Abstract

发明领域和发明背景
本发明涉及癌症的检测方法,更具体地讲,本发明涉及优化癌症药物治疗的方法。
在世界范围内,大多数发病率和死亡率是由癌症引起,尽管目前医学技术不断进步。目前的治疗策略主要集中在通过各种外科手术和非手术技术切除患者体内的癌细胞或使癌细胞死亡;但最广泛使用的还是化学治疗和γ辐射。这些方法具有许多显著缺陷,尤其是会破坏患者体内的健康细胞/组织,而且目前生产的用于癌症治疗的化疗药物具有相当大的毒副作用。
“分化治疗(Differentiation therapy)”是一种替代方法,该方法可促进表型从恶性恢复到正常。分化治疗是根据以下概念:癌细胞是被阻止在未成熟或未完全分化状态的正常细胞,缺乏控制其自身生长的能力,因而以异常快的速率繁殖。分化治疗的目的是迫使癌细胞重新恢复成熟过程。尽管分化治疗不能杀死癌细胞,但是可阻止其生长并可允许使用更多常规治疗(例如化疗),以消除剩余恶性细胞。
与旨在癌/肿瘤细胞死亡的常规治疗策略相比,分化治疗具有众多优势。一开始,就不需要用化疗药或γ辐射来破坏患者体内的健康细胞/组织,同时也就没有其相关不良副反应。在许多情况下,通过γ辐射或化疗杀死癌细胞能消除患者体内大部分、但并非所有癌细胞,因而导致疾病的缓解。用分化治疗,推测通过诱导癌细胞中的某些细胞进入终末分化途径并最终衰老,这会以某种方式将信号发给其它癌细胞经不同机制而跟随其后。
各种标记物已用作监测正在接受分化治疗的患者的工具。一种这样的标记物就是碱性磷酸酶,其中该酶的表达看来与细胞分化状态有关[Patnaik A等,Clin.Can Research,2002 Jul;8(7):2142-8;Rephaeli A,Zhuk R,Nudelman A,2000,Drug Develop.Res.第50卷:379-391]。

美国专利申请号20060100488公开了在采用交流电之后,通过直接监测细胞的电反应而检测癌细胞。WO 91/15595公开了对癌细胞电导率进行分析,用于监测对治疗和药物筛选的反应性。具体地讲,WO91/15595公开了通过分析其电导率来监测抑制癌细胞体积和数量增加的特定药物的有效性。因此,这两篇专利申请都公开了癌细胞固有电学性质可用作标记物,用于检测和监测癌细胞。
美国专利申请号20040053425公开了对流体中分析物的电流测量分析,其中电极中含有电流产生酶。美国专利申请号20040053425并未公开对胞内标记物的电流测量检测。
美国专利号5,149,629公开了对包括癌细胞标记物在内的标记物的电流测量分析,其中电极中含有能与标记物结合的抗体。分析是通过底物竞争来进行的。美国专利号5,149,629并未检测癌细胞的内源性电流特性。
因此,对分析物进行电流测量检测已经成为有效的检测方法。然而,时至今日,电流测量检测尚未用于分析细胞酶活性。
发明概述
根据本发明的一个方面,提供癌细胞的检测方法,该方法包括:(a)在酶催化细胞与底物反应的条件下,使细胞与酶的底物接触,从而生成能产生电信号的产物;和(b)测定电信号水平,其***号水平与预定阈值之间的差异就是癌细胞的标志。

优选实施方案的描述
本发明涉及癌细胞的检测方法。具体地讲,本发明可用于诊断癌症,监测治疗,确定治疗方案和开发针对疾病的新型治疗模式。
参考附图和所附说明,可以更好地理解本发明方法的原理和操作。
在详细解释本发明的至少一个实施方案之前,应当知道,以下实施例中的描述或举例说明所给出的细节并不限制本发明的应用。可以用其它实施方案或不同方式来实施或实现本发明。另外,应当知道,本文所用的措词和术语都用于描述的目的,不应视为限制性。
