Die vorliegende Erfindung betrifft 4'-Demethyl-1-0[4,6-0 (3-thenyliden)-ss-D-glucopyranosyl]epipodophyllotoxin (Formel I, siehe Formelblatt) sowie Verfahren zu seiner Herstellung und pharmazeutische Zubereitungen davon.
Erfindungsgemäss gelangt man zu der Verbindung der Formel I, indem man 4'-Demethylepipodophyllotoxin- ss-D- glucopyranosid (Formel II) mit 3-Thiophencarboxaldehyd (Formel III) oder seinen niederen Acetalen, vorzugsweise dem Dimethylacetal, in Gegenwart eines geeigneten Katalysators umsetzt.
Als Katalysator werden vorzugsweise Sulfonsäuren, wie z.B.
p-Toluolsulfonsäure verwendet, es kommen aber auch Lewissäuren, wie wasserfreies Zinkchlorid, oder getrocknete Kationenaustauscher mit Sulfonsäuregruppen in der H@-Form in Frage. Die Aldehydkomponente der Formel III setzt man zweckmässig als Acetal ein, neben dem Dimethylacetal kann auch das Diäthylacetal Verwendung finden. Zur Erhöhung der Ausbeute an Kondensationsprodukt kann das gebildete Reaktionswasser bzw. der beim Einsatz von Acetalen entstehende niedere Alkohol durch azeotrope Destillation mit einem Lösungsmittel, vorzugsweise Acetonitril, bei tiefer Temperatur oder durch einen Katalysator, der wasserbindende Eigenschaften hat, entfernt werden.
Das erfindungsgemässe Verfahren wird ausgeführt, indem man 4'-Demethylepipodophyllotoxin- ,B-D-glucopyranosid der Formel II in die Verbindung der Formel III oder das entsprechende niedere Acetal gibt und den Katalysator zusetzt. Man arbeitet dabei unter Feuchtigkeitsausschluss und ggf. in Stickstoffatmosphäre. Die Verbindung der Formel II kann aber auch in einem trockenen inerten Lösungsmittel, z. B. Nitromethan oder Acetonitril, gelöst bzw. suspensiert und durch Zugabe der Verbindung der Formel III oder des entsprechenden niederen Acetals und eines Katalysators, ggf. in Stickstoffatmosphäre zur Reaktion gebracht werden.
Die Umsetzung wird bei Temperaturen von 20 bis 50", vorzugsweise bei Raumtemperatur vorgenommen und ist nach 1,5 bis 8 Stunden beendet. Zur Isolierung nimmt man das Reaktionsgemisch, ggf. nach Abfiltrieren des Katalysators, in einem mit Wasser nicht mischbaren Lösungsmittel, z.B. Chloroform, auf und schüttelt zur Entfernung evt. vorhandener wasserlöslicher Katalysatoren und Nebenprodukte mehrmals mit Wasser aus. Die organische Phase wird anschliessend getrocknet und im Vakuum eingedampft. Der Rückstand kann entweder durch Digerieren bzw. Macerieren mit Pentan, Petroläther oder Hexan oder direkt durch eine Vorchromatographie von einer überschüssigen Menge der Verbindung der Formel III oder eingesetzten Derivaten davon befreit werden.
Das rohe Kondensationsprodukt wird dann in bekannter Weise durch Chromatographie und/oder Umkristallisieren bzw.
Umfällen gereinigt.
Eine vorzugsweise Ausführungsform des Kondensationsverfahrens ist die Umacetalisierung. Sie wird durchgeführt, indem man die Aldehydkomponente der Formel III als Di methylacetal einsetzt, wobei man ca. 3 Mol dieses Acetals mit
1 Mol der Verbindung der Formel II in Gegenwart von p
Toluolsulfonsäure (0,04 bis 0,06 Mol pro Mol der Verbindung
II) bei 20 zur Reaktion bringt. Als Lösungsmittel für diese
Umacetalisierung ist Acetonitril optimal geeignet. Das entste hende Methanol wird durch 2-3-maliges Abdestillieren in Form eines azeotropen Acetonitril-Methanol-Gemisches im partiel len Vakuum bei 20 aus dem Gleichgewicht entfernt. Der
Verlust an Lösungsmittel wird durch Zugabe von frischen
Acetonitril ausgeglichen. Die Reaktion ist nach 1,5 Stunden beendet.
