本发明涉及一种藤本豆肽营养品及其制备方法,藤本豆肽营养品含有1~80%重量百分比的肽,还含亚油酸、亚麻酸、油酸、双歧因子、黄酮类物质、皂甙、VB、VC、VE、VA、胡萝卜素、钙、锌、铁、钾、镁等营养素及水;所述的藤本豆肽营养品是这样制得的:将藤本豆榨油后的粕或藤本豆加纯净水经离心研磨制成浆液,提取蛋白;再经胶体磨及高压均质机多次研磨、细分,均质压力为65~80Mpa,细分至510~600目;将均质后的浆液在124~135℃的温度下脉冲式冲击与加热处理,然后用蛋白酶降解缩合肽,得藤本豆肽原液,进而制成藤本豆肽营养品。

背景技术
藤本豆是人类很好的天然保健品。其营养极其丰富,富含全价优质蛋白和配比均衡的十八种氨基酸,其对人体所必需的氨基酸含量高达50%以上,是一种比大豆蛋白更具营养价值的优质蛋白资源。另外,藤本豆中还含有对人体有益的钾、钙、镁、铁、锰、锌、铜、磷、硒等十多种无机元素。经专家反复试验,藤本豆的药用和保健功能显著,特别是对高胆固醇症,具有调节血清胆固醇水平功能,对心脑血管有很好的保健功效。其蛋白质含量高达23~26%,粗脂肪含量可达15%左右,且含有大量的不饱和脂肪酸,特别是亚油酸等是人体必需而又不能在人体合成的脂肪酸,这些成份都是人体细胞的重要组成物质,参与细胞成份磷脂的合成,形成细胞膜,与胆固醇结合参与体内新陈代谢,亚油酸在动物体内与胆固醇结合成脂而使其降解为胆酸,然后排出体外。故有降低血脂、血压、胆固醇的特殊生理保健功能和医疗作用。
虽然藤本豆含有丰富的营养成分,尤其是所含的蛋白质是一种优质蛋白质,但是目前人们将藤本豆榨油后,均将其榨油后的粕仅仅用作饲料,使得优质的蛋白不能得到很好的利用。

本发明所要解决的问题是提供一种以藤本豆榨油后的粕为原料或以藤本豆为原料,用生物工程技术制备的藤本豆肽(低聚肽、短肽和多肽)营养品及其制备方法,以便使营养丰富的藤本豆能够更方便地为广大消费者带来健康。
以藤本豆榨油后的粕或藤本豆为原料制备藤本豆肽营养品需要解决以下技术难题:一是解决藤本豆蛋白质降解易形成大量氨基酸而产生极少量肽,即转化为肽的转化率低下的难题;二是解决部分未榨完残留的或全部脂肪转化为各种低级脂肪酸的难题;三是解决藤本豆蛋白抗营养性、肠胃不适、易痛风和胀气等不良作用。
本方面是采用生物工程中的酶工程、组织蛋白工程和基因工程及现代物理学纳米技术,将藤本豆(或粕)中的蛋白质链断裂后降解,再进行生物技术缩合而制得的藤本豆肽(低聚肽、短肽、多肽)营养品(口服液、粉、胶囊、片剂和高肽快速增能活性运动饮),其肽的缩合率(即肽与氨基酸的比)达到85%以上。
本发明提供的藤本豆肽营养品含有1~80%重量百分比的肽,还含亚油酸、亚麻酸、油酸、双歧因子、黄酮类物质、磷脂、皂甙、VB、VC、VE、VA、胡萝卜素、钙、锌、铁、钾、镁等营养素及水;所述的藤本豆肽营养品是这样制得的:将藤本豆榨油后的粕或藤本豆加纯净水经离心研磨制成浆液,提取蛋白;再经胶体磨及高压均质机多次研磨、细分,均质压力为65~80Mpa,细分至510~600目;将均质后的浆液在124~135℃的温度下脉冲式冲击与加热处理,然后用脂肪酶降解缩合肽,得藤本豆肽原液(低聚肽、短肽和多肽),进而制成藤本豆肽营养品。
上述藤本豆肽营养品是藤本豆肽口服液、藤本豆肽粉、藤本豆肽胶囊、藤本豆肽片剂或藤本豆肽高肽快速增能活性运动饮料。
本发明提供的藤本豆肽营养品的制备方法,其步骤如下:
(1)、浸泡:取新鲜的、无杂质、无脂肪氧化、无霉变的藤本豆粕或藤本豆,清洗干净后加入纯净水浸泡3~5小时,水温控制在25~35℃;藤本豆粕或藤本豆与水的重量比为1∶6~10;
(2)、提取蛋白:通过将浸泡后藤本豆粕或藤本豆加水制浆,用浆渣分离机提取蛋白,制成蛋白浆液;
