Die Erfindung betrifft Verfahren zur Herstellung von
neuen 3-Azidomethyl-2,9-dioxatricyclo-[4,3,1,03,7]-decane der allgemeinen Formel I

in welcher einer der beiden Reste Rund R2 Wasserstoff und der andere Hydroxy, Acyloxy oder Carbamoyloxy oder beide Reste Rl und R2 zusammen Sauerstoff und der Rest R3 Alkoxy oder Aralkoxy bedeuten.
In einer Reihe älterer Anmeldungen sind 2,9- Di ox atricyclo[4,3, 1 ,037]-decane beschrieben, die sich durch vorteilhafte pharmakologische Wirkungen, insbesondere auf das zentrale Nervensystem auszeichnen.
Darunter sind gemäss der deutschen Offenlegungsschrift 25 47 205 auch solche, die analgetische bzw. sedierende Eigenschaften haben.
Da für viele Indikationen Wirksubstanzen erwünscht sind, die neben analgetischen auch antipyretische und entzündungshemmende Eigenschaften haben, liegt der Erfindung die Aufgabe zugrunde, Verbindungen der Körperklasse der 2,9-Dioxatricycio[4,3, 1,037]-decane zu schaffen, die neben der analgetischen auch antipyretische und antiphlogistische Wirkungen zeigen.
Überraschenderweise wurde dabei gefunden, dass durch Einführung der Azidofunktion in die Position C-3' der bekannten 2,9-Dioxatricyclo[4,3, 1 ,03,7]-decane Verbindungen des gesuchten Wirkungsprofils erhalten werden.
Die neuen Verbindungen sind den auf dem Markt befindlichen Präparaten mit vergleichbarem Wirkungsprofil überlegen. Die Vergleichswerte ergeben sich aus der nachfolgenden Tabelle:
Die Toxizität der neuen Substanzen ist extrem günstig und liegt, i.p. an der Maus bestimmt, in der Grössenordnung von 500 bis 700 mg/kg. Beispielsweise sind für die nachstehenden Verbindungen folgende Werte ermittelt worden.

Zur Charakterisierung der analgetischen Wirkung wurde das PBC-Syndrom (Phenylbenzochinon-Syndrom; Writhing Test: Wertung eines Analgetikums durch seine Wirkung gegen einen mittels Phenylbenzochinon chemisch induzierten Schmerzes [E. Siegmund, R. Cadmus, G. Lu, Proc. Soc. exp.
Biol. Med. 95, 729(1957)]) an der Maus verwendet. Die antiphglogistischen/analgetischen Eigenschaften wurden durch den Randall-Selitto-Test [tierexperimentelle Wertungen von Analgetika anhand der Druckschmerzschwelle an der Rattenpfote [Real-Lexikon der Medizin, Band 5, Urban u. Schwarzenberg, 1973)] an der Ratte bewiesen.
Wie aus der Tabelle zu ersehen ist, sind die ED 50-Werte bis zu einer IOer-Potenz günstiger als bei den Marktpräparaten.

