Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Dithiocarbaminsäureestern der Formel

worin R, einen gegebenenfalls durch Niederalkyl, Halogen, Trifluormethyl, Nitro und/oder Cyan substituierten Phenyloxyphenyl- oder Phenylaminophenylrest darstellt und alk Niederalkyliden oder Niederalkylen bedeutet, und ihrer Salze.
Ein Phenyloxyphenyl- bzw. Phenylaminophenylrest ist in erster Linie ein entsprechender 4-Phenyloxyphenyl- bzw.
4-Phenylaminophenylrest, wobei vorzugsweise die Phenyloxy- bzw. Phenylaminogruppierung substituiert ist, wobei sich ein Substituent in erster Linie in 4-Stellung dieser Gruppierung befindet.
Niederalkyl enthält vorzugsweise 1 bis 4 Kohlenstoffatome und ist z.B. Methyl, Äthyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl oder tert.-Butyl.
Halogen steht vorzugsweise für Halogen mit einer Atomnummer bis 35, insbesondere für Fluor oder Brom und in erster Linie für Chlor.
Niederalkyliden alk enthält insbesondere bis zu 4 Kohlenstoffatome und ist vorzugsweise unverzweigt, kann aber auch verzweigt sein; ein solcher Rest ist insbesondere Methylen, kann aber auch Äthyliden oder Isobutyliden sein.
Niederalkylen alk ist vorzugsweise unverzweigtes Nie- deralkylen, kann aber auch ein verzweigter Niederalkylenrest sein und bedeutet z.B. Äthylen, 1 - oder 2-Methyl-äthylen, 1,3-Propylen, 1-, 2- oder 3-Methyl-l,3-propylen oder 1,4-Butylen.
Salze von Verbindungen der Formel I sind in erster Linie pharmazeutisch verwendbare Salze mit Basen, wie Metalloder Ammoniumsalze, wie Alkali- oder Eralkalimetall, z.B.
Lithium-, Natrium-, Kaliumsalze, oder Salze mit Ammoniak.
Die neuen Verbindungen weisen wertvolle antiparasitäre Wirkungen, insbesondere gegen parasitäre Helminthen auf.
So zeigen sie bei sehr guter Verträglichkeit, z.B. in Versuchstieren, wie Maus, Ratte, Goldhamster, Mongolian Jird, Hund, Affe oder Huhn, Wirkungen gegen Nematoden, wie Ascariden, z.B. Ascaridia galli, Trichostrongyliden, z.B. Nippostrongylus brasiliensis oder Nematospiroides dubius, Ancylostomatiden, z.B. Necator americanus und Ancylostoma ceylanicum, und Strongyliden, gegen Cestoden, wie Hymenolepis nana, Anoplocephaliden und Taeniiden, und insbesondere gegen Trematoden, wie Fascioliden, z.B. Fasciola hepatica, und in erster Linie Schistosomen, z.B. Schistpsoma mansoni, Schistosoma japonicum und Schistosoma hematobium, ferner auch gegen die Erreger der Filariose, z.B.;Di- petalonema witei und Litomosoides carinii, sowie der Malaria, z.B. Plasmodium berghei.
Die Verbindungen der vorliegenden Erfindung weisen z.B. bei der Behandlung von Mäusen mit einer 6 bis 8 Wochen alten Infektion von Schistosoma mansoni, Schistosoma japonicum oder Schistosoma hematobium bei einmaliger Verabreichung p.o. (z.B. mit einer Magensonde} eine ED,, ab etwa 10 mg/kg und eine kurative Dosis ab etwa 25 mg/kg auf. Ferner erweisen sich die neuen Verbindungen bei der Behandlung der Filariose im Mongolian Jird als makro- und microfilarizid und zwar bei einmaliger Verabreichung p.o. in einer kurativen Dosis ab etwa 200 mg/kg bei Infektionen mit Dipetalonema witei und ab etwa 50 mg/kg bei solchen mit Litomosoides carinii.
