发明背景

硫酸长春新碱在鞘磷脂-胆固醇脂质体中的封装一般通过利用酸性脂质体内pH(例如,pH 4)和中性pH(例如,pH 7)的外部介质来实现。这种pH梯度使弱碱性长春新碱以高效率扩散到脂质体内部中(Cullis等,Trends in Biotech 9:268-272,1991;Boman等,Bioch Biophys Acta,1152:253-258,1993)。为使长春新碱在跨膜pH梯度下在脂质体内部中积累,脂质体膜必须对于长春新碱的空间体积是暂时可渗透的。因此,不同于通常可被动地封装到脂质体中的中性或阴离子药物,必须增加鞘磷脂-胆固醇脂质体的温度以使跨膜pH梯度关于长春新碱起作用。脂质体双层,是联锁鞘磷脂和胆固醇分子的取向,需要特殊的瞬态热模式来产生向热性紊乱过渡态(thermotropic disorder transition states)。这些过渡态显著减轻膜脂质之间的弱分子间结合,在联锁脂质中产生间隙,并使脂质体生物层变得暂时可渗透。封装过程利用在冷却回到环境温度时发生的鞘磷脂-胆固醇脂质的自发性重新自组装,恢复了膜完整性。
这种药物封装加热曲线必须与长春新碱对于热暴露的化学不稳定性平衡(Vendrig等,Internatl.J.of Pharmaceutics 50:189-196,1989;Sethi等,Cancer Res。45:5386-5389,1985)。长春新碱是热不稳定的,并且容易在升高的温度下降解成N-脱甲酰基长春新碱。长春新碱的这种甲酰胺水解是公知的降解途径,并且影响硫酸长春新碱注射液(VLSI)的稳定保质期。例如,VLSI溶液由于这种热不稳定性不能被热灭菌,必须在冷藏温度下储存和运送以实现延长的稳定性。
因此,当前硫酸长春新碱脂质体注射液(VSLI)根据FDA批准的标签(www.accessdata.fda.gov;参考ID:3172211,2012)上提供的指导在药房由单独的组分制备。这些指导包括加热程序——需要使用水浴以实现长春新碱在鞘磷脂-胆固醇脂质体中的有效封装和保持长春新碱的化学纯度。水的良好传热性质允许大于95%的长春新碱封装,而无可观的药物化学降解。
由于VSLI是可注射药物,组分的生产和药房制备被严格控制以保持无菌。因此,VSLI制备过程中的开放水浴的使用需要另外的物力、规划和设备(例如,浮动环),包括保持无菌环境的无菌罩或“洁净”间。在一些药房中,VSLI的配成由于保持无菌环境的限制而无法完成。
因此,仍需要可有效且可再现地进行而不需当前所需的另外的物力和设备的制备VSLI的改进方法。
发明概述
本发明至少部分基于避免在封装过程中使用加热水浴的需求的制备VSLI的方法的建立。因此,本发明提供有效的、可再现的制备VSLI的方法,其可广泛应用,并具有预料不到的简便性和减少的污染危险。
一方面,本发明描述了制备包括脂质体封装的无显著降解产物的长春新碱的药学上可接受的液体组合物的方法,该方法包括步骤:(a)在单一瓶中配成(i)第一溶液,包括硫酸长春新碱,浓度为约1mg/ml至约5mg/ml,其中第一溶液的pH为约3.5至约5.5;和(ii)第二溶液,包括低pH的鞘磷脂/胆固醇脂质体;(b)升高单一瓶中的配成溶液的pH至约7.0至7.5的pH;(c)在平衡在约75℃的干热区块中加热包括配成溶液的单一瓶至少约13至约18分钟,其中所述加热区块包括一个或多个孔——其大于单一瓶的平均长度或直径约1-5%,以产生包括配成的脂质体封装式长春新碱的溶液;(d)平衡配成的溶液至室温;(e)用适于静脉内给予的药物稀释剂稀释一定体积的、包括对于患者约1.5至约2.4mg/m2剂量的脂质体封装式长春新碱的配成溶液,以产生药学上可接受的液体组合物;和(f)给予药学上可接受的液体组合物至患者,其中配成的包括脂质体封装式长春新碱的溶液包括(i)小于约2.5%的游离长春新碱;和(ii)小于约1.5%的N-脱甲酰基长春新碱。
在一些实施方式中,包括硫酸长春新碱的第一溶液的pH为约4.5至约4.7。在一个实施方式中,第一溶液进一步包括约100-200mg/ml浓度的甘露醇。

