Description
The invention relates to a process for the preparation of ß-chloroalanine. This process is characterized in that a bisulfite or sulfite addition compound of chloroacetaldehyde in aqueous solution is first reacted with ammonia and then with hydrocyanic acid or its salt to give α-amino-ß-chloropropionitrile and the latter is hydrolyzed under acidic conditions.
ß-Chloralanin ist eine Aminosäure mit physiologischer Wirksamkeit (J. Biol. Chem. 252,3170). ß-Chloralanine is an amino acid with physiological activity (J. Biol. Chem. 252,3170). In addition, ß-chloralanine is an intermediate for the production of, for example, various pharmaceutically active compounds and pesticides, such as antibiotics (e.g. cycloserine) and sulfur-containing amino acids (e.g. cystine) or cysteine.
However, since no economically advantageous industrial process for the production of β-chloro alanine was known, β-chloro alanine was rarely used as an intermediate for the production of pharmaceutically active compounds and pesticides. For example, the hydroxyl group of serine is replaced by chlorine (J. Chem. Soc. Pages 1968, 1969) or chlorinated cystine (Ber. 93, page 782, 1960) by known processes for the production of β-chloroalanine. However, since expensive starting materials are used in these processes, they are unsuitable for the large-scale production of β-chloroalanine.
A so-called Strecker synthesis for the large-scale production of amino acids is also known. However, after the Strecker synthesis, the corresponding aldehyde, i.e. Chloroacetaldehyde, no ß-chloroalanine obtained. That is, by the reaction of chloroacetaldehyde with hydrocyanic acid and ammonia, β-chlorolactonitrile is obtained by adding a cyano group to chloroacetaldehyde. Since the amination of the hydroxyl group of β-chloro-lactonitrile formed with ammonia is very difficult, the intermediate a-amino-β-chloropropionitrile required for the production of β-chloroalanine cannot be obtained. Even if chloroacetaldehyde is first reacted with ammonia to a-amino-ß-chloroethanol and then with hydrocyanic acid or its salts, no a-amino
ß-chloropropionitrile are formed, but by replacing the amino group of a-amino-ß-chloroethanol with a cyano group, ß-chloro-lactonitrile is obtained. When ß-chloro-lactonitrile is reacted with ammonia under extreme reaction conditions to obtain a-amino-ß-chloropropionitrile, the ß-chloro-lactonitrile is decomposed.
The aim of the present invention is therefore to counter the problems of the prior art described above and to provide a process of great selectivity for the preparation of β-chloroalanine from chloroacetaldehyde.
Further objects and advantages of the invention are shown in the description below.
The invention therefore relates to a process for the preparation of β-chloroalanine, which is characterized in that a) a bisulfite or sulfite addition compound of chloroacetaldehyde in aqueous solution with ammonia and then with hydrocyanic acid or its salt to give a-amino-β-chloropropioni tril and b) hydrolysing the a-amino-ß-chloropropionitrile obtained under acidic conditions.
In den Figuren werden dargestellt: The invention is illustrated below using the drawings. The figures show:

The bisulfite addition compound of chloroacetaldehyde can easily be obtained by reacting approximately equimolar amounts of chloroacetaldehyde with bisulfite in an aqueous solution at room temperature. Any water-soluble bisulfite can be used to form the bisulfite addition compound of chloroacetaldehyde. Examples of typical bisulfites are: alkali metal salts such as lithium, sodium, potassium and the like; Alkaline earth metal salts such as calcium, magnesium and the like and an ammonium salt. In particular, ammonium bisulfite is preferred.
Entsprechend sollte, wenn Bisulfït mit Chloracetaldehyd unter alkalischen Bedingungen umgesetzt Although the amounts of bisulfites and chloroacetaldehyde used for the preparation of the bisulfite addition compounds can be varied within a wide range, bisulfite and chloroacetaldehyde are preferably used in a molar ratio of 1: 0.8 to 1.2. Although a further excess of one of the two compounds can be used, such an excess not only does not give an advantageous result, but is also uneconomical. In addition, there is a fear that the use of an excess of one of the two compounds causes undesirable by-products to be formed. Instead of bisulfites, ammonium sulfite can be used for the preparation of the sulfite addition compound with chloroacetaldehyde. The use of other sulfites, for example alkali metal sulfites and alkaline earth metal sulfites, is unsuitable for the formation of the sulfite addition compound with chloroacetaldehyde, since the sulfite addition compound formed does not give the desired α-aminochloroethanesulfonates in good yield in the next reaction step. Accordingly, when bisulfite is reacted with chloroacetaldehyde under alkaline conditions

