Abstract

含氨基酸结构的杯[4]芳烃的一种合成方法,属于化学合成技术领域,本发明反应温度为室温,反应和重结晶中采用无水乙醇为溶剂,无水硫酸钠为助催化剂,原料廉价易得,实现了一锅两步反应,操作简单,条件温和。

含氨基酸结构的杯[4]芳烃的一种合成方法
技术领域
本发明属于化学合成技术领域,特别是涉及化合物4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四(3-氮杂正丁酸-4-)基杯[4]芳烃的制备技术。
背景技术
杯芳烃是一类具有较好生物和催化活性的有机化合物,而氨基酸因其独特的亲酸和亲碱的结构在生命化学和材料化学中应用广泛,一直以来,将氨基酸枝接在杯芳烃上以研究二者共同作用后的理化特性是化学家们的追求,遗憾的是,将氨基酸枝接在杯芳烃上尚未见文献报道有较好的方法。
4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四(3-氮杂正丁酸-4-)基杯[4]芳烃是首次人工合成的含有氨基酸官能团的杯芳烃类有机化合物,该化合物具有八个羟基和四个羧基,极性比较强,同时该化合物具有杯芳烃母体骨架和四个正戊基非极性官能团,因此该化合物具有潜在的药物和材料方面的应用价值。
发明内容
本发明的提供一种合成4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四(3-氮杂正丁酸-4-)基杯[4]芳烃的方法。
本发明技术方案包括以下步骤:
1)将4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四甲酰基杯[4]芳烃、甘氨酸乙酯盐酸盐、三乙胺和无水乙醇置于反应器中,常温搅拌至反应结束;
2)向上述反应器中加入无水硫酸钠,搅拌后过滤出固体,将固体转移到含有硼氢化钠的乙醇溶液中,常温搅拌至反应结束;
3)过滤出固体并转移至氢氧化钠水溶液中,充分搅拌后过滤掉不溶杂质,母液用盐酸调pH至7.0,抽滤出固体,用乙醇重结晶,得到产物4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四(3-氮杂正丁酸-4-)基杯[4]芳烃。
本发明的反应式为:
本发明反应温度为室温,反应和重结晶中采用无水乙醇为溶剂,无水硫酸钠为助催化剂,原料廉价易得,实现了一锅两步反应,操作简单,条件温和。
进一步地,本发明所述步骤1)中反应时间为2~3小时。时间不足时,反应不彻底,实验表明在该时间内,产物产率最高。
所述步骤2)中反应时间为3~5小时。时间不足时,反应不彻底,实验表明在该时间内,产物产率最高。
所述4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四甲酰基杯[4]芳烃、甘氨酸乙酯盐酸盐、三乙胺、硼氢化钠和无水硫酸钠的投料摩尔比为1∶4∶4∶16.4∶2。此配比量得到的产率最高。
所述氢氧化钠水溶液的质量百分数为10%。
所述盐酸的质量百分数为10%。

具体实施方式
二、合成方法:
向含有20mL乙醇的100mL圆底烧瓶中加入1mmo的4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四甲酰基杯[4]芳烃,4mmol甘氨酸乙酯盐酸盐,4mmol三乙胺,常温搅拌3h。向反应器中加入2mmol的无水硫酸钠,搅拌后过滤出固体,将固体转移到含有16.4mmol的硼氢化钠的50mL乙醇溶液中。常温搅拌4h。过滤出固体并转移至30 mL 质量百分数为10%的氢氧化钠水溶液中,充分搅拌后过滤掉不溶杂质,母液用质量百分数为10%的盐酸调pH至7.0,抽滤出固体,用乙醇重结晶,得到产物4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四(3-氮杂正丁酸-4-)基杯[4]芳烃。

二、产物鉴定:
产物氢谱图见图1,从图1可见:和氮原子相连的八组氢全部出现,说明采用本发明方法成功地合成了4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四(3-氮杂正丁酸-4-)基杯[4]芳烃。

Claims (5)

1.含氨基酸结构的杯[4]芳烃的一种合成方法,其特征在于包括以下步骤:
1)将4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四甲酰基杯[4]芳烃、甘氨酸乙酯盐酸盐、三乙胺和无水乙醇置于反应器中,常温搅拌至反应结束;
2)向上述反应器中加入无水硫酸钠,搅拌后过滤出固体,将固体转移到含有硼氢化钠的乙醇溶液中,常温搅拌至反应结束;
3)过滤出固体并转移至氢氧化钠水溶液中,充分搅拌后过滤掉不溶杂质,母液用盐酸调pH至7.0,抽滤出固体,用乙醇重结晶,得到产物4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四(3-氮杂正丁酸-4-)基杯[4]芳烃;
所述4,6,10,12,16,18,22,24-八羟基-2,8,14,20-四正戊基-5,11,17,23-四甲酰基杯[4]芳烃、甘氨酸乙酯盐酸盐、三乙胺、硼氢化钠和无水硫酸钠的投料摩尔比为1∶4∶4∶16.4∶2。

4.根据权利要求1所述的含氨基酸结构的杯[4]芳烃的一种合成方法,其特征在于:所述氢氧化钠水溶液的质量百分数为10%。
