Falls R13 für einen (C1-5)Alkylrest steht, so bedeutet Alkyl die oben angegebenen Reste, insbesondere jedoch Isopropyl, n-Butyl, Isobutyl und 2-Methyl-butyl. Hydroxyalkyl bedeutet vorzugsweise Hydroxymethyl oder Hydroxyethyl. Falls R14 Aminomethylpyridyl bedeutet, so steht es vorzugsweise für Aminomethyl-2-pyridyl.
Die im Anspruch 3 beschriebenen Verfahren a) bis e) zur Herstellung von Verbindungen der Formel I können im einzelnen wie folgt durchgeführt werden: Das Verfahren gemäss Verfahrensstufe a) wird vorzugsweise so durchgeführt, dass man eine Säure der Formel II mit einem Amin der Formel H-B unter Verwendung von in der Peptidchemie bekannten Verfahren z.B. in Gegenwart von N,N min -Dicyclohexylcarbodiimid (unter Zusatz von 1-Hydroxybenzotriazol) in einem geeigneten Lösungsmittel wie z.B. Methylenchlorid bei Temperaturen zwischen 0 DEG und Raumtemperatur umsetzt.
Das Verfahren gemäss Verfahrensstufe b) wird so durchgeführt, dass man einem Alkohol der Formel I mit CrO3.Py2 (Collin's Reagenz) in einem Lösungsmittel wie Methylenchlorid oder DMF zum entsprechenden Keton oxidiert.
Das Verfahren gemäss Verfahrensstufe c) wird vorzugsweise so durchgeführt, dass ein Keton der Formel I mit NaBH4 in einem Lösungsmittel wie Methanol zum entsprechenden Alkohol reduziert wird.
Das Verfahren gemäss Verfahrensstufe d) erfolgt zweckmässigerweise unter Verwendung von Palladium (10% auf Aktivkohle) als Katalysator in einem Lösungsmittel wie z.B. Ethanol bei einem Wasserstoffdruck von 1 bis 5 atm bei Temperaturen von Raumtemperatur bis 60 DEG .
Die Umsetzung gemäss Verfahrensstufe e) erfolgt vorzugsweise so, dass die t-BOC Gruppe in Gegenwart von Trifluoressigsäure bei Temperaturen von 0 bis 5 DEG abgespalten und das freie Amin mit einem aktiven Säurederivat wie z.B. Hydroxysuccinimidestern in einem Lösungsmittel wie Methylenchlorid acyliert wird.
Die in den obigen Verfahren verwendeten Ausgangsverbindungen sind entweder bekannt oder können auf an sich bekannte Weise, beispielsweise wie in den nachfolgenden Beispielen beschrieben, hergestellt werden.
Die erfindungsgemäss hergestellten Verbindungen der Formel I können auf an sich bekannte Weise isoliert und gereinigt werden. Racemische und/oder diastereomere Gemische können auf an sich bekannte Weise aufgetrennt werden.
Falls die Verbindungen der Formel I saure oder basische Gruppen enthalten, so können diese gegebenenfalls auch Salze bilden, beispielsweise Metallsalze wie Natriumsalze oder Säureadditionssalze wie Hydrochloride.
In den nachfolgenden Beispielen sind alle Temperaturen in DEG C angegeben und sind nicht korrigiert.
Die in den Beispielen in abgekürzter Form angegebenen Verbindungen sind im Anspruch 2 in der Reihenfolge der Beispiele voll ausgeschrieben.

Beispiel 1: t-BOC-Cha(O)Bly-F2Cha-NHiBu
140 mg F2ChaNHiBu werden in 2 ml Tetrahydrofuran mit 170 mg t-BOC-Cha(O)Bly-OH in Gegenwart von 120 mg Hydroxybenzotriazol und 97 mg N,N min -Dicyclohexylcarbodiimid 2 Stunden bei 0 DEG , dann 24 Stunden bei Raumtemperatur umgesetzt. Vom ausgefallenen Dicyclohexylharnstoff wird abfiltriert und das Filtrat an Kieselgel mit Ether/Hexan 10-50% chromatographiert. [ alpha ]D = -7.3 DEG (c = 0.4 in Methylenchlorid).