Then the bisulfite or sulfite addition compound of chloroacetaldehyde is reacted with ammonia in an aqueous medium. Ammonia can be used in gaseous, liquid or dissolved form in water. Although the amount of ammonia used may vary depending on the type of bisulfite and sulfite and the form of ammonia used, the molar ratio of ammonia to chloroacetaldehyde is preferably 1 to 5, especially 2 to 4. Although there is no critical reaction temperature or time, the reaction is generally carried out in the temperature range from 0 to 50 ° C., preferably 20 to 30 ° C., for 0.5 to 2 hours. This gives α-amino-β-chloroethanesulfonates ClCH2CH (NH2) SOsM, where M = NH4, Na, K, Vi Ca, Vi Mg etc. These a-amino-ß-chloroethanesulfonates and the a-amino-ß-chloroethanesulfonic acid are new compounds. The a-amino-ß-chloroethane sulfonic acid or its salts can be obtained from the aqueous solution containing them by neutralizing the excess ammonia with a mineral acid, e.g. Sulfuric acid or hydrochloric acid, and lowering the pH of the solution to an isoelectric point. The mass spectrum and the IR spectrum of the α-amino-β-chloroethanesulfonic acid C1CH2CH (NH2) S03H are shown in FIGS. 1 and 2.
The elementary analysis of the a-amino-ß-chloroethanesulfonic acid thus obtained is as follows:
n-Hexan, n-Pentan, Cyclohexan, Diäthyläther, Diisopropyl-äther, Benzol, Toluol, Xylol, Chlorbenzol, Äthylbenzol und dergleichen. Als Lösungsmittel werden bevorzugt Äthylace-tat, Äthylenglykolmonoäthylätheracetat, Propionitril oder 5 Propylencarbonat. n-hexane, n-pentane, cyclohexane, diethyl ether, diisopropyl ether, benzene, toluene, xylene, chlorobenzene, ethylbenzene and the like. Preferred solvents are ethyl acetate, ethylene glycol monoethyl ether acetate, propionitrile or propylene carbonate.
Preferably hydrocyanic acid or its salts, e.g. Sodium cyanide and potassium cyanide, used in this reaction in stoichiometric or larger amounts. In particular, 2.0 to 5.0 10 mol of hydrocyanic acid or its salts are used per mole of a-amino-ß-chloroethanesulfonate. Although there is no critical reaction temperature or reaction time, the reaction is preferably carried out at a temperature of 0 to 30 ° C, especially 5 to 10 ° C, for 5 to 24 hours.
The a-amino-ß-chloropropionitrile obtained and its salts are also new compounds. Since the a-amino-ß-chloropropionitrile obtained is unstable, it is obtained as hydrochloride by passing dry gaseous hydrochloric acid into the organic solution containing the product. The a-amino-ß-chloropropionitrile hydrochloride is precipitated from the organic solution in crystalline form and separated off in a known manner.
The mass spectrum of the α-amino-β-chloropropionitrile obtained is shown in FIG. 3. The IR spectrum of a-amino-ß-chloropropionitrile hydrochloride is shown in FIG. 4 and the NMR spectrum of a-amino-ß-chloropropionitrile in FIG. 5.

