Die Erfindung betrifft ein Verfahren zur Herstellung von heterocyclischen Verbindungen der allgemeinen Formel

Vor- und nachstehend wird unter einem niederen Rest insbesondere ein solcher Rest mit bis zu 7 C-Atomen, vor allem bis zu 4 C-Atomen verstanden.
Niederalkylreste sind Reste mit bevorzugt bis zu 7 C-Atomen, vor allem bis zu 4 C-Atomen und sind z.B. Methyl, Äthyl, n-Propyl, i-Propyl oder unverzweigtes oder an beliebiger Steht gebundenes oder verzweigtes Butyl, Pentyl, Hexyl oder Heptyl.
Gegebenenfalls substituierte Phenylniederalkylreste sind Niederalkylreste mit bis zu 7- C-Atomen, vor allem bis zu 4 C-Atomen, die in beliebiger Stelle durch gegebenenfalls substituierte Phenylgruppen substituiert sind. Als Phenylsubstituenten kommen vor allem Niederalkyl-, Niederalkoxygruppen, der Trifluormethylrest und Halogenatome in Frage.
Als Beispiele sind 3-Phenyl-apropyl oder vor allem Benzyl und 2-Phenyläthyl zu nennen.
Carboxyniederalkylreste sind Niederalkylreste mit bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, die in beliebiger Stelle durch Carboxy substituiert sind, wie z.B.
3-Carboxy-n-propyl, 4-Carboxy-n-butyl und vor allem Carboxymethyl und 2-Carboxyäthyl.
Funktionell abgewandeltes Carboxyniederalkyl ist z.B.
verestertes Carboxyniederalkyl, amidiertes Carboxyniederalkyl oder Cyanoniederalkyl.
Verestertes Carboxyniederalkyl ist z.B. mit einem aliphatischen Alkohol verestertes Carboxyniederalkyl. Der Niederalkylteil des veresterten Carboxyniederalkyls hat bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome. Aliphatische Alkohole sind solche, in denen die Hydroxylgruppe an ein C-Atom gebunden ist, welches nicht Glied eines aromatischen Systems ist. Geeignete aliphatische Alkohole sind z.B. Cycloalkanole, wie solche mit 3-7, insbesondere 5-7 Ringgliedern, z.B.
Cyclopropanol, Cyclopentanol, Cyclohexanol und Cy eloheptanol, Cydoalkylniederalkanole, die z.B. obige Cycloalkylteile enthalten, wie Cyclopentyl-methanol, Cyclohexyl Methanol, 2-Cyclohe,xyl-äthanol und Cycloheptyl-methanol, Phenylniederalkanole, wie 2-Phenyläthanol und Benzylalko hol, wobei Phenylreste auch durch Halogen, Niederalkyl und oder Niederalkoxy, wie die oben genannten substituiert sein können, und insbesondere Niederalkanole, wie n-Propanol, iso-Propanol, geradkettiges oder verzweigtes Butanol, Pentanol, Hexanol oder Heptanol, und insbesondere Methanol oder Äthanol. Verestertes Carboxyniederalkyl ist vor allem Methoxycarbonylmethyl, 2-Methoxycarbonyl-äthyl, Äthoxycarb onylmethyl und 2-Äthoxycarbonyl-äthyl.
Amidiertes Carboxyniederalkyl ist substituiertes oder unsubstituiertes Carbamoylniederalkyl. Der Niederalkylteil des amidierten Carboxyniederalkyls hat bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome. Substituiertes Carbamoyl hat z.B. die Formel -CONR;,R6, worin Rs Wasserstoff oder Niederalkyl ist, R6 Niederalkyl ist oder Rs und R6 zusammen Niederalkylen, Oxaniederalkylen, Thianiederalkylen oder Azaniederalkylen ist. Niederalkylen ist verzweigtes oder insbesondere geradkettiges Niederalkylen mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Oxaniederalkylen ist verzweigtes oder insbesondere geradkettiges Oxaniederalkylen mit insbesondere 4 oder 5 C-Atomen in der Oxaalkylenkette.
Thianiederalkylen ist verzweigtes oder insbesondere geradkettiges Thianiederalkylen mit insbesondere 4 oder 5 C-Atomen in der Thiaalkylenkette. Azaniederalkylen ist verzweigtes oder geradkettiges Azaniederalkylen mit insbesondere 2-6, vor allem 4-6 C-Atomen in der Azaalkylenkette. Amidiertes Carboxyniederalkyl ist demnach vor allem Carbamoylmethyl, 2-Carbamoyläthyl, N,N-Dimethylcarb amoylrnethyl, 2-(N,N-Dimethylcarbamoyl)-äthyl, N,N-Di äthylcarbamoylmethyl, 2- (N,N-Diäthylcarbamoyl) -äthyl, Pyrrolidinocarbonylmethyl, 2-Pyrrolidinocarbonyl-äthyl, Piperidinocarbonylmethyl, 2-Piperidinocarbonyläthyl, Morpholinocarbonylmethyl, 2-MorphoXinocarbonyläthyl, Thiomorpholinocarbonylmethyl, 2-Thiomorpholinocarbonyläthyl,

Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. Die Hauptwirkung der substituierten Pyridine, Pyrazine und Pyrimidine besteht in einer Blockade adrenergischer ;a-Rezeptoren, die sich zum Beispiel als Hemmwirkung gegenüber Effekten von bekannten p-Rezep- toren-Stimulatoren in verschiedenen Organen nachweisen lässt: Hemmung der Isoproterenol.Tachykardie am isolierten Meerschweinchenherzen und der Isoproterenol-Relaxation an der isolierten Meerschweinchentrachea bei Konzentrationen von 0,001 bis 3 ,zg/ml, Hemmung der Isoproterenol Tachykardie und -vasodilatation an der narkotisierten Katze bei intravenöser Gabe von 0,01 bis 30 mg/kg i.v.
Die genannten Verbindungen gehören entweder der Klasse der nicht cardioselektiven p-Rezeptorenblocker an, d.h. sie blockieren die p-Rezeptoren an den Gefässen bzw. in der Trachea in ähnlichen oder sogar kleineren Dosen bzw. Konzentrationen, als die 53-Rezeptoren im Herzen, oder sie gehören der Klasse der sogenannten cardioselektiven -Rezeptorenblocker an, d.h. sie blockieren die p-Rezeptoren des Herzens schon in einem Dosen- bzw. Konzentrationsbereich, welcher noch keine Blockade der p-Rezeptoren in den Gefässen bzw. in der Trachea bewirkt.
Als zusätzliche Eigenschaft besitzt ein Teil dieser Verbindungen eine sogenannte intrinsic sympathominetic activity (ISA) , das heisst, diese Verbindungen bewirken neben der p-BIockade (= Hauptwirkung) eine partielle 48-Stimulation. Die Hauptwirkung von unsubstituiertem Pyrazin und Pyrimidin besteht in einer Stimulation der adre nergischen p-Rezeptoren, die sich z.B. am Herzen als positiv inotrope und positiv chronotrope Wirkung nachweisen lässt.
Die genannten Verbindungen steigern an isolierten Meerschweinchenvorhöfen Herzfrequenz und myocardiale Kon traktionskraft in Konzentrationen von 0,01 bis 1 k tg/ml und an der narkotisierten Katze bei intravenöser Gabe von 0,001 bis 0,1 mg/kg i.v. In Konzentrationen, die deutlich höher sind, als die für eine ss-Stimulation benötigen, weisen diese Verbindungen auch -Rezeptoren-blockierende Eigenschaften auf 2-(2'-Hydroxy-3'-isopropylaminopropoxy)-pyrimidin und 2-(2'-Hydroxy-3'-isopropyl-aminopropoxy)-pyrazin unterscheiden sich jedoch qualitativ deutlich von bekannten ,8-Rezeptorenstimulatoren, indem sie an der narkotisierten Katze erst in einer Dosis von 1 mg/kg i.v.
den arteriellen Blutdruck senken, also in einem Dosenbereich, der deutlich über demjenigen liegt welcher für eine Steigerung von myocardialer Kontraktionskraft und Herzfrequenz benötigt wird.
An der isolierten Meerschweinchentrachea weisen die Verbindungen in einer Konzentration von 10 ,eg/ml noch keine relaxierende Wirkung auf. Aufgrund dieser Eigenschaften können diese Verbindungen als cardioselektive ,8-Rezeptoren- stimulatoren bezeichnet werden.
Die neuen Vebindungen können daher zur Behandlung von Erkrankungen des Herz- und Kreislaufsystems verwendet werden.
Die ,-Rezeptorenblocker können z.B. zur Therapie von Angina pectoris, Hypertonie und Herzrhythmusstörungen Anwendung finden. Die cardioselektiven Präparate bringen gegenüber den nicht cardioselektiven den Vorteil, dass in den Dosen, welche für eine Blockade der p-Rezeptoren des Herzens benötigt werden, noch keine Blockade von ,8-Rezepto- ren in anderen Organen zu erwarten ist. Das Risiko, unerwünschte Nebenwirkungen, wie z.B. einen Bronchospasmus auszulösen, ist demnach sehr gering. Im Gegensatz zu den cardioselektiven blockieren die nicht cardioselektiven Präparate entweder die P-Rezeptoren in allen Organen etwa gleich stark oder in bestimmten Organen, (wie z.B. in den Gefässen) bevorzugt.
Die ss-Rezeptorenstimulatoren können als Cardiotonika zur Behandlung der Herzmuskel-Insuffizienz (allein oder in Kombination mit anderen Präparaten, wie z.B. Herzglycosiden) Anwendung finden. Gegenüber bekannten frRezep- torenstimulatoren weisen diese Verbindungen folgende Vorteile auf: Aufgrund der pharmakologisch nachgewiesenen Cardioselektivität kann erwartet werden, dass die myocardiale Kontraktionskraft gesteigert wird, ohne dass es gleichzeitig zu einer unerwünschten Blutdrucksenkung kommt.
Ferner ist nur mit einem unwesentlichen Anstieg der Herzfrequenz zu rechnen, weil die, als Folge einer Blutdrucksenkung, auftretende reflektorische Tachykardie wegfällt. Sie können aber auch als wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere pharmazeutisch wirksamer Verbindungen verwendet werden.

