另一方面我们的发明是一种增强至少一种抗原在哺乳动物或非哺乳动物中的免疫反应的方法,包括使用包裹在NISV中的至少一种所说的抗原。
我们的发明另一个目的是一种制备疫苗的方法,包括把至少一种抗原,(如合成或重组起源的肽)包裹在NISV中。
用于制备NISV的非离子表面活性剂可以是任何药理学上可接受的具有适当表面活性性质的物质。这种物质优选的例子是甘油酯类。这种甘油酯类可以包含一种或两种高级脂肪族酰基。例如,在每个酰基部分含有至少10个碳原子。优选甘油单酯类,特别是含C12-C20链烷酰基或链烯酰基(例如己酰基,月桂酰基,肉豆蔻酰基,棕榈酰基、油基或硬脂酰基)部分的那些。最佳的表面活性剂是1-棕榈酰基甘油酯。

为了形成有效囊(泡),将需要把非离子型表面活性剂与一种能形成双层结构的适宜的高分子量疏水性物质(特别是一种甾族化合物,如一种甾醇象胆甾醇)混合。这种甾族化合物的存在有助于形成决定囊(泡)生理特性的双层结构。
NISV可以掺合一种产生电荷的两亲物。使NISV带有一个负电荷。酸性物如高级链烷和链烯酸(例如棕榈酸,油酸);或其它含有酸性基团的化合物,例如,磷酸酯象二烷基、最好是联(高级烷基)磷酸酯,例如联十六烷基磷酸酯,或磷脂酸或脂酰丝氨酸;或者磺酸-酯如磺酸高级烷酯,例如,磺酸鲸蜡酯,都可以用于这种目的。
这种甾族化合物可以例如含有非离子型表面活性剂的20-120%(重量百分比),优选60-100%。产生一个负电荷的两亲物可以例如含有非离子型表面活性剂的1-30%(重量百分比)。
产生电荷的两亲物稳定该囊(泡)的结构并提供有效的扩散。
非离子型表面活性剂和形成膜的疏水性物质可以通过剪切力存在下的水合作用转变为非离子型表面活性剂囊(泡)。使用的这种剪切力的设备是已知的,适宜仪器例如在WO 88/06882中提到的。声处理和超声处理也是形成非离子型表面活性剂囊(泡)或改变它们颗粒大小的有效方法。
Collins等在J.Pharm.Pharmocol.42,53(1990)中公开一种生产NISV的有效方法。它包括把非离子型表面活性剂,甾族化合物和两亲物(如果使用的话)的混合物融化并在含水缓冲液存在下强烈搅拌使水合。然后在升高的温度(足以使形成的NISV混合物保持在融化状态)下,把这种混悬液挤压多次通过多微孔的聚碳酸酯膜。
从一种有机溶剂,例如,一种烃或氯化烃溶剂象氯仿中蒸发环形膜也能形成NISV。然后可把所得薄膜在有抗原和任意其它表面活性剂存在的磷酸盐缓冲液中水合(Russell and Alexander,J.Immund、140.1274(1988))。在这种情况当中,NISV形成的同时抗原也被包裹在其中,使用已知分离方法,如用凝胶过滤或离心分离(例如超离心分离法)通过清洗把囊(泡)外的抗原从囊(泡)混悬液中除去。
把抗原包裹进制备好的NISV的方法是脱水-再水合方法(Kirby and Gregoriadis,Biotechnology 2 979(1984)),其中把水相中的抗原包裹进已制备好的囊(泡)中是用急骤冷冻接着升华干燥和冷冻-融化技术(Pick,Arch、Biochem、Biophys、212,195(1981))。在后一种技术中,把囊(泡)与抗原混合并在液氮中反复急骤冷冻并且例如加温到60℃(即,相关表面活性剂的临界温度之上)。
把囊(泡)可作进一步处理来除去任何没有被包裹进去的抗原,例如,冲洗和离心分离。应该注意到我们的结果清楚地表明单独非离子型表面活性剂并不是有效的佐剂,也就是说,囊(泡)的形成对获得理想的有效作用是必要的。如果要达到所要求的佐剂效果必须把抗原包裹在非离子型表面活性剂囊(泡)中。
抗原优选含有例如8-50,最好为10-20个氨基酸单位的合成或重组来源的肽。这种抗原可以模拟病原生物体的一个或多个B细胞,或B细胞和T细胞的表位,从而使疫苗能诱发中和抗体抵抗那种病原生物体(参见,例如,我们的WO 88/10267和WO 91/13909中的HIV合成抗原的说明书)。
