Abstract

Description
技术领域
本发明涉及一种用于表征颗粒的光学方法。
背景技术
在一些技术领域,计算透明介质中小的透明颗粒是熟知的问题。这可以是在材料科学中计算分散相颗粒的数量,在生物培养中计算细胞数量,在化学或聚合物合成中计算乳胶中微胶粒的数量……。
解决这个问题的第一个尝试是使用标准的透明网格(标线),然后手动计算显微照片中各个由网格所限定的每个正方形中颗粒的数量。这种手动计数方法存在一些缺陷。首先,它非常耗时,它在数量很少时具有较差的统计有效性,并且通常具有有限的可靠性。
因此,图像分析有时用于使这种计数自动化。在这些方法中,显微照片通常通过标准程序转换,如利用阈值、分割、膨胀和腐蚀变换、边缘检测……。然后这些利用或多或少复杂的算法计算单独的颗粒。这种自动化方法通常存在一些缺陷。例如,如果获得的照片对比度有限,或者在颗粒重叠的情况下,该算法可能无法精确的将不同的颗粒分离,并且计数仅具有较差的可靠性。
其它已知的用于计算在液体中流动的颗粒的方法是基于阴影或者光漫射效应。这些方法广泛用于,例如制药领域中,也存在一些缺陷。首先,其通常不可能区分气泡和固体颗粒。这在评估包含大量干扰测量的溶解CO2的生理缓冲溶液的洁净度时特别关键。在这种测量中,根本不能从结果中得到任何关于颗粒性质的信息。
发明目的
本发明旨在提供一种不存在现有技术缺点的表征和/或计算透明介质中球形透明颗粒的方法。
根据以下描述,本发明的优点将是显而易见的。
发明内容
本发明涉及一种用于表征显示为焦点区(焦点)的透明物体至少一种性质的方法,所述物体在透明介质中,所述方法包括以下步骤:
-通过定向光源照射包含待表征物体的样品,从而在所述透明物体的
焦点区诱导光强度峰。测定光强度峰的至少一个特性,
由所述光强度峰的至少一个特性测定所述物体的至少一种性质。
对于显示为焦点区(焦点)的透明物体,在本文中其意指当被定向光源照射时,能够在所述焦点区集中光的物体,该集中的光形成光强度峰。在本发明中,所述光强度峰直接由物体和独立于成像装置的定向光源之间的相互作用诱导(即,光峰即使没有被观测到也存在)。
根据特别优选的实施方式,本发明的方法包括以下特征中的一个或者至少两个的适当组合:
该方法进一步包括以下步骤:
记录被照射样本的全息再现像(holographic representation);
由所述全息再现像重建由所述样本诱导的光场强度的三维再现像;
扫描该光场强度的三维再现像用来测定给出高于预定阈值的强度的光峰面积,所述光峰的每一个相应于一个颗粒;
-至少一种物体特性包括所述物体的数量,光强度峰的决定的特性(其包括峰的数量);
-这些物体选自由以下各项组成的组:乳液、固体珠、活细胞、死细胞以及它们的混合物中的气泡、液泡;
-这些物体选自由活细胞、死细胞和它们的混合物组成的组;

具体实施方式
本发明涉及一种利用许多透明物体在被光照射时共有的光学特征的分析方法。该共有特征为许多透明物体充当透镜,使光集中在所述物体之后的实焦点区(5)或在所述物体之前的虚区(6)。
这样的焦点区可以由例如具有不同于周围介质的折射率的球形或椭圆形透明颗粒(2-3)形式的物体诱导。这样的颗粒可以是例如分散在水溶液中的油滴,液体、活细胞或者死细胞中的气泡(3),气流中的液滴……。
对于透明,在本文中意指介质保持足够的光定向性以观测聚焦峰。这样的透明度例如可以通过雾度测量(ASTM D1003)来表征。由高雾度引起的问题是光背景增加,并且增加了区分背景和峰的难度。
为了观测光聚焦强度峰,优选颗粒表现大出于入射光波长的尺寸,更优选大于光波长三倍。在可见光的情况下,优选颗粒具有大于1μm的尺寸。
诱导焦点区的透明颗粒不限于自由流动的球形颗粒,而是还可以包括在平面上捕获的颗粒,如玻璃等接触的液滴或气泡。
存在这样的聚焦点/区的结果是这些颗粒中的每一个在被照射时将会产生光强度峰,通过扫描三维光强度分布可以容易地探测到该光强度峰。
有利的是,本发明的方法首先通过记录包含待分析颗粒的照射样品的数字全息再现像实施。然后优选地,在重建的由照射样品诱导的光场的3D再现像上实施扫描和分析。
即使可以通过本发明的方法表征单一颗粒,但是该方法自动化的可能性使得其对大组的颗粒特别适合。待表征的颗粒可以同时存在于再现像的范围内或者可以存在于时间序列内。
优选地,数字全息再现像通过数字全息显微镜记录(DHM)。所述DHM可以有利地是在EP1399730中描述的类型,其通过引用合并于此。
作为优选的替代,DHM可以是差分全息显微镜,如在EP1631788中所描述的,其通过引用合并于此。
有利的是,如在WO/2010/037861中所描述的DHM以暗场模式操作的。这样的暗场模式的优点是通过减少平均光背景使得容易检测光峰。
如国际专利申请号PCT/EP2010/64843中所描述的,使用离轴DHM具有的优点是快速记录动态事件(dynamical event),如流体中流动颗粒。