Die neuen Verbindungen können deshalb zur Behandlung von Warmblütern bei Infektionen mit parasitären Helminthen, wie den obgenannten, insbesondere bei der Behandlung der Schistosomiase und der Filariose verwendet werden.
Die Erfindung betrifft in erster Linie Verbindungen der Formel I, worin R1 gegebenenfalls durch Methyl, Chlor, Trifluormethyl, Cyan und/oder besonders Nitro substituiertes 4-Phenyloxyphenyl oder 4-Phenylaminophenyl darstellt, wobei ein Substituent sich in erster Linie im Phenyloxy- bzw.
Phenylaminorest und vorzugsweise in 4-Stellung dieses Restes befindet und alk für den Rest der Formel -(CH2)n- steht, worin n für 1, 2 oder 3 steht, sowie Salze, in erster Linie pharmazeutisch verwendbare Salze, wie Alkalimetall-, Erdalkalimetall- oder Ammoniumsalze davon.
Die Erfindung betrifft insbesondere Verbindungen der Formel I, worin R1 4-(4-Nitro-phenyloxy)-phenyl oder 4-(4 -Nitro-phenylamino)-phenyl darstellt und alk für den Rest der Formel -(CH21,- steht, worin n die obige Bedeutung hat und insbesondere für 2 steht, sowie Salze, insbesondere pharmazeutisch verwendbare Salze, wie Alkalimetall-, Erdalkalimetall- oder Ammoniumsalze davon.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man eine an der Carboxylgruppe funktionell abgewandelte Verbindung der Formel I solvolysiert. Die Solvolyse ist insbesondere eine Hydrolyse, welche vorzugsweise in saurem Medium, wie z.B. in Salzsäure oder verdünnter Schwefelsäure, denen ein mit Wasser mischbares organisches Lösungsmittel, z.B. ein Niederalkanol, wie Methanol oder Äthanol, oder Dioxan beigefügt sein kann, nötigenfalls unter Erwärmen durchgeführt wird.
Zur Solvolyse geeignete an der Carboxylgruppe funktionell abgewandelte Verbindungen der Formel I sind z.B. deren durch Umsetzung eines Isothiocyanats der Formel Rl-N=C=S und einem Niederalkylester einer Verbindung der Formel HS-alk-COOH oder eines Alkalimetall-, z.B. Natrium-N-R,-dithiocarbaminates mit einem Niederalkylester einer Säure der Formel X-alk-COOH, worin X reaktionsfähig verestertes Hydroxy ist, oder einem solchen der Acrylsäure zugängliche Niederalkylester, weiter die analog zum zweitgenannten Verfahren, z.B. durch Addition von Alkalimetall-N-R1-dithiocarbaminaten an Acrylnitril oder Umsetzung der genannten Salze mit Halogenniederalkansäurenitrilen in an sich bekannter Weise herstellbaren Nitrile, sowie die aus den Nitrilen durch Addition von Wasser bzw.
partielle Hydrolyse oder durch Umsetzung von Alkalimetall-N-R1-dithiocarbaminaten mit Halogenniederalkansäureamiden oder mit Acrylamid ebenfalls in an sich bekannter Weise erhältlichen Amide.
Erhaltene Salze von Verbindungen der Formel I können durch Behandeln mit einer Säure, wie einer Mineralsäure, z.B. Salz- oder Schwefelsäure, in die freien Verbindungen übergeführt werden.
Erhaltene freie Verbindungen der Formel I können in an sich bekannter Weise in Salze übergeführt werden, z.B. durch Behandeln der freien Säure mit der äquivalenten Menge einer Base, wie einem Alkalimetall- oder Erdalkalimetallhydroxid oder Ammoniak, in einem geeigneten Lösungsoder Verdünnungsmittel, wie Dimethylformamid oder Dimethylsulfoxid, und Eindampfen des Reaktionsgemisches.
Das Verfahren umfasst auch diejenigen Ausführungsformen, wonach man einen Ausgangsstoff in Form eines unter den Reaktionsbedingungen erhältlichen rohen Reaktionsgemisches oder eine Reaktionskomponente in Form eines ihrer Salze einsetzt.