Beispiel 2: t-BOC-Cha(O)Bly-F2Cho-NHiBu
22 mg des Produktes aus Beispiel 1 werden in 1.8 ml Methylenchlorid gelöst und mit 84 mg Collins Reagenz (CrO3.Py2) versetzt. Nach 30 min. wird Essigester zugesetzt, über Hyflo und Kieselgel filtriert und das Filtrat aus Benzol lyophilisiert. [ alpha ]D = +4.6 DEG (c = 0.17 in Methylenchlorid).
Beispiel 3: t-BOC-Cha(O)Nle-F2Cha-NHiBu
12 mg des Produktes aus Beispiel 1 werden in 1.2 ml Äthanol gelöst und während 4 Stunden in Gegenwart von 1.4 mg Pd/C hydriert. Vom Katalysator wird abfiltriert und das Filtrat eingedampft. [ alpha ]D = -5.7 DEG (c = 0.14 in Methylenchlorid).
Beispiel 4: t-BOC-Leu(O)Leu-F2Cha-Leu-2-Picolin
Analog Beispiel 1 werden 115 mg F2Cha-Leu-2-Picolin, 78 mg t-BOC-Leu(O)Leu-OH, 60 mg Hydroxybenzotriazol und 47 mg N,N min -Dicyclohexylcarbodiimid umgesetzt. Das Rohprodukt wird mit Ether an Kieselgel chromatographiert. [ alpha ]D = +8.0 DEG (c = 0.2 in Methylenchlorid).
Beispiel 5: t-BOC-Cha(O)Phe-Cha(OH)Phe-NHBu
Analog Beispiel 1 werden 34 mg Cha(OH)Phe-NHBu, 40 mg t-BOC-Cha(O)Phe-OH, 25 mg Hydroxybenzotriazol und 20 mg N,N min -Dicyclohexylcarbodiimid umgesetzt. Das Rohprodukt wird mit Ether in Hexan (40 bis 100%) an Kieselgel chromatographiert. [ alpha ]D = +13.7 DEG (c = 0.1 in Methylenchlorid).
Beispiel 6: t-BOC-Cha(O)Bly-Cha(OH)Bly-NHBu
Analog Beispiel 1 werden 37 mg Cha(OH)Bly-NHBu, 40 mg t-BOC-Cha(O)Bly-OH, 17 mg Hydroxybenzotriazol und 22 mg N,N min -Dicyclohexylcarbodiimid umgesetzt. Das Rohprodukt wird mit Ether in Hexan (40-100%) chromatographiert. [ alpha ]D = -11.4 DEG (c = 0.1 in Methylenchlorid).
Beispiel 7: t-BOC-Cha(O)Nle-Cha(OH)Nle-NHBu
30 mg des Produktes aus Beispiel 6 werden in 2 ml Ethanol gelöst und 3 Stunden in Gegenwart von 3 mg Pd/C hydriert. [ alpha ]D = -13.0 DEG (c = 0.1 in Methylenchlorid).
Beispiel 8: t-BOC-Cha(OH)Bly-Cha(OH)Bly-NHBu
10 mg des Produktes aus Beispiel 6 werden in 0.5 ml Methanol gelöst und mit 5 mg Natriumborhydrid umgesetzt. Nach 1/2 Stunde wird mit Ether verdünnt, mit wässr. Weinsäure (2N)-Lösung gewaschen, die org. Phase getrocknet über Magnesiumsulfat und eingedampft. Man erhält das Produkt als Diastereomerengemisch ca. 2:1.
Beispiel 9: Z-Cha(O)Bly-Cha(OH)Bly-NHBu
30 mg des Produktes aus Beispiel 6 werden 1 Stunde in einem Gemisch von Trifluoressigsäure und Methylenchlorid 1:1 (0.5 ml) stehen gelassen, wobei die Schutzgruppe abgespalten wird. Dann wird mit Methylenchlorid verdünnt, mit wässriger Sodalösung (2N) extrahiert, die org. Phase mit Kaliumcarbonat getrocknet, eingedampft und mit 15 mg N-(Benzyloxycarbonyloxy)-succinimid in 0.5 ml Tetrahydrofuran bei 50 DEG umgesetzt. Das Rohprodukt wird in Ether gelöst, mit Wasser gewaschen, getrocknet und eingedampft. [ alpha ]D = -14.6 DEG (c = 0.05 in Methylenchlorid).
