xo Die Erfindung betrifft ein Verfahren zur Herstellung neuer cycloaliphatischer Sulfide der allgemeinen Formel I
25 worin R1 H,
R2 = O oder (H, OH),
R3 H oder OH,
R1 gegebenenfalls durch F, OH, Phenyl oder p-Tolyl substi-30 tuiertes Alkyl mit bis zu 12 C-Atomen, und A gegebenenfalls ein- oder mehrfach durch F substituiertes
Alkylen mit bis zu 8 C-Atomen bedeuten, und die durch gekennzeichnete Bindung a- oder ß-Konfiguration anzeigt, mit der Massgabe, dass min-35 destens eine Hydroxygruppe vorhanden ist, sowie deren physiologisch unbedenkliche Salze.
Es ist bekannt, dass prostaglandinanaloge Verbindungen, in denen die Seitenketten variiert oder ein oder mehrere Kohlenstoffatome durch Heteroatome ersetzt wurden, eine 40 bessere pharmakologische Wirksamkeit haben können als die natürlichen Prostaglandine. Bisher ist aber nur die Substitution von Kohlenstoffatomen durch Sauerstoffatome beschrieben.
Der Erfindung lag die Aufgabe zugrunde, ein Verfahren 45 zur Herstellung neuer Verbindungen mit wertvollen Eigenschaften aufzufinden, insbesondere solche, die zur Herstellung von Arzneimitteln verwendet werden können. Diese Aufgabe wurde durch die Bereitstellung der Verbindungen der Formel I gelöst.
50 Es wurde gefunden, dass die Verbindungen der Formel I bei guter Verträglichkeit neben einer blutdrucksenkenden Wirkung auch prostaglandinartige und antiprostaglandin-artige Wirkungen besitzen. So besitzen sie vasodilatorische, antiphlogistische, diuretische, bronchienentkrampfende, die 55 Magensaftsekretion, die Thrombozyten-Aggregation, den Lipidabbau und die Noradrenalin Freisetzung hemmende, sowie Nasenschleimhaut abschwellende Eigenschaften. Ausserdem beeinflussen sie die Funktion des corpus luteum, den Eitransport durch den Eileiter, die Nidation und die Fertilität 60 des Mannes. Die Verbindungen der Formel I können daher als Arzneimittel und auch als Zwischenprodukte zur Herstellung anderer Arzneimittel verwendet werden.
Die Verbindungen der Formel I enthalten mindestens zwei asymmetrische C-Atome im Fünfring. Ist R2 ungleich O 65 oder R3 ungleich H, so ist ein drittes C-Atom im Ring asymmetrisch; ist R2 ungleich O und Ra ungleich H, so sind im Ring vier Asymmetriezentren vorhanden. Handelt es sich bei A bzw. R4 um substituiertes Alkylen bzw. Alkyl, so können

in den beiden Seitenketten weitere Asymmetriezentren auftreten.
Die Verbindungen der Formel I können daher in einer Vielzahl stereoisomerer Formen auftreten und liegen in der Regel als Gemische von Racematen vor.
Die Verfahrensprodukte umfassen auch optisch aktive Verbindungen der Formel I sowie deren Racemate. Besonders bevorzugt sind die optisch aktiven Stereoisomeren der Formeln la - Ih, ihre optischen Antipoden und ihre Racemate:

Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I, dadurch gekennzeichnet, dass man in einer Verbindung, die sonst der Formel I entspricht, worin aber wenigstens eine der vorhandenen Hydroxygruppen mit einer Carbonsäure mit 1 bis 18 C-Atomen oder einer Sulfonsäure mit 1 bis 10 C-Atomen oder einer anorganischen Säure verestert ist oder mit einer Aralkylgruppe mit 7 bis 19 C-Atomen, einer Alkylgruppe mit bis zu 6 C-Atomen, der Tetrahydropyranyl- oder einer Tri-alkylsilylgruppe veräthert ist und gegebenenfalls die Carbo-nylgruppe in geschützter Form als Ketal, Hemiketal, Thio-. ketal, Hemithioketal, Enoläther, Schiffsche Base, Oxim,

Hydrazon, Semicarbazon oder Azin vorliegt und gegebenenfalls die Carboxylgruppe in geschützter Form vorliegt, mindestens eine veresterte oder verätherte Hydroxygruppe und ge-60 gebenenfalls eine geschützte Carbonyl- und/oder Carboxylgruppe durch Behandeln mit solvolysierenden oder hydro-genolysierenden Mitteln in Freiheit setzt.
Eine erhaltene Verbindung der Formel I kann durch Behandeln mit einem veresternden Mittel in eine andere Ver-65 bindung der Formel I überführt,
und eine Verbindung der Formel I kann in ihre Racemate und/oder optischen Antipoden gespalten werden.
Ferner kann eine Verbindung der Formel I durch Be

handeln mit einer Base in eines ihrer physiologisch unbedenklichen Salz überführt oder durch Behandeln mit einer Säure aus einem ihrer Salze in Freiheit gesetzt werden.
