培养组分的配比,提高了脱毒种苗的增殖效率、移栽成活率,从而达到了建立高品质和高数量快繁体系的目的。本发明具体技术方案如下:
一种昙花脱毒种苗的工厂化快繁方法,其特征在于,包括以下操作步骤:
(1)脱毒种苗增殖培养:
将已获得的昙花脱毒种苗在无菌条件下接种于增殖培养基中进行培养,培养室温度控制为22~26℃、光照强度控制为1500-2000Lux、相对湿度控制为60~70%、光周期控制为光12h/暗12h,待种苗长至3~4厘米大小并且长出新的不定芽从时,将不定芽逐个完整的切下,再转入新的增殖培养基中继续增殖,每28天增殖1代,增殖4~6代后可获得大量脱毒幼苗;
(2) 脱毒种苗生根培养:
将步骤(1)获得的脱毒幼苗,转移到生根培养基上进行培养,控制光照强度为1500~2000Lux,环境温度为22~26℃,相对湿度为65~75%,培养18~23天长成生根的试管苗;
(3)炼苗驯化:
当步骤(2)中试管苗株高长到 5cm~8cm,具有4~5条根系且根长3cm~5cm时,将植物组织培养瓶的无菌瓶盖打开,将组织培养瓶放在光照培养架上培养,逐渐将室内恒温空调关闭,连续炼苗7d左右,然后将试管苗从组培瓶中移出并洗净根部培养基,移栽到消毒后的驯化基质中,定期浇水并施加特定的营养液,移至室外遮阳网下生长;
(4)移栽:
待步骤(3)中的试管苗在驯化基质上生长2-3周后,再移栽到消毒后的栽培基质中,栽培密度为株行距18 cm×25 cm,在室外自然光照、温度条件下常规栽培管理。
所述步骤(1)中增殖培养基的配方为:硝酸钾2400~2600mg/L、磷酸二氢铵310~330mg/L、硫酸镁192~200mg/L、无水氯化钙295~315mg/L、碘化钾0.8~0.9mg/L、硼酸4.0~5.0mg/L、一水硫酸锰10.0~12.0mg/L、硫酸锌0.8~1.2mg/L、钼酸钠0.11~0.14mg/L、五水硫酸铜0.23~0.27mg/L、氯化钴0.09~0.12mg/L、乙二胺四乙酸铁钠19.6~20.0mg/L、肌醇90~110 mg/L、维生素B6 0.4~0.6 mg/L、维生素B1 0.2~0.4 mg/L、维生素 C 0.4~0.6mg/L、烟酸0.2~0.5 mg/L、甘氨酸1.4~1.7 mg/L、蛋白胨0.7~0.9mg/L、酵母浸提液45~55ml/L、三十烷醇0.08~0.11mg/L、胺鲜脂0.45~0.55mg/L、激动素0.17~0.22mg/L、异戊烯基腺嘌呤0.18~0.23mg/L、白砂糖 25~30g/L、卡拉胶8~12g/L,pH调节为5.6~6.0。
所述步骤(2)中生根培养基的配方为:硝酸钾 1100~1300mg/L、硝酸钙170~185mg/L、磷酸二氢钾150~170mg/L、硝酸铵440~465mg/L、硫酸镁91~94mg/L、碘化钾0.3~0.5mg/L、氯化钾28~34mg/L、硼酸1.0~1.3mg/L、七水硫酸锰14.0~17.0mg/L、硫酸锌0.5~1.0mg/L、钼酸钠0.10~0.15mg/L、硫酸铁12.0~15.0mg/L、氯化钴0.10~0.14mg/L、乙二胺四乙酸铁钠16.0~20.0mg/L、肌醇90~110 mg/L、维生素B6 0.4~0.6 mg/L、维生素B1 0.2~0.4 mg/L、维生素 C 0.4~0.6mg/L、烟酸0.2~0.5 mg/L、甘氨酸1.4~1.7 mg/L、蛋白胨0.7~0.9mg/L、活性炭 2~5g/L、酵母浸提液45~55ml/L、茉莉酸甲酯 0.02~0.05mg/L、氯化胆碱0.2~0.4mg/L、白砂糖25~30g/L 、花生壳粉8~11g/L、卡拉胶3~5g/L,pH调节为5.6~6.0。
所述步骤(3)中特定的营养液的配方为过磷酸钙0.8g/L、硝酸钙0.5g/L、硝酸钾0.9g/L、尿素0.3 g/L、硫酸镁0.28g/L、硫酸铁0.12g/L、硼酸0.06mg/L、硫酸锰0.03mg/L、硫酸锌0.06mg/L、硫酸铜0.06mg/L、钼酸铵0.06mg/L、甘露醇1.0~1.2 mg/L、氯化胆碱1.7~2.3mg/L、亚精胺3.2~3.7mg/L 。