从另一方面来说,这种肽能够诱发抗其它生物活性物质,特别是具有激素活性的物质的免疫反应。后一类型的例子是诱导内源黄体化激素(endogenous luteinising hormone)-释放激素(LHRH)的免疫反应。能例如被用于农场动物雄激素抑制或免疫去势的治疗以及雄激素敏感的或雌(甾)酮敏感的恶性肿瘤的治疗(参见我们的GB-B-2196969)。包裹在NISV中用作疫苗的这种肽抗原自身常具有足够的免疫原性;特别是在该肽含有很好识别的表位并包含大约至少12个,最好是至少15个氨基酸残基时的情况下。
在有些情况下,最好是把这种肽与一种载体结合来增强它的免疫原性。适宜的载体是本领域熟知的,例如,蛋白载体象结核病菌素的净蛋白衍生物(PPD),破伤风类毒素,霍乱毒素质和它的B亚单位,卵清蛋白,牛血清清蛋白,大豆胰蛋白酶抑制剂,胞壁酰二肽和它们的类似物,以及Braund′s脂蛋白。当使用结核菌素净蛋白衍生物(PPD)作载体时,如果疫苗受体借助早期BCG接种已对结核菌素敏感,就会得到较高的抗体效价。
使用药学上常规的方法能把这种包裹在NISV中的抗原制备成疫苗,如悬浮在一个无菌的非肠道可接受的含水载体中。
尽管合成或重组肽是用于本发明最好的抗原,但是当把蛋白抗原包裹在NISV中时也能观察到一个强的佐剂作用。例如,使用牛血清清蛋白(BSA)作抗原已获得强阳性结果,更加显著的是,通过免疫接种兔弓形虫的破裂细胞上清液,用非离子型表面活性剂囊(泡)佐剂接种已证明有抗弓形体病的有效保护作用。
我们已经发现皮下接种通常是注射给药的最有效的方法,特别是常比腹膜内给药更有效。肌肉内和表皮内注射也是有效的,也许因为它导致了在注射部位形成抗原贮存。尽管这样,注射部位没有发生由FCA经常产生的慢性炎症反应。注射给药能诱发一个全身免疫反应,也就是,既有体液又有细胞调节免疫。在一个粘膜表面(如鼻)接种预料在其它粘膜表面(如支气管)引起起一种粘膜免疫反应。按照本发明该疫苗也能粘膜线路给药。
按照我们发明的其它意外且有价值的特征,当口服包裹在NISV中的抗原时通常会产生一种有效水平的免疫反应。Niosomes似乎至少在某种程度上能保护抗原不被胃肠道消化,穿过肠壁不发生变化,这样获得迄今仅是胃肠外给药所能获得的结果。
因此,本发明提供了一种制备一种作为口服活性疫苗的抗原的方法,包括把所说的抗原包裹在NISV中。
通过口服例如一种合成肽获得佐剂效果的能力是本发明疫苗的最大有效特点,也是现有技术中从未完成的特征。口服途径给药比现有的注射途径给药有许多优点,避免了伴随注射的象源于使用没有灭菌的针头引起的注射危害。除诱导一个全身免疫反应外,口服也可以诱导一种粘膜反应。这种粘膜反应在抗许多病原体如HLV,弓形体的免疫保护作用中被认为是重要的。此外,对患者来说对口服的组合物有更高的可接受性。
在制备用作疫苗特别是口腔给药的囊(泡)中,优选含酯键的表面活性剂,尽管也可使用含醚键的表面活性剂特别是1-鲸蜡酸甘油醚和二甘醇鲸蜡酸醚。已经发现醚键比相应的酯键酸稳定性更大并被认为它在哺乳动物的胃的酸性环境中有更大的经受分解的能力。
由本发明新疫苗引起的免疫反应的进一步分析意外地表明比例较高的抗体是与释放IL-2和IFN-r有关的IgG2a型抗体。这些细胞***素由CD4+T淋巴细胞的Th1子集分泌。高水平的IgG2a被以为与细胞调节免疫的发生有关。另一方面,氢氧化铝优选刺激Th2细胞,引起IgG1抗体的产生,而且通常与提高细胞调节免疫有关的弗氏完全佐剂也产生较高的长期性的IgG1抗体效价。Th2细胞不仅是IgG1而且是IgE合成的主要调节节(由IL-4分泌)。因而持续产生原生质中的高水平IgG1是不理想的因为它也表明IgE的过度产生。
近来离体研究已经证实Th1和Th2细胞具有不同的处理抗原的要求。Th1细胞需由巨噬细胞提供抗原而Th2细胞则可最好被B-细胞提供的抗原所刺激(Gajewski等,Immund,Rer,Ⅲ,79(1989)和Gajewski等,J.Immund,146,1750(1991)。巨噬细胞似乎能摄取脂质体并因此包裹在囊(泡)中的抗原将有助于Th1的激活和ThG2a抗体的合成,导致高水平的CMI。
