Abstract

本发明涉及医疗护理领域,具体涉及一种带凝血酶的海藻酸盐伤口敷贴的制备方法,具体步骤为:配置凝血酶的乙醇水溶液,然后将海藻酸盐长束纤维放入含凝血酶乙醇水溶液的反应槽中恒温浸泡,浸泡后经干燥、裁切制成海藻酸盐短纤维,并将短纤维卷曲,再经开松、梳理、铺网、针刺等无纺工艺制成无纺布,并裁切成需要尺寸的敷芯,将敷芯与带孔硅酮凝胶薄膜、聚氨酯薄膜胶带、防粘层经敷贴机复合,最后经包装、灭菌,即可制得带凝血酶的海藻酸盐伤口敷贴成品。本发明所制得的复合敷贴,能够最大限度的保持凝血酶的活性,具有优秀的止血和吸液性能,同时可防止敷贴与伤口的粘连,为创面提供湿润的愈合环境,促进伤口愈合。

一种带凝血酶的海藻酸盐伤口敷贴的制备方法
技术领域
本发明涉及医疗护理领域,具体涉及一种带凝血酶的海藻酸盐伤口敷贴的制备方法。
背景技术
皮肤是人体最大的器官,也是人体与外界环境设置的第一道防线,其对保护机体内环境的稳定,抵御外界环境的刺激和病原菌入侵起到重要的作用。
在医疗护理领域,研究者越来越重视如何更好的护理皮肤,产品的实用性,以及便利性就尤为重要。伤口敷贴对伤口具有很好的保护作用,免于受到外界的二次污染,减轻了患者感染的风险,给人们也带来了诸多方便。
但大多数敷贴产品的敷芯还是比较单一,这种复合材料用于伤口仅起到吸收伤口渗液的作用,无法起到伤口保湿、止血并存的功效。
随着功能性敷料的发展,伤口敷贴不断的在改进,产品的单一功能并不能满足临床医患人员的需求,针对出血性的伤口,如何更有效的止血,并有效的促进伤口的愈合,促进伤口肉芽的生长,仍然没有一款更有效的产品出现。
因此,如何克服以上缺点,提供一种可有效止血、愈合效果显著、减轻患者痛苦的伤口敷贴,是一件很有必要且具有意义的事情。
发明内容
本发明的目的在于克服上述现有技术中的不足,提供一种带凝血酶的海藻酸盐伤口敷贴的制备方法。为实现上述目的,本发明提供的技术方案为:
一种带凝血酶的海藻酸盐伤口敷贴的制备方法,具体包括以下步骤:
S1. 配置凝血酶的乙醇水溶液,乙醇与水体积比为(2~3)﹕1,凝血酶含量为50~100单位/ml;
S2. 将海藻酸盐长束纤维放入含凝血酶乙醇水溶液的反应槽中恒温浸泡0.5~1h,温度为30~35℃,反应槽设置“S”型通道;
S3. 浸泡过后,将长束纤维拉出反应槽,经烘箱干燥,干燥温度为35~37℃,干燥时间为1~2h;
S4. 干燥后,经裁切工艺,制成长度为38~65mm的带有凝血酶的海藻酸盐短纤维,并将短纤维卷曲,卷曲数不少于3个/cm;
S5. 将短纤维经开松、梳理、铺网、针刺等无纺工艺织成无纺布;
S6. 将无纺布经裁切机进行裁切成需要的敷芯;
S7. 将敷芯与带孔硅酮凝胶薄膜、聚氨酯薄膜胶带、防粘层经敷贴机复合;
S8. 经包装、灭菌,即得到带凝血酶的海藻酸盐伤口敷贴成品。
进一步的,所述的凝血酶是人体凝血酶、牛凝血酶或重组凝血酶。
进一步的,所述的带凝血酶的海藻酸盐无纺布针刺频率为250~500针/min。
更进一步的,所述的带凝血酶的海藻酸盐无纺布克重为120~150g/m2
