本发明公开了一种山核桃叶总黄酮提取物的制备工艺,将将用80‑90℃的醋液温煮10‑20min,将经过煮制后的山核桃叶放置于烘箱中在70‑75℃下烘焙4‑5h后粉碎,将20目以上的山核桃叶粉末与水混合均匀,再加入重量为山核桃粉末0.05‑0.08%的复合酶,混合均匀进行酶解,酶解结束后,将复合酶失活,得到酶解液;将酶解液以200目滤膜过滤,滤液利用截留分子量为10‑15K道尔顿的超滤膜过滤得到超滤液;将超滤液在70℃条件下,真空浓缩,得到浓缩液;浓缩至相对密度为1.05‑1.15;将浓缩液利用加压MCI‑GEI小孔树脂柱纯化;将收集的洗脱液先在62‑67℃条件下减压浓缩,浓缩至相对密度为1.2‑1.5,浓缩液经真空冷冻干燥,即得山核桃叶总黄酮提取物。本发明具有操作简单、效率高、能耗低、产品纯度高等优点。
Description
技术领域
本发明属于中药或天然药物化学领域,涉及一种总黄酮的制备工艺,特别涉及一
种利用酸解、酶解后的山核桃叶总黄酮的制备工艺。
背景技术
黄酮类化合物是一种天然存在的有机化合物。在自然界中,黄酮类化合物广泛存
在于多种植物的器官(如种子、花、果实、和叶子)和其他中药材原料中,例如
回心草、甜茶、金莲花、益母草、银杏叶、山楂、甘蔗、板栗花、蜂胶等里面都
含有丰富的黄酮类化合物。目前,黄酮类化合物的研究已经非常广泛而深入,其
生理及药理作用和活性研究也相当深入。有文献表明,黄酮类化合物有抗炎镇痛
、抗过敏、扩张血管、抗氧化、抗心律失常、改善微循环、抗脂肪肝、防止动脉
硬化、抗病毒、降血脂、减少糖尿病并发症、协同增进抗肿瘤药物疗效等药理作
用。作为一种天然存在的有机化合物,其在治疗心血管疾病领域有着很重要的地
位。故从天然植物和中药材原料中提取、分离纯化黄酮类化合物,进而进行有效
地药理实验研究是一项极有必要性和有意义的工作。
目前实验室多采用聚酰胺树脂法精制纯化黄酮类化合物,不仅方法简单、成本低
、效率高、稳定性好和容易再生等特点,而且其安全性也高,没有重金属和有毒
有机试剂残留等问题。但是,若是用这种方法分离富集黄酮苷元却存在很多问题
。采用聚酰胺传统分离、富集工艺根本难以做到,原因在于:其一,待分离的黄
酮苷元脂溶性较强,用蒸馏水稀释根本无法溶解;其二,虽然可以先用适量的乙
醇溶解,然后大量蒸馏水稀释能够溶解,但其体积将非常巨大;其三,聚酰胺在
水溶液中对黄酮类物质的吸附能力最强,稀释后的乙醇上样液是否会影响其黄酮
苷元吸附仍有待考证。在实际中,无论是用聚酰胺分离富集黄酮苷元还是黄酮苷
,我们都有类似问题,然而,检索文献资料,未有相关文献论述这一问题,而且
几乎所有文献都只分离富集总黄酮。鉴于前述现象和问题,我们认为有必要采用
新的方法工艺,对聚酰胺分离富集黄酮进行改进,以适应实际需求。
发发明内容
为了克服现有技术中存在的问题,本发明公开了一种山核桃叶总黄酮提取物的制
备工艺,本发明具有操作简单、效率高、能耗低、产品纯度高等优点。
一种山核桃叶总黄酮提取物的制备工艺,是通过如下技术方案实现的:
(1)将山核桃叶用80-90℃的醋液温煮10-20min,将经过煮制后的山核桃叶放置
于烘箱中在70-75℃下烘焙4-5h后粉碎,将20目以上的山核桃叶粉末与水混合均
匀,再加入重量为山核桃粉末0.05-0.08%的复合酶,混合均匀进行酶解,酶解结
束后,将复合酶失活,得到酶解液;
(2)将步骤(1)所得酶解液以200目滤膜过滤,滤液利用截留分子量为10-15K
道尔顿的超滤膜过滤得到超滤液;
(3)将步骤(2)超滤液在70℃条件下,真空浓缩,得到浓缩液;浓缩至相对密
度为1.05-1.15;
(4)将步骤(3)所得浓缩液利用加压MCI-GEI小孔树脂柱纯化,小孔树脂柱的
柱内径与柱高的比值为1:(6-10);纯化过程:先以纯化水为洗脱剂,洗脱1-3
个柱体积,弃去;再以5-10%醇水溶液淋洗,洗脱1-2个柱体积,弃去;再以30-