分化治疗集中在通过迫使癌细胞重新恢复成熟过程而诱导癌细胞变为正常细胞。各种标记物已用作监测接受分化治疗的患者的工具。一种这样的标记物就是碱性磷酸酶,其中该酶的表达看来与细胞分化状态有关。一般而言,正常酶活性表明,作为特定药物治疗结果,细胞正确地分化;而缺乏酶活性则表明,对于特定癌症和/或特定患者而言是无效药物治疗。
然而,对这类标记物,尚无方法能快速简便而高灵敏度、高选择性和高精度地进行检测。
本发明的发明人已经开发出新的电化学方法,用于灵敏而高通量地检测癌细胞对分化治疗的反应。
在将本发明付诸实施的时候,本发明的发明人就已经知道可以使用p-APP等电化学底物来进行酶的电流测量法,以便确定分化剂的有效性。如图1所示,用本发明方法评价了3种分化剂对人结肠癌细胞的有效性。使用含小型电化学电解池阵列的芯片,本发明的发明人能够以高灵敏度、高精度和快速方式来监测这些分化治疗剂对极少量细胞样品(少于~15个细胞)的有效性(图2A-F和图3)。
在5分钟内,可用分辨率为10毫微安培的电流反应测定药物效果。此外,这些结果表明所诱导的电流信号与纳升体积的小室内癌细胞计数之间的数量关系。
在临床相关样品中采用纳升体积的分析设备很令人感兴趣,因为只需要很少量组织进行诊断,而且还能进行高通量分析并比较不同药物对同一肿瘤样品的效果,同时还能保持均一的生物条件和环境条件。另外,这一新方法有助于针对个别患者来定制癌症药物和治疗,成为“个性化医疗”。
因此,根据本发明的一个方面,提供癌细胞的检测方法。方法包括在酶催化细胞与底物反应的条件下,使细胞与酶的底物接触,从而生成能产生电信号的产物,然后测定电信号水平,其***号水平与预定阈值之间的差异就是癌细胞的标志。
本文所用的术语“检测”是指检测、诊断、检查、发现、揭示、观察或鉴定细胞的行动。
本文所用的术语“细胞”是指哺乳动物细胞,优选人体细胞。根据本发明,可使用单细胞以及多个细胞。优选多个细胞包含不少于10个细胞,且不多于500个细胞。优选地,细胞是在单一悬浮液中,这样可以对细胞进行计数,尽管也可检测粘附细胞和聚集物。多个细胞可来自任何生物样品,例如细胞系、原代培养物和细胞样品,例如活检样品(手术活检样品,包括切除或截除的活检物、细针吸出物等)、完整切除物或体液。提取活检样品的方法是本领域众所周知的。
无需任何预处理,就可以评价生物样品中细胞的功能酶。因此,生物样品中的细胞优选是完整的(即整个细胞),并且优选是活的,尽管可以理解,本发明也包括对细胞进行预处理,例如产生细胞提取物或非完整细胞。
“癌细胞”,本文亦称“恶性细胞”,是指已从正常细胞***控制中释放出来的细胞,因而其特性是异常生长并趋向于以非受控方式增殖,在某些情况下还会转移。因此,癌细胞可以是赘生性细胞、恶性前细胞(pre-malignant cell)、转移性细胞、肿瘤细胞、致癌细胞、具有癌基因型的细胞、恶性表型的细胞、癌基因转染细胞、病毒转化细胞、表达癌基因的细胞、表达癌症标记物的细胞或其组合。

根据本发明该方面的优选实施方案,癌细胞是结肠癌细胞。
如上所述,本发明的方法是通过使酶的底物与细胞接触、引起细胞反应而进行的,其中酶促反应产物能够产生电信号。
本文所用的术语“细胞反应”是指底物与细胞所表达的内源性酶之间发生的反应,而不是与外源性酶发生的反应。
根据需要检测的酶(在本文也称为“标记酶”)来选择底物。酶可位于细胞内(即胞内酶)或细胞膜上(即膜结合酶)。根据另一实施方案,酶是分泌酶。可以理解,当待检测的酶是胞内酶时,优选底物是膜渗透性的。此外,优选选择底物,使得在催化后,所生成的产物也是膜渗透性的,使得它可从细胞中扩散出来并到达检测电极。