(3)、研磨、细分:将蛋白液浆投入胶体磨中研磨2~3遍,细度调至310~400目;
(4)、均质、再细分:经过胶体磨处理后的浆液再进入高压均质机中均质3~4遍,均质压力为65~80Mpa,使藤本豆粕或豆中的蛋白质细分在510~600目,使其分散的更均匀,从而达到:a、克服藤本豆的有害副作用;b、有利于藤本豆蛋白和脂肪与尔后的各种酶制剂充分接触交汇;c、有利于放大酶制剂的生化作用;
(5)、去害、变性:将步骤(4)均质处理的浆液在124~135℃的温度下脉冲式冲击与加热处理10~12分钟,使浆液中的蛋白质充分变性,进一步克服藤本豆的前述等有害副作用和使其在下一工序中更易打开蛋白链;
(6)、生化:将上述浆液经换热器通冷却至35~55℃,在冷却后的浆液中加入脂肪酶搅拌均匀,静置生化3~5小时,然后将物料温度调整至40~55℃,加入中性蛋白酶,以2700~2750转/分钟搅拌,使酶制剂与浆液充分混合,保持恒温40~55℃生化4~7个小时;通常情况下搅拌6~8分钟即可,中性蛋白酶的加入量为物料重量的0.03~0.25%;脂肪酶的加入量为物料重量的0.01~0.03%;
(7)、灭酶:将生化后的浆液升温至68~85℃灭酶,灭酶时间15~35秒;
(8)、析出:先给板框析出***中预涂助滤剂,助滤剂采用食品级硅藻土,物料析出处理的温度为70~85℃,即得藤本豆肽原液,进而制成藤本豆肽营养品。
析出的藤本豆肽原液含肽量为1.2~3.0%(重量百分比)左右,为了使其保健功能满足人们的需要,可经过多级浓缩***浓缩处理,可得含肽量大于6%的藤本豆肽营养液。经多级过滤可制作含肽量≥6%的藤本豆肽口服液、亦可得含肽量≥8%的藤本豆高肽快速增能活性运动饮料,也可将其打入真空喷雾干燥***制成藤本豆肽粉或藤本豆肽胶囊、藤本豆肽片剂等。
步骤(8)中含食品级硅藻土助滤剂的板框析出***中涂覆在板框上的食品级硅藻土助滤剂的重量按板框滤网面积计算为0.5~0.8公斤/平方米。
本发明运用现代生物工程技术中的酶工程和组织蛋白工程技术及现代物理学纳米技术将藤本豆粕或藤本豆的蛋白质及部分残留或全部的脂肪降解,制备出能为人体直接吸收的藤本豆肽营养品,其特殊的工艺方法步骤和有效参数所产生的显著性技术进步集中体现在把藤本豆由只能榨油等初加工的低水平阶段提高到深加工制成藤本豆肽(低聚肽、短肽和多肽)营养品的高层次水平,为保健品市场又增加了新的生力军。由于其不仅具有为人体直接、完全吸收且不会对肝、肾、脾、大脑等各器官造成高渗透压,从而具有迅速增强人体生命活力、快速补充能量、增强免疫力、抗氧化性、清除自由基、抗癌、非变应性促进脂代谢、降血脂、降血压、降胆固醇、促进双歧杆菌生长繁殖、恢复体力、抗疲劳和消炎、美容、抗衰老、改善人体新陈代谢等独特的营养保健效果,实为细胞营养“运载机”、细胞“修复器”和人体活力的“核能发动机”,且大大的提高了藤本豆的营养保健功能和经济的附加值,也带动了农副产品的发展,另藤本豆肽营养品所含有的亚麻酸、亚油酸、油酸、双歧因子、黄酮类物质、皂甙、磷脂、VB、VC、VA、VE、胡萝卜素、钙、镁、锌、铁、钾等营养成分对人体的健康也是必不可少的。
具体实施方式
以下结合实施例对本发明的内容加以详细说明。
实施例:
(1)、选取新鲜的、无杂质、无脂肪氧化、无霉变的藤本豆粕100公斤,用纯净水清洗后加入600kg纯净水浸泡3小时,水温为25℃;
(2)、制浆,通过浆渣分离机制取藤本豆蛋白液650Kg;
(3)、将上述蛋白液投入胶体磨中研磨2遍,细度达310目;
(4)、将胶体磨处理后的浆液再进入压力为65MPa(即650公斤压力)的高压均质机中进行高压均质,从而使藤本豆蛋白质细分得颗粒更小,达到510目,使其更均匀;