Das als Ausgangsmaterial für die Umacetalisierung verwen dete 3-Thiophencarboxaldehyddimethylacetal wird aus dem Aldehyd der Formel III durch Umsetzung mit Orthoameisensäuretrimethylester und Methanol in Gegenwart katalytischer Mengen Chlorwasserstoff gewonnen.
Die neue Verbindung der Formel I zeigt in der pharmakologischen Prüfung wertvolle Eigenschaften.
4'-Demethyl- 1 -0[4,6-0 -(3 -thenyliden)- ss-D -glucopyranosyl]epipodophyllotoxin zeigt ausgeprägte cytostatische Wirkung, wie sie in vitro an einem aus einem Maustumor herrührenden Zellstamm (P-815-Mastocytom) sowie in vivo an den transplantierbaren Nagertumoren, der Leukämie L-1210 der Maus und dem Walker-Tumor der Rate, gezeigt werden konnte.
Die Verbindung zeigt in vitro eine 50%ige Hemmung der Zellvermehrung bei einem Gehalt von 0,014 mg/l, in vivo werden befriedigende Resultate erreicht mit einem Dosenbereich von 1-8 mg pro kg Körpergewicht des Testtieres, wobei die zu verwendende Dosis naturgemäss variiert je nach Administration und des zu behandelnden Zustandes.
Die neue Verbindung zeichnet sich zudem durch immunosuppressive Eigenschaften aus und zwar hinsichtlich humoraler wie auch zellgebundener Immunitätsreaktionen.
Die Wirkung betreffend humorale Immunität konnte an der Maus durch lokale Hämolyse im Gel sowie im Hämagglutinationstest festgestellt werden.
Die Beeinflussung der zellgebundenen Immunität konnte an der Ratte durch wirksame Unterdrückung der experimentellen allergischen Encephalomyelitis bei mehrmaliger Applikation (intraperitoneal oder peroral) in Dosen zu je 4 mg/kg pro Tag gezeigt werden; weiterhin durch den Tuberkulintest beim Meerschweinchen, der auf der zellulären Immunität vom verspäteten Überempfindlichkeitstypus beruht. Hierbei hat eine einmalige subcutane Behandlung der Meerschweinchen mit dem Wirkstoff eine deutliche Hemmung der Hautreaktion sowohl nach 24 Stunden als auch nach 48 Stunden zur Folge.
Die Verbindung hemmt die Bildung von Antikörpern und sensibilisierten Zellen und ist demnach zur Unterdrückung unerwünschter Immunreaktionen geeignet. Im allgemeinen gelangt man bei Testtieren zu guten Ergebnissen mit einer Dosis von 3-10 mg pro kg Körpergewicht. Die zu verabreichende Dosis hängt dabei von der gewünschten Wirkung und der Applikationsart ab. Die Tagesdosis für grössere Säugetiere liegt bei etwa 25-500 mg. Für orale Applikationen enthalten die Teildosen etwa 10-250 mg des Wirkstoffes neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Die neue Verbindung kann als Arzneimittel allein oder in entsprechenden Arzneiformen für orale, enterale oder parenterale Verabreichung verwendet werden. Zwecks Herstellung geeigneter Arnzeiformen wird diese mit den üblichen organischen oder anorganischen, pharmakologisch indifferenten Hilfsstoffen verarbeitet.
In dem nachfolgenden Beispiel, welches die Ausführung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelzpunkte sind auf dem Koflerblock bestimmt und korrigiert. Dünnschichtchromatogramme wurden auf Kieselgel G-Platten mit dem Fliessmittel Chloroform/Methanol (95:5) ausgeführt; Sichtbarmachung erfolgte durch Besprühen mit einer Lösung von 0,2% Cer(IV)-sulfat in 50proz. Schwefelsäure und Erwärmen auf 110-130". Präparative Chromatogramme wurden an Kieselgel Merck, Korngrösse 0,05-0,2 mm, durchgeführt.