in welcher Hal Chlor, Brom oder Jod, vorzugsweise Jod, R2 Acetoxy und R3 Alkoxy oder Aralkoxy, vorzugsweise Methoxy, Äthoxy oder Butoxy bedeuten, (a) die 10,11 Doppelbindung zur 10ss-ständigen Methyl- gruppe hydriert, (b) die 3-Halogenmethylgruppe mit einem Metallazid in die 3-Azidomethylgruppe umwandelt und (c) die 4ss-Acyloxygruppe zur 4-Hydroxygruppe hydrolysiert.
In der derart erhaltenen Verbindung der Formel I kann die 4ss-Hydroxygruppe in eine 4ss-Acyloxy- oder 4ss-Carbamoy- loxygruppe umgewandelt oder die 4ss-Hydroxygruppe zum 4-on oxidiert und das 4-on selektiv zur 4a-Hydroxygruppe reduziert werden, worauf gewünschtenfalls die 4a-Hydroxygruppe in eine 4a-Acyloxy- oder 4a-Carbamoyloxygruppe umgewandelt werden kann.
Die Ausgangsverbindungen sind nach dem in der deutschen Offenlegungsschrift 21 29 507 beschriebenen Verfahren herstellbar.
Die Hydrierung der 10,11 -Doppelbindung gelingt normalerweise mittels Wasserstoff in Gegenwart von Platinoxid in einem Lösungsmittel, vorzugsweise Essigester oder Äthanol, ohne dass dabei die Halogenfunktion in C-3'-Position beeinträchtigt wird. Es hat sich dabei als vorteilhaft erwiesen, von den 3-Jodmethyl-4ss-Acetoxy-Verbindungen auszugehen, weil von den nach der Hydrierung erhaltenen epimeren 1 0-Methyl-Verbindungen die 10ss-Methyl-3jod- methyl-4ss-Acetoxy-Verbindungen in Methanol besonders gut kristallisieren und sich so leicht von den zu etwa 10% entstehenden 1 Oa-Verbindungen abtrennen lassen.
Die Umwandlung der 3-Halogenmethylgruppe in die 3-Azidomethylgruppe gelingt z.B. mittels Alkaliaziden, vorzugsweise Natriumazid, bevorzugt in aprotischen polaren Lösungsmitteln, vorzugsweise Hexamethylphosphorsäuretriamid oder Dimethylformamid, insbesondere bei Temperaturen bis etwa 200 C, vorzugsweise bei 80 C bis 150"C.
Die Umwandlung der aus den Ausgangsverbindungen stammenden 4ss-Acetoxygruppe in die 4ss-Hydroxygruppe bzw.4ss-Acyloxygruppe (falls Acyloxy nicht Acetoxy sein soll) bzw. 4P-Carbamoyioxygruppe ist unproblematisch und gelingt mittels konventioneller Methoden.
Die Oxydation der 4ss-Hydroxygruppe zum Decanon erfolgt z.B. mittels einem Chromtrioxid-Oxyxdationsreagenz in Äther oder Aceton und ist an sich in der deutschen Offenlegungsschrift 25 47 205 schon beschrieben.
Die selektive Reduktion des Decanons zur 4a-Hydroxygruppe, ohne dabei die Azidofunktion in Mitleidenschaft zu ziehen, gelingt insbesondere mittels komplexer Metallhydride, wie das ebenfalls in der deutschen Offenlegungsschrift 25 47 205 beschrieben ist. Als besonders günstig hat sich dabei Lithiumborhydrid erwiesen.
Für die Umwandlung der 4a-Hydroxygruppe in Ester bzw.
Carbamate gilt das für die 4ss-Hydroxygruppe Gesagte ent sprechend.
In der nachfolgenden Beschreibung der Ausführungsbeispiele ist das 2,9-Dioxatricyclo[4,3,1,03,7]-decan -Gerüst mit 2,9-DTD abgekürzt.
Soweit bei einzelnen Verbindungen als Schmelzpunkt der Wert < 00C angegeben ist, bedeutet das, dass es sich um bei Zimmertemperatur ölige Substanzen handelt.
Beispiel
1. Herstellung der Ausgangsverbindungen
Je nachdem, in welchem Alkohol als Lösungsmittel und mit welcher Halogenwasserstoffsäure man arbeitet, lassen sich gemäss dem in der deutschen Offenlegungsschrift 21 29 507 beschriebenen Verfahren die gewünschten Ausgangsverbindungen erhalten. So wurden aus je 850 g etwa 70% Didrovaltratum enthaltendem Extrakt aus Valeriana wallichii DC, gelöst in Methanol, Äthanol bzw.
Butanol, mittels Jodwasserstoffsäure, gelöst in demselben Alkohol, wobei sich bei den höheren Alkoholen nach der Zugabe der Jodwasserstoffsäure ein 2stündiges Stehenlassen bei etwa 80 C als vorteilhaft erwiesen hat, erhalten: 1.1. 3-Jodmethyl-4ss-acetoxy-8-methoxy-10-methylen- 2,9-DTD (III)