Niederalkenyl hat bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome, wie Vinyl, 2-Methylvinyl, Methallyl und besonders Allyl.
Geeignete Substituenten für gegebenenfalls substituiertes Phenyl sind Halogen, Trifluormethyl, Niederalkyl, Niederalkenyl, Niederalkoxymethyl, Niederalkoxy und Niederalkenyloxy. Gegebenenfalls substituiertes Phenyl ist somit vor allem Phenyl, Chlorphenyl, Bromphenyl, Trifluormethylphenyl, Tolyl, Methoxymethylphenyl, Methoxyphenyl, Ätoxyphenyl und Allyloxyphenyl.
Niederalkoxyreste haben bevorzugt bis zu 7 C-Atome vor allem bis zu 4 C-Atome, wie z.B. Äthoxy, Propoxy, i-propoxy, gerades oder verzweigtes, an beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl- oder HeptyIoxy oder vor allem Methoxy.
Niederalkoxyniederalkyl hat im Niederalkylteil des Niederalkoxyteils bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie iso- oder n-Propyl, gerades oder verzweigtes, in beliebiger Stelle gebundenes Butyl, Pentyl, Hexyl oder Heptyl, besonders Äthyl und vor allem Methyl. Der den Mederalkoxyteil tragende Niederalkylteil hat bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome. Beispielsweise ist Niederalkoxyniederalkyl Methoxy, 2-Äthoxyäthyl, 3 -Methoxy-n-propyl, 3-Äthoxy-n-propyl, 4-Methoxy-n-butyl oder vor allem 2-Methoxyäthyl und Äthoxymethyl.
NiederalkoxyniederalkenyP sind Niederalkenylreste mit bevorzugt bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen, die an beliebiger Stelle durch Niederalkoxy substituiert sind. Mederalkoxy hat im Niederalkylteil bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome. Niederalkoxyniederalkenyl ist somit bevorzugt 2-Methoxyvinyl, 2-Äthoxyvinyl und vor allem 3-Methoxyallyl und 3-Äthoxyallyl.
Niederalkenyloxyreste sind Reste mit bevorzugt bis zu 7 C-Atome, insbesondere mit 3 oder 4 C-Atome, wie der Methallyloxy- oder vor allem Allyloxyrest.
Niederaikoxyniederalkoxv hat in den Niederalkylteilen je bis zu 7 C-Atome, vor allem bis zu A C-Atome und ist z.B. Methoxymethoxy, Äthoxymethoxy, 2-Methoxy-äthoxy, 4-Methoxy-n-butoxy und insbesondere 3-Methoxy-n-propoxy.
Niederalkylthioreste haben bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie z.B. Äthylthio, n-Propylthio, n-Butylthio, i'-Propylthio oder insbesondere Methylthio.
Niederalkylthioniederalkoxyreste haben in den beiden Niederalkylteilen bevorzugt je bis zu 7 C-Atome, vor allem je bis zu 4 C-Atome, wie z.B. Methylthiomethoxy, Äthylthiomethoxy, Propylthiomethoxy, 3-Methylthiopropoxy, 3 -Äthylthiopropoxy, 3 -Propylthiopropoxy und vor allem 2-Methylthioäthoxy, 2-Äthylthioäthoxy und 2-Propylthiopropoxy.
Niederalkylaminoreste sind Reste mit bevorzugt bis zu 7 C-Atomen, insbesondere bis zu 4 C-Atomen, wie z.B.
Äthyl-, Propyl-, i-Propyl-, gerades oder verzweigtes, an beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl- oder Heptylamino oder vor allem Methylamino.
Diniederalkylaminoreste sind Reste mit bevorzugt bis zu 7 C-Atomen, insbesondere bis zu 4 C-Atomen in jedem der Niederalkylteile. Die beiden Niederalkylreste sind voneinander unabhängig und bilden zusammen mit dem Stickstoffatom Reste wie z.B. Diäthyl-, Methyläthyl-, Diäthyl-, Dipropyl-, Dibutylamino oder insbesondere Dimethylamino.
Niederalkyleniminoreste sind Reste mit z.B. 4 bis 8 Ringgliedern, wobei der Niederalkylenteil verzweigtes oder insbesondere geradkettiges Niederalkylen ist mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Beispiele sind Pyrrolidino oder Piperidino.
Hydroxyniederalkyleniminoreste sind Reste mit z.P 4-8 Ringgliedern, wobei der hydroxysubstituierte Niederalkylen- teil verzweigtes oder insbesondere geradkettiges Niederalkylen ist, mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Beispiele sind 4-Hydroxypiperidino oder 3-Hydroxypyrrolidino.
Oxaniederalkylenimino ist verzweigtes oder insbesondere geradkettiges Oxaniederalkylenimino mit insbesondere 4 oder 5 C-Atomen in der Oxaalkylenkette. Als Beispiel ist insbesondere Morpholino zu nennen.
Thianiederalkylenimino ist verzweigtes oder insbesondere geradkettiges Thianiederalkylenimino mit insbesondere 4 oder 5 C-Atomen in der Thiaalkylenkette. Als Beispiele sind insbesondere Thiomorpholino und 2,6-Dimethylthiomorpholino zu nennen.
Azaniederalkylenimino ist verzweigtes oder insbesondere geradkettiges Azaniederalkylenimino mit insbesondere 2-6, vor allem 4-6 C-Atomen in der Azaalkylenkette. Beispiele sind Piperazino, N'-Methylpiperazino oder N'-(P-Hydroxy- äthyl)-piperazino.
Acylamino ist z.B. niederes Alkanoylamino mit bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen im Niederalkylteil oder gegebenenfalls substituiertes Aroylamino, wie gegebenenfalls substituiertes Benzoylamino oder gegebenenfalls substituiertes Arylniederalkanoylamino oder Niederalkoxycarbonylamino.
Niedere Alkanoylaminoreste sind z.B. n-Propionylamino, n-Butyrylamino, n-Valeryiamino, n-Hexanoylamino, n-Heptanoylamino oder vor allem Acetylamino. Gegebenenfalls substituierte Benzoylaminoreste sind z.B. Benzoylamino oder Niederalkoxybenzoylamino, wie n-Propoxybenzoylamino und vor allem Methoxy- und Äthoxybenzoylamino, oder Niederalkylbenzoylamino, wie n-Propyl-, gerades oder verzweigtes, in beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl- oder Heptylbenzoylamino und vor allem Methyl- und Äthylbenzoylamino, oder Trifluormethylbenzoylamino oder Halobenzoylamino, wie Fluor-, Brom- und ganz besonders Chlorbenzoylamino.
Gegebenenfalls substituierte Arylniederalkanoylaminoreste sind Niederalkanoyiaminoreste, die an beliebiger Stelle des Niederalkylteils z.B. gegebenenfalls substituierte Phenylgruppen tragen. Die Phenylgruppen können die gleichen Substituenten tragen, wie oben für gegebenenfalls substituierte Benzoylaminoreste erwähnt. Als Beispiele für gegebenenfalls substituiertes Arylniederalkanoylamino sind vor allem Phenylacetylamino, 3 -Phenyl-n-propionyl- amino, 4-Phenyl-n-butyrylamino, Chlorphenylacetylamino und Bromphenylacetylamino zu nennen. Niederalkoxycarboylamino hat im Niederalkylteil von Niederalkoxy vorzugsweise bis zu 7 C-Atome und vor allem bis zu 4 C-Atome und ist z.B. n-Propoxycarbonylamino, n-Butoxycarbonylamino, i-Propoxycarbonylamino, tert.-Butoxycarbonylamino und vor allem Methoxycarbonylamino und Äthoxycarbonylamino.
Acylaminoniederalkylreste sind Niederalkylreste mit vorzugsweise bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen, die in beliebiger Stelle durch Acylaminoreste substituiert sind. Acylamino ist z.B. niederes Alkanoylamino mit bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen im Mederalkylteil oder gegebenenfalls substituiertes Aroylamino, wie gegebenenfalls substituiertes Benzoylamino oder gegebenenfalls substituiertes Arylniederalkanoylamino oder Niederalkoxycarbonylamino.
Niedere Alkanoylaminoreste sind z.B. n-Propionylamino, n-Butyrylamino, n-Valerylamino, n-Hexanoylamino, n-Heptanoylamino oder vor allem Acetylamino. Gegebenenfalls substituierte Benzoylaminoreste sind z.B.
Benzoylamino oder Niederalkoxybenzoyiamino, wie n-Propoxybenzoylami- no und vor allem Methoxy- und Athoxybenzoylamino, oder Niederalkylbenzoylamino, wie n-Propyl-, gerades oder ver- zweigtes, in beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl- oder Heptylbenzoylamino und vor allem Methyl und Athyllbenzoylamino, oder Trifluormethylbenzoylamino oder Halobenzoylaminv, wie Fluor-, Brom- und ganz besonders Chlorbenzoylamino. Gegebenenfalls substituierte Arylniederalkanoylaminoreste sind Niederalkanoylamino- reste,
die an beliebiger Stelle des Niederalkylteils z.B. gegebenenfalls substituierte Phenylgruppen tragen. Die Phenyl- gruppen können die gleichen Substituenten tragen, wie oben für gegebenenfalls substituierte Benzoylaminoreste erwähnt.
Ale Beispiele für gegebenenfalls substituiertes Arylniederalkanoylamino sind vor allem Phenylacetylamino, 3-Phenyl -n-propionylamino, 4-Phenyl-n-butyrylamino, Chlorphenylacetylamino und Bromphenylacetylamino zu nennen. Nieder alkoxycarbonylamino hat im Niederalkylteil von Niederalkoxy vorzugsweise bis zu 7 C-Atome und vor allem bis zu 4 C-Atome und ist z.B. n-Propoxycarbonylamino, n-Butoxycarbonylamino, i-Propoxycarbonylamino, tert.-Butoxycarbonylamino und vor allem Methoxycarbonylamino und Äthoxycarbonylamino. Als Beispiele für Acylaminoniederalkylreste sind demnach vor allem Acetylaminomethyl,
2-Acetylamino-äthyl, Benzoylaminomethyl, 2-Benzoylamino- äthyl, Methoxybenzoylaminomethyl, 2-Methoxybenzoyl- aminoäthyl, Athoxybenzoylaminomethyl, 2-Äthoxybenzoyl- aminoäthyl, Methylbenzoylaminomethyl, 2-Methylbenzoyl- aminoäthyl, Athylbenzoylaminomethyl, 2-Athylbenzoylami- noäthyl, Chlorbenzoylaminomethyl, 2-Chlorbenzoylamino- äthyl, Phenylacetylaminomethyl, 2-Phenylacetylaminoäthyl, Chlorphenylacètylaminomethyl,

Acylaminoniederalkenyl trägt im Niederalkenylteil bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome, während Acylamino z.B. Niederalkanoylamino, gegebenenfalls substituiertes Benzoylamino oder gegebenenfalls substituiertes Phenylniederalkanoylamino oder Mederalkoxy- carbonylamino ist. Als Beispiele für Acylaminoniederalkenyl sind vor allem 2-Acetylaminovinyl, Benzoylaminovinyl, Phenylacetylaminovinyl, 3-Acetylaminoallyl, 3-Benzoylami noallyl, 3-Phenylacetylaminoallyl und 3-Methoxycarbonylaminoallyl zu nennen.

