Abstract
硫辛酸作为兔巴氏杆菌病疫苗佐剂的应用,能够显著提高免疫效率降低死亡率,并且安全、有效、不会引起毒副作用。
Description
技术领域
本发明涉及硫辛酸作为疫苗佐剂的应用。
背景技术
佐剂是一种能够增强对疫苗组分抗原特异性免疫应答或改变免疫反应的物质。在疫苗制剂中,佐剂的功能主要包括:增强疫苗抗原的免疫原性;促进细胞免疫和体液免疫,优化免疫应答,促进免疫能力较弱人群中的免疫应答;增进抗原与黏膜之间的传递以及免疫接触;减少疫苗成分中抗原的需求量以及在实施过程中的免疫接种次数;优化抗原结构,维持抗原构象等。佐剂的种类丰富,根据其功能,可分为运载传递***和免疫刺激性佐剂。前者包括运载体即能与半抗原结合的一类具有免疫原性质的物质,还包括对整个疫苗***起运输传递和衬托作用的一类物质。根据佐剂的来源,可分为细菌性、非细菌性、微生物性、细胞因子类以及合成体类等。根据理化性质,可分为颗粒性、凝胶性、乳胶佐剂等。跟据其性能,可分为治疗性佐剂、免疫刺激性佐剂、黏膜佐剂以及基因佐剂等。
硫辛酸是一种存在于线粒体的辅酶,类似维他命,能消除加速老化与致病的自由基。硫辛酸在体内经肠道吸收后进入细胞,兼具脂溶性与水溶性的特性,因此可以在全身通行无阻,到达任何一个细胞部位,提供人体全面效能,是具脂溶性与水溶性的万能抗氧化剂。目前硫辛酸主要用于急性及慢性肝炎、肝硬变、肝性昏迷等。B族维生素,用于治疗急性及慢性肝炎、肝硬化、肝性昏迷、脂肪肝、糖尿病等。
发明内容
本发明的目的一方面是拓展硫辛酸的新的应用领域,另一方面是提供一种更高效的疫苗佐剂,增强对兔巴氏杆菌病疫苗抗原特异性免疫应答。
为了实现上述目的,本发明采用了以下技术方案:

与现有技术相比较,本发明具备的有益效果:
将硫辛酸作为兔巴氏杆菌病疫苗佐剂能够显著提高免疫效率降低死亡率,并且安全、有效、不会引起毒副作用。
具体实施方式
下面通过实施例对本发明的技术方案作进一步阐述。
实施例1
(1)简介
兔巴氏杆菌病是由Fo型多杀性巴氏杆菌引起的,其血清型为7:A、5:A,家兔中较常发生,一般无季节性,以冷热交替、气温骤变,闷热、潮湿多雨季节发生较多。改病的传统防治方法主要是平时加强饲养管理,改善环境卫生,改善兔子食欲为主,注意保暖防寒,防治寄生虫病等;定期进行检疫;兔舍、用具要严格消毒。预防时,可用兔巴氏杆菌氢氧化铝菌苗肌肉注射或禽巴氏杆菌菌苗免疫肌肉注射,每年两次,对预防本病有一定效果。病兔可用链霉素、诺氟沙星、增效磺胺及头孢菌素等治疗。
(2)免疫试验
将30只生长健康状态良好的平均体重1.2千克的中国白兔随机分配成三组:对照组、疫苗组和疫苗+硫辛酸组;对照组每只白兔肌肉注射1毫升生理盐水;疫苗组每只白兔肌肉注射1毫升市售兔用巴氏杆菌灭活菌苗;疫苗+硫辛酸组每只白兔肌肉注射1毫升市售兔用巴氏杆菌灭活菌苗和内含硫辛酸200μmoL的混合物。第一次注射后第8天再次进行一次注射,第一次注射后第20天,采用多杀巴氏杆菌攻毒,剂量为450cfu/只,观察15天,统计白兔的死亡情况,并计算相对免疫保护率,结果如表1所示。
表1试验结果
表1中相对免疫保护率一是与对照组相比的计算结果,二是与疫苗组相比的计算结果,从表1可以看出,试验白兔接受市售兔用巴氏杆菌灭活菌苗免疫后,与对照组相比,白兔死亡率有所下降。而疫苗+硫辛酸共同免疫后,比疫苗组的相对免疫保护率提高了66.67%,说明硫辛酸能够提高市售兔用巴氏杆菌灭活菌苗的保护效率。
