caractérisé en ce que l'on fait réagir du chlorure mercurique aziridine avec la N-cyano-N',S-diméthylisothio-urée pour obtenir la 1- (NH-méthyl-N'-cyanocarboxamidino)aziridine, et que l'on fait réagir cette dernière avec le 4-méthyl-5-méthylmercapto-imidazole.
La présente invention concerne un procédé de préparation de composés de formule I telle que définie dans la revendication I et, notamment, du composé selon la formule II de la revendication 2.
Ces composés connus, notamment celui selon la formule I, ont des qualités thérapeutiques, notamment contre les ulcères de l'estomac, en bloquant les récepteurs de l'histamine H2.
Des procédés de fabrication de ces composés connus sont décrits dans les DT-OS Nos 2344779 et 2344833. Le but de la présente invention est de fournir des nouveaux procédés pour la préparation de ces composés connus. Les procédés selon l'invention sont définis dans les revendications 1, 4 et 5. Le caractère principal de ces procédés réside dans le fait que le groupe Y est une structure possédant un carbone électrophile en position '3par rapport à l'atome d'azote.
En se référant aux formules dans la revendication 1, il est à remarquer que le substituant Rl désigne l'hydrogène ou un groupe alkyle avec 1 à 4 atomes de carbone, par exemple, le méthyl, l'éthyl, le propyl ou le butyl, et le substituant Het, dans la formule III, est un reste d'un hétérocycle, substitué ou non, comportant 1 ou 2 atomes d'azote et en tout 5 ou 6 atomes dans le cycle, par exemple,
I'imidazolyle, etc.
Le produit préféré de la présente invention est la NH-méthyl-N' cyano-N"2-[(t-méthyl-5-imidazolyl)méthylthio]éthyl}guanidine qui est obtenue par réaction de chlorhydrate de 4-méthyl-5-méthylmercapto-imidazole et de N-(N'H-méthyl-N"-cyanoamidino)aziridine. Cette réaction est exécutée de préférence en solution aqueuse et sous atmosphère inerte. De préférence, on ajoute sous courant d'azote le NaOH et maintient l'atmosphère inerte durant toute la réaction. Comme décrit dans l'exemple, sous D, on obtient un produit cristallin de bonne qualité.
Les composés de formule III et IV sont des produits nouveaux qui sont obtenus selon le procédé décrit dans l'exemple, sous A à C (composés III) et C (composé IV). Le 4-méthyl-5-méthylmercaptoimidazole est préparé en trois étapes.
Le produit de départ est l'éthoxydithioformyl-2,3-dicétobutane, qui est transformé en 4-méthyl-5-(éthoxydithioformyl)imidazole, qui est transformé en 4-méthyl-5-méthylmercapto-imidazole.
Le produit intermédiaire de formule IV est aussi un produit nouveau et obtenu par le procédé décrit dans l'exemple, sous C.
Selon ce procédé, on obtient un produit cristallisé de bonne qualité.
L'exemple suivant décrit la préparation du composé préféré ainsi que des produits intermédiaires. L'invention n'est cependant pas limitée à ces formes d'exécution.
Exemple:
A. Ethoxydithioformyl-2,3-dicétobutane
A une solution, refroidie dans de la glace, de 74,4 g d'éthylxanthogénate de potassium dans 200 ml de méthanol, on ajoute goutte à goutte, sous agitation, 76,7 g de l-bromobutane-2,3-dione.
Après 16 h, on sépare le KBr insolubilisé par filtration et on élimine le solvant sous pression réduite; on reprend le résidu par de l'éther de pétrole, filtre et évapore le solvant; liquide résiduel: 97,7 g;
Eb. 98-100 C sous 0,1 Torr.

B. 4-Méthyl-5- (éthoxydithioformyl) imidazole
Une solution de 13,2 g d'éthoxydithioformyl-2,3-dicétobutane, 1,66 g d'hexaméthylénetétramine, 32 g d'acétate d'ammonium dans 200 ml d'acide acétique glacial est portée à 50 C durant 7 h. On évapore le solvant sous pression réduite et reprend le résidu par du chloroforme. Après filtration, la solution est extraite 2 fois par une solution de 15 ml d'acide chlorhydrique 12N dans 250 ml d'eau. La phase aqueuse est neutralisée par de l'hydrogénocarbonate de sodium, puis extraite par du chloroforme. La solution organique est séchée sur du sulfate de sodium déshydraté. Le résidu d'évaporation du solvant fournit un solide cristallin qui est recristallisé dans de l'acétone,F= 139-141 C.

