2. Verfahren zur Herstellung von Dihydro-cyclosporin G nach Anspruch 1, dadurch gekennzeichnet, dass man das durch Züchtung des Stammes NRRL 8044 der Pilzspecies Tolypocladium infiatum Gams herstellbare Cyclosporin G hydriert.
3. Heilmittel, dadurch gekennzeichnet, dass es Dihydrocyclosporin G nach Anspruch 1 enthält.
Die vorliegende Erfindung betrifft die Verbindung Dihydro-cyclosporin G.
Erfindungsgemäss gelangt man zum Dihydro-cyclosporin G durch Hydrierung des Cyclosporin G.

Der Molekularpeak von Dihydro-cyclosporin G ergibt sich aus dem Felddesorptions-Massenspektrum zu 1217 (Cf,3Hi 5ei 1012).
Im Dünnschichtchromatogramm auf Kieselgel-Fertigplatten Merck , Schichtdicke 0,25 mm, werden mit dem Fliessmittel Chloroform-Methanol (96:4), Laufstrecke 10 cm, folgende Rr-Werte beobachtet: Dihydro-cyclosporin G 0,45 Cyclosporin G 0,45
Die Detektion erfolgt mit Joddampf (wobei Cyclosporin G eine intensive, Dihydro-cyclosporin G hingegen eine schwache Anfärbung zeigt) oder mit Dragendorff-Sprühreagenz nach Munier.
Löslichkeit: Dihydro-cyclosporin G ist leicht löslich in Methanol, Äthanol, Aceton, chlorierten Kohlenwasserstoffen; mässig löslich in Äther und Petroläther; praktisch unlöslich in Wasser.
Das erfindungsgemässe Verfahren lässt sich nach an sich bekannten Methoden ausführen, z.B. durch katalytische Hydrierung.
Als Lösungsmittel kommen vorzugsweise Äthylacetat oder niedere aliphatische Alkohole, wie z.B. Methanol, Äthanol, Isopropanol, in Frage. Die Hydrierung erfolgt zweckmässigerweise im neutralen Bereich bei Temperaturen zwischen 20 und 30 und bei Atmosphärendruck oder wenig erhöhtem Druck. Als Katalysator kommt Platin, vorzugsweise Palladium, z.B. Palladium auf Kohle, in Frage. Das so erhaltene Hydrierungsprodukt von Cyclosporin G wird hierauf in an sich bekannter Weise gereinigt, z.B. durch Chromatographie.
Das als Ausgangsprodukt benützte Cyclosporin G kann erhalten werden, indem man einen Cyclosporin G produzierenden Stamm der Pilzspecies Tolypocladium inflatum Gams in Gegenwart eines Nährmediums züchtet und Cyclosporin G aus der Fermentationsbrühe auf an sich bekannter Weise durch extraktive und/oder adsorptive Arbeitsmethoden isoliert und hierauf chromatographisch oder mittels Gegenstromverteilung reinigt.
Ein bevorzugter Cyclosporin G produzierender Stamm ist der frei zugängliche Stamm NRRL 8044 der Pilzspecies Tolypocladium inflatum Gams. Eine Kultur davon wurde beim United States Department of Agriculture (Northern Research and Development Division), Peoria, I11., USA deponiert.
Dieser Stamm wurde vormals def Pilzspecies Trichoderma polysporum (Link ex. Pers.) zugeordnet und ist z.B. in der DOS 2,455,859 beschrieben.
Dihydro-cyclosporin G zeichnet sich durch interessante pharmakologische Eigenschaften aus und kann daher als Heilmittel verwendet werden.
Insbesondere zeichnet sich die Substanz durch eine antiarthritische Wirkung aus. So bewirkt sie im Freund-Adjuvans Arthritis-Latenzzeitversuch an der Ratte in Dosen von ca.
30 mg/kg p.o. Körpergewicht/Tag eine starke Hemmung der primären und sekundären Entzündung.
Die zu verwendenden Dosen variieren naturgemäss je nach Art der Administration und des zu behandelnden Zustandes.
Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von 10 bis 300 mg/kg lSörperge- wicht erzielt. Diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 300 bis 900 mg. Für orale Applikationen können die Teildosen beispielsweise etwa 150 bis 300 mg Dihydro-cyclosporin G neben festen und flüssigen Trägersubstanzen enthalten.