Beispiel 10: t-BOC-Leu(O)Leu-F2Cho-Leu-2-Picolin
Analog Beispiel 2 werden 65 mg Produkt aus Beispiel 4 mit 150 mg Collin's-Reagenz umgesetzt. [ alpha ]D der erhaltenen Titelverbindung = -16.2 DEG (c = 0.1 in Methylenchlorid).
Zwischenprodukte:

293 mg t-BOC-Cha(OH)Bly-OH werden in 7 ml Dimethylformamid gelöst und mit 1.54 g Collins Reagenz umgesetzt. Das Reaktionsgemisch wird zwischen Äther und wässriger Kaliumbisulfat-Lösung verteilt, die org. Phase mit Wasser gewaschen, über Magnesiumsulfat getrocknet und eingedampft.
Beispiel 11: BOC-Cha(O)Bly-Cha(OH)Bly-Leu- alpha -Picolin
Analog Beispiel 1 werden 66 mg H-Cha(OH)Bly-Leu- alpha -Picolin mit 52 mg BOC-Cha(O)Bly-OH, 20 mg HOBT und 30 mg DCC umgesetzt. Das Rohprodukt wird mit 3% Methanol in Methylenchlorid chromatographiert. [ alpha ]D = -26.0 DEG (c = 0.1 in CH2Cl2).
Beispiel 12: BOC-Cha(O)Bly-Cha(OH)Ala-NH-Bu
Analog Beispiel 1 werden 0.84 g H-Cha(OH)Ala-NHBu mit 1.09 g BOC Cha(O)Bly-OH, 0.5 g HOBT und 0.6 g DCC in Methylenchlorid umgesetzt. Das Rohprodukt wird mit Äther/Methylenchlorid (20-60%) an Kieselgel chromatographiert. [ alpha ]D = -16.9 DEG (c = 0.2 in CH2Cl2).
Die erfindungsgemässen Verbindungen weisen pharmakologische Aktivität auf. Sie können als Arzneimittel verwendet werden.
Wie Resultaten von Standard-Tests entnommen werden kann, weisen sie insbesondere für Enzymhemmer typische Wirkungen auf. Die hemmende Wirkung in bezug auf ein spezifisches Enzym hängt selbstverständlich von der Peptidstruktur gesamthaft ab. Die obigen, besonders als Hemmer der Reninaktivität geeigneten Verbindungen bewirken am humanen synthetischen Tetradekapeptidsubstrat bei einer Konzentration von 10-<5>M bis 10-<1><1>M eine 50%ige Hemmung der Enzymaktivität von reinem Humanrenin nach der Methode von F. Cumin et al. (Bioch. Biophys. Acta 913, 10-19 (1987) oder mittels Renin Binding Assay.
In der "antibody-trapping"-Methode von K. Poulsen und J. J rgensen (J. Clin. Endocrin. Metab. 39 [1974] 816-825) hemmen sie die Humanplasmareninaktivität bei einer Konzentration von 10-<5>M bis 10-<1><1>M.
Die erfindungsgemässen Verbindungen sind daher zur Verwendung für die Prophylaxe und Behandlung von Zuständen geeignet, die durch eine enzymatische Dysfunktion charakterisiert sind und für die eine Hemmung der enzymatischen Aktivität angezeigt ist.
Als Reninhemmer sind sie z.B. zur Verwendung bei der Prophylaxe und Behandlung der Hypertonie und der Herzinsuffizienz ("congestive heart failure") geeignet.
Bevorzugt für die Prophylaxe und Behandlung der Hypertonie und der Herzinsuffizienz sind die Titelverbindungen der Beispiele 1 und 4 bis 12, insbesondere der Beispiele 5, 6 und 8 bis 10, und ganz besonders der Beispiele 5, 6, 8 und 10.