70%醇水溶液进行洗脱,洗脱2-4个柱体积,收集洗脱液;
(5)将步骤(4)收集的洗脱液先在62-67℃条件下减压浓缩,浓缩至相对密度
为1.2-1.5,浓缩液经真空冷冻干燥,即得山核桃叶总黄酮提取物。
优选地,步骤(1)中的醋液为1.015-1.030kg/L的竹醋液;水的体积与山核桃叶
粉末质量的比为(15-18)mL:1g。
作为一种优选实施方式,步骤(1)调酶解反应的条件为:pH值为3.0-8.0,在
40-80℃条件下进行酶解20-30min,酶解后在90-100℃处理2-5min使酶失活,所
述复合酶是由纤维素酶和果胶酶按1:1-2的质量比混合而成的。
优选地,步骤(4)中MCI-GEI小孔树脂柱采用加压处理,压力为0.1-0.5MPa。
其中,步骤(5)所得的山核桃叶总黄酮提取物采用高效液相色谱法测得提取物
中总黄酮含量为96-98%。
在本发明的技术方案中:先将山核桃叶进行酸化处理,再进行烘焙、粉碎得到山
核桃叶粉末,然后将山核桃叶粉末酶解提取,药材成分提取较为彻底,节约提取
溶剂,提取温度低,处化效率快、总黄酮含量可高达96-98%,是一种切实有效的
总黄酮制备工艺。
具体实施方式
以下描述用于揭示本发明以使本领域技术人员能够实现本发明。以下描述中的优
选实施例只作为举例,本领域技术人员可以想到其他显而易见的变型。
实施例1
一种山核桃叶总黄酮提取物的制备工艺,是通过如下技术方案实现的:
(1)将山核桃叶用80℃的1.030kg/L的竹醋液温煮20min,将经过煮制后的山核
桃叶放置于烘箱中在70℃下烘焙5h后粉碎,将20目以上的山核桃叶粉末与水混合
均匀,再加入重量为山核桃粉末0.05%的复合酶(由纤维素酶和果胶酶按1:1的
质量比混合而成的),混合均匀,在pH值为3.0-3.5,40℃条件下进行酶解30min
,酶解后在90℃处理5min使酶失活,得到酶解液,其中,水的体积与山核桃叶粉
末质量的比为15mL:1g;
(2)将步骤(1)所得酶解液以200目滤膜过滤,滤液利用截留分子量为10K道尔
顿的超滤膜过滤得到超滤液;
(3)将步骤(2)超滤液在70℃条件下,真空浓缩,得到浓缩液;浓缩至相对密
度为1.05-1.06;
(4)将步骤(3)所得浓缩液利用加压MCI-GEI小孔树脂柱纯化,压力条件为
0.1MPa,小孔树脂柱的柱内径与柱高的比值为1:6;纯化过程:先以纯化水为洗
脱剂,洗脱1个柱体积,弃去;再以5%醇水溶液淋洗,洗脱1个柱体积,弃去;再
以30%醇水溶液进行洗脱,洗脱2个柱体积,收集洗脱液;
(5)将步骤(4)收集的洗脱液先在62℃条件下减压浓缩,浓缩至相对密度为
1.2,浓缩液经真空冷冻干燥,即得山核桃叶总黄酮提取物,所得的山核桃叶总
黄酮提取物采用高效液相色谱法测得提取物中总黄酮含量为96%。
实施例2
一种山核桃叶总黄酮提取物的制备工艺,是通过如下技术方案实现的:
(1)将山核桃叶用90℃的1.015kg/L的竹醋液温煮10min,将经过煮制后的山核
桃叶放置于烘箱中在75℃下烘焙4h后粉碎,将20目以上的山核桃叶粉末与水混合
均匀,再加入重量为山核桃粉末0.08%的复合酶(由纤维素酶和果胶酶按1:2的
质量比混合而成的),混合均匀,在pH值为8.0,80℃条件下进行酶解20min,酶
解后在100℃处理2min使酶失活,得到酶解液,其中,水的体积与山核桃叶粉末
质量的比为18mL:1g;
(2)将步骤(1)所得酶解液以200目滤膜过滤,滤液利用截留分子量为15K道尔
顿的超滤膜过滤得到超滤液;
(3)将步骤(2)超滤液在70℃条件下,真空浓缩,得到浓缩液;浓缩至相对密
度为1.13-1.15;
(4)将步骤(3)所得浓缩液利用加压MCI-GEI小孔树脂柱纯化,压力条件为
0.5MPa,小孔树脂柱的柱内径与柱高的比值为1:10;纯化过程:先以纯化水为洗