我们的结果表明按照本发明的新疫苗能诱发IgG2a的效值,开始远大于由弗氏完全佐剂所产生的,但过一段时间最终与弗氏完全佐剂所产生的效价相类似。按照本发明对疫苗的初始反应与由弗氏完全佐剂所得到的相比似乎相对于Th2淋巴细胞来说优先激发Th1淋巴细胞。我们的结果还表明使用本发明疫苗所获得的诱发的IgG1效价是与弗氏完全佐剂所产生的效价同级的,因此本发明疫苗在调节体液免疫方面也是有用的。
因此我们发明的另外一方面是配制至少一种抗原以通过Th1 T淋巴细胞通道刺激抗体产生的方法,它包括把所说的至少一种抗原包裹在非离子型表面活性剂囊(泡)中。
按照本发明的抗原配方也能经Th2 T淋巴细胞通道刺激抗体产生。这种抗原可配方成与经Th2 T淋巴细胞通道刺激抗体产生相比优先经Th1 T淋巴细胞通道刺激抗体的产生,特别是与使用弗氏完全佐剂产生的抗体水平相比较时。
而且,用NISV强化的疫苗免疫接种激发的抗体效价实际上高于使用弗氏完全佐剂(FCA)所获得的,而完全避免了弗氏完全佐剂的强毒性作用。所以我们的发明的另一方面是把至少一种抗原的免疫原性提高到至少等同于使用弗氏完全佐剂所获得的水平的方法,包括把所说的至少一种抗原包裹进NISV中。
NISV可认为是用于抗那些需要细胞或体液免疫的疾病的疫苗的理想佐剂。作为佐剂的NISV(特别是含有醚键的表面活性剂的那些)的另一个主要优点是在一般的外界环境条件下的稳定性。特别是它们不经历与蛋卵磷脂为基础的脂质体相同方式的破坏性自氧反应。
NISV作为佐剂用于疫苗的另一个重要优点是与现有技术中佐剂相反,NISV基本上无毒。我们已经发现给小鼠腹膜内注射包裹在含1-单棕榈酰基甘油酯(monopalmitoyl glycerol ester)的囊(泡)中的LHRH类似物LHRH-Gly-Gys(如我们的EP-A-0293530中所述的)没有引起任何毒副作用,特别是与弗氏完全佐剂或甚至铝凝胶有关的毒副作用。在实验动物中没有观察到可察觉的病状。
按本发明完成的疫苗主要适用于哺乳动物,因而在人用和兽用药物领域中能够应用。我们也观察到NISV对一些非哺乳动物象,鱼和家禽的接种,也是一个有效的佐剂。
以下实施例详细说明NISV的佐剂特性。
实施例1-用LHRH-Gly-Cys-PPD NISV组成物免疫小鼠
摩尔比为5∶4∶1的非离子型表面活性剂,胆甾醇和磷酸联十六烷酯(Sigma,Poole,Dorset,OK),混合制备囊(泡)。下列显示微摩尔数和重量毫克数:

用Collins等(在上)的技术制备囊(泡)。接着在Metter电子水浴近距离声波***(50
,Pasadena,(A)中把囊(泡)制剂在20℃下声处理5分钟。在我们的EP-A-0293530中描述了抗原LHRH-Gly-Cys,一种促黄体激素(Luteinising hormone)一释放激素的类似物。用于Kirby和Gregoriadis描述的脱水一再水合技术(Biotechndogy,2,979(1984))。把抗原包入制备好的囊(泡)中。简单地说,在聚丙烯离心管(Elkay Produets,Inc.,Shrewsbury,MA)中把5ml(150μ mol)囊(泡)溶液与1ml PBS中的抗原(0.5mg/ml)混合,在液氮中用旋转方法急骤冷冻为薄层。然后在冷冻干燥器中把制剂在0.1乇下冻干过夜,再用0.5ml蒸馏水
水合。剩余试样持续30分钟,然后用蒸馏水加至6ml。
a)用有和没有BCG起动及佐剂的LHRH-Glys-Cys-PPD免疫
给人组雄性Copenhagen Fisher F,杂交鼠腹膜内注射0.2ml的LHRH-Cly-Cys-PPD磷酸缓冲生理盐水、包裹在NISV中的LHRH-Gly-Cys-PPD、在FCA中的LHRH-Gly-Cys-PPD乳化液和吸附在氢氧化铝上的LHRH-Gly-Cys-PPD。每支注射液载有的LHRH-Gly-Cys的量在生理盐水,FCA/FIA和钒中是50μg,在NISV中是5μg。
