技术领域
本公开大体上涉及用于分离类胡萝卜素的材料和方法。
背景技术
类胡萝卜素是促成活生物体中观察到的许多鲜艳的黄色、橙色和红色的天然色素。类胡萝卜素广泛分布于自然界中并且执行许多重要的生物功能,例如充当光合作用中的捕光色素以及保护生物体免于光氧化损伤。
在人类中,类胡萝卜素提供强大的抗氧化作用,保护机体抵抗可由于正常代谢以及暴露于污染和其它环境危害而产生的自由基。类胡萝卜素还在食品和化妆品中用作天然色素,在营养制剂/制药产业中作为维生素A的膳食前体。
到2015年,预计类胡萝卜素的全球市场高达$12亿美元。目前,市场上接近90%的类胡萝卜素是通过化学合成生产的。然而,研发用于从天然来源分离类胡萝卜素的方法的兴趣正在增长。众所周知,某些植物含有大量的类胡萝卜素。
棕榈油是已知最丰富的天然植物类胡萝卜素来源之一。取决于其来源,棕榈油可具有500mg/kg到7,000mg/kg的类胡萝卜素。
已知许多用于从棕榈油回收类胡萝卜素或浓缩类胡萝卜素的方法。这些方法包括油的化学修饰和蒸馏、液-液萃取以及色谱术。
在一种已知方法中,对棕榈油进行酯交换,然后通过萃取和蒸馏来浓缩。在这种方法中,首先使棕榈油与醇系溶剂(例如形成脂肪酸酯的甲醇和甘油)进行酯交换。一旦使反应混合物分离,就用醇与水的混合物洗涤由具有溶解的类胡萝卜素的脂肪酸酯组成的上相,然后单独用水洗涤。通过蒸馏完全或部分去除脂肪酸酯,留下类胡萝卜素浓缩物。在真空中在最高达150℃的温度进行蒸馏步骤,所述蒸馏步骤可耗时约5小时。在这种方法中,棕榈油必定被转化成非食用酯以帮助分离棕榈油的组分。因此,被处理的棕榈油材料不能用于制备食品。另外,类胡萝卜素在蒸馏温度下容易分解,这降低了留在类胡萝卜素浓缩物中的类胡萝卜素的分离产率和纯度。
还已经使用超临界CO2从棕榈油分离类胡萝卜素。在这种方法中,将棕榈油添加到高压萃取容器中并且使超临界二氧化碳通过所述容器,在所述方法中萃取所述材料中的类胡萝卜素。收集萃取物,然后使二氧化碳从类胡萝卜素萃取物蒸发。虽然这种方法以良好的纯度产生了类胡萝卜素萃取物,但其缺点在于所述方法的高成本、高操作压力和类胡萝卜素在超临界二氧化碳中的极低溶解度。
鉴于现有分离方法的上述限制,因此需要研发用于从例如棕榈油的液体分离类胡萝卜素的高效的且有成本效益的方法,所述方法不将所述油修饰成非食用化合物。另外,还需要研发用于分离类胡萝卜素的材料。本公开满足了这些需要并且具有相关优点。
发明内容
根据第一方面,提供用于可逆地结合类胡萝卜素的材料,所述材料包含:
使得能够与所述类胡萝卜素可逆结合的量的偶合到银离子的支持物,并且其中,除银离子之外,所述支持物基本上不含过渡金属。
有利地,由于支持物基本上不含过渡金属,因此发生在类胡萝卜素与所述支持物之间的可逆结合的程度相较于相同的反应条件下含有过渡金属的支持物更大。在一个实施方案中,所述支持物是基本上不含过渡金属且偶合到或掺杂有银离子的合成粘土。
在实施方案中,所述支持物相对于天然粘土具有更大的表面积,这导致类胡萝卜素与所述支持物的更大结合。所述支持物可占约400m2/g到约700m2/g的表面积。在实施方案中,所述表面积可选自由以下组成的组:400m2/g、425m2/g、450m2/g、475m2/g、500m2/g、525m2/g、550m2/g、575m2/g、600m2/g、625m2/g、650m2/g、675m2/g和700m2/g。
根据第二方面,提供用于可逆地结合类胡萝卜素的方法,所述方法包括以下步骤:提供使得能够与所述类胡萝卜素可逆结合的量的偶合到银离子的支持物,并且其中,除银离子之外,所述支持物基本上不含过渡金属;使所述支持物与所述类胡萝卜素在结合条件下接触以使所述类胡萝卜素结合所述支持物;使所述类胡萝卜素在解离条件下解离以释放所述类胡萝卜素。
有利地,已发现,已与银离子偶合的支持物能够实现与类胡萝卜素的可逆结合。有利地,类胡萝卜素与掺杂银离子的支持物的可逆结合使得类胡萝卜素可容易在适当的有机溶剂中被萃取,而无需苛刻的萃取条件,例如,高温。有利地,萃取条件不使类胡萝卜素变性。
本文还公开了包括以下步骤的方法:使含有至少一种类胡萝卜素的液体与包含至少一种亲π键物质的组合物接触,由此形成包含类胡萝卜素络合物和所述液体的混合物;和从所述液体去除所述类胡萝卜素络合物。
所述方法可进一步包括以下步骤:使所述类胡萝卜素络合物与洗涤溶剂接触,由此形成洗过的类胡萝卜素络合物;和用有机溶剂从所述洗过的类胡萝卜素络合物萃取类胡萝卜素以形成含有至少一种类胡萝卜素的萃取相。所述方法可进一步包括以下步骤:通过蒸发从含有至少一种类胡萝卜素的所述萃取相去除所述有机溶剂,由此形成浓缩的类胡萝卜素萃取物。
有利地,所述方法可用于以高度有效且选择性的方式从含有类胡萝卜素的液体(例如棕榈油)分离类胡萝卜素。低至2重量%的组合物(相对于液体的重量)可用于有效地分离存在于液体中的大部分类胡萝卜素。
此外,可在本文所述的分离过程期间有效地使可存在于含有至少一种类胡萝卜素的液体中的化合物例如生育酚、三烯生育酚和胆固醇与回收的类胡萝卜素分开。因此,使用本文所述的方法分离的类胡萝卜素可基本上不含所述化合物。本文所述的方法的又一优点是当处理棕榈油时,回收的棕榈油未被分离过程修饰,因此可用于利用存在于棕榈油中的甘油酯的食品和其它应用中。
根据第三方面,提供合成锂蒙脱石粘土,其中存在于锂蒙脱石粘土中的至少一些钠离子已与银离子交换。在一个实施方案中,存在于锂蒙脱石粘土中的至少70mol%到100mol%的钠离子已与银离子交换。
在一个实施方案中,使用合成的掺杂银离子的锂蒙脱石粘土作为上文所公开的方法中的支持物。在实施方案中,已与银离子交换的钠离子的mol%可选自由以下组成的组:70mol%、75mol%、80mol%、85mol%、90mol%、95mol%和100mol%。
可容易地从市售的且廉价的起始材料制备根据第一方面的材料。所述材料还可再用,这导致处理成本的进一步降低。