In den vorstehenden Formeln bedeutet R1 Wasserstoff. Aus den freien Säuren kann man durch entsprechende Veresterung Verbindungen der Formel I erhalten, worin R1 einen Alkylrest mit bis zu 12 C-Atomen darstellt. Vor allem handelt es sich um gerades Alkyl mit insbesondere bis zu 6 C-Atomen, z.B. um Methyl-, Äthyl-, n-Propyl-, n-Butyl-, n-Pen-tyl-, n-Hexyl; aber auch um Isopropyl-, Isobutyl-, sec.-Butyl-, tert.-Butyl-, Isopentyl-, Pent-2-yl-Pent-3-yl-, tert.-Pentyl-, Neopentyl-, Hex-2-yl-, Hex-3-yl- oder Isohexylgruppen, oder beispielsweise auch um n-Heptyl-, n-Octyl-, n-Nonyl-, n-De-cyl-, n-Undecyl- oder n-Dodecylreste.
R2 bedeutet neben = O auch (H, OH), wobei die OH-Gruppe a- oder ß-ständig sein kann.
R3 ist neben H auch OH, wobei die OH-Gruppe a- oder ß-ständig sein kann.
R4 bedeutet ein gegebenenfalls durch F, OH, Phenyl oder p-Tolyl substituiertes, gerades oder verzweigtes Alkyl mit bis zu 12 C-Atomen.
Als Alkylreste kommen vorzugsweise in Frage: Methyl, Äthyl, Propyl, Butyl, Pentyl, Hexyl, Heptyl, Octyl, Nonyl oder Decyl; 2,2-Dimethylpropyl, 3,3-Dimethylbutyl, 4,4-Di-methylpentyl, 5,5-Dimethylhexyl, 6,6-Dimethylheptyl, 7,7-Dimethyloctyl, 8,8-Dimethylnonyl, 9,9-Dimethyldecyl; aber auch Isopropyl, Isobutyl, Isopentyl, Isohexyl, Isoheptyl, Iso-octyl, 2-Butyl, 2-Pentyl, 2-Hexyl, 2-Heptyl oder 2-Octyl. Besonders bevorzugt sind 3,3-Dimethylpentyl, 3,3-Dimethyl-hexyl, 3,3-Dimethylheptyl, 3,3-Dimethyloctyl oder 3,3-Di-methylnonyl.
Besonders bevorzugt sind auch Alkylgruppen, welche in 2-Stellung durch OH substituiert sind, wie 2-Hydroxypropyl, 2-Hydroxybutyl, 2-Hydroxypentyl, 2-Hydroxyhexyl, 2-Hy-droxyheptyl, 2-Hydroxyoctyl, 2-Hydroxy-6,6-dimethyIheptyl, 2-Hydroxy-2-methylheptyl, 2-Hydroxy-7,7-dimethyloctyl, 2-Hydroxynonyl, 2-Hydroxy-2-methylnonyl, 2-Hydroxy-3,3--dimethylnonyl, 2-Hydroxy-8,8-dimethylnonyl, sowie, soweit hier noch nicht genannt, die in 2-Stellung durch OH substituierten oben aufgezählten Alkylgruppen.
R4 kann auch durch F substituiertes Alkyl mit bis zu 12 C-Atomen sein, vorzugsweise ein endständiges Trifluoralkyl, wie 5,5,5-Trifluorpentyl, 6,6,6-Trifluorhexyl, 7,7,7-Trifluor-heptyl oder 8,8,8-Trifluoroctyl. Wenn R4 ein durch Phenyl substituierter Alkylrest ist, so handelt es sich vor allem um omega-Phenyl-alkylgruppen wie 3-Phenylpropyl-, 4-Phenyl-butyl-, 5-Phenylpentyl-, 6-Phenylhexyl-, 7-Phenylheptyl-oder 8-Phenyloctylgruppen. R4 kann auch ein durch p-Tolyl substituierter Alkylrest sein, vorzugsweise ein omega-p-To-lylalkylrest, wie 2-p-Tolyläthyl, 3-p-Tolylpropyl oder 4-p-To-lylbutyl.
Selbstverständlich kann R4 auch ein Alkylrest mit bis zu 12 C-Atomen sein, der mehrere verschiedene Substituenten trägt. Bevorzugte Beispiele für solche Reste R4 sind: 2-Hy-droxy-7,7,7-trifluorheptyl-, 2-Hydroxy-8,8,8-trifluoroctyl-; 2-Hydroxy-9,9,9-trifluornonyl, 2-Hydroxy-2-methyl-9,9,9-tri-fiuornonyl, 2-Hydroxy-3,3-dimethyl-9,9,9-trifIuornonyl.
A ist vorzugsweise ein unsubstituiertes Alkylen mit bis zu 8 C-Atomen wie Methylen-, Äthylen-, Trimethylen-, Tetra-methylen-, Pentamethylen-, Hexamethylen-, Heptamethylen-oder Octamethylenreste. Ist A ein substituiertes Alkylen mit bis zu 8 C-Atomen, so handelt es sich um Fluoralkylengrup-pen wie -CH..CHF-, -CH2CF2-, -CH2CH2CHFCH2-, -CH2CH2CH2CF2-, -CH2CH2CH2CHFCHF-, -CH2CH2CF2-,
-ch2ch2ch2cf2ch2-, -ch2ch2cf2ch2ch2-,
-CH2CH„CH2CF2CH2CH2-, -(CH2)5CF2- oder -(CH2)4-CHFCHF-.