在首次注射(LHRH-Gly-Cys-PPD结合的前4周给四组小鼠首先注射BCG。四次结合液的注射总共有两周的间隔。
抗LHRH抗体的测定
用ELISA方法测定间隔获得的血浆试样中的抗LHRH抗体。在96孔级的组织培养微量滴定板上以0.1μg/孔涂LHRH-Gly-Cys-BSA(在PBS中,pH7.2)并在37℃培养1小时。把这些孔按上述方法清洗并把用PBS稀释的50μl试样加到相同孔中。把试样在37℃下培养1小时,如前清洗这些孔。把50μl的用PBS稀释到1∶5000的辣根过氧化物酶羊抗鼠IgG轭合物(Sigma)的等分试样加到每个孔上在如上清洗前在37℃下培养45分钟。底物溶液的制备是把4μl过氧化氢和250μl的贮备在二甲基亚砜中的四甲基联苯胺(6mg/ml)加到25ml的0.1M乙酸构橼酸缓冲液(pH5.5)中。
在室温黑暗中培养5分钟前每个孔中盛有100μl所加的底物物。加入10%硫酸中止反应并测量在450nm处的吸收。ELISA的结果用阳性对照的百分值表示。整个测定中使用的恒定标准以便能进行内和处对照。这个恒定标准是一个许多用在FCA中的LHRH-Gly-Cys-PPD免疫的大量鼠的血清池。这个阳性对照有一个100%的任意值。
结果
ELISA的结果在图1和图2中显示。图1描述了用LHRH-Gly-Cys-PPD与生理盐水(A),niosomes(B),弗氏完全和不完全佐剂(C)以及氢氧化铝(b)的轭合物免疫的Copenhagen-Fisher雄性鼠的抗体反应。尽管使用FCA/FIA或钒时对LHRH-Gly-Cys的抗体反应似乎较大,但使用这些佐剂的肽的用量比掺到NISV中时多10倍。因此,NISV表现出比氢氧化铝或弗氏完全佐剂在辅助较低量的肽轭合物上有更强的作用。图2显示了起动对LHRH-Ghy-Cys-PPD轭合物的抗体反应的BCG的作用。用和不用起动均可看到相似数量的免疫反应。而且用FCA和钒所得到的抗体效价比较高。然而,使用这些佐剂时免疫原的给药量是用NISV的十倍。
b)为了更精确地证实NISV的佐剂作用,在被注射在FCA中的LHRH-Ghy-Cys-PPD和在如上所述制备的NISV囊(泡)中的LHRH-Ghy-Cys-PPD的BALB/C鼠中测量了等量抗原的抗体反应。
用LHRH-Ghy-Cys-PPD免疫
给3只雄性BALB/C小鼠皮下注射0.2ml在FCA中的LHRH-Gly-Cys-PPD(PBS中轭合物:FCA的体积比为1∶1)。两周后用皮下注射0.2ml在FIA中的LHRH-Gly-Cys-PPD(PBS中的共轭物∶FIA体积比为1∶1)增加小鼠用药量。
给5只雌性BALB/C小鼠类似地皮下注射0.2ml包裹在NISV中的LHRH-Gly-Cys-PPD并在两周后再用0.2ml增加剂量。没有使用BCG起动。每次注射给药等量于100μgLHRH-Gly-Cys-PPD。
抗-LHRH抗体的测定
在第二次免疫接种后一周取尾部放出的血液,把血清试样汇集起来,用基本上如上所述的ELISA方法测定总抗体反应,但是用系列稀释液来确定终止效价。
结果
结果如图3所示。能清楚地看到在同等免疫试样下,NISV是一个强烈的佐剂,并能诱发比用FCA所获得的更高的特异抗体水平。

实施例2 用牛血清清蛋白(BSA)免疫鼠
从实施例1所描述组分用实施例1所描述的方法或者用旋转薄膜蒸发法从氯仿中制备囊(泡)。在PBS和抗原(5mg/ml)中或用洗涤剂(1%辛基-β-D-吡喃葡萄糖苷,Sigma)和PBS中的抗原(5mg/ml)把所得到的膜水合。(如Rllseel和Alexander(见上)所描述的有关脂质体的方法)。为除去洗涤剂(如使用了的话)和任何未包裹的抗原,把制备物以84000克40-45分钟离心并使颗粒悬浮于PBS中。重复清洗两次。