Als Heilmittel kann Dihydro-cyclosporin G allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
In dem nachfolgenden Beispiel, das die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel
700 mg Palladium-Kohle (10% Palladium) werden in 20 ml Äthanol während 30 Minuten vorhydriert. Zu dieser Suspension des Palladiumkatalysators wird die Lösung von 4,55 g Cyclosporin ci in 60 ml Äthanol zugegeben und darauf bei 21 und einem Druck von 742 mm Quecksilbersäule bis zur beendeten Wasserstoffaufnahme hydriert. Anschliessend filtriert man vom Katalysator ab und dampft das Filtrat im
Vakuum bei 20 bis 40 zur Trockene ein. Der Rückstand besteht aus dünnschichtchromatographisch einheitlichem Dihydro-cyclosporin G in Form eines weissen amorphen Pulvers vom Smp. 150-153 (nach Trocknen im Hochvakuum während 4 Stunden bei 80 ).
Das als Ausgangsmaterial verwendete Cyclosporin G wird wie folgt hergestellt:
500 Liter einer Nährlösung, die pro Liter 40 g Glucose, 2,0 g Natriumcaseinat, 2,5 g Ammoniumphosphat, 5 g MgSO 7H2O, 2 g KH2PO4, 3 g NaNOs, 0,5 g KCI, 0,01 g FeSO4 und entmineralisiertes Wasser enthält, werden mit 50 Liter einer Vorkultur des Stammes NRRL 8044 angeimpft und in einem Stahlfermenter unter Rühren (170 UPM) und Belüftung (1 Liter Luft/Min./Liter Nährlösung) 13 Tage bei 27 inkubiert (siehe DOS 2,455,859).
Die Kulturbrühe wird mit der gleichen Menge n-Butylacetat ausgerührt, nach Abtrennung der organischen Phase wird diese im Vakuum konzentriert und der Rohextrakt durch 3-stufige Verteilung zwischen Methanol-Wasser (9:1) und Petroläther entfettet. Die methanolische Phase wird abgetrennt, im Vakuum konzentriert und das Rohprodukt durch Zugabe von Wasser ausgefällt. Das nach der Filtration gewonnene Material wird an der 5- bis 7-fachen Menge Sephadex LH-20 mit Methanol als Elutionsmittel chromatographiert. Die Spitzenfraktionen werden anschliessend an Kieselgel 60, Korngrösse 0,063-0,2 mm (Merck) mit Hexan Aceton (2:1) chromatographiert, wobei die zuerst eluierten Fraktionen vorwiegend Cyclosporin A und Cyclosporin D enthalten, die später eluierten Anteile vorwiegend Cyclosporin C.
Zur weiteren Reinigung werden die Cyclosporin Aund D-haltigen Fraktionen aus der 2- bis 2,5-fachen Menge Aceton bei - 15 kristallisiert und anschliessend durch zweimalige Chromatographie an Kieselgel 60, Korngrösse 0,0630,2 mm (Merck) weiter aufgetrennt, wobei die mit Hexan Aceton (2:1) zuerst eluierten Fraktionen Cyclosporin D in stark angereicherter Form enthalten. Diese werden in der doppelten Menge Aceton gelöst und bei - 15 kristallisieren lassen. Das dabei erhaltene Rohkristallisat besteht aus sehr stark angereichertem Cyclosporin D, die Mutterlauge enthält neben Cyclosporin D noch weitere Komponenten, so Cyclosporin G.
Zur Gewinnung von Cyclosporin G wird die zur Trockne verdampfte Mutterlauge an Kieselgel 60, Korngrösse 0,063-0,2 mm (Merck) mit wassergesättigtem Essigeester chromatographiert, wobei die zuerst eluierten Fraktionen Cyclosporin D enthalten, und die später eluierten Fraktionen Cyclosporin G im Gemisch neben weiteren Komponenten.
Zur weiteren Anreicherung wird das Gemisch zuerst an Kieselgel 60, Korngrösse 0,063-0,2 mm (Merck) mit Chloroform-Methanol (98:2) und anschliessend zur Auftrennung mit Hexan-Aceton (2:1) chromatographiert; dabei enthalten die zuerst eluierten Fraktionen Cyclosporin G in sehr stark angereicherter Form. Zur Reingewinnung werden die Cyclo sporin-G-Anteile zweimal aus Äther-Petroläther (1:1) bei Raumtemperatur kristallisiert; sie liefern dünnschichtchromatographisch-einheitliches, reines Cyclosporin G als farblose Polyeder vom Smp. 193-194" (nach Trocknen der Kristalle im Hochvakuum bei 80" während 2 Stunden).

The crude crystallizate obtained in this way consists of very strongly enriched cyclosporin D, the mother liquor contains, in addition to cyclosporin D, further components, so cyclosporin G.