进一步的,所述的带孔硅酮凝胶薄膜,孔径为0.5~1mm,剥离强度为1~3N/cm。
进一步的,所述的医用聚氨酯薄膜胶带上表面涂覆有压敏胶。
进一步的,所述的压敏胶为丙烯酸压敏胶、聚氨酯压敏胶和医用聚异丁烯压敏胶中的任意一种。
更进一步的,所述的带孔硅酮凝胶薄膜与医用聚氨酯薄膜胶带等面积,且带孔硅酮凝胶薄膜覆盖敷芯层与医用聚氨酯薄膜胶带粘贴,做成袋装将敷芯包裹在其中。
进一步的,所述的防粘层为格拉辛硅油纸,具有易剥离的优点;所述防粘层为两页,分别从敷贴两端往中间延伸直到相互覆盖,并与敷贴外缘粘结固定。
更进一步的,所述的灭菌方式为钴-60辐照灭菌,辐照剂量为25kGy~50kGy,辐照时间为2h~3h。
与现有技术相比,本发明具有以下有益效果:
(1)本发明通过“S”型通道浸泡法在海藻酸盐纤维上负载凝血酶,“S”型通道增大了纤维与凝血酶的接触面积,同时确保恒定的反应温度,能最大限度的保证凝血酶附着于纤维上的均匀性,解决了蛋白质在敷料基材上难以固定的问题。
(2)海藻酸盐接触伤口渗出液后,释放钙离子,与凝血酶双重作用,加强对伤口的止血效果。
(3)带孔硅酮凝胶薄膜与敷芯复合,可以避免海藻酸盐纤维碎屑遗落在伤口上,起到防粘连作用,揭开敷贴时不会对伤口造成二次伤害,且其为惰性疏水材料,可减少对皮肤的致敏率。
(4)聚氨酯薄膜防水透气,呈透明状,方便观察伤口情况,同时为创面提供湿润的密闭环境,促进伤口愈合。
具体实施例
下面将结合具体实施例对本发明做进一步详细说明。
实施例1:
一种带凝血酶的海藻酸盐伤口敷贴的制备方法,其制备过程如下:
(1)配置凝血酶的乙醇水溶液,乙醇与水体积比为2:1,凝血酶含量为50单位/ml;
(2)将海藻酸盐长束纤维放入含凝血酶乙醇水溶液的反应槽中恒温浸泡,反应槽设置“S”型通道,浸泡时间为0.5h,温度为30℃;
(3)浸泡过后,将长束纤维拉出反应槽,经烘箱干燥,干燥温度为35℃,干燥时间为1h;
(4)干燥后,经裁切工艺,制成一定长度的带有凝血酶的海藻酸盐短纤维,并将短纤维卷曲,裁切短纤维长度为38mm,卷曲数为3个/cm;
(5)将短纤维经开松、梳理、铺网、针刺等无纺工艺织成无纺布,针刺频率为250针/min,无纺布克重为120g/m2
(6)将无纺布经裁切机进行裁切成需要的敷芯;
(7)将敷芯与带孔硅酮凝胶薄膜、聚氨酯薄膜胶带、防粘层经敷贴机复合,带孔硅酮凝胶薄膜孔径为0.5mm,剥离强度为1N/cm;
(8)经包装、灭菌,即得到带凝血酶的海藻酸盐伤口敷贴成品,辐照剂量为28kGy,辐照时间为2h。
实施例2:
一种带凝血酶的海藻酸盐伤口敷贴的制备方法,其制备过程如下:
(1)配置凝血酶的乙醇水溶液,乙醇与水体积比为3:1,凝血酶含量为100单位/ml;
(2)将海藻酸盐长束纤维放入含凝血酶乙醇水溶液的反应槽中恒温浸泡,反应槽设置“S”型通道,浸泡时间为1h,温度为35℃;
(3)浸泡过后,将长束纤维拉出反应槽,经烘箱干燥,干燥温度为37℃,干燥时间为2h;
(4)干燥后,经裁切工艺,制成一定长度的带有凝血酶的海藻酸盐短纤维,并将短纤维卷曲,裁切短纤维长度为60mm,卷曲数为4个/cm;