Vorzugsweise werden solche Ausgangsstoffe verwendet und die Reaktionsbedingungen so gewählt, dass man zu den vorstehend als besonders bevorzugt aufgeführten Verbindungen gelangt.
Die neuen Verbindungen werden vorzugsweise in Form von pharmazeutischen Präparaten, die sich in erster Linie zur oralen Verabreichung eignen, verwendet.
Die pharmazeutischen Präparate, die ebenfalls einen Gegenstand der vorliegenden Erfindung darstellen, enthalten von etwa 10% bis etwa 95% des pharmakologischen Wirkstoffs. Bevorzugt sind Präparate zur olaren Verabreichung, wie Dragees, Tabletten oder Kapseln, ferner Suspensionen, wobei die festen Präparate pro Dosiseinheitsform von etwa 0,1 g bis etwa 1,5 g, vorzugsweise von etwa 0,25 g bis etwa 1,0 g Wirkstoff enthalten, während flüssige Präparate von etwa 0,5% bis etwa 15% Wirkstoff aufweisen.
Die pharmazeutischen Präparate werden in an sich bekannter Weise, z.B. mittels konventioneller Misch-, Granulierund Dragierverfahren hergestellt. So kann man pharmazeutische Präparate, die zur oralen Anwendung geeignet sind, erhalten, indem man den Wirkstoff, der, falls erwünscht, in mikronisierter Form vorliegen kann, mit festen Trägerstoffen kombiniert, ein erhaltenes Gemisch gegebenenfalls granuliert, und das Gemisch bzw. Granulat, wenn erwünscht und/oder gegebenenfalls durch Zugabe von geeigneten Hilfsstoffen, zu Tabletten oder Dragee-Kernen verarbeitet.
Geeignete Trägerstoffe sind insbesondere Füllstoffe, wie Zucker, z.B. Lactose, Saccharose, Mannit oder Sorbit, Cellulosepräparate und/oder Calciumphosphate z.B. Tricalciumphosphat oder Calciumhydrogenphosphat, ferner Bindemittel, wie Stärkekleister unter Verwendung z.B. von Mais-, Weizen-, Reis- oder Kartoffelstärke, Gelatine, Traganth, Methylcellulose, Hydroxypropyl-methylcellulose, Natriumcarboxymethylcellulose und/oder Polyvinylpyrrolidon, und/oder, wenn erwünscht, Sprengmittel, wie die obgenannten Stärken, ferner Carboxymethylstärke, quervernetztes Polyvinylpyrrolidon, Agar, Alginsäure oder ein Salz davon, wie Natriumalginat. Hilfsmittel sind in erster Linie Fliessregulier- und Schmiermittel, z.B.
Kieselsäure, Talk, Stearinsäure oder Salze davon, wie Magnesium- oder Calciumstearat, und/oder Polyäthylenglykol. Dragee-Kerne werden mit geeigneten, gegebenenfalls Magensaft-resistenten Überzügen versehen, wobei man u.a. konzentrierte Zuckerlösungen, welche gegebenenfalls arabischen Gummi, Talk, Polyvinylpyrrolidon, Polyäthylenglycol und/oder Titandioxid enthalten, Lacklösungen in geeigneten organischen Lösungsmitteln oder Lösungsmittelgemischen oder, zur Herstellung von Magensaftresistenten Überzügen, Lösungen von geeigneten Cellulosepräparaten, wie Acetylcellulosephthalat oder Hydroxypropylmethylcellulosephthalat, verwendet. Den Tabletten oder Dragee-Überzügen können Farbstoffe oder Pigmente, z.B. zur Identifizierung oder zur Kennzeichnung verschiedener Wirkstoffdosen, beigefügt werden.