2. Hydrierung der 10,11 -Doppelbindung 2.1 3-Jodmethyl-4ss-acetoxy-8-methoxy- 1 Oss-methyl- 2,9-DTD (VI) Eine Lösung von 800 g 3-Jodomethyl-4ss-acetoxy-8-methoxy10-methylen-2,9-dioxatricyclo[4,3,1,03,7]decan (III, Beispiel 1.1) in 3 1 Essigester wurde zu einer Suspension von 35 g vorhydriertem Platinoxid in 300 ml Essigester gegeben. Es wurde bei Raumtemperatur und Normaldruck hydriert. Die Wasserstoffaufnahme war anfangs sehr schnell, gegen Ende verlief sie jedoch sehr langsam. Nach Aufnahme der theoretischen Menge an Wasserstoff (47,21) wurde die Reaktionsmischung unter Stickstoff-Atmosphäre über Asbest filtriert.
Nach Einengen erhielt man 804 g Rohprodukt (A 100% d.Th.). Nach mehrmaligem Umkristallisieren aus Methanol erhielt man 542 g reines 10ss-Epimeres (# 67% d.Th.).

3. Einführung der Azidofunktion
3.1 10ss-Methylverbindungen 3.1.1 3-Azidomethyl-4ss-acetoxy-8-methoxy- 10ss-methyl- 2,9-DTD (X) 50 g 3-Jodmethyl-4ss-acetoxy-8-methoxy- lOss-methyl- 2,9-DTD (VI, Beispiel 2.1) plus 100 g Natriumazid in 150 ml Hexamethylphosphorsäuretriamid (HMPA) wurden bei 100 C 1 Stunde gerührt. Anschliessend wurde die Reaktionslösung mit 600 ml Äther versetzt und 5mal mit je 150 ml Wasser geschüttelt. Nach Extrahieren der Wasserphasen mit Äther wurden die vereinten organischen Phasen über Na2SO4 getrocknet, filtriert, i.V. eingeengt und über Kieselgel mittels n-Hexan/Äther gereinigt.

4. Hydrolysierung der 4ss-Acetoxygruppe
4.1 1 Oss-Methylverbindungen 4.1.1 3-Azidomethyl-4ss-hydroxy-8-methoxy- 1 Oss-methyl- 2,9-DTD (XIV) 16 g 3-Azidomethyl-4ss-acetoxy-8-methoxy- 1 Oss-methyl- 2,9-DTD (X, Beispiel 3.1.1) in 200 ml Äther und 1,6 g NaOH in 50 ml Methanol wurden 10 Minuten bei Raumtemperatur gerührt. Anschliessend wurde mit Eiswasser versetzt und mit Eisessig neutralisiert. Die Lösung wurde dann mit Äther extrahiert. Die vereinten organischen Phasen wurden über Na2SO4 getrocknet, filtriert und i.V. eingeengt.
Nach Reinigung über Kieselgel mittels n-Hexan/Äther erhielt man 12,5 g farbloses Öl (#90,6% d.Th.)

Oxidation der 4ss-Hydroxygruppe zum Decan-4-on
5.1 10ss-Methylverbindungen 5.1.1 3-Azidomethyl-8-methoxy- 10ss-methyl-2,9-DTD-4-on (XVIII) 16 g 3-Azidomethyl-4ss-hydroxy-8-methoxy-10ss-methyl- 2,9-DTD (XIV, Beispiel 4.1.1) in 300 ml Aceton wurden bei 0 C langsam unter Rühren mit 40 ml CrO3-Lösung (Jones Reagenz) versetzt. Anschliessend wurden 10 ml Isopropanol hinzugegeben, die ausgefallenen Chromsalze abfiltriert und der Niederschlag mit CH2Ch gewaschen. Die vereinigten organischen Phasen wurden mit Na2CO3-Lösung neutralisiert und dann mit Wasser gewaschen. Nach Trocknen über Na2SO4 und Filtrieren wurde i.V. eingeengt.
Nach Reinigung über Kieselgel mittels n-Hexan/Äther wurden 13,8 g erhalten #76,5% d.Th.)