所述步骤(3)中驯化基质消毒方法为:用75%百菌清可湿性粉剂500倍液进行消毒,驯化基质按重量配比为蛭石20~25g、活性炭2~5g、蔗渣 18~20g、油茶壳13~20g、椰子壳15~20g、玉米秸秆25~30g、谷壳灰30~35g、腐殖土16~24g。
所述步骤(4)中栽培基质消毒方法为用75%百菌清可湿性粉剂500倍液进行消毒,栽培基质按重量配比为:蛭石10~15g、蔗渣18~22g、油茶壳11~20g、木屑13~18g、玉米秸秆20~25g、谷壳灰30~40g、虾壳粉15~18 g、腐殖土45~50g。
本发明的有益效果是 :
1.本发明使用了一些价格较为低廉的原材料,从而降低了生产成本;
2.本发明改良了培养基配方,使用了高效植物激素并合理调节激素用量及相互间的配比,大大提高昙花脱毒种苗的增殖率和生根率,最终扩繁效率提高了70%左右;
3.本发明不仅提供了昙花脱毒种苗的快繁方法,而且获得的种苗质量高,弱苗或死苗率仅有1.5%左右;种苗的移栽成活率达到98%以上。
综上所述,本发明能充分满足产业化要求,即生产成本低、生产周期短、操作简便、种苗增殖率高、移栽成活率高、苗壮有活力,从而更高效、节能的达到扩繁效果,适合大规模的推广应用。

以下具体的采用不同实施例进行对比,进一步证明本发明的效果:
实施例1
本实施例的昙花脱毒种苗的工厂化快繁方法,实施时间为2017年至2018年,实施地点为江苏省东海县花卉试验基地,详细步骤如下:
(1)脱毒种苗增殖培养:
将已获得的昙花脱毒种苗在无菌条件下接种于增殖培养基中进行培养,培养室温度控制为22~26℃、光照强度控制为1500-2000Lux、相对湿度控制为60~70%、光周期控制为光12h/暗12h,待种苗长至3~4厘米大小并且长出新的不定芽从时,将不定芽逐个完整的切下,再转入新的增殖培养基中继续增殖,每28天增殖1代,增殖5代后可获得大量脱毒幼苗。所用增殖培养基配方为:硝酸钾 2550mg/L、磷酸二氢铵326mg/L、硫酸镁195mg/L、无水氯化钙300mg/L、碘化钾0.83mg/L、硼酸5.0mg/L、一水硫酸锰12.0mg/L、硫酸锌1.0mg/L、钼酸钠0.13mg/L、硫酸铜0.27mg/L、氯化钴0.10mg/L、乙二胺四乙酸铁钠19.80mg/L、肌醇100 mg/L、维生素B6 0.5mg/L、维生素B1 0.1 mg/L、维生素C 0.5 mg/L、烟酸0.3mg/L甘氨酸1.5mg/L、蛋白胨0.7~0.9mg/L、活酵母浸提液50ml/L、三十烷醇0.1mg/L 、胺鲜脂0.5mg/L、 激动素 0.2mg/L、异戊烯基腺嘌呤0.2mg/L、白砂糖 27g/L、卡拉胶10g/L,pH调节为5.7。
(2)脱毒种苗生根培养:
将步骤(1)获得的脱毒幼苗,转移到生根培养基上进行培养,控制光照强度为1500~2000Lux,环境温度为22~26℃,相对湿度为65~75%,培养18~23天长成生根的试管苗。所用生根培养基的配方为;硝酸钾 1200mg/L、硝酸钙175mg/L、磷酸二氢钾160mg/L、硝酸铵450mg/L、硫酸镁92.5mg/L、碘化钾0.4mg/L、氯化钾30mg/L、硼酸1.0mg/L、七水硫酸锰16.0mg/L、硫酸锌0.8mg/L、钼酸钠0.13mg/L、硫酸铁14.0mg/L、氯化钴0.10mg/L、乙二胺四乙酸铁钠19.0mg/L、肌醇 100 mg/L、维生素 B6 0.5 mg/L、维生素 B1 0.4 mg/L、维生素 C 0.5 mg/L、甘氨酸1.5 mg/L、蛋白胨0.7~0.9mg/L、活性炭 3g/L、酵母浸提液50ml/L、茉莉酸甲酯0.03mg/L、氯化胆碱0.3mg/L、白砂糖 20g/L、花生壳粉10g/L、卡拉胶3.5 g/L,pH调节为5.8。