Bei Hydroxygruppen, welche in funktionell abgewandelter Form vorliegen, handelt es sich vorzugsweise um z.B. mit einer gesättigten oder ungesättigten aliphatischen, cycloali-phatischen oder aromatischen, substituierten oder unsubsti-5 tuierten Carbonsäure oder Sulfonsäure oder auch einer anorganischen Säure veresterten OH-Gruppen. Bevorzugte Carbonsäuren sind Fettsäuren mit 1-18, vorzugsweise 1-6 C-Ato-men, wie Ameisensäure, Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, Valeriansäure, Isovaleriansäure, Caio pronsäure, Isocapronsäure, Önanthsäure, Caprylsäure, Pelar-gonsäure, Caprinsäure, Laurinsäure, Myristinsäure, Palmitinsäure, Stearinsäure, ferner Crotonsäure, Ölsäure, Cyclo-hexancarbonsäure, Cyclohexylessig- und propionsäure, Benzoesäure, Phenylessig- und -propionsäure, Dicarbonsäuren 15 wie Oxal-, Nalon-, Bernstein-, Malein-, Glutar-, Dimethyl-glutar-, Adipin-, Pimelin-, Acetondicarbon-, Phthal-, Tetra-hydrophthal-, Hexahydrophthal- oder Diglykolsäure, Hy-droxycarbonsäuren wie Glykolsäure, Halogenfettsäuren wie Chloressigsäure, Dichloressigsäure, Trichloressigsäure oder 20 Trifluoressigsäure.
Bevorzugte Sulfonsäureester sind solche, die abgeleitet von Alkylsulfonsäuren mit 1-6 C-Atomen sind, z.B. Methanoder Äthansulfonsäure, und Arylsulfonsäuren mit 6-10 C-Atomen, z.B. Benzol-, p-Toluol-, 1- und 2-NaphthaIinsul-25 fonsäure. Bevorzugte anorganische Säuren sind Schwefelsäure und Phosphorsäure.
Funktionell abgewandeltes OH kann weiterhin eine ver-ätherte OH-Gruppe bedeuten, z.B. Aralkoxy mit vorzugsweise 7-19 C-Atomen, wie Benzyloxy, p-Methylbenzyloxy, 30 1- und 2-Phenyläthoxy, Diphenylmethoxy, Triphenylmeth-oxy, oder 1- oder 2-Naphthylmethoxy; Alkoxy mit vorzugsweise bis zu 6 C-Atomen, insbesondere tert.-Butoxy; Tetra-hydroxypyranyloxy; oder Trialkylsilyloxy, vorzugsweise Tri-methylsilyloxy.
35 Bei Ketogruppen, welche in funktionell abgewandelter Form vorliegen, handelt es sich um die üblichen Gruppen, vorzugsweise um -C(OH) (OR14)-, -C(OR14)2-, -C(OCOR14)2-, -C(=NOH)-, -C(=NNH2)-, -C(=NH)-, -C(=NR)-, =C(-OR14)-, =C(-OCOR14)- oder = C(-NR14R15)-. 40 R14 und R15 sind gleich oder ungleich und bedeuten neben H einen organischen Rest, beispielsweise Alkylgruppen, vorzugsweise unverzweigt, mit insbesondere bis zu 4 C-Ato-men; durch Cl, Br, J, OH, Alkoxy mit bis zu 4 C-Atomen, Acyloxy mit bis zu 4 C-Atomen, Phenyl oder Naphthyl sub-45 stituiertes Alkyl mit bis zu 4 C-Atomen, Cycloalkyl mit bis zu 6 C-Atomen, gegebenenfalls ein- oder mehrfach durch Cl, Br, N02, OH, Alkoxy mit bis zu 4 C-Atomen, Acyloxy mit bis zu 4 C-Atomen, substituiertes Aryl mit bis zu 10 C-Atomen; gemeinsam auch Polymethylen mit bis zu 6 C-Atomen, 50 vorzugsweise unsubstituiertes Polymethylen mit bis zu 6 C-Atomen wie -(CH2)4-, -(CH2)5- oder -(CH2)G-; oder gemeinsam auch ein durch O, N oder S-Atome unterbrochenes Polymethylen mit vorzugsweise bis zu 6 C-Atomen, wie -(CH2)2-0-CH2-, -(CH2)2-0-(CH2)2-, -(CH2)3-0-CH2-, 55 -(CH2)2-NH-CH2-, -(CH2)2-NH-(CH2)2-, -(CH2)3-NH-CH2-, -(CH2)2-S-CH2-, -(CH2)2-S-(CH2)2- oder -(CH2)3-S-CH2-.
Bei Carboxylgruppen, welche in funktionell abgewandelter Form vorliegen, handelt es sich vorzugsweise um eine Halogencarbonylgruppe wie -COCl oder COBr; eine stick-60 stoffhaltige funktionell abgewandelte Carboxylgruppe, insbesondere -CON3, -CONRI4R15, -CONHNH2, C=N, oder eine nur Sauerstoff enthaltende funktionell abgewandelte Carboxylgruppe, insbesondere -C(OR14)3, -COOCOR14 oder -COOR14.

Eine Verbindung, die sonst der Formel I entspricht, worin aber wenigstens eine der vorhandenen Hydroxygruppen und gegebenenfalls die Ketogruppe und gegebenenfalls die Carboxylgruppe in funktionell abgewandelter Form vorliegen, kann ebenfalls in an sich bekannter Weise hergestellt werden.
