本发明公开了一种连翘挥发油的提取工艺,包括如下步骤:取连翘药材,粉碎筛选;加入纤维素酶酶解,喷洒pH4.5‑5.5的醋酸/醋酸钠缓冲溶液,达到手握成团即可;采用连续逆流超声提取,得到连翘挥发油。本发明制得的连翘挥发油25℃时的相对密度为0.6000‑0.92000,折光率为1.2000‑1.6000,旋光度为‑2°—+3°。本发明的提取方法简单易行,不需要大量的有机溶剂,安全无毒,成本低,污染小,适用于工业化生产。

技术领域
本发明涉及中药技术领域,具体涉及一种连翘挥发油的提取工艺。
背景技术
连翘是木犀科连翘属植物,主要含苯乙醇苷类、木脂体及其苷类、五环三萜类、挥发油类等化合物。药理研究认为其具有抗菌、抗炎、解热、镇吐、利尿强心、抗肝损、镇痛、抑制磷酸二酯酶、抗病毒、降血压、抑制弹性蛋白酶活力、抗内毒素等作用。除果实入药,具清热解毒、消结排脓之效外,药用其叶,对治疗高血压、痢疾、咽喉痛等效果较好。
连翘对多种革兰阳性及阴性细菌均有抑制作用。其浓缩煎剂在体外可抑制伤寒杆菌、副伤寒杆菌、大肠杆菌、痢疾杆菌、白喉杆菌及霍乱弧菌、葡萄球菌、链球菌等,连翘在体外的抑菌作用与金银花大体相似,为银翘散中抗菌的主要成分。
发明内容
本发明的目的是提供一种连翘挥发油的提取工艺。
一种连翘挥发油的提取工艺,其特征在于,包括如下步骤:
a.取连翘药材,粉碎,过40目筛,再过100目筛,取截留物;
b.加入原料质量1-3%的纤维素酶,喷洒相当原料0.6-1.8倍的pH4.5-5.5的醋酸/醋酸钠缓冲溶液,达到手握成团即可;
c.取发酵后的原料,加入相当于原料质量4-6倍的60-70%的乙醇溶液,采用连续逆流超声提取,过滤,滤液经有机溶剂萃取、减压浓缩等步骤,得到连翘挥发油。
步骤b中所述的纤维素酶的活性为35-60U/mg。
步骤c中所述的超声频率45-60KHZ,于70~80℃的条件下提取2-3h。
步骤c中所述的有机溶剂为石油醚或***中的一种。
本发明具有安全无毒、耗时短、提取率高、工艺简单、可连续操作的优点。
具体实施方式:
以下通过实例对本发明进行阐述,但本发明的保护并不局限于此。
实施例1:
取连翘药材,粉碎,过40目筛,再过100目筛,取截留物,加入原料质量1%的纤维素酶(活性为30U/mg),喷洒相当原料0.6倍的pH4.5的醋酸/醋酸钠缓冲溶液,达到手握成团即可;取发酵后的原料,加入相当于原料质量4倍的60%的乙醇溶液,采用连续逆流超声提取法,于频率45KHz、温度70℃的条件下连续提取2h,过滤,滤液经石油醚萃取、减压浓缩等步骤,得到黄色的连翘挥发油,得率为2.1%。
实施例2:
取连翘药材,粉碎,过40目筛,再过100目筛,取截留物,加入原料质量3%的纤维素酶(活性为50U/mg),喷洒相当原料1.8倍的pH5.5的醋酸/醋酸钠缓冲溶液,达到手握成团即可;取发酵后的原料,加入相当于原料质量6倍的70%的乙醇溶液,采用连续逆流超声提取法,于频率60KHz、温度80℃的条件下连续提取3h,过滤,滤液经***萃取、减压浓缩等步骤,得到黄色的连翘挥发油,得率为3.2%。

1.一种连翘挥发油的提取工艺,其特征在于,包括如下步骤:
a.取连翘药材,粉碎,过40目筛,再过100目筛,取截留物;
b.加入原料质量1-3%的纤维素酶,喷洒相当原料0.6-1.8倍的pH4.5-5.5的醋酸/醋酸钠缓冲溶液,达到手握成团即可;
c.取发酵后的原料,加入相当于原料质量4-6倍的60-70%的乙醇溶液,采用连续逆流超声提取,过滤,滤液经有机溶剂萃取、减压浓缩等步骤,得到连翘挥发油。
2.根据权利要求1所述的一种连翘挥发油的提取工艺,其特征在于,步骤b中所述的纤维素酶的活性为35-60U/mg。
3.根据权利要求1所述的一种连翘挥发油的提取工艺,其特征在于,步骤c中所述的超声频率45-60KHZ,于70~80℃的条件下提取2-3h。
4.根据权利要求1所述的一种连翘挥发油的提取工艺,其特征在于,步骤c中所述的有机溶剂为石油醚或***中的一种。
