Abstract
The invention belongs to the field of medicines, and particularly relates to a method for constructing a trauma animal model and promoting wound healing. The method for promoting wound healing takes phenytoin silver, dendrobium officinale and hyaluronic acid as active ingredients, is externally applied to the wound, has the effects of stopping bleeding, easing pain, relieving itching, inhibiting bacterial infection of the wound, promoting wound healing and reducing scars, can be widely used for treating various traumas such as incised wound, traumatic injury, burn and scald, frostbite and the like, can obviously shorten the healing time of various traumas, relieves pain and pruritus of the wound and reduces the area of scars.

The invention belongs to the field of medicines, and particularly relates to a method for constructing a trauma animal model and promoting wound healing.
Background
The skin is the protective tissue of the body surface of the animal, is composed of epidermis, dermis and subcutaneous tissue, is an important barrier of the animal body, can prevent various tissues and organs in the animal body from being invaded and damaged by physical, chemical or pathogenic bacteria, and maintains the relative stability of the environment in the animal body. Trauma is one type of skin injury, and when the skin is injured, regardless of the size of the wound, bleeding, bacterial infection, suppuration, ulceration, even systemic infection, tetanus and the like can be caused if the skin is not effectively treated, so that the life of an animal can be seriously threatened. In addition, severe pain is often caused when trauma is serious, itching is also accompanied in wound infection, ulceration and healing processes, the daily life of a patient is seriously influenced, when large-area epidermis or dermis is damaged due to trauma, the skin cannot be regenerated, scars are easily generated even if the wound is healed, and troubles are brought to the life of the patient after the wound is healed. Therefore, the trauma should be treated timely and effectively to avoid more damage to the animal.
Although the existing medicines for trauma are of various types, including Yunnan white medicines, band-aid and the like, the medicines are relatively single in function, are usually only one of the functions of easing pain, promoting blood circulation to remove blood stasis, inhibiting infection and the like, and cannot simultaneously improve the life quality of a patient, shorten the healing time of a wound and reduce the scar area after the wound is healed. Accordingly, the present invention is directed to providing a pharmaceutical composition effective in promoting wound healing.
Disclosure of Invention
An object of the present invention is to provide a method for promoting wound healing, which has hemostatic, analgesic, antipruritic, wound bacterial infection inhibiting, wound healing promoting and scar reducing effects by topically coating a traumatic wound with phenytoin silver, dendrobium officinale and hyaluronic acid as active ingredients.
The invention also aims to provide a pharmaceutical composition for promoting wound healing, which comprises the active ingredients of phenytoin silver, dendrobium officinale and hyaluronic acid.
The wound is trauma, preferably, the trauma is selected from knife wound, traumatic injury, burn and scald, frostbite and the like.
The weight ratio of the phenytoin silver to the dendrobium officinale to the hyaluronic acid is 1: 10-20: 10-25, and preferably the weight ratio of the phenytoin silver to the dendrobium officinale to the hyaluronic acid is 1: 15: 20.
The pharmaceutical composition for promoting wound healing is a topical preparation, preferably, the topical preparation can be a spray, an ointment, a patch, a powder, granules or a lotion and the like, and more preferably, the pharmaceutical composition for promoting wound healing further comprises a film-forming agent and/or a film-forming auxiliary agent.
Still another object of the present invention is to provide the use of phenytoin silver, dendrobium officinale and hyaluronic acid for preparing a pharmaceutical composition for promoting wound healing.
The wound is trauma, preferably, the trauma is selected from knife wound, traumatic injury, burn and scald, frostbite and the like.
The weight ratio of the phenytoin silver to the dendrobium officinale to the hyaluronic acid is 1: 10-20: 10-25, and preferably the weight ratio of the phenytoin silver to the dendrobium officinale to the hyaluronic acid is 1: 15: 20.
Phenytoin is an antiepileptic and antiarrhythmic drug and has a certain analgesic effect, silver ions are one of common antibacterial drugs in the field, and CN 104016923A discloses a phenytoin derivative, wherein sodium phenytoin and a silver ammonia solution are subjected to chemical reaction to obtain a silver salt of a compound 5, 5-diphenyl-2, 4 imidazolidinedione, and the silver salt is named as phenytoin silver.
The Dendrobium officinale is a plant of the genus Dendrobium of the family Orchidaceae, and can nourish yin, nourish body fluid, tonify spleen and stomach, and nourish skin, and modern pharmacological studies also show that the Dendrobium officinale also has the efficacy of hemostasis and can be used for trauma hemostasis.
Hyaluronic acid, also known as uronic acid and hyaluronic acid, is widely present in various parts of the human body, wherein the skin contains a large amount of hyaluronic acid, and has the effects of heat preservation, skin tendering, wrinkle removal and aging prevention, and meanwhile, hyaluronic acid is also an effective transdermal absorption enhancer which can effectively promote the transdermal absorption of nutrient components, so that the hyaluronic acid is widely applied to various cosmetics.

The preparation method comprises the following steps:
(1) respectively crushing the dendrobium officinale, the hyaluronic acid and the powder auxiliary materials, and then sieving the crushed materials with a 100-mesh sieve, wherein the dendrobium officinale, the hyaluronic acid and the powder auxiliary materials are respectively equally divided into 4 parts for later use;

The lotion for promoting wound healing comprises the following components in parts by weight:
10g of phenytoin silver, 200g of dendrobium officinale, 250g of hyaluronic acid and 1540g of lotion auxiliary materials.
The lotion for promoting wound healing of the invention is prepared according to the conventional preparation method of the lotion in the field.
Comparative examples 1 to 3: powder for promoting wound healing
Powders for promoting wound healing of comparative examples 1 to 3 were prepared according to the method of example 1 except that comparative example 1 contains phenytoin silver (20g) and dendrobium officinale (190g) as active ingredients, comparative example 2 contains phenytoin silver (20g) and hyaluronic acid (340g) as active ingredients, and comparative example 3 contains only powder excipients, and the amounts and preparation methods of the ingredients were the same as those of example 1.
Example of effects: effect of the pharmaceutical composition of the present invention on the healing time of traumatic wound in rats