本文提供用于从含有类胡萝卜素的液体分离类胡萝卜素的方法。合适的液体包括但不限于棕榈油、粗制棕榈油、粗制棕榈油精、红棕榈油/红棕榈油精、棕榈硬脂精、中和的红棕榈油、中和的红棕榈油精、向日葵油、椰子油、小麦胚芽油、胡萝卜油、大豆油、菜籽油、橄榄油和所述油的衍生物。
在一个实施方案中,所公开的方法包括以下步骤:提供使得能够与类胡萝卜素可逆结合的量的偶合到银离子的支持物,并且其中,除银离子之外,所述支持物基本上不含过渡金属;使所述支持物与所述类胡萝卜素在结合条件下接触以使所述类胡萝卜素结合所述支持物;使所述类胡萝卜素在解离条件下从所述支持物解离以释放所述类胡萝卜素。
所述支持物可被选择为相对于天然粘土具有更高表面积。所述支持物可被选择为具有400m2/g到700m2/g的表面积。
在一个实施方案中,选择用于上文公开的方法中的支持物是合成粘土。有利地,合成粘土在其化学组成上可为基本上均质的。合成粘土还可相对于非合成粘土支持物材料有利地展现更大的用于使类胡萝卜素与其结合的表面积。
所述方法可进一步包括以下步骤:在接触步骤之前,用银离子部分地或完全地交换支持物的固有阳离子的步骤。
在一个实施方案中,所述支持物被选择为具有下式:

含有至少一种类胡萝卜素的液体与掺杂银的支持物保持接触的时间量可为约5分钟到约12小时。在某些情况下,含有至少一种类胡萝卜素的液体与掺杂银的支持物接触达选自由以下组成的组的持续时间:约10分钟、约30分钟、约1小时、约2小时、约3小时、约4小时、约5小时、约6小时、约7小时、约8小时、约9小时、约10小时、约11小时和约12小时。在一个实施方案中,优选地,使类胡萝卜素与掺杂银的支持物接触的时间为约10分钟到约6小时。
在某些情况下,使含有至少一种类胡萝卜素的液体在连续过程中经过或通过掺杂银的支持物。
含有至少一种类胡萝卜素的液体与掺杂银的支持物可在接触时间期间混合,例如,搅动或搅拌,或静置,和其组合。可能的搅动元件包括传统搅拌器、旋转磁力棒、整合有容器的结构的囊装置(bladder device),以及用于操作混合容器的形状的不同装置。
在支持物与类胡萝卜素可逆结合后,可任选地通过蒸发、离心、过滤、倾析和/或通过用洗涤溶剂洗涤去除过量液体。

通常将洗涤溶剂相对于含有至少一种所用类胡萝卜素的液体以约1:1到约50:1(v/v)的比率添加到类胡萝卜素。可将洗涤溶剂相对于含有至少一种所用类胡萝卜素的液体以约1:1到约40:1、约1:1到约30:1、约1:1到约20:1、约1:1到约15:1、约1:1到约10:1或约5:1到约10:1(v/v)的比率添加到类胡萝卜素。
可使结合的类胡萝卜素与洗涤溶剂接触1次到10次,或直到已去除至少一部分或基本上全部与类胡萝卜素存在的甘油酯。在实施方案中,使类胡萝卜素与洗涤溶剂接触5-10次。可使用本领域技术人员已知的任何分析方法来监测洗涤溶剂中的甘油酯,所述分析方法例如IR光谱术、LCMS、GCMS、H-NMR、C-NMR等。
然后可通过与合适的有机溶剂接触,由此形成含有至少一种类胡萝卜素的萃取相并且使包含至少一种亲π键物质的支持物再生,从洗过的类胡萝卜素萃取类胡萝卜素。包含至少一种亲π键物质的再生支持物然后可再用于随后的类胡萝卜素回收过程中。所述方法进一步包括在使洗过的类胡萝卜素与所述有机溶剂接触的步骤之前,去除洗涤溶剂的步骤。

所要求保护的方法可用于回收含有至少一种类胡萝卜素的液体中的约10%到约99%的类胡萝卜素。所要求保护的方法可用于分离存在于含有至少一种类胡萝卜素的液体中的约10%到约90%、约10%到约85%、约10%到约80%、约10%到约75%、约10%到约70%、约20%到约70%、约30%到约70%或约50%到约70%的类胡萝卜素。
在实施方案中,本文中的方法包括以下步骤:

在一个实施方案中,所述支持物具有基本上均匀的化学组成。例如,所述支持物可为基本上不含过渡金属且偶合到银离子的合成粘土。在一个实施方案中,优选地,银离子是单价、二价或三价。
在相同的反应条件下,已有利地发现,掺杂银的合成粘土将相对于含有除银以外的过渡金属的天然粘土更容易结合类胡萝卜素和从类胡萝卜素解离。更有利地,由于所述支持物不存在过渡金属,因此可在较不极端的反应条件下(即,在不损害或不显著降解类胡萝卜素的结构的温度下)进行类胡萝卜素的解离。例如,如果类胡萝卜素为β-胡萝卜素,则相对于结合也含有除银以外的过渡金属的支持物的胡萝卜素有利地更容易在解离期间保持β-胡萝卜素的双键。
在一个实施方案中,基本上全部类胡萝卜素均能够在有机溶剂中从支持物解离。有利地,可能在随后的类胡萝卜素回收过程中再用所述支持物。
在其中所述支持物进一步包含粘土的情况下,所述粘土可为天然粘土,即从自然界分离的,或合成粘土。
粘土表示一大群特征在于存在层状硅酸铝的矿物质,并且还可包含其它元素,例如铁、镁、碱金属、碱土金属和其它阳离子。
合适的粘土可包含来自高岭土族的矿物质,例如高岭石、地开石、叙永石和珍珠陶土;来自绿土族的矿物质,例如蒙脱石和绿脱石;来自伊利石族的矿物质,其包括粘土-云母;来自绿泥石族的矿物质;和混合层粘土变化形式。

金属盐的金属可为多价金属。此金属可选自由碱金属、碱土金属或元素周期表的第IIIA族、第VIIA族和第VIII族的金属组成的组。示例性的金属包括钠、钾、锂、镁、钙、铝、铁和锰。
金属盐的阴离子可为氢氧根;卤离子,例如氯离子、溴离子、碘离子或氟离子;羧酸根,例如乙酸根;或硝酸根。

用于制备合成粘土的传统方法通常采用加热反应混合物。然而,使用辐射源进行加热允许在其整个容积内以基本上相同的速率来加热材料,也就是说,其使得能够进行容积式加热。来自辐射源的热能通过加热过的材料以电磁方式进行转移。因此,加热速率并不会受到在对流加热或传导加热期间出现的通过材料转移热的速率的限制,并且极大地改进了热分布的均匀性。加热时间可减少到小于使用对流加热或传导加热所需的加热时间的1%。

添加到含有至少一种类胡萝卜素的液体中的包含至少一种亲π键物质的支持物的量可取决于存在于所述液体中的类胡萝卜素的量和类型、所述液体的性质、所用亲π键物质的类型和接触条件,例如,所述支持物将与所述液体保持接触的时间长度和所述材料与所述液体接触的温度。
允许含有至少一种类胡萝卜素的液体与包含至少一种亲π键物质的支持物保持接触足以允许存在于所述液体中的至少一些类胡萝卜素结合所述支持物的时间量,由此形成类胡萝卜素络合物。
类胡萝卜素的可逆结合可由于与亲π键物质和/或支持物的络合、吸附、吸收和其组合而形成。在其中所述液体中存在超过一种类型的类胡萝卜素(例如,α-胡萝卜素和β-胡萝卜素两者)的情况下,类胡萝卜素可包括存在于液体中的超过一种类型的结合亲π键物质的类胡萝卜素。在优选的实施方案中,类胡萝卜素是α-胡萝卜素、β-胡萝卜素和其组合。

实施例
将进一步描述本公开的非限制性的实施例,其不应被解释为以任何方式来限制本公开的范围。
实施例1.合成粘土的制备
为了合成粘土,将MgCl2·6H2O(104g)、LiCl(1.3g)、Na2Si3O7(166g)和Na2CO3(29.5g)溶解于H2O(1800ml)中。在将其搅拌30分钟后,在家用微波炉中处理(2400W,60分钟)溶液混合物。将反应产物过滤,用水(2500ml)洗涤并且在110℃(16小时)干燥,获得60g期望的合成粘土。
合成粘土的X-射线衍射分析指示其为锂蒙脱石粘土(单斜晶系,空间群C 2/m),层间距为约制备的合成粘土的X-射线荧光分析指示所述粘土以wt%计由Na2O(1.91%)、MgO(17.76%)、SiO2(47.37%)和H2O组成。
实施例2.银插层合成粘土的制备
向实施例1中制备的分散于H2O(500ml)中的合成粘土(5g)中添加AgNO3盐(1g)的溶液,之后搅拌24小时。然后将Ag-插层粘土过滤并且用去离子H2O洗涤,之后在80℃(16小时)干燥。将干燥的Ag-粘土用于类胡萝卜素分离。
合成粘土的X-射线衍射分析指示Ag插层合成粘土的结构为Ag1.2[Mg4.8Li1.2Si8O20(OH)4](图1)。X-射线荧光分析指示银插层粘土由Ag(30-52wt%)与MgO(5.02-12.28wt%)、SiO2(16.64-30.37wt%)和H2O组成(表1)。

