Abstract

本发明公开了一种式(Ⅰ)所示的含吡啶的噁二唑类化合物及其制备方法与防治番茄细菌性斑点病、黄瓜枯萎病、番茄灰霉病中的应用,本发明化合物为具有杀菌活性的新化合物,为新农药的研发提供了基础。

Description

一种含吡啶的噁二唑类化合物及其制备与应用
技术领域
本发明涉及一种含吡啶的噁二唑类化合物即2-(4-吡啶-基)-5-(取代烷硫基)-1,3,4-噁二唑衍生物及其制备与应用。
背景技术
杂环化合物已是新农药发展的主流,而在杂环化合物中,又以含氮杂环为主。1,3,4-噁二唑类化合物也有很重要的生物活性,包括抗炎、抗癌、抗病毒、抑菌和除草等活性。如合成的2-取代吡唑-5-甲硫基-1,3,4-噁二唑,在100 mg/L时对水稻纹枯病菌表现出良好的抑制活性;2-甲基-5-取代苄硫基-1,3,4-噁二唑在100ppm下对油菜具有较好的抑制效果。
然而,尚未见有关2-(4-吡啶-基)-5-(取代烷硫基)-1,3,4-噁二唑衍生物的合成及其生物活性研究的文献报道。
发明内容
本发明目的是提供一种具有杀菌活性的2-(4-吡啶-基)-5-(取代烷硫基)-1,3,4-噁二唑衍生物及其制备方法与应用。
本发明的目的是这样实现的:
式(I)所示的含吡啶的噁二唑类化合物:
(I)
式(I)中:R为C1-C6烷基、C1-C6烷氧羰基、C2-C6烯基、任选取代的苯基、任选取代的5元或6元杂芳基,所述取代选自一个或多个氢、C1-C6烷基、C1-C6烷氧基、C1-C6烷硫基、氰基、卤素。
本发明所述式(I)化合物可由如下方法制得:将式(II)化合物和式(III)化合物与有机溶剂混合,在碱性物质的存在下,0~25℃(优选25℃)搅拌反应,TLC跟踪反应(以体积比1:1~5的石油醚和乙酸乙酯混合溶液为展开剂,跟踪原料点消失为反应完全),反应完全后,将反应液置于冰水中,析出固体,过滤,滤液进行硅胶柱层析(以体积比为5:1的石油醚和乙酸乙酯混合溶液为洗脱剂,收集含目标组分的洗脱液,干燥)或用重结晶溶剂进行重结晶,获得式(I)所示的含吡啶的噁二唑类化合物;
重结晶溶剂为石油醚、乙酸乙酯、正己烷或乙醇中的一种或多种,优选石油醚与乙酸乙酯的混合溶液;
有机溶剂为N,N-二甲基甲酰胺、四氢呋喃、1,4-二氧六环或丙酮中的一种或多种,优选为N,N-二甲基甲酰胺(DMF);
碱性物质为碳酸钾、碳酸氢钠、氢氧化钠、氢氧化钾或氢化钠中的一种或多种,优选为碳酸钾;
式(II)化合物、式(III)化合物和碱性物质的物质的量之比为1:1.0~1.5:1.0~1.5,优选为1:1.0:1.2;有机溶剂体积用量以式(II)化合物物质的量计为2-10ml/mmol,优选4ml/mmol。
式(I)和式(III)中:R的定义如前所述,X为卤素。
本发明还提供式(I)所示的含吡啶的噁二唑化合物在防治番茄细菌性斑点病、黄瓜枯萎病、番茄灰霉病中的应用。
本发明还提供含有式(I)所示的含吡啶的噁二唑化合物的杀菌剂,其还包含至少一种农药上可接受的助剂。
与现有技术相比,本发明的有益效果主要体现在:本发明提供了一种含吡啶的噁二唑类化合物及中间体的其制备方法及应用,该化合物为具有杀菌活性的新化合物,为新农药的研发提供了基础。
具体实施方式
下面结合具体实施例对本发明进行进一步描述,但本发明的保护范围并不仅限于此:
本发明的含吡啶的噁二唑类化合物(I)可以如下方法合成:
25 mL圆底烧瓶中加入2 mmol化合物II、2 mmol取代氯苄、8mL DMF和2.4mmol的K2CO3,室温搅拌,TLC检测反应进度。反应完毕后,反应液倾入30mL冰水中,析出固体,过滤,脱溶,硅胶柱层析,得固体产物。
实施例1
2-(4-吡啶-基)-5-(4-甲氧基苄硫基)-1,3,4-噁二唑(1):产率 88%,熔点126-128℃; 1H NMR (CDCl3, 400 MHz), d:3.77(s, 3H, OMe), 4.50(s, 2H, SCH2), 6.84(d, J=8.6Hz, 2H, ArH), 7.36(d, J=8.6Hz, 2H, ArH), 7.82(d, J=4.8Hz, 2H, Py), 8.77(d,J=2.8Hz, 2H, Py).
实施例2
2-(4-吡啶-基)-5-(2-氟苄硫基)-1,3,4-噁二唑(2):产率 84%,熔点119-122℃;1H NMR (CDCl3, 400 MHz), d:4.51(s, 2H, SCH2), 7.30(s, 2H, ArH), 6.99-7.05(m,2H, ArH), 7.19-7.25(m, 1H, ArH). 7.46-7.49(m, 1H, Py), 7.83(s, 2H, Py), 8.74(s, 2H, Py).
实施例3