Funktionelle Carbonylderivate von Verbindungen der Formel I erhält man in an sich bekannter Weise z.B. aus den Cyclopentanonen der Formel I durch Umsetzen mit einem entsprechenden N-haltigen Carbonylreagens, beispielsweise einem Hydroxylamin wie H2NOH oder O-AIkyl-hydroxyl-aminen; einem Hydrazin, wie H2NNH2, N-Methylhydrazin, N,N-Dimethylhydrazin, Phenylhydrazin, 4-Nitrophenylhy-drazin oder 2,4-Dinitrophenylhydrazin; Semicarbazid; oder einem primären Amin, vorzugsweise einem aliphatischen, ali-cyclischen oder aromatischen primären Amin, wie Methylamin, Äthyl amin, n-Propylamin, Isopropylamin, n-Butylamin, Cyclohexylamin, Anilin, p-Toluidin, a-Naphthylamin oder ß-Naphthylamin. Diese Umsetzungen erfolgen unter Wasserabspaltung, im allgemeinen unter Verwendung eines inerten Lösungsmittels, vorzugsweise niedrigen Alkoholen mit bis zu 4 C-Atomen, wie Methanol oder Äthanol und in Gegenwart eines sauren oder basischen Katalysators, vorzugsweise einer anorganischen oder organischen Säure, wie HCl, H2S04 oder Essigsäure; oder auch eines Alkalihydroxids, wie NaOH oder KOH, oder eines basisch reagierenden Salzes, wie K2COs, Na,C03 oder Na-Acetat. Die Reaktionstemperaturen liegen meist zwischen Raumtemperatur und 120°, vorzugsweise arbeitet man beim Siedepunkt des Reaktionsgemisches.
Die N-haltigen Carbonylderivate der Verbindungen der Formel I werden aus diesen Verbindungen entweder zur Isolierung aus den Reaktionsgemischen bzw. zur Reinigung hergestellt, da sie im allgemeinen besser kristallisieren als die — in der Regel öligen — Verbindungen der Formel I selbst; man kann sie aber auch zur Herstellung anderer Verbindungen der Formel I heranziehen.
Verbindungen, die sonst der Formel I entsprechen, in denen aber R2 eine funktionalisierte, insbesondere ketalisierte Oxogruppe ist, sind vorzugsweise Ketale, insbesondere Äthy-lenketale, Hemiketale, Thioketale, Hemithioketale; aber auch 5 Oxime, Hydrazone, Semicarbazone oder Schiffsche Basen.
Die funktionellen Derivate der Verbindungen der Formel I können nach in der Literatur beschriebenen Methoden zu den Verbindungen der allgemeinen Formel I hydrolysiert werden. Die Hydrolyse wird in saurem oder gegebenenfalls io alkalischem Medium bei Temperaturen zwischen —20° und 300°, vorzugsweise bei der Siedetemperatur des Reaktionsgemisches, durchgeführt. Als saure Katalysatoren eignen sich beispielsweise Salz-, Schwefel-, Phosphor- oder Bromwasserstoffsäure, oder organische Säuren wie Oxalsäure, Weinsäure, 15 p-Toluolsulfonsäure; als basische Katalysatoren verwendet man zweckmässig Natrium-, Kalium- oder Calciumhydroxid, Natrium- oder Kaliumcarbonat. Als Lösungsmittel wählt man vorzugsweise Wasser; niedere Alkohole wie Äthanol, Methanol; Äther wie Dioxan, Tetrahydrofuran; Amide wie Dime-20 thylformamid; Nitrile wie Acetonitril; Sulfone wie Tetramethylensulf on; oder deren Gemische, besonders die Wasser" enthaltenden Gemische.
Äther, die sonst der Formel I entsprechen, in denen die Reste R2 und R3 aber die nachfolgende Bedeutung haben, 25 R2 = [H, OR16], R3 = H; R2 = [H, OR16], R3 = OR16; R2 = [H, OR16], R3 = OH; oder R2 = O, R3 = OR16 können nach den aus der Literatur bekannten Ätherspaltungs-Methoden in Alkohole der Formel I (R2 = [H, OH] und/ oder R3 = OH; oder R2 = O und Ra = OH) umgewandelt 30 werden. Zum Beispiel kann man die Äther spalten durch Behandeln mit Bromwasserstoff oder Jodwasserstoff in wässeriger oder essigsaurer Lösung, durch Erhitzen mit Lewis-Säuren wie A1C13 oder Bortrihalogeniden oder durch Verschmelzen mit Pyridin- oder Anilin-Hydrohalogeniden bei ca. 200°. 35 Aus erhaltenen Säuren der Formel I (R1 = H) können Ester der Formel I (R1 = Alkyl mit bis zu 12 C-Atomen)
nach in der Literatur beschriebenen Methoden hergestellt werden. So kann man beispielsweise einet Säure der Formel I (R1 = H) mit dem betreffenden Alkohol in Gegenwart einer 40 anorganischen oder organischen Säure, wie HCl, HBr, HJ, H2S04, H3P04, Trifluoressigsäure, einer Sulfonsäure wie Ben-zolsulfonsäure oder p-Toluolsulfonsäure, oder eines sauren Ionenaustauschers gegebenenfalls in Gegenwart eines inerten Lösungsmittels, wie z.B. Benzol, Toluol oder Xylol, bei Tem-45 peraturen zwischen etwa 0° und vorzugsweise Siedetemperatur umsetzen. Der Alkohol wird bevorzugt im Überschuss eingesetzt. Bevorzugte Alkohole sind solche der Formel
R17OH
worin R17 Alkyl mit bis zu 12 C-Atomen 50 bedeutet.
Insbesondere handelt es sich dabei um unverzweigte primäre Alkohole mit bis zu 12 C-Atomen, wie Methanol, Äthanol, Propanol, Butanol, Hexanol, Octanol, Decanol oder Do-decanol; es kann sich aber auch z.B. um Isopropylalkohol, 55 sec.-Butylalkohol oder tert.-Butylalkohol handeln.