Niederalkylreste sind Reste mit bevorzugt bis zu 7 C-Atomen, vor allem bis zu 4 C-Atomen und sind z.B. Methyl, äthyl, n-Propyl, i-Propyl oder unverzweigtes oder an beliebiger Stelle gebundenes oder verzweigtes Butyl, Pentyl, Hexyl oder Heptyl.
Hydroxyniederalkyl hat im Niederalkylteil vorzugsweise bis zu 7 C-Atome, vor allem bis zu 4 Ç-Atome und ist z.B.
3-Hydroxy-propyl, 2-Hydroxy-propyl, l-Methyl-2-hydroxy äthyl oder unverzweigtes oder an beliebiger Stelle gebunde nes oder verzweigtes Hydroxybutyl, Hydroxypentyl, Hydroxyhexyl, Hydroxyheptyl und vor allem Hydroxymethyl und 2-Hydroxyäthyl.
Niederalkenyl hat bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome, wie Vinyl, 2-Methylvinyl, Methallyl und besonders Allyl.
Geeignete Substituenten für gegebenenfalls substituiertes Phenyl sind Halogen, Trifluormethyl, Niederalkyl, Niederalkenyl, Niederalkoxymethyl, Niederalkoxy und Niederalkenyloxy. Gegebenenfalls substituiertes Phenyl ist somit vor allem Phenyl, Chlorphenyl, Bromphenyl, Trifluormethylphenyl, Tolyl, Methoxymethylphenyl, Methoxyphenyl, Äth- oxyphenyl und Allyloxyphenyl.
Niederalkoxyreste haben bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie z.B. Äthoxy, Propoxy, i-Propoxy, gerades oder verzweigtes, an beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl- oder Heptyloxy oder vor allem Methoxy.
Niederalkoxyniederalkyl hat im Niederalkylteil des Niederalkoxyteils bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie iso- oder n-Propyl, gerades oder verzweigtes, in beliebiger Stelle gebundenes Butyl, Pentyl, Hexyl oder Heptyl, besonders Äthyl und vor allem Methyl. Der den Niederalkoxyteil tragende Niederalkylteil hat bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome. Beispielsweise ist Niederalkoxyniederalkyl, Methoxymethyl, Äthoxymethyl, 3-Methoxy-n-propyl, 3-Äthoxy-n-propyl, 4-Methoxy-n-butyl oder vor allem 2-Methoxyäthyl und 2-Äthoxyäthyl.
Niederalkoxyniederaikenyl sind Niederalkenylreste mit bevorzugt bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen, die an beliebiger Stelle durch Niederalkoxy substituiert sind. Niederalkoxy hat im Niederalkylteil bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome. Niederalkoxy niederalkenyl ist somit bevorzugt 2-Methoxyvinyl, 2-Äthoxyvinyl und vor allem 3-Methoxyallyl und 3-Äthoxyallyl.
Niederalkenylxyreste sind Reste mit bevorzugt bis zu 7 C-Atome, insbesondere mit 3 oder 4 C-Atome, wie der Methallyloxy- oder vor allem Allyloxyrest.
Niederalkoxyniederalkoxy hat in den Niederalkylteilen je is zu 7 C-Atome, vor allem bis zu 4 C-Atome und ist z.B.
Methoxymethoxy, Äthoxymethoxy, 2-Methoxy-äthoxy, 4-Methoxy-n-butoxy und insbesondere 3-Methoxy-n-propoxy.
Niederalkylthioniederalkoxy hat in den Niederalkylteilen je bis zu 7 C-Atome, vor allem bis zu 4 C-Atome und ist z.B. Methylthiomethoxy, Äthylthiomethoxy, 3-Methylthiopropoxy, 4-Methylthiobutoxy und insbesondere 2-Methylthioäthoxy, 2-Äthylthioäthoxy oder 2-(n-Propylthio) > äthoxy.
Niederalkylthioreste haben bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie z.B. Äthylthio, n-Propylthio, n-Butylthio, i-Propylthio oder insbesondere Methylthio.
Niederalkylthioniederalkyl hat in den Niederalkylteilen je bis zu 7 C-Atome, vor allem bis zu 4 C-Atome und ist z.B. Methylthiomethyl, Äthylthiomethyl, 3-Methylthiopropyl, 4-Methylthiobutyl und insbesondere 2-Methylthioäthyl, 2-Äthyithioäthylo oder 2-(n-Propylthio)-äthyl.
Niederalkylaminoreste sind Reste mit bevorzugt bis zu 7 C-Atomen, insbesondere bis zu 4 C-Atomen, wie z.B.Äthyl-, Propyl-, i-Propyl-, gerades oder verzweigtes, an beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl oder Heptylamino oder vor allem Methylamino.
Diniederalkylaminoreste sind Reste mit bevorzugt bis zu 7 C-Atomen, insbesondere bis zu 4 C-Atomen in jedem der Niederalkylteile. Die beiden Niederalkylreste sind vonein ander unabhängig und bilden zusammen mit dem Stickstoffatom Reste wie z.B. Diäthyl-, Methyläthyl-, Dipropyl-, Dubutylamino oder insbesondere Dimethylamino.
Niederalkyleniminoreste sind Reste mit z.B. 4 bis 8 Ringgliedern, wobei der Niederalkylenteil verzweigtes oder insbesondere geradkettiges Niederalkylen ist mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Beispiele sind Pyrrolidino oder Piperidino.
Hydroxyniederalkyleniminoreste sind Reste mit z.B. 4-8 Ringgliedern, wobei der hydroxysubstituierte Niederalkylenteil verzweigtes oder insbesondere geradkettiges Niederalkylen, ist mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Beispiele sind 4-Hydroxypiperidino oder 3-Hydroxypyrrolidino.
Oxaniederalkylenimino ist verzweigtes oder insbesondere geradkettiges Oxaniederalkylenimino mit insbesondere 4 oder 5 C-Atomen in der Oxaalkylenkette. Als Beispiel ist insbesondere Morpholino zu nennen.
Thianiederalkylenimino ist verzweigtes oder insbesondere geradkettiges Thianiederalkylenimino mit insbesondere 4 oder 5 C-Atomen in der Thiaalkylenkette. Als Beispiele sind insbesondere Thiomorpholino und 2,6-Dimethylthiomorpholino zu nennen.
Azaniederalkylenimino ist verzweigtes oder insbesondere geradkettiges Azaniederalkylenimino mit insbesondere 2-6, vor allem 4-6 C-Atomen in der Azaalkylenkette. Beispiele sind Piperazino, N'-Methylpiperazino oder N'-(l8-Hydroxy- äthyl)-piperazino.
Acylamino ist z.B. niederes Alkanoylamino mit bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen im Niederalkylteil oder gegebenenfalls substituiertes Aroylamino, wie gegebenenfalls substituiertes B enzoylamino oder gegebenenfalls substituiertes Arylniederalkanoylamino oder Niederalkoxycarbonylamino.
Niedere Alkanoylaminoreste sind z.B. n-Propionylamino, n-Butyrylamino, n-Valerylamino, n-Hexanoylamino, n-Heptanoylamino oder vor allem Acetylamino. Gegebenenfalls substituierte Arylnieder aikanoylaminoreste sind Niederalkanoylaminoreste, die an beliebiger Stelle des Niederalkylteils z.B. gegebenenfalls substituierte Phenylgruppen tragen. Die Phenylgruppen können die gleichen Substituenten tragen, wie oben für gegebenenfalls substituierte Benzoylaminoreste erwähnt.
Als Beispiele für gegebenenfalls substituiertes Arylniederalkanoylamino sind vor allem Phenylacetylamino, 3-Phenyl-n-propionylamino, 4-Phenyl-n-butyrylamino, Chlorphenylacetylamino und Bromphenylacetylamino zu nennen. Niederalkoxycarbonyl- amino hat im Niederalkylteil von Niederalkoxy vorzugsweise bis zu 7 C-Atome und vor allem bis zu 4 C-Atome und ist z.B. n-Propoxycarbonylamino, n-Butoxycarbonylamino, i-Propoxycarbonylamino, tert.-Butoxycarbonylamino und vor allem Methoxycarbonylamino und Äthoxycarbonylamino.
Acylaminoniederalkylreste sind Niederalkyfreste mit vorzugsweise bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen, die in beliebiger Stelle durch Acylaminoreste substituiert sind. Acylamino ist z.B. niederes Alkanoylamino mit bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen im Niederalkylteil oder gegebenenfalls substituiertes Aroylamino, wie gegebenenfalls substituiertes Benzoylamino oder gegebenenfalls substituiertes Arylniederalkanoylamino oder Niederalkoxycarbonylamino.