发明详述
本发明克服了与当前采用的制备VLSI的方法过程中的水浴使用相关的缺陷。惊奇地发现,加热区块联合适应用于配成VSLI的容器而设计的定制设计***物的应用提供了实现均匀封装长春新碱而无显著降解所需的热曲线。
另外,本文描述的制备VSLI的方法证明参数释放的提高能力显著。由于VSLI的稳定性保证了“适时”制备,配成过程一定是高效的和药剂师可再现的。药剂师需要知道(即,参数方面)封装已经通过热诱导过程实现,因为其提供“基于配成过程中收集的信息的复查和参数释放相关具体GMP要求的顺从性确认产物具有预期质量的释放***”(Annex 17EU指南)。
本发明的加热区块程序提供了便于和适于实现以大于95%的封装效率配成VSLI的工艺。由于其涉及较少的步骤,操作失误的可能性减少。该工艺总体上更直接,用时更少,耗用较少物力,并且便于常规制药操作。另外,制备VSLI的个体无需处理水浴中生长的微生物的潜在微生物污染或加热水浴的水蒸气。

硫酸长春新碱
硫酸长春新碱是从长春花植物(Catharanthus roseus)原始分离的长春花生物碱家族成员。硫酸长春新碱具有细胞周期特异性抗癌活性。长春花生物碱结合于微管蛋白,改变微管蛋白聚合,导致中期停顿、细胞有丝***抑制和细胞死亡。作为细胞周期特异性试剂,其治疗响应被脂质体封装促进,脂质体封装保持了延长的药物水平。细胞至长春新碱(以及其他细胞周期特异性药物)的延长暴露已显示增强药物的体外细胞毒性(Bfurris等,JNCI 84;1816-1826,1992;Georgiadis等,Clin Cancer Res 3:449-454,1997;Jackson和Bender,Cancer Res 39:4346-4349,1979)。

VSLI制备

在一些实施方式中,在三个单独的容器中提供硫酸长春新碱溶液、脂质体溶液和高pH缓冲溶液。在某些实施方式中,将这三种溶液组合到一个无菌容器,该无菌容器具有包容溶液组合体积的容量,例如,约20-50mL、约25-40mL、或约30-35mL。