在每组中使用4到5只8-10周龄的BALB/C鼠。在第一天,各组小鼠皮下或腹膜内注射包裹BSA的NISV(NISV/BSA),或BSA的FCA乳化液(FCA+BSA∶Sigma),在每只注射液中的BSA的最终浓度为0.5mg/ml并且每只鼠接受100μg剂量的BSA。在第二次激发之前14天给小鼠初次注射。在图4的柱状图中的14、28等天是指第二次激发后的时间。
用ELISA测定抗BSA抗体水平。图4中,(a)部分代表IgG1抗体效价,和(b)部分表示IgG2a的效价。画有斜线的柱状图是指NISV/BSA而实心柱状图是指BSA+FCA。在(a)和(b)每一种情况中图左边方柱表示腹膜内注射所得到的结果,而右边方柱表示皮下注射所获得的结果。
将会看到用腹膜内接种,FCA+BSA产生了非常高的IgG1效价,维持到70天;NISV/BSA的效价则随时间下降。用皮下给药,NISA/BSA免疫开始产生高于FCA的效价,但到70天,这些效价已下降到远低于用FCA+BSA所获得的效价。
与此相反,在图(b)部分对IgG 2a的试验表明不论用腹膜内还是皮下方式NISV/BSA诱发的最初效价远远高于由FCA+BSA所得到的,但是到70天,由于由FCA+BSA产生的IgG 2a水平随时间增加,两种佐剂配方产生的效价相似。
引人注意的是,在皮下注射FCA后观察到的肉芽瘤的形成在注射NISV后完全没有出现。
实施例3 从各种表面活性剂中制备的NISV的佐剂活性
从(a)1-单棕榈酰基甘油酯(MPG)(如实施例1和2中的)或(b)聚氧乙烯-3-月桂醚(POE)二者之一,按实施例1所述的表面活性剂∶胆甾醇∶磷酸联十六烷酯5∶4∶1制备NISV。按实施例2中所述的方针对8-10周龄雌性BALB/C小鼠组(每组5只),在第一天皮下注射包裹于NISV中的BSA或在PBS乳化液中的BSA二者之一。在第二次激发后14、35和70天用ELISA测定总Ig,IgG 1和IgG 2a的水平。为便于分析,把血浆稀释到1/8000分析总抗体效价,1/15000分析IgG 1,1/1000分析IgG 2a,并且个体鼠血浆中存在的抗体效价用由ELISA测得的吸收值表示。图5中显示出了所得的结果,其中图5a显示了第二次接种后在不同时间测得的总免疫球蛋白;图5b和5c分别显示了IgG 1和IgG 2a的水平。
将能看出在从每一个表面活性剂生产的NISV包裹BSA与BSA没有佐剂相比显著地提高了抗体效价。对被分析的所有抗体亚类,在第二次注射后14天BSA包裹在以MPG为基质的囊(泡)中比BSA包裹在以POE为基质的囊(泡)中产生更高的抗体效价。到第二次感染后35天,用MPG比用POE为基质的囊(泡)有较高的Ig和IgG 1和效价,然而IgG 2a的水平相似。到70天,两者的囊(泡)制剂在总抗体效价;或两者的分析的免疫球蛋白亚类上没有显著性区别。
因此能够看出NISV的佐剂活性是广泛适用的并不依赖于任何一种表面活性剂。
实施例4 在同类品系鼠中NISV的佐剂活性
已知现有技术中的某些佐剂因个体间免疫反应上遗传决定的不同应用上受到限制。为了证明NISV的佐剂活性不被遗传性所限制,测定了在三种BALB鼠(BALB/C,BALB/B和BALB/K)中对单独给药的BSA(即,在PBS中),包裹在NISV中或在弗氏完全佐剂乳化液中的BSA的抗体反应。按照实施例1和2制备NISV和给这些小鼠组皮下注射。用ELISA测得抗体水平并在图6A、B和C中显示。在图6A中,表示的是从在第二次接种后14天(图a),35天(图b)和70天(图c)收集的血液制备的血浆所测得的特异性BSA总抗体效价,其中结果用在450nm测得的平均吸收值来表示(+/-S.E.)。在IgG 1(图6B)和IgG 2a(图6e)中显示了相似数据。为了ELISA,血浆试样被稀释到1/15000,所用轭合物稀释度为1/1000(6A),血浆和轭合物的相应的图分别是1/30000和1/8000(6B)和1/1000和1/800(6C)。