实施例3.AgO的制备
向硝酸银(2g)溶液(水,50ml)中添加NaOH的水溶液(0.7g,于5ml水中),在室温下恒定搅拌15分钟。将沉淀在80℃干燥2小时并且通过XRD和BET表面积(产量1.2g)来表征。
X-射线衍射分析指示所形成的材料为具有少量AgO的Ag2O(图2)。
实施例4.硅酸银的制备
向硝酸银(1g)溶液(水,30ml)中添加Na2Si3O7的水溶液(33wt%),在室温下恒定搅拌10分钟。添加丙酮(20ml)以加速期望产物的沉淀。将沉淀用水(100ml)洗涤,之后用丙酮(50ml)洗涤并且在50℃干燥。X-射线衍射分析显示形成的材料为具有一些非晶相的Ag2SiO4(图3)。
实施例5.从棕榈油分离类胡萝卜素
对于胡萝卜素萃取来说,在恒定搅拌下将Ag-粘土(0.5g)添加到粗制棕榈油(10g)中。发现最佳搅拌为2小时。通过离心、之后用有机溶剂(5X 10ml)洗涤,将所述油与吸收了胡萝卜素的粘土分离开,所述溶剂可优选丙酮、己烷。进行洗涤直到出现吸附的油,并且通过IR甘油酯C=O峰来鉴别。
然后将十八碳烯或1,5-己二烯(5ml)添加到洗过的类胡萝卜素络合物中并且进行混合。然后通过过滤出固体来收集十六烷。然后在减低压力下去除烯烃,获得分离的类胡萝卜素。通过UV-可见光谱术(图4)和液体色谱术(图5)来确认分离。如图4中所示,可以看出,在为类胡萝卜素的特征吸收峰的472nm、447nm和426nm处观察到吸收。另外,从图5中的色谱图可以看出,从粗制棕榈油分离的含有胡萝卜素的分离的类胡萝卜素(B)的保留时间与标准胡萝卜素(A)的保留时间相同。
类胡萝卜素的典型产量为0.6-2.2mg,其由约35%的α–胡萝卜素、54%的β-胡萝卜素、3%的γ-胡萝卜素和番茄红素构成。这代表了最初存在于粗制棕榈油中的类胡萝卜素的约10-70%回收率。这些实验的结果列于表2中。