Weiterhin kann man in Gegenwart wasserbindender Agen-tien arbeiten, z.B. von wasserfreien Schwermetallsulfaten oder von Molekularsieben. Man kann auch das Reaktionswasser azeotrop entfernen, wobei man vorteilhaft Kohlen-60 Wasserstoffe (z.B. Benzol oder Toluol) oder chlorierte Kohlenwasserstoffe (z.B. Chloroform oder 1,2-Dichloräthan) zusetzt. Unter milden Bedingungen verläuft die Veresterung, wenn man das Reaktionswasser chemisch durch Zusatz von Carbodiimiden (z.B. N,N'-Dicyclohexylcarbodiimid) bindet, 65 wobei man inerte Lösungsmittel wie Äther, Dioxan, 1,2-Di-methoxyäthan, Benzol, CH2C12 oder CHC13 verwendet und Basen wie Pyridin zusetzen kann. Die Methylester (bzw. Äthylester) können auch durch Umsetzen der freien Säuren

mit Diazomethan (bzw. Diazoäthan) in einem inerten Lösungsmittel wie Äther, Benzol oder Methanol hergestellt werden. Man erhält Ester der Formel I (R1 ungleich H) auch durch Anlagerung der Carbonsäuren (I, R1 = H) an Olefine, (z.B. Isobutylen, Cyclohexen), vorzugsweise in Gegenwart von Katalysatoren (z.B. ZnCl2, BF3, H2S04, Arylsulfonsäuren, Pyrophosphorsäure, Borsäure, Oxalsäure) bei Temperaturen zwischen etwa 0 und etwa 200°, Drucken zwischen 1 und 300 at und in inerten Lösungsmitteln wie Äther, Tetrahydro-furan, Dioxan, Methylenchlorid, Benzol, Toluol oder Xylol.
Weiterhin kann man Ester der Formel I (R1 = ungleich H) herstellen durch Umsetzen von Metallsalzen der Carbonsäuren der Formel I (R1 = H), vorzugsweise der Alkalimetall-, Blei- oder Silbersalze, mit Alkylhalogeniden, z.B. solchen der Formel R17C1, gegebenenfalls in einem inerten Lösungsmittel, z.B. Äther, Benzol, DMF oder Petroläther, oder mit Alkylchlorsulfiten, z.B. solchen der Formel R17-OSOCl und nachfolgende Thermolyse der erhaltenen Addukte.
Die Verbindungen der Formel I haben, wie bereits erläutert, im allgemeinen mehrere Asymmetriezentren, stets aber wenigstens zwei. Sie werden daher meist als Gemisch verschiedener stereoisomerer Formen erhalten, d.h. als Racemate oder in der Regel als Gemische von Racematen. Da verschiedene Racemate zueinander diastereomer sind, können sie aufgrund ihrer unterschiedlichen physikalischen Eigenschaften aus ihren Gemischen isoliert und rein erhalten werden, beispielsweise durch Umkristallisieren aus geeigneten Lösungsmitteln (wobei insbesondere anstelle der Verbindungen selbst, gut kristallisierende Derivate eingesetzt werden können), durch destillative Trennung, insbesondere aber mit Hilfe chromatographischer Methoden, wobei sowohl adsorp-tionschromatographische oder verteilungschromatographische Methoden als auch Mischformen in Frage kommen.
Die Racemate können nach einer Vielzahl bekannter Methoden, wie sie in der Literatur angegeben sind, in ihre optischen Antipoden getrennt werden. Die Methode der chemischen Trennung ist zu bevorzugen. Danach werden aus dem racemischen Gemisch durch Umsetzung mit einem optisch aktiven Hilfsmittel Diastereomere gebildet. So kann man gegebenenfalls eine optisch aktive Base mit der Carboxylgruppe einer Verbindung der Formel I (R1 = H) umsetzen. Zum Beispiel kann man diastereomere Salze der Verbindungen der Formel I mit optisch aktiven Aminen, wie Chinin, Chinchonidin, Bruein, Cinchonin, Hydroxyhydrindamin, Morphin, 1-Phenyläthylamin, 1-Naphthyläthylamin, Phenyl-oxynaphthylmethylamin, Chinidin, Strychnin, basischen Aminosäuren, wie Lysin, Arginin oder den Aminosäureestern bilden. In ähnlicher Weise lassen sich Ester-Diastereomere durch Veresterung von Verbindungen der Formel I (R1 = H) mit optisch aktiven Alkoholen, wie Borneol, Menthol, Octa-nol-2, herstellen. Der Unterschied in der Löslichkeit der anfallenden diastereomeren Salze bzw. Ester erlaubt die selektive Kristallisation der einen Form und die Regeneration der jeweiligen optisch aktiven Verbindungen aus dem Gemisch.