Weitere oral anwendbare pharmazeutische Präparate sind Steckkapseln aus Gelatine, sowie weiche, geschlossene Kapseln aus Gelatine und einem Weichmacher, wie Glycerin oder Sorbitol. Die Steckkapseln können den Wirkstoff in Form eines Granulats, z.B. im Gemisch mit Füllstoffen, wie Maisstärke, Bindemitteln und/oder Gleitmitteln, wie Talk oder Magnesiumstearat, und gegebenenfalls von Stabilisatoren, enthalten. In weichen Kapseln ist der Wirkstoff vorzugsweise in geeigneten Flüssigkeiten, wie fetten ölen, Paraffin öl oder flüssigen Polyäthylenglykolen, gelöst oder suspendiert, wobei ebenfalls Stabilisatoren zugefügt sein können.
Suspensionen für die orale Verwendung sind in erster Linie wässrige Suspensionen, die z.B. Stabilisatoren zur Erhöhung der Viskosität und Verzögerung der Sedimentation, wie wasserlösliche Celluloseäther, z.B. Carboxymethylcellulose und deren Salze, wie das Natriumsalz, oder Methylcellulose, und Netzmittel, wie Natriumlaurylsulfat oder Dioctylnatriumsulfosuccinat, oder nicht-ionogene Netzmittel, wie Polyoxyäthylensorbitanfettsäureester oder Polyäthylenglycol -polypropylenglycol-Copolymere enthalten und denen, falls erwünscht oder notwendig, Konservierungsmittel, wie 4-Hydroxybenzoesäure-niederalkylester, wie die entsprechenden Methyl-, Äthyl- oder n-Propylester, und/oder Aroma- und/ oder Süssstoffe beigemischt sein können.
Vorzugsweise verabreicht man einem Warmblüter von etwa 70 kg Gewicht eine einmalige Dosis von etwa 0,25 g bis etwa 1,5 g, vorzugsweise von etwa 0,5 g bis etwa 1,0 g des Wirkstoffs.
Die nachfolgenden Beispiele illustrieren die oben beschriebene Erfindung; sie sollen jedoch diese in ihrem Umfang in keiner Weise einschränken.
2 g N-[4-(4-Nitroanilino)-phenyl] -dithiocarbaminsäure- -(2-äthoxycarbony]äthyl)-ester werden mit 40 ml Dimethylformamid und 20 ml 2n-Natronlauge 8 Stunden bei 50 gerührt. Nach Abkühlen wird mit 2n-Salzsäure angesäuert und der Niederschlag nach Verdünnen mit Wasser abgenutscht.
Der erhaltene N-[4-(4-Nitroanilino)-phenyl]-dithiocarb aminsäure-(2-carboxyäthyl)-ester schmilzt bei 163-5 .
Das Ausgangsmaterial kann beisipelsweise folgendermassen hergestellt werden:
Zu einer Lösung von 4,7 g 4-(4-Nitroanilino);phenyliso- thiocyanat in 40 ml Dimethylformamid werden 11 g ;;-Mer- captopropionsäureäthylester eingetropft. Nach zweitägigem Rühren wird die Lösung in 500 ml Wasser eingerührt. Der entstandene Niederschlag von N-[4-(4-Nitroanilino)-phenyl]- -dithiocarbaminsäure-(2-äthoxycarbonyläthyl);ester wird abgenutscht, gut mit Wasser gewaschen und getrocknet; Smp. 177-9".

In analoger Weise wie in Beispiel 1 kann man unter Verwendung der geeigneten Ausgangsstoffe folgende Verbindungen herstellen: N-[4-(4-Nitroanilino)-phenyl-dithiocarbaminsäure-(carb- oxymethyl)-ester, z.B. durch Behandeln von 4-Isothiocyana to-4'-nitroester diphenylamin mit mercaptoessigsäureäthylestei und Hydrolyse des so erhältlichen N-[4-(4-Nitroanilino)-phe- nyl] -(äthoxycarbonylmethyl)-esters, N-[4-(4-Nitroanilino)-phenylj-dithiocarbaminsäure-(1 - -carboxyäthyl)-ester, N- [4-(4-Nitroanilino)-phenyl]-dithiocarbaminsäure-(3 - -carboxypropyl)-ester, N-[4-(2-Nitroanilino > -phenylj-dithiocarbaminsäure-(2- -carboxyäthyl)-ester,