实施例6.温度对从棕榈油分离类胡萝卜素的作用
在另一组实验中,如实施例5中所述的,从粗制棕榈油分离类胡萝卜素,但在40℃加热粗制棕榈油和粘土的混合物。胡萝卜素分离量相较于室温过程更高(0.6-4.2mg)。
实施例7.使用AgO从棕榈油分离类胡萝卜素
在另一组实验中,在粗制棕榈油(10g)的处理中使用Ag2O(0.2g)代替Ag-粘土。所述过程与如上文所讨论的实施例5中所述的相同。使用Ag2O,从10g粗制棕榈油萃取0.6-1.8mg类胡萝卜素。
实施例8.使用硅酸银从棕榈油分离类胡萝卜素
在另一组实验中,在粗制棕榈油(6g)的处理中使用硅酸银(0.2g)代替Ag-粘土。所述过程与如上文所讨论的实施例5中所述的相同。使用硅酸银,从10g粗制棕榈油萃取0.5-1.5mg类胡萝卜素。
实施例9.粗制棕榈油浓度对从棕榈油回收胡萝卜素的作用
在另一组实验中,研究了棕榈油浓度对类胡萝卜素回收的作用。当使用0.5g Ag-粘土时,用丙酮和己烷稀释粗制棕榈油(在15ml丙酮或己烷中稀释的2g粗制棕榈油)导致类胡萝卜素吸收增加。己烷作为溶剂相较于丙酮溶剂导致更高的胡萝卜素结合。这一变化导致浓缩的萃取相中分离的类胡萝卜素从45ppm增加到50ppm。
通过用其它萃取溶剂(十六碳烯、1,5己二烯、苧烯、香叶醇、山梨酸、异戊二烯、α-蒎烯、香叶烯、法呢烯、法尼醇、柠檬醛、石竹烯、姜烯,5ml)处理从类胡萝卜素络合物分离络合的类胡萝卜素。所述过程涉及在溶剂存在下搅拌类胡萝卜素络合物(15-30分钟)。
萃取溶剂与金属离子具有高结合效率,因此置换胡萝卜素-金属离子键结;胡萝卜素出现于溶剂介质中。通过IR和UV-光谱术来确认萃取的胡萝卜素。
上述过程通常得到约0.1mg分离的类胡萝卜素,所述类胡萝卜素代表了最初存在于粗制棕榈油中的类胡萝卜素的20-80%回收率。这些实验的结果列于表3中。

应用
本文所述的用于可逆地结合类胡萝卜素的材料和类胡萝卜素分离方法可结合从各种各样的含有类胡萝卜素(例如α-胡萝卜素和β-胡萝卜素)的液体回收和/或分离所述类胡萝卜素使用。示例性的液体包括棕榈油、粗制棕榈油、粗制棕榈油精、红棕榈油/油精、棕榈硬脂精、中和的红棕榈油、中和的红棕榈油精、向日葵油、椰子油、小麦胚芽油、胡萝卜油、大豆油、菜籽油、橄榄油和所述油的衍生物。
所述材料和类胡萝卜素分离方法可用于从源自产类胡萝卜素的真菌、产类胡萝卜素的细菌、产类胡萝卜素的微藻和产类胡萝卜素酵母菌的液体萃取物回收和/或分离类胡萝卜素。
所述材料和类胡萝卜素分离方法还可用于回收、分离和/或纯化通过化学合成制备的类胡萝卜素。可在有机溶剂中稀释通过化学合成制备的粗制类胡萝卜素。所得到的包含类胡萝卜素的溶液可经受本文所述的方法以至少部分地纯化其中存在的类胡萝卜素。
本文所述的材料和分离方法还可用于从天然源回收和/或分离其它烯烃或聚烯烃,或结合所述化合物的化学合成使用。
通过本文所述的材料和分离方法提供的类胡萝卜素由于它们的生物功能而对人类健康具有各种各样的益处。作为抗氧化剂,类胡萝卜素由于它们能够防止细胞组分的氧化损害而对细胞健康至关重要。支持类胡萝卜素对于慢性和退化性疾病(例如癌症)产生的保护作用的流行病学证据已经大幅度增长。
由于本文所述的类胡萝卜素的营养保健性质,因此它们也通常用于食品和生物燃料工业。例如,它们用于增强鱼的颜色,这增加了消费者的质量感觉。更重要地,类胡萝卜素已作为可促使微藻生物燃料生产在经济上可行的增值化合物被提出。高油价、食品与其它生物燃料源之间的竞争性需求和世界粮食危机已经激起了将微藻作为用于生物燃料的有前景的原料的兴趣。这些光合微生物在将二氧化碳转化成富碳脂质方面的生产率大大地超过了农业油料作物的生产率,而且不竞争耕地。因此,富集类胡萝卜素的微藻生产正急剧成为有吸引力的商业。

2.根据权利要求1所述的材料,其中所述支持物相对于天然粘土具有更大的表面积。
3.根据权利要求1或2所述的材料,其中所述支持物具有400m2/g到700m2/g的总表面积。
4.根据前述权利要求中任一项所述的材料,其中所述支持物具有基本上均匀的化学组成。
5.根据前述权利要求中任一项所述的材料,其中所述支持物是合成粘土。