Die Hydroxysäuren und Hydroxyester der Formel I (R2 = [H, OH], R3 = H, R2 = [H, OH], R3 = OH; R2 = O, R3 = OH) können ferner durch Veresterung mit einer optisch aktiven Säure wie (+)- und (—)-Weinsäure, Dibenzoyl-(+)- und (—)-weinsäure, Diacetyl-(+)- und (—)-Weinsäure, Camphersäure, ß-Camphersulfonsäure, (+)- und (—)-Mandelsäure, (+)- und (—)-ÄpfeIsäure, (+)- und (—)--2-Phenylbuttersäure, (+)-Dinitrodiphenylsäure oder (+)-und (—)-MiIchsäure in geeignete diastereomere Ester umgewandelt werden, die sich aufgrund ihrer unterschiedlichen Löslichkeiten trennen lassen. Die optisch aktiven Verbindungen der Formel I werden dann jeweils durch Verseifen des reinen Diastereomeren gewonnen. Man kann aber auch zunächst mit Phthalsäure- oder Bernsteinsäureanhydrid die sauren Phthalsäure- bzw. Bernsteinsäureester darstellen und die so gewonnenen zweibasigen Säuren bzw. deren Monoester mit einer der oben angegebenen, optisch aktiven Basen in die diastereomeren Salze überführen und aus diesen die reinen s Enantiomeren gewinnen. Aus den Ketosäuren und den Keto-estern der Formel I (R2 = O) können durch Umsetzung mit optisch aktiven Keton-Reagentien, wie z.B. Menthylhydrazin oder Menthylsemicarbazid, die entsprechenden diastereomeren Hydrazone bzw. Semicarbazone dargestellt werden, aus io denen man ebenfalls die reinen Enantiomeren gewinnen kann. Besonders vorteilhaft ist die Trennung der Racemate bzw. Racemat-Gemisch mit Hilfe von chromatographischen Methoden. Man kann entweder optisch aktive Trägermaterialien, wie z.B. Weinsäure, Stärke, Rohrzucker, Cellulose oder 15 acetylierte Cellulose und optisch inaktive und/oder optisch aktive Laufmittel zur Trennung in die reinen Enantiomeren verwenden oder ein optisch inaktives Trägermaterial, wie z.B. Kieselgel oder Aluminiumoxid in Kombination mit einem optisch aktiven Laufmittel. Die optischen Antipoden können 20 auch biochemisch unter Anwendung selektiver, enzymati-scher Reaktionen getrennt werden. So kann man die racemischen Säuren der Formel I (R1 = H) einer Oxidase oder gegebenenfalls Decarboxylase aussetzen, die durch Oxidation oder Decarboxylierung eine Form zerstört, während die andere 25 Form unverändert bleibt. Möglich ist ferner die Verwendung einer Hydrolase bei einem funktionellen Säurederivat des racemischen Gemischs zur bevorzugten Bildung einer optisch aktiven Form. So kann man Ester oder Amide der Säuren der Formel I (R1 = H) der Einwirkung einer Hydrolase aus-30 setzen, welche das eine Enantiomere selektiv verseift und das andere unverändert lässt.
Weiterhin ist es natürlich möglich, optisch aktive Verbindungen nach den beschriebenen Methoden zu erhalten, indem man Ausgangsstoffe verwendet, die bereits optisch ak-35 tiv sind.
Die Reste -A-COOR1 und -S-R4 sind in der Regel transständig, d.h. wenn die Alkylthiogruppe ß-ständig ist, so ist die omega-Carboxy- oder Carbalkoxyalkylgruppe a-ständig und umgekehrt.
40 Wird nun eine Verbindung der Formel I oder eine Vorstufe mit bestimmten sterischen Verhältnissen an einem oder mehreren C-Atomen in eine Verbindung der Formel I bzw. eine andere Verbindung der Formel I unter Ausbildung eines neuen Asymmetriezentrums umgewandelt, so lässt sich durch 45 geeignete Reaktionsführung erreichen, dass dieses C-Atom überwiegend eine bestimmte, vorzugsweise die gewünschte Konfiguration aufweist.
Verbindungen der Formel I (R1 = H) können durch Umsetzung mit einer Base in eines ihrer physiologisch unbedenk-50 liehen Metall- bzw. Ammoniumsalze übergeführt werden. Als Salze kommen insbesondere die Natrium-, Kalium-, Magnesium-, Calcium- und Ammoniumsalze in Betracht, ferner substituierte Ammoniumsalze, wie z.B. die Dimethyl-und Diäthylammonium-, Monoäthanol-, Diäthanoi- und Tri-55 äthanolammonium- Cyclohexylammonium-, Dicyclohexyl-ammonium- und Dibenzyläthylendiammonium-Salze.
Umgekehrt können Verbindungen der Formel I aus ihren Metall- und Ammoniumsalzen durch Behandlung mit Säuren, vor allem Mineralsäuren wie Salz- oder Schwefelsäure, in 60 Freiheit gesetzt werden.
Die neuen Verbindungen können im Gemisch mit festen, flüssigen und/oder halbflüssigen Arzneimittelträgern als Arzneimittel in der Human- oder Veterinärmedizin verwendet werden. Als Trägersubstanzen kommen solche organischen 65 oder anorganischen Stoffe in Frage, die für die parenterale, enterale oder topikale Applikation geeignet sind und die mit den neuen Verbindungen nicht in Reaktion treten, wie beispielsweise Wasser, pflanzliche Öle, Benzylalkohole, Poly-

äthylenglykole, Gelatine, Lactose, Stärke, Magnesiumstearat, Talk, Vaseline, Cholesterin. Zur parenteralen Applikation dienen insbesondere Lösungen, vorzugsweise ölige oder wäss-rige Lösungen, sowie Suspensionen, Emulsionen oder Implantate. Für die enterale Applikation eignen sich Tabletten, Dragees, Sirupe, Säfte oder Suppositorien, für dit topikale Anwendung Salben, Cremes oder Puder. Die angegebenen Zubereitungen können gegebenenfalls sterilisiert oder mit Hilfsstoffen, wie Gleit-, Konservierungs-, Stabilisierungsoder Netzmitteln, Emulgatoren, Salzen zur Beeinflussung des osmotischen Druckes, Puffersubstanzen, Färb-, Geschmacks-und/oder Aromastoffen und/oder anderen Wirkstoffen, beispielsweise Vitaminen, versetzt werden.