VSLI
根据本文描述的方法生成的VSLI呈现为白色至米白色、半透明悬浮液,基本上不含可见异物和聚集物。一般,大于约95%、约96%、约97%、约98%或更多的硫酸长春新碱被封装在脂质体中。
根据本文描述的方法生成的VSLI包含的杂质总量小于约4.0%、3.5%、3.4%、3.2%、3.1%或3.0%。在一些实施方式中,VSLI包含小于约2.0%、1.8%、1.7%、1.6%、1.5%、1.4%或1.3%的N-脱甲酰基长春新碱。
根据本文描述的方法生成的VSLI的平均体外释放速率(IVR)或体内释放速率为在72小时内至少约75%、76%、77%、78%、79%、80%、81%、82%、83%、84%或约85%。
确定长春新碱封装水平、杂质水平和长春新碱自脂质体的释放速率的试验在本领域已知。参见,例如,美国专利5543152和5837282;Zhigalstev等,J.ControlledRelease 104:103-111,2005);Puscalau等,Am.J.Health-Syst.Pharm.62:1606-1612,2005)。
总体而言,根据本文描述的方法生成的VSLI包含约0.1mg/mL至约0.5mg/mL的硫酸长春新碱。在某些实施方式中,硫酸长春新碱以约0.15mg/mL至约0.2mg/mL存在。在一个实施方式中,硫酸长春新碱以约0.16mg/mL存在。在一个实施方式中,VSLI包含5mg硫酸长春新碱、500mg甘露醇、73.5mg鞘磷脂、29.5mg胆固醇、36mg柠檬酸钠、38mg柠檬酸、355mg磷酸钠、和225mg氯化钠。
剂量和给药
根据本文描述的方法制备的VSLI可用于治疗任何类型的癌症,包括原发性、复发性和顽固性癌症。用VLSI治疗的患者或对象可以是多种动物,包括人、非人灵长类、鸟类、马类、犬类、猫类、牛类、猪、兔类、啮齿类及类似物。在某些实施方式中,VSLI用于治疗血液和淋巴***的癌症,包括但不限于,淋巴瘤、白血病和骨髓瘤。在某些实施方式中,VSLI用于***,包括但不限于,神经母细胞瘤和脑癌。
VSLI可作为单剂使用,或组合其他化疗剂使用,如环磷酰胺、阿霉素和/或***。在一个实施方式中,VSLI与环磷酰胺、阿霉素和***一起作为脂质体CHOP制剂(“lipo-CHOP)给予。在另一实施方式中,VSLI与至少一种另外的抗肿瘤剂被共同给予。在另一实施方式中,另外的抗肿瘤剂是抗肿瘤单克隆抗体,如OncolyTM、RituxanTM、或BexxarTM。在另一实施方式中,另外的抗肿瘤剂是反义药物或抗肿瘤疫苗。在另一实施方式中,VSLI与神经毒性的预防性或治疗性治疗,如加巴喷丁(gabapentin)(NeurontinTM),被共同给予。
一般,在给予患者约24小时之内制备VSLI,并储存在室温下(15℃至30℃)或冷藏(2-8℃)。
将VSLI通过静脉内递送全身地给予患者。在一个实施方式中,经例如约30分钟、约45分钟、约60分钟、约90分钟或更长的时间通过静脉内灌注给予VSLI。
一般周期性给予VSLI,例如,每7-28日一次。在某些实施方式中,VSLI每3、5、7、10、14、21或28日给予一次。在一个实施方式中,每14日通过静脉内灌注给予VSLI。在另一实施方式中,每7日通过静脉内灌注给予VSLI。如本文所用,每次给予VSLI都被认为是治疗的一个“疗程”。
每剂量给予的VSLI量取决于多个因素,如患者医疗史、其他治疗手段的应用、和疾病性质(例如,原发性(first line)、复发性或顽固性癌症)。一般,根据本文描述的方法制备的VSLI以约1.4至约2.4mg/m的剂量给予。在某些实施方式中,VSLI以约1.5mg/m2、约1.8mg/m2、约2.0mg/m2、2.1mg/m2、2.2mg/m2、2.3mg/m2或2.4mg/m2(即,mg长春新碱/m2身体表面积)的剂量给予。在一个实施方式中,VSLI每7日一次通过静脉内灌注经约60分钟以2.25mg/m2的剂量给予。
在其他实施方式中,VSLI的剂量可在治疗过程中被暂时中断和/或减少。例如,在一个实施方式中,给予表现为3级外周神经病或持续性2级外周神经病的患者的VSLI剂量可中断上至约7日,然后在恢复至1级或2级后减少至约2mg/m2的剂量。在另一实施方式中,给予甚至在接受减少剂量后还表现为持续性2级外周神经病的患者的剂量可中断上至7日,然后减少至1.825mg/m2的剂量、或1.5mg/m2的剂量。
VSLI的剂量通过根据公知的方法计算对象的身体表面积(BSA)来确定。例如,根据Mosteller公式,其中BSA等于以kg表示的对象重量乘以以cm表示的身高的乘积除以3600的平方根。人的“标准”BSA通常取为1.7m2,但也取决于其他因素,包括个体年龄和性别。例如:

实施例1

程序
通过记录置于加热设备(即,干燥区块或水浴)后小瓶内部的溶液温度生成温度曲线测量结果。零时是小瓶置于加热设备的时间点。采用下列程序:
1.将组分VSI和SCLI组合到单个SPI小瓶(Swiss Precision Instruments,Inc.)中。将数字热电偶***小瓶的隔垫,并保持在距小瓶底面大约5mm且定位在液体溶液中央。
2.使用干燥区块加热器,其具有包含1.480英寸小瓶容器孔的区块。通过将温度计置于区块的温度计孔中来监测区块温度,该温度计孔的位置接近小瓶容器孔。将区块加热器设定至75℃温度,并允许其升温直到区块温度计读数为75±2℃。使加热区块在75±2℃平衡最少15分钟。然后将小瓶***75±2℃平衡区块的孔中14分钟±15秒,然后取出。通过置于环境条件中约60分钟使配成的小瓶达到环境温度。利用同一小瓶,程序重复两次。
3.以1分钟间隔(或按照说明)记录小瓶内部液体温度和区块温度,并列表于表1。
4.作为比较,允许加热水浴直到水温为65±5℃。使水浴平衡最少15分钟。然后将包含配成的VSLI的小瓶***水浴10分钟±1分钟,然后取出。使配成的小瓶达到环境温度。
5.记录小瓶内部液体温度、时间和水温度,并列表于表1。
结果和讨论
用平衡在75℃±2℃的干燥区块加热配成的VSLI小瓶的液体内含物的温度曲线证明:加热速率均匀且渐进至65℃±5℃。表1和图1列出的结果显示,干燥区块以平均速率3.26℃/分钟加热液体小瓶内含物,与之相比,水浴以平均速率4.21℃/分钟加热小瓶内含物。65℃±5℃的期望温度在干燥区块中在14分钟内实现,与之相比水浴耗费10分钟。关于这两种加热设备,在将小瓶从加热源取出后,存在逐渐冷却。从干燥区块中取出后,温度保持在59-65℃内3-4分钟,而从水浴取出后,保持1-2分钟。关于这两种设备,利用干燥区块,小瓶暴露于50-65℃至少20分钟,而利用水浴,小瓶暴露15分钟。在任一设备中加热后,溶液保持视觉一致:白色至米白色、半透明悬浮液,基本上不含可见异物和聚集物。