C. Chlorhydrate de 4-méthyl-5-mérhylmercapto-imidazole
On porte à reflux sous courant d'azote, durant 6 h, une solution de 8,7 g de 4-methyl-5-(éthoxydithioformyl)imidazole dans 100 ml d'acide chlorhydrique 12N. On élimine le solvant sous pression réduite et cristallise à froid le résidu solide dans du butanol-2; recristallisation dans du butanol-2; F = 208-210 C.

Dans une solution de 15,8 g du ligand-chlorure mercuriqueaziridine (1:1) dans 60 ml de diméthylformamide, on dissout 3,1 g de
N-cyano-N',S-diméthylisothio-urée et porte la température à 55" C durant 5 h, puis on filtre; le solvant est évaporé sous pression réduite.
Le résidu est repris par une solution aqueuse contenant 184,4 mmol d'acide éthylènediaminotétraacètîque et 737,6 mmol de
NaOH. Cette solution est extraite 6 fois du chloroforme. Le résidu d'évaporation du solvant est cristallisé dans du tétrahydrofuranne puis recristallisé dans de l'acétate d'éthyle; F = 133-135 C.

The present invention relates to a process for the preparation of compounds of formula I as defined in claim I and, in particular, of the compound according to formula II of claim 2.
These known compounds, in particular that according to formula I, have therapeutic qualities, in particular against stomach ulcers, by blocking the histamine H2 receptors.
Processes for the manufacture of these known compounds are described in DT-OS Nos 2344779 and 2344833. The object of the present invention is to provide new methods for the preparation of these known compounds. The methods according to the invention are defined in claims 1, 4 and 5. The main character of these methods lies in the fact that the group Y is a structure having an electrophilic carbon in position '3 relative to the nitrogen atom .
Referring to the formulas in claim 1, it should be noted that the substituent R1 denotes hydrogen or an alkyl group with 1 to 4 carbon atoms, for example, methyl, ethyl, propyl or butyl, and the substituent Het, in formula III, is a residue of a heterocycle, substituted or unsubstituted, containing 1 or 2 nitrogen atoms and in total 5 or 6 atoms in the ring, for example,
Imidazolyle, etc.
The preferred product of the present invention is NH-methyl-N 'cyano-N "2 - [(t-methyl-5-imidazolyl) methylthio] ethyl} guanidine which is obtained by reaction of 4-methyl-5- hydrochloride methylmercaptoimidazole and N- (N'H-methyl-N "-cyanoamidino) aziridine. This reaction is preferably carried out in aqueous solution and under an inert atmosphere. Preferably, NaOH is added under a stream of nitrogen and the atmosphere is kept inert throughout the reaction. As described in the example, under D, a good quality crystalline product is obtained.
The compounds of formulas III and IV are new products which are obtained according to the process described in the example, under A to C (compounds III) and C (compound IV). 4-methyl-5-methylmercaptoimidazole is prepared in three stages.
The starting product is ethoxydithioformyl-2,3-diketobutane, which is transformed into 4-methyl-5- (ethoxydithioformyl) imidazole, which is transformed into 4-methyl-5-methylmercapto-imidazole.
The intermediate product of formula IV is also a new product and obtained by the process described in the example, under C.
According to this process, a good quality crystallized product is obtained.
The following example describes the preparation of the preferred compound as well as intermediate products. The invention is however not limited to these embodiments.
Example:
A. Ethoxydithioformyl-2,3-diketobutane
To a solution, cooled in ice, of 74.4 g of potassium ethyl xanthogenate in 200 ml of methanol, 76.7 g of l-bromobutane-2,3-dione are added dropwise with stirring.
After 16 h, the insoluble KBr is separated by filtration and the solvent is removed under reduced pressure; the residue is taken up in petroleum ether, filtered and the solvent evaporated; residual liquid: 97.7 g;
Eb. 98-100 C under 0.1 Torr.

The solvent is removed under reduced pressure and the solid residue is cold crystallized from 2-butanol; recrystallization from butanol-2; F = 208-210 C.