根据一个实施方案,需要检测的酶是碱性磷酸酶(或分泌型碱性磷酸酶(SEAP)),而底物是17,4-氨基苯磷酸(p-APP)。碱性磷酸酶使p-APP转化为电化学产物,即对氨基苯酚(PAP)。
p-APP是一种普通的市售产品,可得自Sigma-Aldrich公司(www.sigmaaldrich.com)、Bio-world公司(www.Bio-world.com)和许多其他公司。
碱性磷酸酶存在于正常细胞中,但在癌细胞中减少(或甚至不存在)。因此,对细胞的碱性磷酸酶活性水平进行分析,可用作评价特定药物有效性的标记,可用于治疗癌症乃至用于诊断目的。
本文所用的术语“接触”是指在底物可被酶催化的条件下,使底物接近细胞。因此,例如应在缓冲剂条件下,在允许催化底物并生成电化学电解池所检测的足够的产物的温度和时间下进行接触。例如,当p-APP用作底物时,优选在室温下接触至少5分钟-参见以下实施例2和实施例3。
可在体外、离体或体内进行接触。可在容器中进行接触,该容器也能检测酶促反应产物(即在电化学电解池中),这样就可在线检测电信号。下文将进一步介绍这样的容器。或者,可在分离的容器中进行接触,从中进行检测,使得能够在特定时间点连续取样并将所取样品放入电化学电解池内。因此,可在试管、烧瓶、组织培养器、芯片、阵列、板、微量板、毛细管等中进行接触。可将细胞放在振动板上,再加入底物,用于连续充分混合细胞内容物。
如上所述,在化学电解池的电极上对能经历氧还反应(即能进行电子转移)而得到电信号的产物(即电化学产物)进行电化学测量,通常是在电化学电解池中进行。
本文所用的术语“电信号”是指电子或电化学活性物。
本文所用的术语“电化学测量”是指通过使用溶液中的、尤其是电化学电解池中的电极来进行的测量。可通过例如定时电流法、定时电位法、循环伏安法、定时库仑法或方波伏安法,来进行测量。在这样的测量中可检测的信号,与在这些电化学测量中来自对照的信号是不同的。
对于在线测量,本发明的电化学电解池包括测量电极、返回电极(return electrode)、参考电极和容纳电解池的小室。
工作电极具有各种不同类型,例如可由碳(包括玻璃碳、活性碳布电极、碳毡、铂化碳布、平纹碳布)制成,也可由金、铂或银制成。对电极也可由与工作电极相同的材料制成。参考电极可以是例如饱和甘汞电极,可以是Ag/AgCl电极。此外,电极可以是丝网印刷电极,其可***到含电解池的容器中,而无需取出样品并转移到单独的电化学电解池中。
用于按照本发明方法测定产物的电极可以是可重复使用的电极或一次性电极。可重复使用的电极可以是例如由玻璃碳制成的盘状或棒状电极,其可嵌入特氟隆中。一次性电极可以是例如呈碳纸、碳布、碳毡形式的电极或上述种类的丝网印刷电极。
根据一个实施方案,电化学电解池是三电极电解池。根据另一实施方案,电化学电解池是两电极电解池。根据优选的实施方案,可以阵列(即芯片)形式提供电化学电解池,所述阵列包含多个这样的电解池即多孔阵列,其中各孔的大小是纳升体积。
用于测定由反应产物产生的电信号的***还可包括控制模块,其可以是计算机、恒电位仪和复用器模块(multiplexer module),复用器模块在同时测量多个电化学电解池的典型实施方案的情况下是需要的。
参考定时电流模式,将描述在电解池中进行的电化学测量。应当理解,对其加以必要的修正,也可用于上述其它电化学测量模式。此外,参考使用多电极***(包含多个电极的***),可进行描述,并且显然也可用于含单个电解池的***。
