Description

Oral care composition for children and preparation method thereof
Technical Field
The invention relates to the technical field of oral care, in particular to a children oral care composition and a preparation method thereof.
Background
Common oral diseases include oral ulcer, periodontal disease, and dental caries. Clinically, oral ulcer, commonly known as aphtha, is a common ulcerative injury disease occurring in oral mucosa and is divided into catarrhal, aphthous, vesicular, cellulitis, pseudomembranous, ulcerative and papular types, and the incidence rate reaches 70-90%. The common clinical symptoms are manifested by salivation, loss of appetite or slow ingestion, oral malodor, flushing of the oral mucosa, warming, swelling, and pain. Periodontal disease refers primarily to gingivitis and periodontitis; gingivitis refers to acute and chronic inflammation occurring in gingival tissues, which is commonly manifested as gingival bleeding, redness and swelling, swelling and pain, and repeated outbreak of dental ulcer, the gingival bleeding, swelling and pain can affect each other to form a vicious circle, and if effective treatment is not carried out, periodontitis possibly developed to a deep layer can cause more serious problems of tooth loosening, tooth falling and the like. Periodontitis is a chronic inflammation that attacks the gingival and periodontal tissues, is a destructive disease, and is mostly a gingival inflammation and swelling caused by plaque, tartar, physical impaction, poor restoration, bite wound, etc., and meanwhile, the plaque is accumulated and aggravated and extends from the upper part of the gum to the lower part of the gum. Because of the characteristics of the subgingival micro-ecological environment, a large number of periodontal pathogenic bacteria with high toxicity, such as gingival bacteroides, intermediate bacteroides, spirochetes and the like, are bred in the subgingival bacterial plaque, so that the inflammation of gingiva is aggravated and expanded, alveolar bone formed by a periodontal pocket is absorbed, and periodontitis is caused, and the main inducement of periodontitis is also caused by the accumulation of the bacterial plaque.
At present, more Chinese herbal medicines can be used for treating oral ulcer and periodontal disease, and most of the raw materials used in the traditional Chinese medicine are plant or mineral components. Wherein, the plant is the extract of the root, stem and leaf, etc. after being practiced and screened for thousands of years, the plant is listed as the Chinese herbal medicine component in the Chinese pharmacopoeia, and can be called as the treasure of Chinese nation with the unique medicinal value and safety. With the continuous improvement of the technological level, nearly 3800 raw materials can be analyzed to determine the active ingredients of the raw materials, and qualitative or quantitative detection can be carried out at the same time. The efficacy of the active ingredient is also continually being demonstrated. The development and research of oral products such as Chinese herbal medicine toothpaste for treating oral ulcer and periodontal disease also become a new subject of recent research. The traditional Chinese herbal medicine toothpaste or other oral products are not available in the market, but the effect on treating oral ulcer and gingivitis is not ideal, most of the existing oral products only aim at cleaning oral cavity and transiently refreshing breath, do not substantially improve oral care, even damage enamel and tooth surface to make teeth fragile, stimulate and further damage oral mucosa, are not good for oral health, and are particularly not suitable for children.

Figure BDA0001608638890000101
As can be seen from the results in table 1, the treatment effect of the children's oral care compositions obtained in examples 1 to 6 of the present invention is significantly better than that of the children's oral care compositions obtained in comparative examples 1 to 4, wherein the treatment effect of the children's oral care composition obtained in example 6 of the present invention is the best, and the children's oral care composition obtained in example 5 of the present invention is the next. The raw materials adopted in comparative examples 1 to 4 are only slightly different from the raw materials of examples 1 to 6 of the invention, but the treatment effects are far from each other, which shows that the proportion of the raw materials of the invention is not set randomly, but has synergistic effect, and the components have the best effect only when being matched, especially the matching of traditional Chinese medicine components (licorice extract, aloe barbadensis leaf extract, panax japonicus extract, lilac extract and mint extract) and cationic surfactants (domiphen, chlorhexidine and cetylpyridinium chloride).
In addition, stability tests show that the stability of the oral care composition for children is the best when the raw materials are in the weight percentage range of the invention of the embodiment 4-6.
Effect example 2
In order to further illustrate the beneficial effects of the invention, 100 children patients with oral problems such as dental caries, halitosis and ulcer in 9-12 years old are selected, the oral care compositions for children obtained in the embodiments 1-6 and the comparative examples 1-4 of the invention are used for 15 days and 2 times a day, and then the tested patients are tested, wherein the specific test steps are as follows:
saliva of 100 children patients who did not use the oral care compositions for children obtained in examples 1 to 6 and comparative examples 1 to 4 of the present invention and saliva of 15 days after using the oral care compositions for children obtained in examples 1 to 6 and comparative examples 1 to 4 were collected using sterile test tubes, and after the saliva samples were diluted with sterile PBS solutions, 0.2mL of the diluted saliva samples were applied to BHI agar plates, and after anaerobic culture at 37 ℃ for 72 hours, the BHI agar plates were eluted with 2mL of the sterile PBS solutions, and eluates were collected.
Blank control group: saliva obtained without using the oral care compositions for children obtained in examples 1 to 6 and comparative examples 1 to 4 of the present invention was diluted 10-fold with sterile PBS solution to 1X 10-6~5×10-6Obtaining bacterial suspension at mol/L, sucking 1mL of bacterial suspension, properly diluting, carrying out anaerobic culture at 37 ℃ for 72h, and then calculating the number of colonies.
Test groups: the eluates obtained by the above procedures were applied to the saliva of 15 days after the use of the children's oral care compositions obtained in examples 1 to 6 and comparative examples 1 to 4 of the present invention, respectively, and sterilized byPBS solution was diluted 10-fold to 1X 10-6~5×10- 6Obtaining bacterial suspension at mol/L, sucking 1mL of bacterial suspension, properly diluting, carrying out anaerobic culture at 37 ℃ for 72h, and then calculating the number of colonies.
The sterilization rate was calculated according to the following formula:

The above results show that the number of bacteria in saliva of children was significantly inhibited after 15 days using the children's oral care compositions of examples 1 to 6 of the present invention, wherein the children's oral care composition obtained in example 6 of the present invention had the best inhibitory effect on anaerobic bacteria in saliva, and the children's oral care compositions obtained in examples 4 to 5 of the present invention. The raw materials adopted in comparative examples 1 to 4 are only slightly different from the raw materials of examples 1 to 6 of the present invention, but the bactericidal effects are far from each other, which indicates that the mixture ratio of the raw materials of the present invention is not set at will, but has a synergistic effect, and the bactericidal effect is the best when the components are used in combination, especially the combination of the traditional Chinese medicine components (licorice extract, aloe barbadensis leaf extract, panax japonicus extract, lilac extract, mint extract) and the cationic surfactants (domiphen bromide, chlorhexidine, cetylpyridinium chloride). In conclusion, the children oral care composition provided by the embodiments 1 to 6 can effectively prevent dental caries, prevent and treat periodontitis and ulcer, clean oral cavity and strengthen teeth.