(5)、将上述浆液进行脉冲式冲击,并在124℃的温度下加热处理12分钟,使浆液中的蛋白质充分变性,为尔后的降解缩合奠定基础;
(6)、将上述变形处理后的藤本豆蛋白浆液经管式换热器40秒时间冷却至55℃后,加入占物料重量0.03%的脂肪酶搅拌均匀,静置生化3小时,再将物料温度调整至40℃,加入0.1%(重量比)的中性蛋白酶,经过2700转/分钟6分钟搅拌,然后保持恒温40℃生化4个小时;
(7)、将上述生化的浆液经过管式换热器将物料升温至68℃,进行灭酶处理15分钟;
(8)、将物料通过预涂好食品级硅藻土的板框析出***进行析出处理即得含肽量1.2~3.0%的肽原液500kg;预涂的食品级硅藻土的用量按板框析出***板框面积计为0.5公斤/平方米,物料温度必须调至70℃;
(9)、通过多级浓缩***浓缩可得150kg肽含量≥6%的藤本豆肽营养液;
(10)、将上述浓缩后的藤本豆肽营养液经多级过滤后,可直接进真空喷雾塔得20kg藤本豆肽粉或将粉,制成藤本豆肽片剂或者制成藤本豆肽胶囊,亦可将该液再浓缩制成100Kg含肽≥8%的藤本豆高肽快速增能活性运动饮料。
Claims (5)
1.一种藤本豆肽营养品,其特征是含有1~80%重量百分比的肽,还含亚油酸、亚麻酸、油酸、双歧因子、黄酮类物质、皂甙、磷脂、VB、VC、VE、VA、胡萝卜素、钙、锌、铁、钾、镁等营养素及水;所述的藤本豆肽营养品是这样制得的:将藤本豆榨油后的粕或藤本豆加纯净水经离心研磨制成浆液,提取蛋白;再经胶体磨及高压均质机多次研磨、细分,均质压力为65~80Mpa,细分至510~600目;将均质后的浆液在124~135℃的温度下脉冲式冲击与加热处理,然后用蛋白酶降解缩合肽,得藤本豆肽原液,进而制成藤本豆肽营养品。
2.根据权利要求1所述的藤本豆肽营养品,其特征是是藤本豆肽口服液、藤本豆肽粉、藤本豆肽胶囊、藤本豆肽片剂或藤本豆肽高肽快速增能活性运动饮料。
3.一种藤本豆肽营养品的制备方法,其步骤如下:
(1)、浸泡:取新鲜的、无杂质、无脂肪氧化、无霉变的藤本豆粕或藤本豆,清洗干净后加入纯净水浸泡3~5小时,水温控制在25~35℃;
(2)、提取蛋白:通过将浸泡后藤本豆粕或藤本豆加水制浆,用浆渣分离机提取蛋白,制成蛋白浆液;
(3)、研磨、细分:将蛋白液浆投入胶体磨中研磨2~3遍,细度调至310~400目;
(4)、均质、再细分:经过胶体磨处理后的浆液再进入高压均质机中均质3~4遍,均质压力为65~80Mpa,使藤本豆粕或豆中的蛋白质细分在510~600目;
(5)、去害、变性:将步骤(4)均质处理的浆液在124~135℃的温度下脉冲式冲击与加热处理10~12分钟;
(6)、生化:将上述浆液经换热器通冷却至35~55℃,在冷却后的浆液中加入脂肪酶搅拌均匀,静置生化3小时,然后将物料温度调整至40~55℃,加入中性蛋白酶,以2700~2750转/分钟搅拌,使酶制剂与浆液充分混合,保持恒温40~55℃生化4~7个小时;
(7)、灭酶:将生化后的浆液升温至68~85℃灭酶;
(8)、析出:先给板框析出***中预涂助滤剂,助滤剂采用食品级硅藻土,物料析出处理的温度为70~85℃,即得藤本豆肽原液,进而制成藤本豆肽营养品。
4.根据权利要求3所述的制备方法,其特征是步骤(6)中性蛋白酶的加入量为物料重量的0.03~0.25%;脂肪酶的加入量为物料重量的0.01~0.03%。
5.根据权利要求3所述的制备方法,其特征是步骤(8)中含食品级硅藻土助滤剂的板框析出***中涂覆在板框上的食品级硅藻土助滤剂的重量按板框滤网面积计算为0.5~0.8公斤/平方米。