优选地,通过检测3D全息像的容积内高于预定阈值的光强度来测定光峰强度。
这种聚焦点测定的第一应用是一种用于计算流动介质中球形颗粒的方法。在这种方法中,光峰强度的数量相当于样品中球形颗粒的数量。这种计数方法的优点是聚光区比颗粒尺寸小得多,使得即使在颗粒的密度较高的情况下,也可以容易地分辨并很好地区分光峰。相比于2D再现像中重叠颗粒的计算方法,这是关键的优点。
计算颗粒仅使用峰的检测,但是其它的光峰特征,如形状、强度和位置也可以有利地利用。此类信息是相当于每个颗粒的透镜的特征。那些透镜特征本身是通过这些颗粒的几何形状和折射率而测定的。
例如,在液体中的气泡将充当凹透镜,在气泡前方产生焦点,这与将充当会聚透镜的高折射率的颗粒相反,其在该颗粒后方产生焦点。因此,在优选的方法中,这些颗粒是根据相应光峰区的相对位置而分类的。该分类使得容易区分不同种类的颗粒,如气泡和高折射率颗粒。
对于高或低的折射率,在本文中意指分别高于或低于颗粒周围介质折射率的折射率。
作为通过颗粒光学性质而区分的颗粒的另外实例,出乎意料地显示出,本发明的方法能够基于与细胞相应的光峰特征来区分流体介质中流动的死细胞和活细胞。
在下文中示出的实验性实施例中,如在图5中示出的,峰的尺寸用作区分标准(即,其中光强度高于阀值的体积或面积)。也发现可以使用其他标准,如绝对峰强度(即,峰内最大强度或光强度的积分)。
由于该方法可以容易地自动化并以连续的时间序列自动实施,因此本发明的方法可以有利地用于研究单个细胞的位移。
作为本发明进一步的动力学应用,可以利用一些峰特征对喷雾干燥过程进行研究。在这类研究中,喷雾颗粒通过上述方法计数,可以精确地测定他们在流体中的单独移动,同时可以通过分析重构颗粒(“在焦点上”图像)来测定作为时间函数的颗粒尺寸,并且该溶液浓度可以由颗粒折射率计算,由颗粒形状和位于焦点区的光强度峰之间的相互关系测定。这种方法带来的一个优点将是例如测定过饱和现象和相应的成核和生长过程的能力。
测定透明物体焦点的另一个有利应用是准确测定透明平面上液滴的几何参数。然后可以将那些几何参数用于例如准确计算接触角。
更一般地,以下参数可以是感兴趣的:峰数量、峰形状和尺寸、峰强度(积分值和/或最大值)、与相应颗粒的相对位置。
那些峰参数可以有利地与相应颗粒的形状和位置相关。
与源于颗粒的相应荧光数据的相关性也可以有利地用于表征颗粒。这种荧光相关性可以使用在文献US2004/156098中描述的方法,其通过引用合并于此。
由那些参数,以下特征可以有利地推论:活细胞的活力、溶液浓度、细胞类型、颗粒运动等等。
实施例
使用本发明的方法表征细胞培养物。
如图3所示的显微镜用于记录数字全息图。全息图采样速率是2.5Hz,并且在一个序列中拍摄200张全息图。
如图1所示,样品架4是流动室,其中流体样品处于流体8中,并且颗粒是动态观测的。
图4a示出所获得的全息记录之一的重建图。图4b示出聚焦峰的相应投影。在图4b中可以看到,当考虑用光峰代替相应细胞再现像(细胞图像,cell representation)时,即使接触细胞也很好地分辨。注意到为了简化图像,在二维上示出这些峰。在真实图像中,也可以在深度上分辨它们。
通过给出的方法测定的细胞的数量是三百七十二万细胞/ml。作为对比,在bürker计数细胞中手动计数为三百七十一万细胞/ml。即使在非常高的细胞浓度的情况下,也保持良好的一致性。在最后一种情况下,常用的自动计数方法给出了不精确的结果。
在进一步的实验中,本发明的方法首先用于计算在培养物种的细胞数。在第二步中,培养介质在42.5℃下经受3小时热处理。已知这种处理可以降低细胞的活力。
然后在热处理前后测定观测到的光峰尺寸的分布。该结果如图5所示。如在该图中可以看到的,峰尺寸的分布与细胞活力有很大的相关性。
Claims (14)
1.用于表征在透明介质(1)中透明物体(2、3)的方法,所述透明物体(2、3)显示为光学焦点区(5、6),所述方法包括以下步骤:
-通过定向光源(7)照射包含待表征的所述透明物体(2、3)的样品,从而在所述透明物体的所述焦点区诱导光强度峰(5、6);
-测定诱导的所述光强度峰(5、6)的至少一个特征;
由所述光强度峰(5、6)的所述至少一个特征测定所述透明物体(2、3)的至少一种性质。
2.根据权利要求1所述的方法,进一步包括以下步骤:
-记录被照射的所述样品的全息再现像;
-由所述全息再现像重建由所述样品诱导的光场强度的三维再现像;
-扫描所述光场强度的所述三维再现像,用来测定显示为高于预定阀值的强度的光强度峰(5、6),所述光强度峰(5、6)的每一个相应于一个透明物体(2、3)。
3.根据权利要求2所述的方法,其中,所述全息再现像通过全息显微镜获得。
4.根据权利要求3所述的方法,其中,所述全息显微镜根据差分模式操作。
