Abstract

The invention belongs to the technical field of traditional Chinese medicines, and particularly relates to a traditional Chinese medicine composition with a whitening effect and a preparation method thereof, wherein the traditional Chinese medicine composition comprises the following fresh raw materials in parts by weight: 100-750 parts of fresh radix astragali, 50-350 parts of fresh astragalus membranaceus, 30-350 parts of raw fleece-flower root, 20-350 parts of soft date, 30-350 parts of radix polygonati officinalis, 30-500 parts of pawpaw, 30-400 parts of fig, 30-400 parts of raspberry, 30-400 parts of dandelion and 10-300 parts of black propolis. The invention has the beneficial effects that: (1) compared with the dry traditional Chinese medicine composition, the extract of the fresh traditional Chinese medicine composition is obviously improved in clinical efficacy; (2) the addition of the soft jujube into the Chinese medicinal composition obviously improves the whitening effect.

Description

Traditional Chinese medicine composition with whitening effect and preparation method thereof
Technical Field
The invention belongs to the technical field of traditional Chinese medicines, and particularly relates to a traditional Chinese medicine composition with a whitening effect and a preparation method thereof.
Background
With the development of society and the improvement of material culture level, people have better and better life, so that the desire of people for beauty and health care is stronger and the requirements are more diversified. Melanin is a protein present in the basal layer of the skin of each person, and irradiation with ultraviolet rays changes melanin to produce a substance for protecting the skin, which reaches the epidermal layer of the skin via layer-by-layer movement of cellular metabolism, thereby causing skin problems such as color spots, uneven skin color, and yellowish complexion.

The present invention has been made in view of this situation.

The whitening effect of the traditional Chinese medicine composition is obviously improved by adding the soft jujube.
The fresh raw materials with the following parts by weight are also included: 40-300 parts of fresh kudzuvine root, 40-300 parts of white paeony root, 50-400 parts of white poria, 50-500 parts of cortex dictamni, 30-400 parts of bletilla striata, 20-350 parts of black sesame and 30-400 parts of roxburgh rose.
The Chinese medicinal materials are added, so that the traditional Chinese medicine has an obvious effect of treating facial acne and pustule sore caused by stomach cold and qi stasis.
The traditional Chinese medicine composition also comprises pharmaceutically acceptable auxiliary materials or carriers.
The composition is prepared into different medicament forms by adding pharmaceutically acceptable auxiliary materials or carriers, is convenient to carry and take, is easy to transport and store, has stable quality, and does not deform or mildew at low temperature and high temperature.

In the step 1), water is added for soaking for 1-4 hours, and the low-temperature extraction temperature T is more than or equal to 35 ℃ and less than 60 ℃;
preferably, the low-temperature extraction is divided into two times; the first extraction is carried out at the temperature of more than or equal to 35 ℃ and less than 45 ℃, and the second extraction is carried out at the temperature of more than or equal to 45 ℃ and less than 60 ℃.
In order to extract the effective components completely, the number of extraction times may be more than two.
In the step 2), the specific gravity of the fluid extract obtained by concentrating the extracting solution is 1.20-1.35.

step 1): weighing the above fresh raw materials according to a formula ratio, adding water, soaking for 1 hour, performing low-temperature extraction twice, wherein the first extraction is performed at 35 ℃, and the second extraction is performed at 45 ℃ and filtering to obtain an extracting solution and dregs;
step 2): concentrating the extracting solution obtained in the step 1) under reduced pressure, wherein the pressure of the reduced pressure concentration is 0.06MPa, the temperature is 30 ℃, and a fluid extract with the specific gravity of 1.20 is obtained;
step 3): drying the medicine residues in the step 1), and crushing into fine powder;
step 4): mixing the fluid extract obtained in step 2) with the fine powder of the residue obtained in step 3), stirring, and freeze drying at-55 deg.C to obtain a composition.
In order to ensure that the fresh traditional Chinese medicine extract has better quality, selecting the fresh traditional Chinese medicine with qualified quality, analyzing the content of the effective components of the raw medicinal materials by using a national standard measuring method, washing the qualified raw medicinal materials by using water until the color of the water is clear, and then airing or draining the raw medicinal materials. The traditional Chinese medicine composition has obvious effect on treating facial acne and pustule sore caused by stomach cold and qi stasis by adding the traditional Chinese medicines on the basis of the first embodiment.
EXAMPLE III
This example differs from example one in that the first extraction in step 1) is carried out at 20 ℃ and the second extraction is filtered at 30 ℃.
Example four
This example differs from example one in that the first extraction in step 1) was carried out at 70 ℃ and the second extraction was filtered at 80 ℃.
Comparative example 1
The difference between the comparative example and the first example is that the raw materials of the traditional Chinese medicine composition of the comparative example are dry traditional Chinese medicine components, the weight of the traditional Chinese medicine composition is that the dry traditional Chinese medicine components are obtained after drying the components of the first example, and the preparation method is the same as that of the first example.
Comparative example 2
The difference between this embodiment and the first embodiment is that the Chinese medicinal composition of this embodiment does not contain soft date.
Experimental example 1 in vitro evaluation of whitening function of extract
1. Preparation of the test specimens
The traditional Chinese medicine formula preparations prepared in the first to fourth examples and the comparative examples 1 to 2 are taken as samples. A sample (1 g) was dissolved in dimethyl sulfoxide (99 g), and the mixture was stirred well to obtain a supernatant, which was used for the following experiment.
The supernatant is mainly prepared from corresponding Chinese medicinal composition.
2. Evaluation experiment of inhibitory effect on non-activity of tyrosine
Taking the supernatant of the traditional Chinese medicine, Phosphate Buffer Solution (PBS) with pH of 6.8 and dopa solution with mass fraction of 1% according to the table 1, fully mixing, incubating at 30 ℃ for 5min, adding tyrosinase solution with activity of 110U/mL, incubating at 30 ℃ for 10min, rapidly transferring to a cuvette, and measuring absorbance at 475 nm.