结论
本研究证明,干燥区块验证了使VSLI小瓶内部液体温度在14分钟内实现65℃±5℃的适当温度加热曲线,并提供了20分钟的50-65℃范围内的加热暴露。与之相比,在水浴中加热小瓶在10分钟内实现了65℃±5℃的内部液体温度和15分钟的50-65℃范围内的总体加热暴露。两种方法的加热速率均是渐进的和均匀的。设定在75℃±2℃的干燥区块产生3.26℃每分钟的加热速率,而设定在65℃±5℃的水浴产生4.21℃每分钟的速率。利用水浴加热平稳,而干燥区块中的加热速率以稳定速率持续至75℃±2℃设定点。在将小瓶从任一加热设备取出后,冷却至环境温度耗费约60分钟。这些加热曲线表明,长春新碱的接近的定量封装是个热力学过程,其取决于暴露于促进膜封装而非实现那些条件的动力学的温度的总体暴露。
总之,干燥区块和水浴可提供允许将长春新碱封装到鞘磷脂胆固醇脂质体中以有效制备VSLI的热力学特征。
实施例2
考察利用干热区块配成的VSLI的长春新碱降解物产物水平。
设备和材料:
·Techne DB-3配备有1.480”直径孔和温度计套。
·直径不大于7mm并且在0-100℃范围内精确至±1℃的温度计。

结果和讨论
利用Dri-Block加热器、利用直径为1.4305和1.4545英寸的配成小瓶来实现VSLI的配成。这些代表最接近小瓶直径容许的1.41英寸下限和1.47英寸上限的小瓶。在加热区块中14分钟温育VSLI小瓶的过程中,区块温度由于区块孔直径和小瓶任一直径之间的间隙之间的温度平衡造成的下降不大于1度。这种传热动力学不影响VSLI的制备。暴露于平衡在75℃的Dri-Block热14分钟后,从两个小瓶的所得的配成的VSLI均实现大于97%的长春新碱封装。利用Dri-Block的温育导致封装效率平均为2.175%游离长春新碱。通过Dri-Block加热曲线没有观察到新的或增加的杂质。观察到主要降解物,N-脱甲酰基长春新碱,平均为1.33%,无其他大于0.574%的杂质,并且杂质总量不大于3.10%。颗粒尺寸分布与VSLI规格一致,平均直径为直径107.5nm,平均D25为90.5nm,和平均D90为138.5nm,通过IVR分析,通过Dri-Block加热制备的VSLI通过72小时释放大约84%的长春新碱。
总之,利用平衡在75±2℃的干燥区块加热器、以14分钟±15秒温育期配成的VSLI使大于99%的长春新碱封装在鞘磷脂-胆固醇脂质体中,并且此实验过程中无异常记录。数据建立了导致产物有效封装长春新碱并且证明干燥区块加热器在VSLI配成时可代替水浴的干燥区块配成温度曲线。
实施例3
在本研究中,测量长春新碱降解物产物水平,以提供利用干燥区块程序的配成时间范围。
设备和材料
·配备有1.476”(±0.004)直径孔和温度计套。
·温度计,直径不大于7mm,并且在0-100℃的范围内精确至±1℃。
·校准秒表或计时器
·试剂盒,Lot#TTX0611(Talon Therapeutics,Inc.)
程序
随机选择三种试剂盒,并通过在干燥区块中在75℃下以三个不同时间(分别为13、14和15分钟)加热来配成。利用适当的硫酸长春新碱注射液USP方法测试配成的小瓶的总量和游离硫酸长春新碱、相关化合物、颗粒尺寸和分布。
利用上述指导配成本研究试剂盒,除了用具有包含1.476英寸(±0.004”)小瓶容器孔的区块的加热器代替水浴之外。通过将温度计置于位于小瓶容器孔邻近的区块温度计孔中来监测区块温度。将区块设定至75℃温度,并使其加热,直到区块中的温度计读数为75±2℃。然后使加热区块平衡最少15分钟。条件记录在表9中。然后将各小瓶***75±2℃加热区块分别13分钟±15秒、14分钟±15秒、和15分钟±15秒。然后取出小瓶,并置于环境温度。使配成后的小瓶在环境温度下冷却至少60分钟,然后测试。