在电化学测量开始,一起操作所有电极,计算机通过并联端口扫描所有电极,并通过计算机记录针对各电极施用电位时的背景响应。在一段长时间中,可进行完整的电化学测量程序,同时测量因产物浓度改变而产生的电流。在电极表面因天然变异性所导致的不相同的情况下,可通过测量电活性物(尤其是与电化学电解池中的酶促反应产物相同种类)的氧化或还原并比较所有电极结果,来标定***。
在进行测定时,可将电极与恒电位仪相连接,并同时也通过复用器聚集到微机并联端口。
将各电极***到含参考电极和反电极的电化学电解池中,这些电极也与恒电位仪相连接。通过恒电位仪将特定电位施加在电极上(所有电极可以施加相同电位,或者每个电极施加不同电位)并检测各电极的电流。在计算机屏幕上可实时观察电信号。
因为酶促反应的电化学产物所产生的电信号反映出细胞中的酶水平,而且酶(其存在、不存在或水平)是癌症的标记物,所以信号可用于确定细胞是否癌变(即恶性)。具体地讲,如果所产生的电信号水平与预定阈值不同,则表明细胞是癌细胞。通常,预定阈值是通过对照细胞所产生的电信号来确定的。
对照细胞可以是正常分化细胞、非癌变细胞,优选与怀疑癌变或未分化表型的被测细胞同属相同组织和样本。与对照细胞相比,优选差异至少为10%、20%、30%、40%、50%、80%、100%(即2倍)、3倍、5倍或10倍。
根据本发明的一个实施方案,癌细胞中的酶含量(和由此而来的电信号)低于非癌细胞中的酶含量(和由此而来的电信号)。
根据本发明的另一个实施方案,癌细胞中的酶含量(和由此而来的电信号)高于非癌细胞中的酶含量(和由此而来的电信号)。
可以理解,本发明的方法可用于诊断癌症患者。
本文所用的术语“诊断”是指对癌症进行分类,确定癌症的严重程度(分级或分期),监测癌症的发展,预测癌症的结果和/或痊愈的希望。
受试者可以是经过常规健康检查的健康动物或人类受试者。或者,受试者可以是处于癌症危险中的患者(例如具有遗传倾向的受试者,具有病史和/或家族史的受试者,已接触致癌物、职业危害、环境危害的受试者,和/或表现出可疑癌症临床症状(例如便血或黑便、不明原因的疼痛、出汗、不明原因的发热、不明原因的体重减轻至厌食、肠习性改变(便秘和/或腹泻)、里急后重(感觉排便不完全,尤其是对于直肠癌而言)、贫血和/或全身乏力)的受试者。
可以理解,本发明具有涉及以下的各种用途:个性化优化癌症治疗,监测患者的抗癌治疗,测定患者的抗癌治疗和鉴定能逆转细胞恶性表型的药物。
因此,根据本发明的另一方面,提供能逆转细胞恶性表型的药物的鉴定方法。方法包括使至少一个癌细胞接触药物并测定抗癌药的效果,即按照本发明的方法来监测标记酶(例如碱性磷酸酶)的活性或表达。
本文所用的术语“逆转恶性表型”是指至少部分地逆转恶性细胞的增殖和/或侵袭性。
本文所用的术语“药物”是指包含生物制剂或化学制剂的试验组合物。
经本发明的方法测试为潜在抗癌药的生物制剂的实例包括但不限于核酸(例如多核苷酸、核酶、siRNA和反义分子(包括但不限于RNA、DNA、RNA/DNA杂合体、肽核酸以及具有改变的主链和/或基本结构(bass structure)或其它化学修饰的多核苷酸类似物);蛋白质、多肽(例如肽)、碳水化合物、脂质和“小分子”候选药物。“小分子”可以例如是天然存在的化合物(例如衍生自植物提取物、微生物肉汤等的化合物)或合成的有机化合物或有机金属化合物,其分子量小于约10,000道尔顿,优选小于约5,000道尔顿,最优选小于约1,500道尔顿。
根据本发明该方面的优选实施方案,药物可以是分化剂,包括但不限于丁酸及其衍生物。
经本发明方法测试为潜在抗癌药的条件的实例包括但不限于辐射暴露(例如γ辐射、紫外辐射、X射线)。
根据本发明该方面的一个实施方案,也可在接触药物之前对“标记酶”进行测定分析,从而可在治疗前后进行比较。