The a-amino-ß-chloroethanesulfonate obtained by reacting the bisulfite or sulfite addition compound of chloroacetaldehyde with ammonia is reacted with hydrocyanic acid or its salts, e.g. Alkali metal, alkaline earth metal salts or ammonium salt converted to a-amino-ß-chloropropionitrile. Since a-amino-ß-chloropropionitrile is a very volatile substance, especially in aqueous solution, special precautionary measures must be taken during the course of the reaction.
For example, the reaction is carried out in the presence of a suitable organic solvent which is immiscible with water, so that the a-amino-β-chloropropionitrile formed is rapidly converted into the organic phase. Once a-amino-ß-chloroethanesulfonate is separated from the aqueous reaction mixture, the separated a-amino-ß-chloroethanesulfonate can of course be reacted with hydrocyanic acid or its salts in a suitable organic solvent. The reaction is preferably carried out in a device in which the reactor is connected to an extractor and a decanter, and the α-amino-β-chloropropionitrile obtained is extracted continuously from the aqueous reaction mixture with a suitable organic solvent.
Beispiele für solche organischen Lösungsmittel sind Tetrachlorkohlenstoff, Chloroform, Methylenchlorid, Trichloräthylen, The organic solvents used for this reaction should be stable under the reaction conditions, they should not react or decompose with the reagents or the product, they should not easily dissolve the reagents, but the product should be readily soluble in them and they should not react with hydrochloric acid. Examples of such organic solvents are carbon tetrachloride, chloroform, methylene chloride, trichlorethylene,

According to the invention, the a-amino-ß-chloropropionitrile obtained in an organic solution is hydrolyzed by the addition of a mineral acid, such as hydrochloric acid, sulfuric acid, nitric acid or the like, after the aqueous phase which may be present has been separated off from the organic phase. The organic solvent is evaporated by heating the solution and the residue is hydrolyzed under acidic conditions at a temperature of, for example, 80 to 100 ° C. for 0.5 to 2 hours. As a result, ß-chloralanine is formed in a solution containing aqueous mineral salt.
The ß-chloralanine can be separated as follows, for example:
The aqueous solution containing ß-chloroalanine is first concentrated, then alcohols such as methanol, ethanol and the like are added. As a result, the ammonium salt of the mineral acids is crystallized out of the solution. The alcoholic solution obtained is separated from the crystallized ammonium salt and a suitable solvent such as ether is added to the separated alcoholic solution to crystallize the mineral acid salt of β-chloroalanine. The crystals are cleaned 60 by known methods, for example recrystallization. If desired, ß-chloralanine can be obtained as a free amino acid by neutralizing the alcoholic solution of the mineral acid salt of ß-chloro alanine with an equimolar amount of, for example, pyridine and triethylamine.

According to the invention, ß-chloroalanine can be obtained with a very high selectivity without the formation of substantial amounts of ß-chloro lactic acid.

It has further been found that if carbon dioxide is added to its solution before the acidic hydrolysis of the a-amino-ß-chloropropionitrile, this compound can be stabilized and ß-chloroalanine of high purity can be obtained in high yield by the hydrolysis. Carbon dioxide can be added either as a gas or in solid form. Although the amount of carbon dioxide added to the aqueous a-amino-β-chloropropionitrile solution is not critical, so much carbon dioxide is generally added that the pH of the aqueous solution is 7 to 6 or that 0.5 to 1.0 mole of carbon dioxide can be absorbed per mole of a-amino-ß-chloropropionitrile. In order to accelerate the absorption of the carbon dioxide, the aqueous solution containing a-amino-ß-chloropropionitrile can be cooled to a temperature of 10 ° C or less. Since the absorption rate of the carbon dioxide increases with the increase in the aminonitrile concentration, the aminonitrile concentration is preferably set to 20 to 60%, in particular 40 to 50%.
The invention is illustrated by the following examples, in which all percentages, unless stated otherwise, are to be understood as% by weight.