结果和讨论
各配成的试剂盒分别被置于平衡在75℃的加热区块13、14和15分钟。全部样本时间均产生封装的长春新碱,并且三个小瓶的测试结果之间不显示显著差异。利用Dri-Block的三个小瓶均观察到大于97%的长春新碱封装,导致封装效率最大为2.3%游离长春新碱。关于Dri-Block加热曲线未观察到新的或增加的杂质。观察到主要降解物N-脱甲酰基长春新碱的最大值为1.51%,无其他大于0.59%的杂质,杂质总量不大于3.3%。颗粒尺寸分布与VSLI规格一致,其平均直径为直径107nm,平均D25为90nm,和平均D90为137nm。
总之,用平衡在75±2℃的Dri-Block代替水浴、以13至15分钟(±15秒)的温育期配成的VSLI有效封装长春新碱,并且实验进行期间无异常记录。数据建立了导致有效VSLI配成和显示Dri-Block加热器在VSLI配成中可代替水浴的Dri-Block配成温度曲线。

Claims (37)
1.制备包括脂质体封装式长春新碱的药学上可接受的液体组合物的方法,所述脂质体封装式长春新碱无显著降解产物,所述方法包括:
(a)在单一瓶中配成(i)第一溶液,包括约1mg/ml至约5mg/ml浓度的硫酸长春新碱,其中所述第一溶液的pH为约3.5至约5.5;和(ii)第二溶液,包括低pH的鞘磷脂/胆固醇脂质体;
(b)将所述单一瓶中的所述配成溶液的pH升高到约7.0至7.5的pH;
(c)在平衡在约75℃的干热区块中加热包括所述配成溶液的所述单一瓶至少约13至约18分钟,其中所述加热区块包括大于所述单一瓶的平均长度或直径约1-5%的一个或多个孔,以生成包括配成的脂质体封装式长春新碱的溶液;
(d)平衡所述配成溶液至室温;
(e)用适于静脉内给予的药物稀释剂稀释一定体积的所述配成溶液,所述配成溶液包括对于患者约1.5至约2.4mg/m2剂量的脂质体封装式长春新碱,以产生所述药学上可接受的液体组合物;和
(f)将所述药学上可接受的液体组合物给予所述患者,其中包括脂质体封装式长春新碱的所述配成溶液包括(i)小于约2.5%的游离长春新碱;和(ii)小于约1.5%的N-脱甲酰基长春新碱。
2.权利要求1所述的方法,其中包括硫酸长春新碱的所述第一溶液的pH为约4.5至约4.7。
3.前述权利要求中任一项所述的方法,其中所述第一溶液进一步包括约100-200mg/ml浓度的甘露醇。
4.前述权利要求中任一项所述的方法,其中包括所述脂质体的所述第二溶液的pH为约4.0。
5.前述权利要求中任一项所述的方法,其中所述第二溶液进一步包括柠檬酸盐缓冲液。
6.前述权利要求中任一项所述的方法,其中通过添加包括pH约9.0的缓冲液的第三溶液,升高所述配成溶液的pH。
7.权利要求6所述的方法,其中所述第三溶液包括磷酸钠缓冲液。
8.前述权利要求中任一项所述的方法,其中所述配成溶液包括约0.1/1.0至约0.2/2.0的硫酸长春新碱/脂质的浓度比。
9.前述权利要求中任一项所述的方法,其中所述配成溶液中的硫酸长春新碱的浓度为约0.1mg/mL至约0.5mg/mL。
10.前述权利要求中任一项所述的方法,其中所述配成溶液中的硫酸长春新碱的浓度为约0.15mg/mL至约0.2mg/mL。
11.前述权利要求中任一项所述的方法,其中所述配成溶液中的硫酸长春新碱的浓度为约0.16mg/mL。
12.权利要求6所述的方法,其中所述第一溶液包括硫酸长春新碱USP(5mg/5mL),其等同于4.5mg/5mL长春新碱游离碱,和500mg/5mL甘露醇;所述第二溶液包括由73.5mg/mL鞘磷脂、29.5mg/mL胆固醇、33.6mg/mL柠檬酸、35.4mg/mL柠檬酸钠组成的鞘磷脂/胆固醇脂质体;和所述第三溶液包括355mg/25mL磷酸氢二钠和225mg/25mL氯化钠。
13.前述权利要求中任一项所述的方法,其中所述脂质体中鞘磷脂与胆固醇的比在约75/25mol%/mol%鞘磷脂/胆固醇至30/50mol%/mol%鞘磷脂/胆固醇之间。
14.前述权利要求中任一项所述的方法,其中所述脂质体包括约70/30mol%/mol%鞘磷脂/胆固醇至40/45mol%/mol%鞘磷脂/胆固醇。
15.前述权利要求中任一项所述的方法,其中所述脂质体包括大约55/45mol%/mol%鞘磷脂/胆固醇。
16.权利要求13所述的方法,其中所述脂质体包括约60/40mol%/mol%鞘磷脂/胆固醇。