Eine reaktionsfähige, veresterte Hydroxylgruppe ist insbesondere eine durch eine starke anorganische oder organische Säure, vor allem eine Halogenwasserstoffsäure, wie Chlorwasserstoffsäure, Bromwasserstoffsäure oder Jodwasserstoffsäure, ferner Schwefelsäure oder eine starke organische Sulfonsäure, wie beispielsweise Benzolsulfonsäure, 4-Brombenzolsulfonsäure, 4-Toluolsulfonsäure oder Methansulfonsäure, veresterte Hydroxylgruppe. So steht Z1 oder Z2 insbesondere für Chlor, Brom oder Jod.
Diese Umsetzung wird in der üblichen Weise durchgeführt. Bei Verwendung eines reaktionsfähigen Esters als Ausgangsmaterial wird vorzugsweise in Gegenwart eines basischen Kondensationsmittels und/oder mit einem Überschuss an Amin gearbeitet. Geeignete basische Kondensationsmittel sind z.B. Alkalihydroxyde, wie Natrium- oder Kaliumhydroxyd, Alkalicarbonate, wie Kaliumcarbonat, und Alkalialkoholate, z.B. Alkaliniederalkanolate, wie Natriummethylat, Kaliumäthylat und Kaliumtertiärbutylat.
Die genannten Reaktionen werden in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensationsund/oder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säureadditionssalze. So können beispielsweise basische, neutrale oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesquioder Polyhydrate davon, erhalten werden. Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung übergeführt werden, z.B. mit basischen Mitteln, wie Alkalien oder Ionenaustauschern. Andererseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere solche Säuren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind.
Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefel- säuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbon oder Sulfonsäuren, wie Ameisen-, Essig-, Propion, Bernstein-, Glykols, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Fumar- oder Brenztraubensäure; Phenylessig-, Benzoe-, Anthranil-, p-Hydroxybenzoe-, Salicylsäure, Embonsäure, Methansuifon-, Athansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäure, Sulfanilsäure oder Cyclohexyl aminsulfonsäure.
Diese oder andere Salze der neuen Verbindungen, wie z.B. die Pikrate oder Perchlorate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freisetzt. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmässig, gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, bei denen eine Reaktionskomponente in Form ihrer Salze vorliegt.
Die neuen Verbindungen können je nach der Wahl der Ausgangsstoffe und Arbeitsweisen, als optische Antipoden oder Racemate oder, sofern sie mindestens zwei asyflime- trische Kohlenstoffatome enthalten auch als Isomerengemische (Racematgemische) vorliegen.
Erhaltene Isomerengemische (Racematgemische) können aufgrund der physikalisch-chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden stereoisomeren (diastereomeren) reinen Racemate aufgetrennt werden, beispielsweise durch Chromatographie und/oder fraktionierte Kristallisation.
Erhaltene Racemate lassen sich nach bekannten Methoden, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen oder durch Umsetzen mit einer, mit der racemischen Verbindung Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltenen Salze, z.B. aufgrund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, aus denen die Antipoden durch Einwirkung geeigneter Mittel freigesetzt werden können, zerlegen. Besonders gebräuchliche optisch akive Säuren sind z.B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Äpfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden.
Zweckmässig verwendet man für die Durchführung der erfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs besonders erwähnten Gruppen von Endstoffen und besonders zu den speziell beschrieben oder hervorgehobenen Endstoffen führen.
Die Ausgangsstoffe sind bekannt oder können, falls sie neu sind, nach an sich bekannten Methoden erhalten werden.
Verbindungen der Formeln IIa, Leib, IIc oder XI können z.B. erhalten werden, wenn man (R3)(R4)-Pyrazinole, bzw.
-Pyridazinole, bzw. -Pyrimidinole, bzw. (R3)n-Pyridinole mit Epichlorhydrin umsetzt.
Die neuen Verbindungen können als Heilmittel, z.B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem z.B. für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z.B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche öle, Benzylalkohole, Gummi, Polyaikylenglykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z.B. Tabletten, Dragees, Kapseln, Suppositorien, Salben, Crem oder in flüssiger Form als Lösungen (z.B. als Elixier oder Sirup), Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die Präparate welche auch in der Veterinärmedizin Verwendung finden können, werden nach üblichen Methoden gewonnen.
Die tägliche Dosis beträgt etwa 40-150 mg im Falle eines Warmblüters von etwa 75 kg Körpergewicht.
Die folgenden Beispiele erläutern die Erfindung ohne sie jedoch einzuschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I
39,7 g 5-Brom-3-morpholinyl-2-(3'-brom-2'-hydroxyprop- oxy)-pyrazin werden mit 41 g Isopropylamin in 500 ml Methanol 15 Stunden zum Rückfluss erwärmt. Anschliessend wird das Reaktionsgemisch im Wasserstrahlvakuum eingedampft. Der Rückstand wird zwischen 2-n. Salzsäure und Äther verteilt. Die wässrige Phase wird mit konzentrierter Natronlauge alkalisch gestellt und mit Äther ausgeschüttelt. Die vereinigten Ätherextrakte werden über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft.
Man erhält so die rohe Base und daraus mit ätherischer Salzsäure in Methanol das 5-Brom-3-morpholinyl-2-(3'-iso- propylamino-2'-hydroxy-propoxy)-pyrazin-hydrochlorid, F. 202.2030.