32 g of the sodium bisulfite addition compound of chloroacetaldehyde was dissolved in a mixture of 50 ml of 28% concentrated ammonia water and 50 ml of water, and the resulting mixture was allowed to react at room temperature for 1 hour. 100 ml of ethyl ether were then added to the reaction mixture. Thereafter, 10 g of sodium cyanide were dissolved in 30 ml of water and added dropwise over 15 minutes to the reaction mixture, which was kept at a temperature of -10 ° C. during this time. Then the mixture was allowed to react at -5 to + 5 ° C for 6 hours with vigorous stirring.
After the reaction was completed, the mixture was allowed to stand, separating the ether solution from the aqueous solution. The former was removed and the aqueous solution was extracted with 100 ml of fresh ether. The two ether solutions were combined and 50 ml of concentrated hydrochloric acid were slowly added. The ether was distilled off and the resulting aqueous solution was warmed and concentrated on a boiling water bath for 1 hour. 30 ml of ethanol were added to the concentrated aqueous solution, whereby ammonium chloride was precipitated. 100 ml of ether were added to the separated ethanol solution, as a result of which 14 g of β-chloroalanine hydrochloride were precipitated. This means a molar yield of ß-chloralanine of 50%.

The mixture was left at room temperature for 2 hours. 60 ml of 28% ammonia water was added to the mixture and the mixture was allowed to react at room temperature for 1 hour.
The reaction mixture obtained was treated as described in Example 1, with the difference that in the last step, instead of ethyl ether, triethylamine was added to the alcoholic solution of β-chloroalanine hydrochloride, as a result of which the crystals of β-chloroalanine were precipitated by neutralization. The yield of β-chloroalanine was 22 g.
Beispiel 4 Example 4
54 g of ammonium bisulfite addition compound of chloroacetaldehyde were dissolved in 50 ml of water and mixed with 60 ml of ammonia water. The mixture was allowed to react at room temperature for 1 hour. Thereafter, 100 ml of ethyl ether was added to the reaction mixture, and 50 ml of an aqueous solution containing 14.7 g of sodium cyanide was added dropwise over 15 minutes while the temperature of the mixture was kept at 10 ° C or less. The reaction was continued for 15 hours at 10 ° C with vigorous stirring.
After the completion of the reaction, the separated ether solution was removed from the aqueous solution and the aqueous solution was extracted with 100 ml of fresh ether. The two ether solutions were combined and the combined solution was dried over anhydrous magnesium sulfate.
After the desiccant had been removed, dry hydrochloric acid was introduced into the ether solution until saturated with simultaneous cooling with ice water. As a result, a-amino-ß-chloropropionitrile hydrochloride was precipitated. The crystals of the precipitate were separated from the ether and dissolved in 30 ml of concentrated hydrochloric acid solution. The solution was warmed and concentrated on the boiling water bath for 1 hour. 50 ml of ethanol were added to the concentrate and the insoluble ammonium chloride was separated off. 150 ml of ethyl ether were added to the ethanolic solution. This gave 30 g of crystalline β-chloroalanine hydrochloride.

7 g of chloroacetaldehyde hemihydrate crystals were completely dissolved in 16 g of a 50% aqueous ammonium bisulfite solution. 25 ml of a 30% aqueous ammonia solution were added to this solution. Thereafter, the mixture was allowed to react at room temperature for 2 hours while introducing ammonia gas to saturation. After the amination reaction was completed, the mixture was cooled to a temperature of 10 ° C or less, 15 ml of an 80% aqueous hydrocyanic acid solution was added dropwise while simultaneously introducing gaseous ammonia. After dropwise addition of the hydrocyanic acid, the mixture was allowed to react at a temperature of 10 ° C or less for 3 hours. Then the reaction mixture was concentrated on a water bath at 40 ° C under reduced pressure of 4 to 6 mmHg for 5 minutes.
Etwa 1 g des Kohlen4 The weight of the reaction mixture was reduced by about 15 g and the pH changed to 7-8. Unreacted ammonia and unreacted hydrocyanic acid were essentially evaporated. While the mixture was cooling, gaseous carbon dioxide was introduced for 30 minutes with vigorous stirring. About 1 g of coal4

