实施例22.通过农杆菌-介导的转化将grg12转化进玉米植物细胞中
穗最好在授粉8-12天后收集。从玉米穗中分离出胚,并且将那些0.8-1.5mm大小的胚优选用于转化。将胚盾片侧朝上涂布于合适的孵育培养基上,并在25℃下于黑暗中孵育过夜。然而,在本质上是不必将胚孵育过夜的。将胚与农杆菌菌株接触约5-10分钟,所述农杆菌菌株含有用于Ti质粒介导的转移的合适载体,并随后涂布于共培养培养基上约3天(在25℃下于黑暗中)。在共同培养后,将外植体转移至恢复阶段培养基中约5天(在25℃下于黑暗中)。将外植体在选择培养基中孵育达到8周,这取决于所采用的特定选择的性质和特征。在选择阶段后,将得到的愈伤组织转移至胚成熟培养基中,直到观察到成熟的体细胞胚形成。随后,将得到的成熟的体细胞胚置于低光照下,并且通过本领域熟知的方法起始再生过程。