脱剂,洗脱3个柱体积,弃去;再以10%醇水溶液淋洗,洗脱2个柱体积,弃去;
再以70%醇水溶液进行洗脱,洗脱4个柱体积,收集洗脱液;
(5)将步骤(4)收集的洗脱液先在67℃条件下减压浓缩,浓缩至相对密度为
1.5,浓缩液经真空冷冻干燥,即得山核桃叶总黄酮提取物,所得的山核桃叶总
黄酮提取物采用高效液相色谱法测得提取物中总黄酮含量为98%。
实施例3
一种山核桃叶总黄酮提取物的制备工艺,是通过如下技术方案实现的:
(1)将山核桃叶用86℃的1.025kg/L的竹醋液温煮14min,将经过煮制后的山核
桃叶放置于烘箱中在72℃下烘焙4.5h后粉碎,将20目以上的山核桃叶粉末与水混
合均匀,再加入重量为山核桃粉末0.06%的复合酶(由纤维素酶和果胶酶按2:3
的质量比混合而成的),混合均匀,在pH值为6.0,60℃条件下进行酶解25min,
酶解后在95℃处理3min使酶失活,得到酶解液,其中,水的体积与山核桃叶粉末
质量的比为16mL:1g;
(2)将步骤(1)所得酶解液以200目滤膜过滤,滤液利用截留分子量为12K道尔
顿的超滤膜过滤得到超滤液;
(3)将步骤(2)超滤液在70℃条件下,真空浓缩,得到浓缩液;浓缩至相对密
度为1.10-1.12;
(4)将步骤(3)所得浓缩液利用加压MCI-GEI小孔树脂柱纯化,压力条件为
0.3MPa,小孔树脂柱的柱内径与柱高的比值为1:8;纯化过程:先以纯化水为洗
脱剂,洗脱2个柱体积,弃去;再以8%醇水溶液淋洗,洗脱2个柱体积,弃去;再
以50%醇水溶液进行洗脱,洗脱3个柱体积,收集洗脱液;
(5)将步骤(4)收集的洗脱液先在65℃条件下减压浓缩,浓缩至相对密度为
1.4,浓缩液经真空冷冻干燥,即得山核桃叶总黄酮提取物,所得的山核桃叶总
黄酮提取物采用高效液相色谱法测得提取物中总黄酮含量为97%。

Claims (6)
1.一种山核桃叶总黄酮提取物的制备工艺,其特征在于是通过如下技术方案实现的:
(1)将山核桃叶用80-90℃的醋液温煮10-20min,将经过煮制后的山核桃叶放置于烘箱中在70-75℃下烘焙4-5h后粉碎,将20目以上的山核桃叶粉末与水混合均匀,再加入重量为山核桃粉末0.05-0.08%的复合酶,混合均匀进行酶解,酶解结束后,将复合酶失活,得到酶解液;
(2)将步骤(1)所得酶解液以200目滤膜过滤,滤液利用截留分子量为10-15K道尔顿的超滤膜过滤得到超滤液;
(3)将步骤(2)超滤液在70℃条件下,真空浓缩,得到浓缩液;浓缩至相对密度为1.05-1.15;
(4)将步骤(3)所得浓缩液利用加压MCI-GEI小孔树脂柱纯化,小孔树脂柱的柱内径与柱高的比值为1:(6-10);纯化过程:先以纯化水为洗脱剂,洗脱1-3个柱体积,弃去;再以5-10%醇水溶液淋洗,洗脱1-2个柱体积,弃去;再以30-70%醇水溶液进行洗脱,洗脱2-4个柱体积,收集洗脱液;
(5)将步骤(4)收集的洗脱液先在62-67℃条件下减压浓缩,浓缩至相对密度为1.2-1.5,浓缩液经真空冷冻干燥,即得山核桃叶总黄酮提取物。
2.如权利要求1所述的制备工艺,其特征在于:步骤(1)中的醋液为1.015-1.030kg/L的竹醋液。
3.如权利要求1所述的制备工艺,其特征在于:步骤(1)中水的体积与山核桃叶粉末质量的比为(15-18)mL:1g。
4.如权利要求1所述的制备工艺,其特征在于:步骤(1)调酶解反应的条件为:pH值为3.0-8.0,在40-80℃条件下进行酶解20-30min,酶解后在90-100℃处理2-5min使酶失活,所述复合酶是由纤维素酶和果胶酶按1:1-2的质量比混合而成的。
5.如权利要求1所述的制备工艺,其特征在于:步骤(4)中MCI-GEI小孔树脂柱采用加压处理,压力为0.1-0.5MPa。
6.如权利要求1所述的制备工艺,其特征在于:步骤(5)所得的山核桃叶总黄酮提取物采用高效液相色谱法测得提取物中总黄酮含量为96-98%。
