Abstract
Prepn. of new pleuromutilin derivs. of formula Q-O-CO-CH2-X-(CH2)n-NR2'R3' (I) comprises reacting a pleuromutilin deriv. of formula Q-O-CO-CH2-X-(CH2)n-NR2R3 (II) with an acylating agent of formula A-R5 (III) and opt. converting the prod. into an acid-addn. or quat. ammonium salt. In the formulae, Q is a gp. of formula (IV); R1 is ethyl or vinyl; n = 2-5; X is S, NR4 or Y-Ar-Z; R4 is H or QOCOCH2 Y and Z are both S or one is S and the other is O; Ar is phenylene; R2 and R3 form a 4-hydroxyalkyl-1-piperazinyl gp. or, when X = NR4 and n = 2, R2 and R4 form a piperazinyl gp. and R3 is lower hydroxyalkyl; R2' and R3' form a 4-acyloxyalkyl-1-piperazinyl gp. or, when X = NR4 and n = 2, R2' and R4 form a piperazinyl gp. and R3' is a lower (lower acyloxy) alkyl gp.; A is haloformyl or R5-CO-O-CO; R5 is lower alkyl. (I) have the same pharmacological properties as the cpds., described in the Parent Patent. A typical cpd. is 14-desoxy-14- 2- 4-(2-propionyloxyethyl)-1-piperazinyl -ethylthio- acetoxy mutilin. In an example, this is prepd. from 14-desoxy- 2- 4-(2-hydroxyethyl)-1-piperazinyl ethylthioacetoxy) mutilin and propionyl chloride.