26.前述权利要求中任一项所述的方法,其中所述包含所述配成溶液的单一瓶的直径在约35.8至约37.3mm之间。
27.权利要求26所述的方法,其中所述加热区块的所述口径孔是圆柱形的,其直径在37.2至37.8mm之间。

30.前述权利要求中任一项所述的方法,其中从灌注容器取出患者计算剂量的体积,并更换为所述配成的VSLI溶液的计算体积。
31.前述权利要求中任一项所述的方法,其中将包括脂质体封装式长春新碱的所述药学上可接受的液体组合物在步骤(c)后约24小时内给予所述患者。
32.前述权利要求中任一项所述的方法,其中所述患者患有癌症。
33.权利要求32所述的方法,其中所述癌症选自淋巴瘤、白血病、骨髓瘤、脑癌和神经母细胞瘤。
34.前述权利要求中任一项所述的方法,其中通过静脉内灌注、经约30至60分钟的时间给予包括脂质体封装式长春新碱的所述药学上可接受的液体组合物。
35.前述权利要求中任一项所述的方法,其中通过静脉内灌注、每7-28日一次给予包括脂质体封装式长春新碱的所述药学上可接受的液体组合物。
36.前述权利要求中任一项所述的方法,其中通过静脉内灌注、每7日一次给予包括脂质体封装式长春新碱的所述药学上可接受的液体组合物。
37.制备包括脂质体封装式长春新碱的药学上可接受的液体组合物的方法,所述脂质体封装式长春新碱无显著降解产物,所述方法包括:
(a)在单一瓶中配成(i)第一溶液,包括约1mg/ml浓度的硫酸长春新碱和约100mg/ml浓度的甘露醇,其中所述第一溶液的pH为约4.5至约4.7;(ii)第二溶液,包括在pH约4.0的柠檬酸盐缓冲液中的具有60/40mol%/mol%鞘磷脂/胆固醇的脂质体;和(iii)第三溶液,包括pH约9.0的磷酸钠缓冲液,其中所述配成溶液的最终pH为约7.0至约7.6,并且硫酸长春新碱的浓度为约0.16mg/ml;
(b)在平衡在75℃±2℃的干热区块中加热包括所述配成溶液的所述单一瓶14分钟±30秒,以生成包括配成的脂质体封装式长春新碱的溶液,其中所述单一瓶的直径在约35.8mm至约37.3mm之间,和所述加热区块包括直径在约37.2mm至约37.8mm之间的一个或多个圆柱形孔;
(d)平衡所述配成溶液至室温至少30分钟;
(e)用药物稀释剂稀释一定体积的包括所述脂质体封装式长春新碱的所述配成溶液,以生成药学上可接受的液体组合物,所述药学上可接受的液体组合物适于静脉内给予约1.5至约2.4mg/m2剂量的长春新碱,其中包括脂质体封装式长春新碱的所述配成溶液包括(i)小于约2.5%的游离长春新碱;和(ii)小于约1.5%的N-脱甲酰基长春新碱。