(5)将短纤维经开松、梳理、铺网、针刺等无纺工艺织成无纺布,针刺频率为300针/min,无纺布克重为140g/m2
(6)将无纺布经裁切机进行裁切成需要的敷芯;
(7)将敷芯与带孔硅酮凝胶薄膜、聚氨酯薄膜胶带、防粘层经敷贴机复合,带孔硅酮凝胶薄膜孔径为0.8mm,剥离强度为2N/cm;
(8)经包装、灭菌,即得到带凝血酶的海藻酸盐伤口敷贴成品,辐照剂量为38kGy,辐照时间为3h。
实施例3:止血性能测试
试验组:选取实施例2制得的产品做为试验组;
对照组:市面上纱布止血产品为对照组。
测试步骤如下:
(1)按照1ml/kg,在兔子耳缘静脉缓慢注入3%戊巴比妥钠,进行麻醉,然后在兔子左腹侧部铺上创巾,用碘酒、75%酒精消毒手术部位。
(2)沿左侧内缘,腹中线左侧约1cm做5.0cm纵向切口,切开腹肌至腹膜层,剪开腹膜。
(3)用2个手指或镊子,找出兔子的脾脏,并用生理盐水保持脾脏湿润。
(4)空白对照组:在脾脏的脏面,用手术刀片沿脾脏长轴划开长1cm,深1mm的两条对称性伤口,创面出血活跃。观察创面出血情况,并记录兔子自动止血的时间。
(5)对照组:在脾脏的脏面,用手术刀片沿脾脏长轴划开长1cm,深1mm的伤口,创面出血活跃。10秒后敷以普通纱布止血,手指轻压1分钟,以无活动出血为度。1分钟后,观察创面出血情况,若2分钟内未再出血,即认为止血成功,否则去除创面血凝块和纱布,重新敷药,并手指轻压1分钟,如此反复,直至成功止血。记录止血时间,并观察止血制剂与创面的粘合情况。
(6)试验组:在脾脏的脏面,用手术刀片沿脾脏长轴划开长1cm,深1mm的伤口,创面出血活跃。10秒后敷以带凝血酶的藻酸盐伤口敷贴止血,手指轻压1分钟,以无活动出血为度1分钟后,观察创面出血情况,若2分钟内未再出血,即认为止血成功,否则去除创面血凝块和带凝血酶的藻酸盐伤口敷贴,重新敷药,并手指轻压1分钟,如此反复,直至成功止血。记录止血时间,并观察止血制剂与创面的粘合情况。
试验结果如表1所示:

从试验结果可以看出,本发明产品与现有市场上的止血纱布相比,具有优秀的快速止血性能。

S1. 配置凝血酶的乙醇水溶液,乙醇与水体积比为(2~3)﹕1,凝血酶含量为50~100单位/ml;
S2. 将海藻酸盐长束纤维放入含凝血酶乙醇水溶液的反应槽中恒温浸泡0.5~1h,温度为30~35℃,反应槽设置“S”型通道;
S3. 浸泡过后,将长束纤维拉出反应槽,经烘箱干燥,干燥温度为35~37℃,干燥时间为1~2h;
S4. 干燥后,经裁切工艺,制成长度为38~65mm的带有凝血酶的海藻酸盐短纤维,并将短纤维卷曲,卷曲数不少于3个/cm;
S5. 将短纤维经开松、梳理、铺网、针刺无纺工艺织成无纺布;
S6. 将无纺布经裁切机进行裁切成需要的敷芯;
S7. 将敷芯与带孔硅酮凝胶薄膜、聚氨酯薄膜胶带、防粘层经敷贴机复合;S8. 经包装、灭菌,即得到带凝血酶的海藻酸盐伤口敷贴成品。
2.根据权利要求1所述的一种带凝血酶的海藻酸盐伤口敷贴的制备方法,其特征在于,所述的凝血酶是人体凝血酶、牛凝血酶或重组凝血酶。