Die Substanzen werden vorzugsweise in einer Dosierung von 0,1 bis 2 000 mg pro Dosierungseinheit verabreicht.
Die im folgenden angegebenen IR-Spektren wurden mit Perkin Elmer 6 als Film aufgenommen, und die NMR-Spek-tren mit Varian HA 100 oder A 60 in CDC13 gegen Tetra-methylsilan als inneren Standard gemessen.

Man kocht 5,0 g la-p-ToIuoIsuIfonyloxy-2-(6-carboxy-hexyl)-3-heptylthiocyclopentan (erhältlich aus 2-(6-Carboxy-hexyl)-3-heptylthiocyclopentanol-(la) und p-Toluolsulfon-säurechlorid) 2 Stunden in 60 ml 2n wässeriger NaOH, giesst nach dem Erkalten in 100 ml 2n wässerige HCl, extrahiert zweimal mit je 50 ml CH2C12, wäscht die organische Phase mit Wasser, trocknet über Na2S04, destilliert das Lösungsmittel ab und erhält nach chromatographischer Reinigung des Rückstandes (Kieselgel/Chloroform) 2-(6-Carboxyhexyl)-3--hepty lthio-cyclopentanol-( lß). NMR-Spektrum: Signale bei 1,2 ppm, 2,35 ppm (Triplett),
5 ppm (breites Multiplett) und 7,3 ppm,
sowie ein Stereoisomeres dieser Verbindung als Öl, IR-Spektrum: Banden bei 1600, 1745 und 1750 cm-1, breite
Bande bei 3400 cm-1,
NMR-Spektrum: Signale bei 1,2 ppm, 2,25 ppm (Triplett),
4,25 ppm, 5,2 ppm (breites Multiplett) und 7,3 ppm; 2-(6-Carboxyhexyl)-3-(2-hydroxy-2-methyl-4-phenyl-butyl-
thio)-4-hydroxy-cyclopentanon als Öl,
IR-Spektrum: Banden bei 1710 und 1730 cm-1, breite Bande bei 3400 cm-1,
NMR-Spektrum: Signale bei 1,35 ppm, 5,3 ppm u. 7,25 ppm.
Beispiel 6
0,6 g l,4-Bis-trimethylsilyloxy-2-(6-carboxyhexyl)-3-(2-tri-methylsilyloxy-2-methyl-2-p-tolyläthylthio)-cyclopentan werden in 10 ml Acetonitril gelöst und mit 1 g Tetrabutylammo-niumfluorid versetzt. Man rührt 16 Stunden bei 20°, erhitzt anschliessend noch 3 Stunden zum Sieden und engt die Lösung ein. Der Rückstand wird zwischen Wasser und Methylenchlorid verteilt, die organische Phase wird gewaschen, getrocknet und eingeengt. Nach chromatographischer Reinigung des Rückstandes erhält man
2-(6-Carboxyhexyl)-3-(2-hydroxy-2-methyl-2-p-tolyläthylthio)-
-1,4-cyclopentandiol als Öl,
IR-Spektrum: Banden bei 1505 und 1705 cm-1, breite Bande bei 3350 cm-1,
NMR-Spektrum: Signale bei 1,23 ppm, 2,3 ppm, 4,21 ppm (Quartett), 4,6 ppm (breites Multiplett) und zwischen 7,0 und 7,3 ppm.
Analog sind aus entsprechend geschützten Cyclopentan-Derivaten durch Solvolyse erhältlich: 2-(6-Carboxyhexyl)-3-(2-hydroxy-2-methyl-4-phenylbutyl-
thio)-l,4-cyclopentandiol als Öl,
IR-Spektrum: Banden bei 1700 und 3400 cm-1, NMR-Spektrum: Signale bei 1,33 ppm, 4,4 ppm u. 7,25 ppm, sowie ein Stereoisomeres dieser Verbindung als Öl, IR-Spektrum: Banden bei 1700 und 3400 cm-1, NMR-Spektrum: Signale bei 1,33 ppm, 5,62 ppm (breites
Multiplett) und zwischen 7,12 und 7,30 ppm, 2-(6-Carboxyhexyl)-3-(2-hydroxy-2-methylheptylthio)-l,4-
-cyclopentandiol als Öl,
IR-Spektrum: Banden bei 1705, 2680 und 3340 cm-1, NMR-Spektrum: Signale bei 0,89 ppm, 1,22 ppm, 2,3 ppm,
4,18 ppm und 5,03 ppm,
sowie ein Stereoisomeres als Öl,
IR-Spektrum: Banden bei 1705,2680 und 3340 cm-1, NMR-Spektrum: Signale bei 0,9 ppm, 1,28 ppm, 2,3 ppm, 4,0 ppm, 4,1 ppm und 4,8 ppm.