根据本发明该方面的另一实施方案,使药物接触癌细胞足够长时间,以产生抗癌效果。因此,例如如果是分析丁酸和/或其衍生物的话,优选这些药物接触癌细胞的时间至少为1天,更优选为3天。
可以理解,可以使药物在体外、离体或体内接触癌细胞。如果接触是在体内进行,通常在接触本发明底物之前,将细胞从患者体内取出。
在理论上,尽管可用单个电化学电解池实施本发明,但是这样的方法既无效果又不合乎要求。优选本发明的方法使用多个电化学电解池同时筛选不同药物,用于高通量药物筛选。电解池可以是芯片的组成部分,所述芯片例如以下实施例1所述的硅芯片。
因此,根据一个实施方案,采用高通量工具实施本发明的方法。因此,可使用例如自动取样装置、液体处理装置、分配器、电极阵列、机器人或其任何组合,来实施本发明的方法。
目前已经知道,仅根据肿瘤类型和解剖学位置是无法预测肿瘤治疗反应的。可以理解,可以修改能逆转细胞恶性表型的药物的鉴定方法,使得特定患者细胞可用于测定***,因而可定制针对特定患者的治疗药。此外,可以理解,用本发明的方法不仅可选择具体药物,而且还可根据本发明的方法来鉴定最佳剂量和最佳治疗方案。采用这样的方式,可以确定药物的治疗有效量。
可根据本发明方法所选择的最佳治疗条件来治疗患者,并任选在合适的时间周期之后进行复检。采用这样的方式,可以在治疗进行期间连续监测患者的反应。
可以理解,在治疗期间,对酶水平的分析和给药步骤可重复多次。例如可以每周分析碱性磷酸酶水平,然后给予药物。如果碱性磷酸酶水平比对照高,就减少药物剂量。如果碱性磷酸酶水平比对照低,就增加药物剂量。
可以理解,可在试剂盒中提供本发明的电化学电解池以及至少一种抗癌药(例如分化剂,例如丁酸),用于测定其对癌细胞的效果。如有必要,本发明的试剂盒可以呈包装形式,其可含有本发明试剂盒的一个或多个单位。包装中可带有试剂盒的使用说明书和抗癌药对具体细胞数的估计剂量。包装中也可带有由政府机构对实验室添加物(laboratory supplement)的制造、使用或销售所规定的、与该包装容器有关的信息,这种信息表明政府机构许可所述组合物的形式。
根据一个实施方案,试剂盒还可包含底物,所述底物被生物细胞(即癌细胞)的标记酶催化而进行酶促反应以生成反应产物,从而在电化学电解池的电极上发生氧还反应。这样的底物在上文已有描述。
在本专利有效期间,预计将会开发出许多相关底物,并且底物这一术语的范围包括所有这类新技术成果(all such new technologies apriori)。

实施例1
纳米生物芯片的制备
利用标准微***技术(micro-system-technology,MST)方法,设计并制造出了纳米生物芯片(nano-bio-chip)。其构造包括小型电化学电解池阵列。将电解池放入纳升体积的小室(即电化学电解池)中。每个圆柱状小室容纳100nL体积。所有阵列都包括阳性和阴性对照小室。
装置分两部分来制备:a)一次性芯片:其具有纳升小室,其中放入了电解池(cell),和b)可重复使用芯片:其与包括复用器、恒电位仪、温度控制和袖珍型PC在内的电子电路相连接,用于传感和数据分析。这套设置允许连续重复使用,用于多次测量。
芯片由硅制成,并含有8个小型电化学电解池阵列。每个电化学电解池由以下3个圆形、周围有绝缘氮化硅层的电极组成:1)金工作电极,2)金反电极和3)Ag/AgCl参考电极。电极是通过金溅射、显微光刻技术(microlithography)和选择性沉积Ag而制成,而对于参考电极,还要使其在含氯溶液中阳极化。小室壁是由光聚合的聚酰亚胺构成(SU-8)(图1A-B)。硅芯片通过导线与塑料芯片连接,后者接入电子电路。