将会看出对各种小鼠来说,NISV/BSA都产生了既对总抗体又对特殊的异型IgG 1和IgG 2a的高抗体效价。尽管相应的效价与用BSA和FCA的效价相比随第二次刺激后的时间以及不同品种而变化,但是NISV在被实验的三个品种中都能够起到辅助作用,诱发的抗体反应或者大于由BSA/FCA所产生的或者至少与其在一个级别。
实施例5 抗先天性弓形体病的疫苗制备
弓形体病是由原虫兔弓形虫引起的一种鸟类和哺乳动物的疾病。在非保护的活动中这种疾病能从母亲到胎儿垂直传染而且这种传递能导致胎儿死亡。但是在感染前用疫苗可产生一定程度的保护性免疫。它被以为这是由CD8+T细胞和Th1 CD4+子集间的协同作用产生的。用BALB/C和BALB/K两种鼠作实验模型研究了特别与T细胞的这些子集有关的NISV包裹抗原的疫苗特性。
小鼠
在常规条件下饲养近亲交配的BALB/C和BALB/K小鼠和远亲交配的Strathclyde A系族小鼠。当小鼠为8-10周龄时使用,并且每个实验组含有5-10只动物。
感染
12周前被用兔弓形虫RRA(Beverly)种感染的A系族小鼠大脑被用作组织囊肿的来源,它是按以前的描述(Roberts和Alexander 1992,Parasitor,104.19-23)被收获和计数的。实验中的感染都用口腔方式以及先天性感染模型也是如以前所描述的(Roberts and Alexander 1992)。简单地说,在受孕11~12天用20个组织囊肿感染BALB/C小鼠。然后存活的后代被转移到感染的养畜母亲,并用ELISA方法在出生后8-9周测定先天性感染的发生率。合适的是用死亡率或者用大脑中总囊肿数监控感染的严重程度。
抗原制备
冷冻-融化杀灭的tachyzoites(KP),tachyzoite***/渗出的抗原(ESAg),膜抗原和可溶性抗原(STAg)都被用于接种研究。从感染的棉花鼠腹膜渗出液中获得RH株的兔弓形虫tachyzoites,并用生理盐水洗3次。采用5×1010tachyzoites接种在40ml PBS中过夜的方法获得ESAg。Tachyzoites被用离心分离法在1000g下除去并且收集上层清液。用Bradford测定法(Bradford.1976 Analyt、Biochem、72,248-254)测得所有的蛋白浓度。按照使用Braun匀浆器(homogeniser)在低渗缓冲液(40ml,10mM tris-HCl,2mMEDTA,pH7.8)中碎裂5×1010寄生物继之以4℃和10000g下离心脱水30分钟得到Tachyzoite可溶性和膜抗原部分。该上层清液含有STAg和丸状膜部分。该膜抗原用1%辛基葡萄糖苷进一步纯化继之以在100,000g下离心。收集上层清液在4℃对PBS透析过滤以除去洗涤剂。
疫苗制备
在感染前2周和4周,所有要接受疫苗制剂的动物皮下接种50μg tachyzoite抗原。用于接种的抗原可以以游离的形式或者乳化于弗氏完全佐剂中或者包裹在非离子型表面活性剂囊(泡)中。

用实施例1中所述的方法将STAg包裹于1-单棕榈氧基甘油NISV中。
结果
所得到的结果显示在图7A-D中。图7A显示了皮下接种包裹在NISV中的STAg,单独STAg或在感染前2和4周与NISV混合的STAg之后通过口腔用20个囊肿在BALB/K鼠感染后4周的囊肿平均数(+/-S.E.)。能够看到,与其它组比较包裹在NISV中的STAg显著降低囊泡数。
图7B显示了平均血浆抗体水平,图7C显示了用20个囊肿口服感染后8周的BALB/C雌鼠大脑中的囊肿平均数。小鼠在受孕11-12天时被感染。接种组已经在感染前2和4周皮下接种包裹在NISV中的STAg(50μg)。能够看到,与对照组相比,用NISV包裹的抗原接种既能显著地降低大脑中的囊肿数又显著地降低抗体水平。
图7D显示了接种和无接种小鼠的出生幼鼠的死亡情况。能够看到,尽管对免疫鼠来说有超过50%的出生的后代被感染,但与非免疫试样相比(多于一些的幼鼠在出生或出生后24小时内死亡),没有死亡在免疫的出生幼鼠中发生。