Das als Ausgangsstoff verwendete 5-Brom-3-morpholinyl-2-(3'-brom-2'-hydroxy-propoxy)-pyrazin kann wie folgt hergestellt werden: la) 149 g 2,3-Dichlorpyrazin werden mit 300 g Morpholin und 400 ml Wasser 16 Stunden bei 40 gerührt. Dann wird das Reaktionsgemisch mit Äther ausgeschüttelt. Die Ätherextrakte werden mit Wasser neutral gewaschen, über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Der Rückstand wird im Wasserstrahlvakuum destilliert. Man erhält so das 2-Chlor-3-morpholinyl-pyrazin, Kp. 162-1650/15 Torr.
lb) 120 g 2-Chlor-3-morpholinyl-pyrazin und 70 g Allylalkohol werden in 640 ml Eexamethylphosphorsäuretri- amid gelöst. In diese Lösung werden bei 0 während 30 Minuten 28,8 g Natriumhydrid eingetragen. Anschliessend rührt man noch 1 Stunde bei 0 , lässt dann bis zumAbklin- gen der Reaktion 1 Stunde bei 30 und 15 Stunden bei Raumtemperatur rühren. Dann wird das Reaktionsgemisch auf 2 Liter Eiswasser gegossen. Nachdem der vorhandene Über- schuss an Natriumhydrid zersetzt ist, wird mit Äther ausgeschüttelt. Die Ätherextrakte werden mit Wasser neutral gewaschen, über Natriumsulfat getrocknet, und im Wasser strahlvakuum eingedampft. Der Rückstand wird im Hochvakuum destilliert.
Man erhält so das 2-Allyloxy-3-morpholinyl-pyrazin, Kp. 105-1060/0,03 Torr.
lc) 32 g 2-Allyloxy-3-morpholinyl-pyrazin werden in 350 ml Dimethylsulfoxyd und 2,7 ml Wasser gelöst. In diese Lösung werden unter Rühren während 30 Minuten 52 g N-Bromsuccinimid eingetragen. Dabei steigt die Reaktionstemperatur auf 35". Man rührt weitere 30 Minuten, verdünnt dann mit ca. 500 ml Wasser und schüttelt das Reaktionsgemisch mit Äther aus. Die Ätherextrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und im Wasser strahivakuum eingedampft. Man erhält so das 5-Brom-3 -morpholinyli2-(3 '-brom-2'-hydroxy-propoxy)-pyrazin.
Beispiel 2
71 g 5-Brom-3-dimethylamino-2-(3'.brom-2'-hydroxy.
propoxy)-pyrazin werden mit 71 g Isopropylamin in 1 Liter Methanol 15 Stunden am Rückfluss gekocht. Anschliessend wird das Reaktionsgemisch im Wasserstrahlvakuum eingedampft. Der Rückstand wird zwischen 2-n. Salzsäure und Äther verteilt. Die wässrige Phase wird mit konzentrierter Natronlauge alkalisch gestellt und mit Äther ausgeschüttelt.
Die vereinigten Ätherextrakte werden über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Man erhält so die rohe Base und daraus mit Fumarsäure in Methanol-Äther das 5-Brom-3-dimethylamino-2-(3'-isopropyl.
amino-2'-hydroxy)-pyrazin.hydrogenfumarat, F. 146-1470.

Das als Ausgangsstoff verwendete 5-Brom-3 -dimethyl- amino-2-(3 -brom-2'-hydroxy-propoxy).pyrazin kann wie folgt hergestellt werden:
2a) Zu 350 ml einer 40%igen wässrigen Lösung von Dimethylamin werden 74 g 2,3-Dichlor-pyrazin unter Rühren innerhalb von 15 Minuten zugetropft. Dabei wird die Reaktionstemperatur durch Kühlen auf 30 gehalten. Nach Abklingen der exothermen Reaktion wird noch 15 Stunden bei Raumtemperatur gerührt. Dann wird das Reaktionsgemisch mit Äther ausgeschüttelt. Die Ätherextrakte werden mit Wasser neutral gewaschen, über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Der Rückstand wird im Wasserstrahlvakuum destilliert. Man erhält so das 2-Chlor -3-dimethylamino-pyrazin, Kp. 100-1020/10 Torr.
2b) 47 g 2-Chlor-3-dimethylamino-pyrazin und 35 g Allylalkohol werden in 300 ml Hexamethylphosphorsäuretriamid gelöst. In diese Lösung werden bei 0 während 30 Minuten 14,4 g Natriumhydrid eingetragen. Anschliessend lässt man noch 1 Stunde bei 0 und 15 Stunden bei Raumtemperatur rühren. Dann wird das Reaktionsgemisch auf 2 Liter Eiswasser gegossen. Nach Zersetzung von überschüssigem Natriumhydrid wird mit Äther ausgeschüttelt. Die Ätherextrakte werden mit Wasser neutral gewaschen, über Natrium sulfat getrocknet und im Wasserstrahlvakuum eingedampft.
Der Rückstand wird bei reduziertem Druck destilliert. Man erhält so das 2-Allyloxy-3-dimethyiamino-pyrazin, Kp. 1101150/10 Torr.
2c) 45 g 2-Allyloxy-3-dimethylamino-pyrazin werden in 600 ml Dimethylsulfoxyd und 9 ml Wasser gelöst. In diese Lösung werden unter Rühren während 30 Minuten 89 g N-Bromsuccinimid eingetragen. Durch Aussenkühlung wird die Temperatur dabei auf 35 gehalten. Anschliessend rührt man noch 1 Stunde bei Raumtemperatur, verdünnt dann mit 2 Liter Eiswasser und schüttelt mit Äther aus. Die Äther extrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Man erhält so das 5-Brom-3-dimethylamino-2-(3'-hydroxy-prop- oxy)-pyrazin.
Beispiel 3
53 g 2-(3'-Brom-2'-hydroxy-propoxy)-3-chlor-pyrazin werden mit 59 g Isopropylamin in 500 ml Methanol 15 Stunde den am Rückfluss gekocht. Dann wird das Reaktionsgemisch im Wasserstrahlvakuum eingedampft. Der Rückstand wird zwischen 2-n. Salzsäure und Äther verteilt. Die salzsaure Phase wird mit konzentrierter Natronlauge alkalisch gestellt und mit Äther ausgeschüttelt. Die Ätherextrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Der Rückstand kristallisiert aus Äther. Man erhält so das 3-Chlor-2-(3'-isopropylamino-2'-hydroxy-propoxy)-pyrazin, F. 99-100".
Das daraus hergestellte Hydrochlorid kristallisiert aus Methanol-Äther, F. 183".

Das als Ausgangsstoff verwendete 2-(3'-Brom-2'-hydroxy -propoxy)-3-chlor-pyrazin kann wie folgt hergestellt werden:
3a) 59,6 g 2,3-Dichlorpyrazin und 92,8 g Allylalkohol werden in 400 ml Dimethylsulfoxyd gelöst. In diese Lösung werden unter Rühren bei 0-5" 9,6 g Natriumhydrid während 30 Minuten eingetragen. Anschliessend wird 15 Stunden bei Raumtemperatur gerührt. Dann wird das Reaktionsgemisch auf 2 Liter Eiswasser gegossen und mit Äther ausgeschüttelt. Die Ätherextrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Der Rückstand wird im Hochvakuum destilliert.
Man erhält so das 2-Allyloxy-3-chlor-pyrazin, Kp. 42-440/ 0,03 Torr.
3b) 34 g 2-Allyloxy-3-chlor-pyrazin und 7,2 ml Wasser werden in 500 ml Dimethylsulfoxyd gelöst. Unter Rühren werden während 30 Minuten 71 g N-Bromsuccinimid eingetragen, wobei die Reaktionstemperatur durch Kühlung auf 30 gehalten wird. Anschliessend wird noch 30 Minuten bei Raumtemperatur gerührt und dann mit 1 Liter Wasser verdünnt. Dann wird das Reaktionsgemisch mit Äther ausgeschüttelt. Die Ätherextrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Man erhält so das rohe 2-(3'-Brom-2' -hydroxy-propoxy)-3-chlor-pyrazin.
Beispiel 4
In analoger Weise wie in Beispiel 1 erhält man aus 39,7 g 5-Brom-3-morpholino-2-(3'-brom-2'-hydroxy-propoxy)-py- razin und 51 g tert.-Butylamin das 5-Brom-3-morpholinyl-2 -(3'-tert.-butylamino-2'-hydroxy-propoxy)-pyrazin, F. 104 105 .