(3) 炼苗驯化:
当步骤(2)中试管苗株高长到 5cm~8cm,具有4~5条根系且根长3cm~5cm时,将植物组织培养瓶的无菌瓶盖打开,将组织培养瓶放在光照培养架上培养,逐渐将室内恒温空调关闭,连续炼苗7d左右,然后将试管苗从组培瓶中移出并洗净根部培养基,移栽到消毒后的驯化基质(按重量配比为蛭石22 g、活性炭10g、蔗渣 18g、油茶壳15g、椰子壳16g、玉米秸秆25g、谷壳灰30g、腐殖土20g)中,定期浇水并施加特定的营养液(配方为磷酸钙0.8g/L、硝酸钙0.5g/L、硝酸钾0.9g/L、尿素0.3 g/L、硫酸镁0.28g/L、硫酸铁0.12g/L、硼酸0.06mg/L、硫酸锰0.03mg/L、硫酸锌0.06mg/L、硫酸铜0.06mg/L、钼酸铵0.06mg/L、甘露醇1.1 mg/L、氯化胆碱2.0mg/L、亚精胺3.6 mg/L),移至室外遮阳网下生长。
(4)移栽:
待步骤(3)中的试管苗在驯化基质上生长2-3周后,再移栽到消毒后的栽培基质(按重量配比为蛭石13 g、蔗渣20g、油茶壳15g、木屑14g、玉米秸秆24g、谷壳灰35g、虾壳粉16g、腐殖土48g)中,栽培密度为株行距18cm×25cm,在室外自然光照、温度条件下常规栽培管理。
实施例2
试验时间地点、操作方法均同实施例1,其中将步骤(1)中增殖培养基的配方改为MS培养基+6-BA 2.0mg/L+NAA 0.2mg/L 。
实施例3
试验时间地点、操作方法均同实施例1,其将步骤(2)中生根培养基的配方改为1/2MS培养基+NAA 0.5mg/L 。
实施例4
试验时间地点、操作方法均同实施例1,其中将步骤(3)中采用的驯化基质改为草炭土:蛭石=1:1,施加的营养液改为山崎营养液,步骤(4)中移栽基质改为草炭土:蛭石=4:1。
试验结果统计:
不同因素影响下生长情况比较:分别采用实施例1-4的方法进行操作,将昙花脱毒种苗接种于增殖培养基继代5次后统计各组平均增殖系数,再转入生根培养基上培养22天后统计平均生根率;将脱毒苗移栽2个月后统计移栽成活率。
从表1的结果可以看出,实施例1的增殖系数、生根率均优于实施例2和3,说明本发明提供的培养基配方、激素种类与配比更适合昙花脱毒种苗的生长。相对于实施例4,实施例1的移栽成活率有明显的提高,说明本发明采用的驯化和移栽营养液及基质对昙花的壮苗效果显著。本发明首次建立了一种昙花脱毒种苗的工厂化快繁方法,本方法通过组培快繁可以在短期内获得整齐一致的优质脱毒苗,从而为企业的种苗规模化生产提供技术支持。
对于本领域的普通技术人员而言,具体实施例只是对本发明进行了示例性描述,显然本发明具体实现并不受上述方式的限制,只要采用了本发明的方法构思和技术方案进行的各种非实质性的改进,或未经改进将本发明的构思和技术方案直接应用于其它场合的,均在本发明的保护范围之内。

Claims (6)

1.一种昙花脱毒种苗的工厂化快繁方法,其特征在于,包括以下操作步骤:
(1)脱毒种苗增殖培养:
将已获得的昙花脱毒种苗在无菌条件下接种于增殖培养基中进行培养,培养室温度控制为22~26℃、光照强度控制为1500-2000Lux、相对湿度控制为60~70%、光周期控制为光12h/暗12h,待种苗长至3~4厘米大小并且长出新的不定芽从时,将不定芽逐个完整的切下,再转入新的增殖培养基中继续增殖,每28天增殖1代,增殖4~6代后可获得大量脱毒幼苗;
(2) 脱毒种苗生根培养:
将步骤(1)获得的脱毒幼苗,转移到生根培养基上进行培养,控制光照强度为1500~2000Lux,环境温度为22~26℃,相对湿度为65~75%,培养18~23天长成生根的试管苗;
(3)炼苗驯化:
当步骤(2)中试管苗株高长到 5cm~8cm,具有4~5条根系且根长3cm~5cm时,将植物组织培养瓶的无菌瓶盖打开,将组织培养瓶放在光照培养架上培养,逐渐将室内恒温空调关闭,连续炼苗7d左右,然后将试管苗从组培瓶中移出并洗净根部培养基,移栽到消毒后的驯化基质中,定期浇水并施加特定的营养液,移至室外遮阳网下生长;