通过接入电子电路的手持式掌上恒电位仪进行信号传导,用于电极信号的调制和检测(Palm Instruments BV-2004)。该电子仪器是由具有温度控制的电化学电解池的8个独立重复电路组成。在各电化学电解池的工作电极与参考电极之间施加电位并测定输出电流。
实施例2-3
通用材料与方法
细胞系和培养物:在37℃/95%空气/5%CO2中,将HT-29人结肠癌细胞在含胎牛血清的DMEM培养基中培养3天,然后进行丁酸处理。将不同浓度的丁酸加入到HT-29细胞培养物中。所用浓度为:0mM、0.078mM、0.156mM、0.3125mM、0.625mM、1.25mM、2.5mM、5mM和10mM。由此求出最佳丁酸浓度、LC50和活力。测定在PBS中进行,用完整细胞而没有对癌细胞进行额外处理(例如溶解细胞)。
丁酸(BA)及其衍生物:将范围在0.08-10mM之内的丁酸(BA)(Sigma,Israel)加入到HT-29结肠癌细胞中并孵育72小时,用于优化。固定浓度为2.5mM的BA用于所有电化学实验。如文献所述,合成BA衍生物即磷酸丁酰基甲酯二乙酯和丁酸新戊酰氧基甲酯[RephaeliA,Zhuk R,Nudelman A,2000,Drug Develop.Res.第50卷:379-391]。将磷酸丁酰基甲酯二乙酯溶于PBS中,丁酸新戊酰氧基甲酯溶于DMSO中,然后用培养基稀释至最终DMSO浓度为≤0.1%。再将固定浓度为50μM的前体药物加入到细胞中并在37℃/95%空气/5%CO2的条件下孵育96小时。与分化剂一起孵育之后,通过锥虫蓝排出法(tripa blue exclusion)进行活细胞计数并检测活力,求出LC50。在所有电化学测量之前,将细胞离心并在PBS中稀释。所有测量都在PBS中进行。通过测定碱性磷酸酶的酶活性,来测定BA、丁酸新戊酰氧基甲酯和磷酸丁酰基甲酯二乙酯的影响。
碱性磷酸酶活性测定:
光学测定:用光学底物对硝基苯磷酸(现成的PNPP试剂盒,Sigma)测定碱性磷酸酶活性。在420nm波长处测定所得酶促产物。
电化学测量:用电化学底物p-APP进行酶的电流测量。在220mV下,酶促反应产物氨基苯酚(p-AP)在金工作电极上被氧化,测量所产生的电流。在8通道100nL体积的电化学小室中加入经BA或其衍生物处理过的HT-29癌细胞。将细胞放入电化学小室中,加入底物至终浓度为1mg/ml,总体积为100nL。测量所产生的电流并在显微镜下对细胞进行计数。分析细胞数与电流密度之间的关系。
实施例2
经本发明生物芯片测定的药物对结肠癌细胞的效果
为了进行个性化癌症治疗,检测各种单一来源的药物对人结肠癌细胞(HT-29)的影响。
结果
使HT-29结肠癌细胞与不同分化治疗剂接触96小时,然后测定所诱导的碱性磷酸酶活性。阵列上的各电化学小室中加入接触过不同试剂的细胞,这些试剂是丁酸、磷酸丁酰基甲酯二乙酯和丁酸新戊酰氧基甲酯。结果见图1A-C。正常酶活性表明,作为特定药物治疗结果,细胞适当地分化。如图1A-C所示,在50μM浓度时,BA和磷酸丁酰基甲酯二乙酯诱导碱性磷酸酶的酶活性,而丁酸新戊酰氧基甲酯却未诱导出任何酶活性,对于HT-29癌细胞,这可能涉及其潜力降低[M.Entin-Meer等,Molecular Cancer Therapeutics,第4卷,第1952-1961页,2005;D.Engel等,Clinical Cancer Research,第11卷,第9052S-9052S页,2005]。另外,设置阳性和阴性对照:在小室中加入经药物处理的细胞,未处理细胞作为阴性对照,而纯化的碱性磷酸酶作为阳性对照。重要的是,注意到磷酸丁酰基甲酯二乙酯与BA的分化效果类似,尽管前者浓度要低1.5个数量级,即磷酸丁酰基甲酯二乙酯和BA的浓度分别为50μM和2.5mM。