Die Ausgangsprodukte sind, soweit nicht beschrieben, ihrer ;Herstellung nach bekannt.

Beispiel 4'-Demethyl-1 -0[4, 6,0- (3-thenyliden)- p-D-gluco pyrano- syllepipodophyllotoxin
7,91 g (50 m Mol) 3 -Thiophencarboxaldehyddimethylacetal werden in 170 ml trockenem Acetonitril gelöst. Man setzt 9,56 g (17 m Mol) 4'-Demethylepipodophyllotoxin- B-D- glucopyranosid und 0,15 g (0,8 m Mol) p-Toluolsulfonsäuremonohydrat zu und rührt das Gemisch unter Feuchtigkeitsausschluss bei 20 . 30 Minuten nach Reaktionsbeginn werden aus der klaren, gelblichen Reaktionslösung bei 60 mm Druck und einer Badtemperatur von 20 10 ml Lösungsmittel abdestilliert und durch frisches Acetonitril ersetzt. Nach weiteren.15 Minuten werden nochmals 25 ml Acetonitril-Methanol-Gemisch abdestilliert und durch frisches Acetonitril ersetzt.
Der Verlauf der Umacetalisierung wird dünnschichtchromatographisch auf Kieselgel G-Platten, Fliessmittel Chloroform/ Methanol (95:5), verfolgt. Nach insgesamt 1,5 Stunden verdünnt man mit 1000 ml Chloroform, wäscht rasch dreimal mit je 150 ml Wasser neutral, trocknet die organische Phase über Natriumsulfat, dampft im Vakuum ein und trocknet den Rückstand im Hochvakuum bei 60". Der so erhaltene gelbe Rückstand wird in 50 ml warmem Aethylalkohol gelöst, man versetzt mit 30 ml Aether und lässt bei 0 kristallisieren. Das Rohkristallisat, Smp. 244-248", wird in 100 ml warmem Chlorform/Methanol (9:1) gelöst, die Lösung klar filtriert und im Vakuum eingedampft. Den resultierenden Schaum löst man in 210 ml siedendem Aethanol und lässt bei 0 kristallisieren.
Man erhält 4'-Demethyl-1-0[4,6-0- (3-thenyliden)- ss-Dglucopyranosyl]epipodophyllotoxin in Form farbloser Polyeder vom 245-250", [n] 20 =-109,3 (c=0,S06inChloro- form/Methanol 9:1). D
Die Mutterlaugen können durch Chromatographie an der 50-fachen Menge Kieselgel und Elution mit Chloroform/Methanol (97:3) aufgearbeitet werden.
Das als Ausgangsmaterial verwendete 3-Thiophencarboxaldehyddimethylacetal kann wie folgt hergestellt werden:
6,1 g (54 m Mol) 3-Thiophencarboxaldehyd, 7,5 g (71 m Mol) Orthoameisensäuretrimethylester, 4,5 g (142 m Mol) abs.
Methanol und 15 mg Chlorwasserstoff (als 1 N Lösung in abs.
Aether) werden zusammengegeben. Man lässt die schwach exotherme Reaktion unter Feuchtigkeitsausschluss während 15 Minuten abklingen. Anschliessend wird 20 Minuten unter Rückfluss erwärmt, dann auf Zimmertemperatur abgekühlt und bei 20 im Vakuum auf ca. 2/3 des ursprünglichen Volumens eingeengt. Durch Zugabe von Kaliumcarbonat wird die Säure neutralisiert, man filtriert die Kaliumsalze ab und unterwirft das Filtrat einer fraktionierten Destillation. Im schwachen Vakuum wird zunächst überschüssiges Methanol und Orthoameisensäuretrimethylester abdestilliert, danach im vollen Vakuum das Acetal. Das 3-Thiophencarboxaldehyddimethylacetal zeigt den Siedepunkt Kp13=830. Es ist frei von 3-Thiophencarboxaldehyd.