Beispiel 6
Eine Lösung von 15,0 g 3-Chlor-2-(2',3'-epoxy-propoxy) -pyridin in 100 mi Isopropanol wird mit 20 g Isopropylamin versetzt und anschliessend 3 Stunden urrter Rückfluss zum Sieden erhitzt. Das Reaktionsgemisch wird im Vakuum eingedampft, in 300 ml Äthylacetat gelöst und mit 100 ml 2-n.
Salzsäure extrahiert. Der Salzsäure-Auszug wird mit 30 ml konzentrierter Natronlauge alkalisch gestellt und mit je 100 ml Äthylacetat 3mal extrahiert. Die organische Phase wird jeweils mit 10 ml Sole gewaschen, vereinigt und über Magnesiumsulfat getrocknet. Man erhält so das 2-(3'-Isopropylamino-2'-hydroxy-propoxy)-3-chlor-pyridin, welches aus Äther-Petroläther umkristallisiert wird und bei 71-73" schmilzt. Das Hydrochlorid schmilzt bei 167-169" (aus Methanol-Aceton).

Das als Ausgangsmaterial verwendete 3-Chlor-2-(2',3' -epoxy-propoxy)pyridin kann auf folgende Weise hergestellt werden:
6a) Zu einer Suspension von 16,5 g Natriumhydrid in 600 ml 1,2-Dimethoxyäthan werden 108 g 2,2-Dimethyl-4 -hydroxymethyl-dioxolan so zugetropft, dass die Reaktion unter Kontrolle bleibt. Hierauf wird 1 Stunde bei einer Badtemperatur von 50-60" gerührt. Zu der entstandenen gallertigen Suspension werden unter Rückfluss portionenweise 100,0 g 2,3-Dichiorpyridin gegeben. Das wieder dünnflüssiger werdende Reaktionsgemisch wird nach beendeter Zugabe noch 3 Stunden unter Rückfluss gerührt. Das Lösungsmittel wird im Vakuum abgedampft und der Rückstand zwischen 2 Liter Äther und 200 ml Wasser verteilt. Die Ätherphase wird über Magnesiumsuifat getrocknet.
Das nach dem Abdampfen des Äthers verbleibende Produkt wird im Hochvakuum destilliert. Das 3-Chlor-2-[2',2'-dimethyl-1',3'-dioxol anyl- (4')]-methoxy-pyridin siedet bei 95-100"/0,01 Torr.

6b) Eine Lösung von 157 g 3-Chlor-2-[2',2'-dimethyl -1',3'-dioxolanyl-(4'}]-methoxy-pyridin in 100 ml Äthanol wird mit 320 ml 2-n. Salzsäure versetzt und 2 Stunden bei Raumtemperatur gerührt. Nach dem Abdampfen des Äthanols wird das Produkt im Minimum Wasser (ca. 100 ml) gelöst, mit 100 ml Äther gewaschen und die wässrige Phase mit konzentrierter Natronlauge alkalisch gestellt. Das sich abscheidende Öl wird mit je 200 ml Äthylacetat 3mal extrahiert. Die Extrakte werden mit je 20 ml Sole gewaschen, getrocknet und eingedampft. Das erhaltene 3-Chlor-2-(2',3' -dihydroxy-propoxy)-pyridin siedet bei 142-145"/0,015 Torr.

6c) Zu einer Lösung von 40,6 g 3-Chlor-2-(2',3'-dihydr oxy-propoxy)-pyridin in 100 ml Pyridin werden unter Rühren und Kühlen bei 0-5" 25,2 g Methansulfonsäurechlorid im Verlauf von 1 Stunde zugetropft. Hierauf wird das Reaktionsgemisch noch 5 Stunden bei Raumtemperatur nachgerührt und anschliessend auf 200 ml Eiswasser gegossen. Das sich abscheidende öl wird wie im Beispiel lb) isoliert. Natronlauge und 5 g Tetrabutylammonium-chlorid werden 15 Stunden bei Raumtemperatur gerührt. Die Methylenchloridphase wird abgetrennt, 2mal mit je 40 ml Wasser gewaschen, getrocknet und eingedampft. Aus dem Eindampfrückstand isoliert man durch Destillation bei 115-130"/0,03 Torr das 3-Chlor-2-(2',3'-epoxy-propoxy)-pyridin als hellgelbes Ö1.

7b) 14,8 g 3-(2',3'-Dihydroxy-propoxy)-2-methoxy-pyri- din gelöst in 100 ml wasserfreiem Pyridin, werden bei - 10 bis - 15" tropfenweise unter Rühren mit 9,0 g Methansulfonsäurechlorid versetzt. Das Reaktionsgemisch wird hierauf noch 3 Stunden bei -10" nachgerührt. Das Pyridin wird sodann am Rotationsverdampfer bei 10 Torr. möglichst vollständig abgedampft und das so erhaltene, rohe 2-Meth oxy-3-(3'-methansulfonyloxy.2'-hydroxy.propoxy)-pyridin ohne weitere Reinigung mit Isopropylamin umgesetzt.
Beispiel 8
33 g rohes 3-(2',3'-epoxy-propoxy)-6-methylpyridin, 350 ml Isopropanol und 75 ml Isopropylamin werden 12 bis 14 Stunden unter Rückfluss zum Sieden erhitzt. Die Aufarbeitung erfolgt analog Beispiel 6. Das so erhaltene 3-(3'-Iso- propylamino-2'-hydroxy.propoxy)-6.methyl-pyridin siedet im Kugelrohr bei 135-1500 /0,2 Torr. Sein neutrales Fumarat schmilzt bei 171-1730 (umkristailisiert aus Methanol/Aceton)

Das als Ausgangsmaterial benötigte 3-(2',3'-epoxy-propoxy)-6-methyl-pyridin wird auf folgende Weise hergestellt:
8a) 22 g 6-Methyl-3-pyridinol, 50 g Kaliumcarbonat und 80 ml Epichlorhydrin werden in 500 ml Aceton unter Rüh- ren und Rückfluss während 18 bis 20 Stunden zum Sieden erhitzt.
Die ungelösten Anteile werden abgenutscht und das Filtrat bei 30 bis 40 Badtemperatur im Vakuum eingeengt.
Das so erhaltene 3-(2',3'-epoxy-propoxy)-6-methyl-pyridin wird ohne weitere Reinigung weiter verarbeitet.
Beispiel 9
25 g rohes 3-(2',3'-Epoxy-propoxy)-2-pyrrolidino-carbonyl-pyridin, gelöst in 350 ml Isopropanol, werden mit 150 ml Isopropylamin 3 Stunden unter Rückfluss gekocht. Nach dem Eindampfen und Aufarbeiten des Rückstandes analog Beispiel 6 erhält man das 3-(3'-Isopropylamino-2'-hydroxy- -propoxy)-2-pyrrolidinocarbonylpyridin, welches bei 108-111 schmilzt, (umkristallisiert aus Äthylacetat-Äther).

Beispiel 10
13,3 g 2-(3'-p-Toluolsulfonyloxy-2'-hydroxy-propoxy)-pyrimidin werden in 100 ml Isopropanol und 22 ml Isopropylamin gelöst und 48 Stunden bei Raumtemperatur aufbewahrt. Dann werden die flüchtigen Anteile unter Vakuum abdestilliert, der ölige Rückstand in Aceton aufgenommen und eine Lösung von 4 g Oxalsäure in Aceton zugefügt.
Man erhält das 2-(3'-Isopropylamino-2'-hydroxy-propoxy) -pyrimidin-hydrogenoxalat, welches aus Methanol-Aceton umkristallisiert wird. Smp. 181-182".