dioxyds wurden im Reaktionsgemisch absorbiert, wodurch ein pH von 6 bis 7 erzielt wurde. dioxides were absorbed in the reaction mixture, whereby a pH of 6 to 7 was achieved.
30 ml of concentrated hydrochloric acid solution was slowly added to the reaction mixture such that its temperature was not raised above 20 ° C. Over time, a white precipitate formed, which was removed by filtration. The filtrate was evaporated to dryness under reduced pressure. 40 ml of concentrated hydrochloric acid solution were added to the residue and hydrolyzed at 95 to 100 ° C. for 1 hour. After hydrolysis, the mixture was evaporated to dryness under reduced pressure. The β-chloralanine hydrochloride obtained was extracted with 100 ml of absolute ethanol and the insoluble ammonium chloride was removed. The ethanol was distilled off, whereupon 8.32 g of crystalline β-chloroalanine hydrochloride were obtained. The yield is 65% of theory.
This procedure was carried out again, with the difference that no carbon dioxide was introduced. 5 g of β-chloroalanine hydrochloride were obtained in the form of a colored residue, which corresponds to 39% of theory.
Beispiel 6 Example 6
As described in Example 5, β-chloroalanine was prepared from 7 g of chloroacetaldehyde hemihydrate crystals, with the difference that 30 ml of concentrated hydrochloric acid solution were not added after the absorption of gaseous carbon dioxide, but gaseous hydrochloric acid was slowly absorbed. After 20 g of hydrochloric acid was absorbed, the precipitated inorganic salt was removed by filtration and the filtrate was evaporated to dryness.
The a-amino-ß-chloropropionitrile hydrochloride obtained was hydrolyzed as described in Example 5. 8 g of β-chloroalanine hydrochloride were obtained in crystalline form, which corresponds to 62.5% of theory.
Beispiel 7 Example 7
26.3 g (0.3 mol) of chloroacetaldehyde hemihydrate crystals were completely dissolved in 62 g (0.31 mol) of a 50% aqueous ammonium bisulfite solution. This aqueous solution was placed in a reaction vessel provided with a perforated plate and connected to a decanter. To this aqueous solution, 100 ml of 30% aqueous ammonia solution was added and ammonia gas was introduced to saturate the solution. The aqueous solution was allowed to react at room temperature for 2 hours. The reaction vessel was cooled to 10 ° C.
Die Methylenchloridschicht wurde unterhalb der perforierten Platte, welche im unteren Teil des Reaktionsgefässes befestigt war, The temperature of the reaction mixture was kept at 10 ° C. and 19.7 ml (0.6 mol) of a 50% hydrocyanic acid solution were added while introducing gaseous ammonia. After adding the hydrocyanic acid, methylene chloride was added with vigorous stirring. The methylene chloride layer was placed below the perforated plate, which was attached in the lower part of the reaction vessel,

In the decanter, the aqueous layer was separated from the methylene chloride layer and returned to the reaction vessel. The separated methylene chloride solution was removed from the decanter and collected in a container.
In the manner described above, the extraction was continued for 3 hours while adding methylene chloride to the reaction mixture at a rate of 5 ml per minute, i.e., 900 ml of methylene chloride was added. The temperature of the reaction mixture was kept at 10 ° C. during the extraction reaction.
The methylene chloride collected in the container was distilled off at a temperature of 20 ° to 30 ° C. under reduced pressure, and the liquid a-amino-β-chloropropionitrile obtained was dissolved in another vessel in 40 ml of distilled water. Gaseous carbon dioxide was introduced over 30 minutes with vigorous stirring. After the introduction of the carbon dioxide, the pH of the mixture was 7. For this reason, 100 ml of concentrated hydrochloric acid solution were added and hydrolyzed at a temperature of 95 ° C. for 1 hour. After the hydrolysis, the reaction mixture was evaporated to dryness. The residue was extracted with 100 ml of absolute ethanol and the insoluble ammonium chloride was removed. The ethanol was distilled off. 31.2 g of β-chloroalanine hydrochloride were obtained, which corresponds to 65% of theory.
This procedure was repeated with the difference that a-amino-ß-chloropropionitrile was not neutralized with gaseous carbon dioxide. 28 g of β-chloroalanine hydrochloride contaminated with brown tar-like substances were obtained. This corresponds to 59.5% of theory.