实施例3
用生物芯片对癌细胞进行定量测定
结果
测定所诱导的电流信号与纳升体积小室内癌细胞计数之间的显著的数量关系。癌细胞计数和相对酶活性见图2A-F。用电化学阵列芯片,得到对碱性磷酸酶活性监测的电流反应曲线。将HT-29结肠癌细胞暴露给丁酸(2.5mM)。将HT-29细胞以及底物PAPP放入芯片中100nL体积的电化学小室内。用电流测量技术在220mV测量电流。上中下3条曲线分别表示小室内计数的约100个细胞、15个和0个细胞的电流反应。多次测量表明小室内细胞计数与碱性磷酸酶活性之间具有高相关性。结果见图3。定量测定酶促反应的能力是因为芯片的小型化设计,并结合电化学检测方法上的优势。
实施例4
经本发明生物芯片测定的磷酸丁酰基甲酯二乙酯对结肠癌细胞的效果
为了进一步检测生物芯片的灵敏度,将数量递增的HT-29结肠癌细胞与磷酸丁酰基甲酯二乙酯一起孵育不同时间。用本发明生物芯片测定内源性碱性磷酸酶的含量。
结果
结果见图4A-C、5A-C和6A-C。磷酸丁酰基甲酯二乙酯对癌细胞的效果随孵育时间的延长而增加。另外,可以观察到,效果的大小与所检测细胞数有关。
结论
已经证明了灵敏而高通量检测响应分化治疗的癌细胞的新方法。就人结肠癌细胞而言,这些细胞用不同分化治疗药物处理,同时在线测量细胞对不同药物的反应。这样的微阵列技术提供了在线检测多种药物的能力并能针对个体来定制有效治疗。
这样的“芯片实验室(Lab-on-a-chip)”***提供了对极少量样品(少于~15个细胞)的灵敏的检测能力,而且在***芯片之前无需进行特别的细胞处理。
可以理解,为了清楚起见,在独立的实施方案内容中所描述的本发明的某些特征,也可以组合方式在单个实施方案中提供。相比之下,为了简明起见,在单个实施方案中所描述本发明的不同特征也可分别或在任何合适的亚组合中提供。

Claims (22)
1.p-APP和高通量工具在制备用于检测癌细胞而设计的***中的用途,其中碱性磷酸酶在细胞中内源性表达并催化与所述p-APP的反应,从而生成能产生电信号的产物;和所述工具用于测定所述电信号水平。
2.p-APP和高通量工具在制备用于诊断癌症患者而设计的***中的用途,其中碱性磷酸酶在受试者细胞中内源性表达并催化与所述p-APP的反应,从而生成能产生电信号的产物;和所述工具用于测定所述电信号水平。
3.p-APP和电化学电解池在制备用于对受试者癌症治疗进行个性化优化的试剂盒中的用途,其中碱性磷酸酶在受试者癌细胞中内源性表达并催化与所述p-APP的反应,从而生成能产生电信号的产物,和所述电化学电解池用于测定在抗癌药的存在下所述细胞所产生的所述电信号水平。
4.p-APP和电化学电解池在制备用于鉴定能逆转细胞恶性表型的药物的试剂盒中的用途,其中碱性磷酸酶在细胞中内源性表达并催化与所述p-APP的反应,从而生成能产生电信号的产物,和所述电化学电解池用于测定在抗癌药的存在下所述细胞所产生的所述电信号水平。
5.权利要求1-2中任一项的用途,其中所述工具选自自动取样装置、液体处理装置、分配器、电极阵列、机器人或其任何组合。
6.权利要求1-4中任一项的用途,其中所述癌细胞是结肠癌细胞。
7.权利要求1-2中任一项的用途,其中所述高通量工具用电化学电解池来进行。
8.权利要求3、4和7中任一项的用途,其中所述测定是在多孔阵列中进行。
9.权利要求8的用途,其中所述多孔阵列的各孔包含电化学电解池。
10.权利要求9的用途,其中所述多孔阵列的各孔是纳升体积的孔。
11.权利要求3-4中任一项的用途,其中所述药物包括试验组合物。