2-(4-吡啶-基)-5-(4-叔丁基苄硫基)-1,3,4-噁二唑(3):产率86%,熔点114-115C; 1H NMR (CDCl3, 400 MHz), d:1.31(s, 9H, Bu), 4.55(s, 2H, SCH2), 7.30(q, J=8.6Hz, 4H, ArH), 7.86(d, J=5.7Hz, 2H, Py), 8.81(d, J=5.3Hz, 2H, Py).
实施例4
2-(4-吡啶-基)-5-(3-氰基苄硫基)-1,3,4-噁二唑(4):产率 88%,熔点110-112℃; 1H NMR (CDCl3, 400 MHz), d: 4.57(s, 2H, SCH2), 7.47(t, J=7.7Hz, 1H, ArH),7.60(d, J=7.7Hz, 1H, ArH), 7.76(d, J=7.7Hz, 1H, ArH), 7.80(s, 1H, ArH), 7.96(s, 2H, Py), 8.84(s, 2H, Py).
实施例5
2-(4-吡啶-基)-5-(2-氯苄硫基)-1,3,4-噁二唑(5):产率 85%,熔点86-88℃; 1HNMR (CDCl3, 400 MHz), d: 4.68(s, 2H, SCH2), 7.28-7.30(m, 2H, ArH), 7.44(d, J=8.4Hz, 1H, ArH), 7.67(d, J=8.4Hz, 1H, ArH), 7.86(d, J=5.9Hz, 2H, Py), 8.82(d,J=5.7Hz, 2H, Py).
实施例6
2-(4-吡啶-基)-5-(2-氯吡啶亚甲硫基)-1,3,4-噁二唑(6):产率 87%,熔点141-143℃; 1H NMR (CDCl3, 400 MHz), d:4.50(s, 2H, SCH2), 7.30(d, J=8.2Hz, 1H,ArH), 7.82-7.85(m, 3H, ArH), 8.50(d, J=2.8Hz, 2H, Py), 8.88(d, J=5.3Hz, 2H,Py).
实施例7
2-(4-吡啶-基)-5-(4-氰基苄硫基)-1,3,4-噁二唑(7):产率 90%,熔点178-180℃; 1H NMR (CDCl3, 400 MHz), d: 4.57(s, 2H, SCH2), 7.64(q, J=8.3Hz, 4H, ArH),7.78(s, 2H, Py), 8.82(s, 2H, Py).
实施例8
2-(4-吡啶-基)-5-(乙氧羰基亚甲硫基)-1,3,4-噁二唑(8):产率 88%,熔点98-100℃; 1H NMR (CDCl3, 400 MHz), d: 1.31(t, 3H, Me), 4.14(s, 2H, SCH2), 4.28(q,2H, CH2), 7.87(s, 2H, Py), 8.82(s, 2H, Py).
实施例9
杀菌活性测试
试验对象:番茄细菌性斑点病、黄瓜枯萎病、番茄灰霉病。
试验方法:采用盆栽试验法。分别将黄瓜种子和番茄种子经过50℃浸泡后,催芽后播种于育苗钵中,待生长至2片真叶供试验。
药剂准备:供试药:取药配成100ppm,取89种化合物各5mg,加丙酮溶解后再加10%土温80,最后加水充分溶解。因100ppm为100mg/L所以加水量 =5mg*1000/100mg=50ml,因有机溶剂最终含量≤1% 所以加丙酮的量=50ml*1%=0.5ml(溶解),因吐温最终含量为0.1% 所以50 ml水里应有吐温 0.05 ml,即:应加10%吐温0.05 ml.
施药方法:番茄细菌性斑点病和番茄灰霉病实验中,番茄苗长出两片复叶时,用喷壶将供试药剂与对照药剂均匀喷施在供试植株上。
黄瓜枯萎病实验中,将黄瓜种子浸泡于各要处理2h后进行接种处理。
接种方法:
番茄细菌性斑点病采用菌悬液喷雾法接种:将培养好的番茄细菌性斑点病菌兑蒸馏水稀释成3×107cfu/ml菌悬液,用喷壶喷雾接种。接种后保湿培养。
番茄灰霉病采用打菌丝接种法接种:通过纱布过滤把液体培养基和菌块分开,用粉碎仪把带有少量液体的培养基的菌块打碎,再用过滤好的液体培养基与打碎的菌块混合配成一定浓度的溶液,并用分光光度计测定溶液透光率(λ=630nm,OD=9.8)。
黄瓜枯萎病采用培根浸种法接种:将在PD培养基中培养好的黄瓜枯萎病菌用纱布过滤菌丝,调节孢子浓度到1×106cfu/ml浸泡种子。
待清水充分发病后进行病情调查,并计算病情指数和防治效果。

1.一种式(I)所示的含吡啶的噁二唑类化合物,
式(I)中,R为4-甲氧基苯基。
2.一种如权利要求1所述的化合物在制备防治番茄细菌性斑点病、黄瓜枯萎病、番茄灰霉病杀菌剂中的应用。
3.杀菌剂,含有权利要求1所述的化合物及至少一种农药上可接受的助剂。