因此我们已证明在NISV中的可溶性抗原的包裹提高了佐剂功效,即提高了这种抗原对成熟鼠的保护性和完全消除了胎儿的死亡。
实施例6用作合成的T细胞表位佐剂的NISV的能力
肽的制备
“Giles”肽,是麻疹F蛋白258-277的残基,用常规的FMOC化学法合成。这种肽具有序列GILESRGI
KARITHVDTESY并含有T和B细胞二者的表位。
按照Collins方法(见上)从1-单棕榈酰基甘油酯中制备NISV,并用Pick的冷冻-融化技术(见上)包裹抗原。
免疫接种
每组4只8-10周龄雌性BALB/C小鼠以25μg在PBS中包裹在NISV中或在FCA中乳化的肽免疫。一周后,摘除它们的脾脏并用胸腺嘧啶脱氧核苷掺合验定法(Corradin等,J.Immurd、119,1048-1053(1977))测量这种肽的特异T细胞增生反应。
结果
该结果显示于图8和图9中。在试验测定的所有浓度中,来源于用NISV包裹抗原的免疫小鼠的脾细胞比源于接受PBS或FCA中肽的鼠的细胞在体外增生程度更大(图8)。
在刺激48小时后移去已用50μg/ml GILES刺激的培养物中的上清液并用ELISA试验IFN-r的存在。从图9能看到,源于用GILES和佐剂一起接种的小鼠脾细胞比用PBS中的GILES接种的鼠的脾细胞明显产生更多的IFN-r,并且NISV包裹的抗原比FCA中的抗原产生更多的IFN-r。因此NISV能作为相应于较大病毒蛋白的肽的佐剂,也能够作为激素的佐剂。而且,NISV比FCA能在更大程度上起动小鼠的抗原特异T细胞增殖到较高程度。在培养物上清液中存在的IFN-r暗示了CD4+Th1细胞和/或CD8+T细胞的活化。
实施例7 口头结医试验
用NISV包裹的BSA对鼠口头给药
囊(泡)按实施例1所描述的以表面活性剂∶胆甾醇,二鲸蜡基磷酸盐5∶4∶1比率从二甘醇鲸蜡基醚(DGCE)或1-单棕榈酰基甘油酯(MPG)组成。囊泡按Rnssell和Alexander(见上)描述的从氯仿中用旋转薄膜蒸发制备。形成薄膜的150μmoles表面活性剂在含有100mg BSA的碳酸盐缓冲液中水合。混合物在60℃用水浴声波处理机处理5分钟之前在60℃振摇2小时。
每一个处理组中使用5支8-10周龄雌性BALB/C鼠,每组接受下列:
(a)在碳酸盐缓冲液中的BSA
(b)在DGCE NISV中的BSA
(c)在MPG NISV中的BSA
鼠在第1天被用管饲养法给药首次接受0.1ml口服药剂(每支鼠240μg)。第12天,第二次口服剂量(每只鼠500μg BSA)被给药。在第20天和24天收集血样品并分析IgG效价。
Claims (42)
1、一种疫苗,包含至少一种包裹于非离子型表面活性剂囊(泡)中的抗原。
2、如权利要求1所要求的疫苗,其中的抗原是一种肽。
3、如权利要求2所要求的疫苗,其中的肽是合成的或重组来源的。
4、如权利要求3所要求的疫苗,其中的肽含有8到50个氨基酸单元。
5、如权利要求4所要求疫苗,其中的肽含有10到20个氨基酸单元。
6、如权利要求3到5中的任一所要求的疫苗,其中的肽是连结到一种载体上。
7、如权利要求1所要求的疫苗,其中的抗原选自兔弓形虫原虫的抗原和黄体化激素释放激素和其分子式为PGln-His-Trp-Ser-Tyr-X-Len-Arg-Pro-Gly-Y-Z的类似物,其中X表示Gly或一个D-氨基酸,Y表示一个或多个氨基酸残基,它们可以相同也可不同,Z表示Cys或Tys,这样所说的类似物的溶液构象基本上相似于弃置的LHRH的构象。
8、如上述权利要求中任一个所要求的疫苗,其中所说的非离子型表面活性剂包含一种甘油酯。
9、如权利要求8所要求的疫苗其中所说的甘油酯是含有C12-C20链烷氧基或链烯氧基部分的甘油单酯。
10、如权利要求9所要求的疫苗,其中的甘油酯是1-单棕榈酰基甘油酯。
11、如权利要求1到7中任一个所要求的疫苗,其中的囊(泡)含有基于甘油或低级脂肪甘醇的醚。