(4)移栽:
待步骤(3)中的试管苗在驯化基质上生长2-3周后,再移栽到消毒后的栽培基质中,栽培密度为株行距18 cm×25 cm,在室外自然光照、温度条件下常规栽培管理。
2.如权利要求1所述的一种昙花脱毒种苗的工厂化快繁方法,其特征在于,所述步骤(1)中增殖培养基的配方为:硝酸钾2400~2600mg/L、磷酸二氢铵310~330mg/L、硫酸镁192~200mg/L、无水氯化钙295~315mg/L、碘化钾0.8~0.9mg/L、硼酸4.0~5.0mg/L、一水硫酸锰10.0~12.0mg/L、硫酸锌0.8~1.2mg/L、钼酸钠0.11~0.14mg/L、五水硫酸铜0.23~0.27mg/L、氯化钴0.09~0.12mg/L、乙二胺四乙酸铁钠19.6~20.0mg/L、肌醇90~110 mg/L、维生素B60.4~0.6 mg/L、维生素B1 0.2~0.4 mg/L、维生素 C 0.4~0.6mg/L、烟酸0.2~0.5 mg/L、甘氨酸1.4~1.7 mg/L、蛋白胨0.7~0.9mg/L、酵母浸提液45~55ml/L、三十烷醇0.08~0.11mg/L、胺鲜脂0.45~0.55mg/L、激动素0.17~0.22mg/L、异戊烯基腺嘌呤0.18~0.23mg/L、白砂糖 25~30g/L、卡拉胶8~12g/L,pH调节为5.6~6.0。
3.如权利要求1所述的一种昙花脱毒种苗的工厂化快繁方法,其特征在于,所述步骤(2)中生根培养基的配方为:硝酸钾 1100~1300mg/L、硝酸钙170~185mg/L、磷酸二氢钾150~170mg/L、硝酸铵440~465mg/L、硫酸镁91~94mg/L、碘化钾0.3~0.5mg/L、氯化钾28~34mg/L、硼酸1.0~1.3mg/L、七水硫酸锰14.0~17.0mg/L、硫酸锌0.5~1.0mg/L、钼酸钠0.10~0.15mg/L、硫酸铁12.0~15.0mg/L、氯化钴0.10~0.14mg/L、乙二胺四乙酸铁钠16.0~20.0mg/L、肌醇90~110 mg/L、维生素B6 0.4~0.6 mg/L、维生素B1 0.2~0.4 mg/L、维生素 C 0.4~0.6mg/L、烟酸0.2~0.5 mg/L、甘氨酸1.4~1.7 mg/L、蛋白胨0.7~0.9mg/L、活性炭 2~5g/L、酵母浸提液45~55ml/L、茉莉酸甲酯 0.02~0.05mg/L、氯化胆碱0.2~0.4mg/L、白砂糖25~30g/L 、花生壳粉8~11g/L、卡拉胶3~5g/L,pH调节为5.6~6.0。
4.如权利要求1所述的一种昙花脱毒种苗的工厂化快繁方法,其特征在于,所述步骤(3)中特定的营养液的配方为过磷酸钙0.8g/L、硝酸钙0.5g/L、硝酸钾0.9g/L、尿素0.3 g/L、硫酸镁0.28g/L、硫酸铁0.12g/L、硼酸0.06mg/L、硫酸锰0.03mg/L、硫酸锌0.06mg/L、硫酸铜0.06mg/L、钼酸铵0.06mg/L、甘露醇1.0~1.2 mg/L、氯化胆碱1.7~2.3mg/L、亚精胺3.2~3.7mg/L 。
5.如权利要求1所述的一种昙花脱毒种苗的工厂化快繁方法,其特征在于,所述步骤(3)中驯化基质消毒方法为:用75%百菌清可湿性粉剂500倍液进行消毒,驯化基质按重量配比为蛭石20~25g、活性炭2~5g、蔗渣 18~20g、油茶壳13~20g、椰子壳15~20g、玉米秸秆25~30g、谷壳灰30~35g、腐殖土16~24g。
6.如权利要求1所述的一种昙花脱毒种苗的工厂化快繁方法,其特征在于,所述步骤(4)中栽培基质消毒方法为用75%百菌清可湿性粉剂500倍液进行消毒,栽培基质按重量配比为:蛭石10~15g、蔗渣18~22g、油茶壳11~20g、木屑13~18g、玉米秸秆20~25g、谷壳灰30~40g、虾壳粉15~18 g、腐殖土45~50g。