Die Verbindungen gemäss der Erfindung zeigen ferner eine antiretrovirale Wirkung und können deshalb zur Behandlung von durch Retroviren, inbegriffen HTLV-I und -III-Viren, hervorgerufenen Krankheiten verwendet werden. Diese Wirkung zeigt sich im FeLV-Katzenmodell [Cerny und Essex, CPC Press IN 1979, Seiten 233-256; Cockerell et al. J. Natl. Cancer Inst. 57, Seiten 1095-1099 (1976); Cotter et al. J. AM.VET.MED.ASSOC. 166, Seiten 449-453 (1975); ESSEX et al. SCIENCE 190, 790-792 (1975)].- einem Modell zur Untersuchung der AIDS-Krankheit. Es wurde berichtet (beispielsweise am 25 ICAAC in Minneapolis vom 30. September bis 2. Oktober), dass bei Verabreichung von 30 mg 3 min -Azido-3 min -deoxy-thymidin während 14 Tagen eine Reduktion des FeLV-Titers um einen Faktor von 10 gezeigt werden konnte, jedoch keine Heilung eintrat.
Nach Verabreichung der Verbindungen gemäss der Erfindung konnte eine Vernichtung der Viren beobachtet werden.
Die zur Erreichung der antiviralen Wirkung benötigten Dosen entsprechen denjenigen die üblicherweise verwendet werden und liegen beispielsweise in der Grössenordnung von 5 bis 20 mg/kg/Tag.
Für obige Anwendungen hängt die zu verabreichende Dosis von der jeweils verwendeten Verbindung, der Art der Verabreichung und der gewünschten Behandlung ab. Im allgemeinen werden zufriedenstellende Resultate erhalten, falls die Verbindungen in einer täglichen Dosis von 0,02 mg/kg bis ca. 20 mg/kg Tierkörpergewicht verabreicht werden. Für grössere Säugetiere beträgt die empfohlene tägliche Dosis von etwa 1 mg bis etwa 500 mg, zweckmässigerweise verabreicht z.B. oral in Dosen von 0.25 mg bis ca. 500 mg 1-4 mal täglich oder in Retard-Form.
Die erfindungsgemässen Verbindungen können in freier Form oder, sofern saure oder basische Gruppen anwesend sind, in pharmakologisch verträglicher Salzform verabreicht werden. Solche Salzformen weisen eine Wirkung in derselben Grössenordnung wie die freien Formen auf und können auf bekannte Weise hergestellt werden. Die vorliegende Erfindung be trifft ebenfalls pharmazeutische Zubereitungen enthaltend eine erfindungsgemässe Verbindung in freier Form oder in pharmazeutisch verträglicher Salzform, gegebenenfalls zusammen mit pharmazeutisch verträglichen Hilfs- und/oder Trägerstoffen. Solche pharmazeutischen Zubereitungen können zur Verwendung bei enteraler, vorzugsweise oraler Verabreichung formuliert werden, z.B. als Tabletten, oder zur Verwendung bei parenteraler Verabreichung, z.B. als injizierbare Lösungen oder Suspensionen.

The compounds according to the invention have pharmacological activity. They can be used as a medicine.
As can be seen from the results of standard tests, they have typical effects, in particular for enzyme inhibitors. The inhibitory effect with regard to a specific enzyme naturally depends on the peptide structure as a whole. The compounds according to the invention are therefore suitable for use for the prophylaxis and treatment of conditions which are characterized by an enzymatic dysfunction and for which an inhibition of the enzymatic activity is indicated.
As renin inhibitors they are e.g. suitable for use in the prophylaxis and treatment of hypertension and heart failure ("congestive heart failure").
Preferred for the prophylaxis and treatment of hypertension and heart failure are the title compounds of Examples 1 and 4 to 12, in particular Examples 5, 6 and 8 to 10, and very particularly Examples 5, 6, 8 and 10.
The compounds according to the invention also show an antiretroviral activity and can therefore be used for the treatment of diseases caused by retroviruses, including HTLV-I and III viruses.