12、如权利要求11所要求的疫苗,其中的醚是甘油醚或基于含C12-C20链烷基或链烯基部分的低级脂肪族甘醇单醚。
13、如权利要求12所要求的疫苗,其中的单醚包含5个以下的甘醇单位。
14、如权利要求12所要求的疫苗,其中的甘油醚是1-单鲸蜡基甘油醚或者其中的低级脂肪族甘醇醚是二甘醇鲸蜡基醚。
15、如上述权利要求中任一个所要求的疫苗,其中的裹(泡)包括带电荷的两亲性分子。
16、如权利要求15所要求的疫苗,其中的带电荷两亲性分子含有选自羧酸盐(酯),磷酸盐(酯)或磺酸盐(酯)的一种酸性基团。
17、如上述权利要求中任一个所要求的疫苗,其形式适合于皮下,肌肉内或表皮肉给药。
18、如权利要求1到16中任一个所要求的疫苗,其形式适合于粘膜给药。
19、如权利要求1到16中任一个所要求的疫苗,其形式适合于口腔给药。
20、一种疫苗的制备方法,包括在非离子型表面活性剂囊(泡)中包裹至少一种抗原。
21、把一种抗原制备为一种口服有活性的疫苗方法,包括在非离子型表面活性剂中包裹所说的抗原。
22、如权利要求20或21所要求的方法,其中的疫苗是如权利要求2到19中任何一个所定义的。
23、制备至少一种抗原经Th1 T淋巴细胞通道刺激抗体产生的方法,包括在非离子型表面活性剂裹(泡)中包裹至少一种所说的抗原。
24、把至少一种抗原的免疫原性增强到至少与使用弗氏完全佐剂所获得的相同水平的方法,包括在非离子型表面活性剂裹(泡)中包裹至少一种所说的抗原。
25、如权利要求23或24所要求的方法,其中所说的抗原是如权利要求2到7中任一个所定义的,所说的囊(泡)是如权利要求8到16中任一个所定义的。
26、如权利要求20到25中任一个所要求的方法,其中的抗原是包裹在制备好的囊(泡)中。
27、具有至少一种包裹的抗原的非离子型表面活性剂囊(泡)在刺激细胞或体液免疫上的用途。
28、如权利要求27中所要求的用途,作为一种可口腔给药的免疫佐剂。
29、如权利要求27到28所要求的用途,其中的抗原是如权利要求2到7中任何一个所定义的。
30、如权利要求28或29中所要求的用途,其中的囊(泡)包含键合酯的表面活性剂分子。
31、在非离子表面活性剂囊(泡)中包裹的至少一种抗原在制备用于在哺乳动物或非哺乳动物对象中增强对所说至少一种抗原的免疫反应的产品方面的用途。
32、包裹在非离子型表面活性剂囊(泡)中的至少一种抗原在制造用于在哺乳运物或非哺乳动物对象中刺激对所说的至少一种抗原的细胞调节和体液免疫应答的产品方面的用途。
33、包裹在非离子型表面活性剂囊(泡)中至少一种抗原在制造用于在哺乳动物或非哺乳动物对象中经Th1 T淋巴细胞通道刺激应答所说的至少一种抗原的抗体产生的产品方面的用途。
34、如权利要求31到33中任一个所要求的用途,其中所说的对象是哺乳动物。
35、如权利要求31到33中任一个所要求的用途,其中所说的抗原是如权利要求2到7中任一个所定义的,所说的囊(泡)是如权利要求8到16中任一个所定义的。
36、一种在哺乳动物或非哺乳动物对象中提高对至少一种抗原的免疫应答的方法,包括给所说的对象使用包裹于非离子型表面活性剂囊(泡)中所说的至少一种抗原。
37、一种在哺乳动物或非哺乳动物对象中刺激应答至少一种抗原的细胞调节和体液免疫的方法,包括给所说的对象服用包裹在非离子型表面活性剂囊(泡)中所说的至少一种抗原。
38、一种在哺乳动物或非哺乳动物对象中经TH1 T淋巴细胞通道刺激应答至少一种抗原的抗体产生方法,包括给所说的对象服用所说的包裹于非离子型表面活性剂囊(泡)中的至少一种抗原。
39、如权利要求36到38中任一个所要求的方法,其中所说的对象是哺乳动物。
40、如权利要求36到39中任一个所要求的方法,其中所说的对象是哺乳动物。
41、如权利要求36到40中任一个所要求的方法,其中所说的抗原是如权利要求2到7中任何一个中所定义的,所说的囊(泡)是如权利要求8到16中任一个中所定义的。
42、一种含有包裹在非离子型表面活性剂囊(泡)中的至少一种抗原的产品,其形式是粉剂,片剂、糖浆、胶囊或颗粒剂。