The relative inhibition rate (I, inhibition rate for short, the same below) of the sample on tyrosinase was calculated according to the formula (1).

In the formula: a1, A2, A3 and A4 are the absorbances of the solutions of groups 1 to 4, respectively.

From the above experimental results it can be seen that:
comparing the experimental results of the first embodiment with those of the comparative example 2, it can be seen that the effect of the combination of the Chinese medicinal composition containing soft date and the Chinese medicinal composition containing no soft date is significantly improved in the inhibition rate of tyrosinase, and the addition of soft date can promote the absorption of whitening components in the extract.
In the embodiment, the fresh traditional Chinese medicine is adopted for extracting the traditional Chinese medicine components, and the comparative example 1 adopts the dry traditional Chinese medicine for extracting, and the experimental result shows that the inhibition rate of the supernatant obtained by adopting the fresh traditional Chinese medicine on tyrosinase is obviously higher than that of the supernatant obtained by adopting the dry traditional Chinese medicine.
Compared with the experimental results of the first embodiment and the second embodiment, it can be seen that the inhibition rate of tyrosinase is affected by the temperature of low-temperature extraction, which is either too high or too low, because the content of effective components in the fresh traditional Chinese medicine is high, and some effective components are easily decomposed at high temperature, the structure of the effective components in the fresh traditional Chinese medicine is damaged by the too high temperature, so that the inhibition rate of tyrosinase is reduced, more and more viscous substances are present between the cell wall and the cell membrane of the fresh traditional Chinese medicine, and the inhibition rate of tyrosinase is lower because the temperature is too low and the effective components are not easily leached, so that the extraction of the fresh traditional Chinese medicine is maintained at 35 ℃ or more and T less than 60 ℃.
3. Experiment for inhibiting melanin synthesis in melanocytes
The B16 melanoma cells were seeded at a density of 1X 105 cells/mL in 96-well plates at 90. mu.L per well, and after 24 hours of incubation in a CO2 incubator, the sample solution was added per well. A blank control group of medium and cells was set up simultaneously.
After the plates were incubated in an incubator for 72 hours, the supernatant was discarded, washed twice with PBS, and then 0.5mL of trypsinized cells were added to each well for 3min, and 2mL of maintenance solution was added to each well to stop digestion. After mixing, 0.5mL of each concentration was taken out and counted. Centrifuging the rest cell suspension at 2500r/min for 5min, discarding supernatant, adding NaOH solution into the precipitate, heating to dissolve melanin, and measuring absorbance at 490nm under enzyme-linked immunosorbent assay. The melanin synthesis inhibition ratio (I') of the sample was calculated according to the formula (2).

In the formula: a5 is drug well absorbance value; p1 is drug well cell density; a6 is control well absorbance value; p2 is control well cell density.

The experimental results show that the extract prepared from the traditional Chinese medicine composition without adding the soft jujube has a significantly lower inhibition rate on melanin synthesis than the extract prepared from the traditional Chinese medicine composition with the soft jujube, as can be seen from the experimental results, the soft jujube is added to the traditional Chinese medicine composition in the first example, and the soft jujube is not added to the comparative example 2.
In the first embodiment, the fresh traditional Chinese medicine is adopted for extracting the traditional Chinese medicine components, and in the comparative example 1, the dry traditional Chinese medicine is adopted for extracting, and the experimental result shows that the traditional Chinese medicine extract prepared by the fresh traditional Chinese medicine is obviously higher than the dry traditional Chinese medicine in the aspect of melanin synthesis inhibition rate, so that the preparation method is more suitable for extracting the fresh traditional Chinese medicine.
Experimental example 2 moisturizing and whitening effects on human skin
And (3) taking 20-30 parts by weight of the composition preparation prepared in the first embodiment, the comparative example 1 and the comparative example 2, adding the composition preparation into milk or egg white, and blending into paste to obtain the mask agent 1, the mask agent 2 and the mask agent 3. The product does not contain chemicals, and has low adverse side effect.
1) Test object
60 subjects, female, aged 18-30 years, with living habits all standing overnight, facing computers for a long time, with dry, dark and dark skin, were selected and randomized into three groups of 20 subjects each.
2) Experimental sample and experimental method
Three groups of subjects use the facial mask agent 1, the facial mask agent 2 and the facial mask agent 3 in sequence, and the subjects use the facial mask once in the morning and at night every day for 30 days continuously.

The experimental results show that the whitening and moisturizing effects of the facial masks prepared in the above examples and comparative examples are remarkably higher than that of comparative example 2 without adding soft jujubes by studying the whitening and moisturizing effects of the facial masks prepared in the above examples and comparative examples, and the whitening and moisturizing effect of the facial mask with the soft jujubes is better.
In the embodiment, the whitening and moisturizing effect of the facial mask prepared from the fresh traditional Chinese medicines is obviously higher than that of the facial mask prepared from the dry traditional Chinese medicine comparative example 1, so that the whitening and moisturizing effect of components extracted from the fresh traditional Chinese medicines is better.
The above ingredients are added with pharmaceutically acceptable auxiliary materials or carriers to prepare a pill preparation, a plaster, an external lotion and a skin membrane agent for human body tests, and the obtained conclusion is consistent with the above conclusion.
The preparation of the invention is convenient to carry and take, easy to transport and store, stable in quality, and does not denature and mildew at low temperature and high temperature.
The control points of pesticide residue and heavy metal indexes of a formula preparation extracted from fresh medicines are zero, and the serious harm of over-standard pesticide residue and heavy metal is reduced.