Beispiel 7
0,56 g 2-(6-Carbo-tetrahydropyranyl-(2)-oxy-hexyl)-3-(2--hydroxy-2-methyl-heptylthio)-4-tetrahydropyranyl-(2)-oxy--cyclopentanon werden in 10 ml Isopropanol gelöst, mit 0,05 g Pyridinium-p-toluolsulfonat versetzt und zwei Stunden unter Rückfluss gekocht. Man engt ein, nimmt in Ethylacetat auf, wäscht und trocknet die organische Phase, zieht das Lösungsmittel ab und erhält nach chromatographischer Reinigung 2-(6-Carboxyhexyl)-3-(2-hydroxy-2-methyl-heptylthio)--4-hydroxy-cyclopentanon
IR-Spektrum: Banden bei 1710 und 1740 cm-1, breite Bande bei 3450 cm-1,
NMR-Spektrum: Signale bei 0,85 ppm, 1,2 ppm, 2,3 ppm (Triplett), 4,2 ppm und 5,8 ppm (Multiplett).

Man gibt zu einer Lösung von 3,9 g 2-(6-Carboxyhexyl)--3-(2-hydroxy-2-methylheptylthio)-4-hydroxy-cyclopentanon in 30 ml trockenem Äthanol bei Raumtemperatur 80 ml einer 0,2 n Diazomethanlösung in Äther, giesst das Reaktionsgemisch in 100 ml Eiswasser, enthaltend 5 ml konzentrierter HCl, rührt, trennt die organische Phase ab, extrahiert die wässerige Phase dreimal mit je 30 ml Chloroform, wäscht die vereinigten organischen Phasen mit Wasser, trocknet über Na2S04, destilliert das Lösungsmittel ab und erhält nach chromatographischer Reinigung des Rückstandes (Kieselgel/ Chloroform)
2-(6-CarbomethoxyhexyI)-3-(2-hydroxy-2-methyI-heptylthio)--4-hydroxy-cyclopentanon,
IR-Spektrum: Banden bei 1740 und 3420 cm-1; NMR-Spektrum: Signale bei 0,86 ppm, 1,21 ppm, 2,25 ppm, 3,61 ppm und 4,22 ppm.

Analog Beispiel 8 erhält man aus 2-(6-Carboxyhexyl)-3--(2-hydroxy-2-methyl-heptylthio)-1,4-cyclopentandiol durch Umsetzen mit Diazomethan 2-(6-Carbomethoxyhexyl)-3-(2--hydroxy-2-methyl-heptylthio)-1,4-cyclopentandiol, 10 IR-Spektrum: Banden bei 1740 und 3490 cm-1, NMR-Spektrum: Signale bei 0,90 ppm, 1,22 ppm, 2,31 ppm, 3,61 ppm und 4,18 ppm.

Claims (8)

- 2. Verfahren zur Herstellung einer Verbindung der Formel I, worin R2, R3, R4 und A die im Patentanspruch 1 angegebene Bedeutung haben und R1 Alkyl mit bis zu 12 C-Atomen bedeutet, mit der Massgabe, dass mindestens eine Hydroxygruppe anwesend ist, dadurch gekennzeichnet, dass man nach dem Verfahren nach Patentanspruch 1 eine Verbindung der Formel I herstellt, worin R1 bis R4 und A die im Patentanspruch 1 angegebene Bedeutung haben und die erhaltene Verbindung anschliessend mit einem veresternden Mittel umsetzt.2PATENTANSPRÜCHE 1. Verfahren zur Herstellung einer Verbindung der allgemeinen Formel Iworin R1 H,R2 = O oder (H, OH),R3 H oder OH,R4 gegebenenfalls durch F, OH, Phenyl oder p-Tolyl substituiertes Alkyl mit bis zu 12 C-Atomen, und A gegebenenfalls ein- oder mehrfach durch F substituiertesAlkylen mit bis zu 8 C-Atomen bedeuten, und die durch gekennzeichnete Bindung a- oder ß-Konfiguration anzeigt, mit der Massgabe, dass mindestens eine Hydroxygruppe vorhanden ist, sowie ihrer physiologisch unbedenklichen Salze, dadurch gekennzeichnet, dass man in einer Verbindung, die sonst der Formel I entspricht, worin aber wenigstens eine der vorhandenen Hy-droxygruppen mit einer Carbonsäure mit 1 bis 18 C-Atomen oder einer Sulfonsäure mit 1 bis 10 C-Atomen oder einer anorganischen Säure verestert ist oder mit einer Aralkyl-gruppe mit 7 bis 19 C-Atomen, einer Alkylgruppe mit bis zu 6 C-Atomen, der Tetrahydropyranyl- oder einer Trialkyl-silylgruppe veräthert ist und gegebenenfalls die Carbonyl-gruppe in geschützter Form als Ketal, Hemiketal, Thioketal, Hemithioketal, Enoläther, Schiffsche Base, Oxim, Hydrazon, Semicarbazon oder Azin vorliegt und gegebenenfalls die Carboxylgruppe in geschützter Form vorliegt, mindestens eine veresterte oder verätherte Hydroxygruppe und gegebenenfalls eine geschützte Carbonyl- und/oder Carboxylgruppe durch Behandeln mit solvolysierenden oder hydrogenolysie-renden Mitteln in Freiheit setzt.
- 3. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel I in ihre Racemate aufspaltet.
- 4. Verfahren nach Patentanspruch 2, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel I in ihre Racemate aufspaltet.
- 5. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel I in ihre optischen Antipoden aufspaltet.
- 6. Verfahren nach Patentanspruch 2, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel I in ihre optischen Antipoden aufspaltet.
- 7. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel Idurch Behandeln mit einer Base in eines ihrer physiologisch unbedenklichen Salze überführt.
- 8. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel I durch Be-5 handeln mit einer Säure aus einem ihrer erhaltenen Salze in Freiheit setzt.