Das als Ausgangsmaterial benötigte 2-(3'-p-Toluolsulf onyioxy-2'-hydroxy-propoxy)-pyrimidin kann wie folgt hergestellt werden: lOa) Zu einer Suspension von 1,2 g Natriumhydrid in 50 ml Dimethoxyäthan gibt man 6,6 g 2,2-Dimethyl-1 ,3-di- oxolan-4-methanol und rührt 2 Stunden bei Raumtemperatur.
Dann werden 5,7 g 2-Chlor-pyrimidin zugegeben. Das Reaktionsgemisch wird 2 Stunden zum Sieden erhitzt und anschliessend unter Vakuum eingedampft. Der Rückstand wird im Kugelrohr destilliert. Nach einem geringfügigen Vorlauf destilliert bei 200 /19 Torr. das 2-[2',2'-Dimethyl-1',3'-di oxoian-(4')]-methoxy-pyrimidin als farbloses Ö1.
lOb) 16 g 2-[2',2'-Dimethyl-1',3'-dioxolan-(4')]-methoxy- -pyrimidin werden mit 20 ml Wasser und 2 ml 2-n. Schwefelsäure 15 Minuten zum Sieden erhitzt. Dann kühlt man ab, fügt die zur Neutralisation der Säure nötige Menge Bariumhydroxyd-Lösung zu und filtriert. Das Filtrat wird eingedampft und das zurückbleibende Öl im Kugelrohr destilliert, wobei das 2-(2',3'-Dihydroxy-propoxy)-pyrimidin bei.
180-190 /0,4 Torr. als farbloses- öliges Destillat erhalten wird.
10c) In eine Lösung von 11,8 g 2-(2',3'-Dihydroxy-propoxy)-pyrimidin in 18 ml Pyrimidin, die auf - 100 gekühlt wird, trägt man unter Rühren 13,6 g p-Toluolsulfochlorid während 15 Minuten ein. Das Reaktionsgemisch wird 15 Stunden bei 0 aufbewahrt. Dann wird Eis zugegeben und, unter Rühren, 20 ml 6-n. Salzsäure. Anschliessend wird zweimal mit je 150 ml Methylenchlorid extrahiert. Die Extrakte werden mit Natriumbicarbonat-Lösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Man erhält das 2-(3'-p-Toluolsulfonyloxy-2'-hydroxy-propoxy).pyrimidin als gelbliches Ö1.
Beispiel 11
19,5 g rohes 3-(2',3'-Epoxy-propoxy)-2-nitro-pyridin, gelöst in 300 ml Isopropanol, werden mit 100 ml Isopropylamin 4 Stunden unter Rückfluss gekocht. Nach dem Aufarbeiten analog Beispiel 6 erhält man 3-(2'-Hydroxy-3'-isopropylamino-propoxy)-2-nitro-pyridin, welches nach Kristal- lisation aus Äther bei 99-1010 schmilzt.
Den Ausgangsstoff erhält man auf folgende Weise: a) 28 g 2-Nitro-3-pyridinol, 200 ml Epichlorhydrin und 60 g Kaliumcarbonat werden unter Rühren in 500 ml Acetonitril 8 Stunden unter Rückfluss gekocht. Filtration und Eindampfen des Reaktionsgemisches ergeben das rohe 3-(2',3' -Epoxy-propoxy)-2-nitro-pyridin als gelbes öL
Beispiel 12
10 g rohes 3-(2,3-Epoxy-propoxy)-6-methyl-pyridin. 1 - -oxid werden in 200 ml Isopropanol mit 25 ml Isopropylamin 6 Stunden unter Rückfluss zum Sieden erhitzt. Das Reaktionsgemisch wird danach im Vakuum eingedampft, in 300 ml Äthylacetat aufgenommen und mit 30 ml gesättigter Kaliumbicarbonat-Lösung extrahiert. Das erhaltene Öl kristallisiert aus Äther und führt so zu 3-(2'-Hydroxy-3'-iso propylamino-propoxy)- 6-methyl-pyridin- 1 -oxid vom Smp.
87-89".
Der Ausgangsstoff kann auf folgende Weise hergestellt werden: a) Zu einer Lösung von 10 g rohem 3-(2,3-Epoxy-propoxy)-6-methyl-pyridin in 100 ml Dichlormethan wird unter Rühren eine Lösung von 12,2 g m-Chlorperbenzoesäure in 100 ml Dichlormethan im Verlauf von ca. 10 Minuten getropft, wobei die Temperatur der Lösung auf 32 steigt.
Das Reaktionsgemisch wird noch weitere 30 Minuten gerührt, hierauf mit 40 ml gesättigter Kaliumbicarbonat-Lösung extrahiert, getrocknet und im Vakuum eingedampft.
Das so erhaltene 3-(2,3-Epoxy-propoxy)-6-methyl-pyridin- 1 - -oxid wird roh weiterverwendet.
Beispiel 13
3,2 g rohes l-(5-Pyrimidinyloxy)-2,3-epoxypropan werden in 50 ml Isopropanol gelöst und nach Zusatz von 15 ml Isopropylamin 18 Stunden unter Rückfluss zum Sieden erhitzt.
Danach wird das Reaktionsgemisch unter Vakuum eingedampft, der Rückstand in 25 ml Aceton aufgenommen, zum Sieden erhitzt und mit 25 ml Essigester versetzt. Dann wird die Lösung von ungelösten Harzen abdekantiert und anschliessend unter Vakuum eingedampft. Der Eindampfrückstand wird in Aceton gelöst und mit einer Lösung von 1,8 g Oxalsäure in 10 ml Aceton versetzt. Es scheiden sich Kristalle ab, welche nach Umkristallisieren aus Methanol-Aceton das 5-(3 -Isopropylamino-2-hydroxypropoxy)-pyrimidin-hy- drogenoxalat vom Smp. 166-1670 ergeben.
Das als Ausgangsstoff verwendete 1-(5-Pyrimidinyloxyk -2,3-epoxypropan kann wie folgt hergestellt werden:
8,6 g 5-Hydroxypyrimidin und 21 g Kaliumcarbonat werden in 300 ml Aceton unter Rühren mit 9 ml Epichlorhydrin versetzt und anschliessend 40 Stunden unter Rückfluss zum Sieden erhitzt. Dann wird der Niederschlag abgesaugt, der Filterrückstand mit Aceton nachgewaschen und das Filtrat unter Vakuum eingedampft. Man erhält als Eindampfrückstand ein braunes Öl, bestehend aus rohem 1-(5-Pyrimidinyloxy)-2,3-epoxypropan, welches ohne Reinigung weiterverwendet werden kann.
Beispiel 14
0,195 g rohes 1-(2-Methylthio-5-pyrimidinyloxy)-2,3- -epoxypropan werden in 10 ml Isopropanol gelöst und nach Zusatz von 1,5 ml Isopropylamin 18 Stunden unter Rückfluss zum Sieden erhitzt und das Reaktionsgemisch anschliessend unter Vakuum eingedampft. Der Eindampfrückstand wird in Aceton gelöst, klar filtriert und das Filtrat mit einer Lösung von 0,1 g Oxalsäure in 0,5 ml Aceton versetzt. Beim Anreiben kristallisiert das 5-(3'-Isopropylamino-2'-hydroxy -propoxy)-2-methylthiopyrimidin-hydrogenoxalat, das aus Methanol-Aceton umkristallisiert wird; Smp. 196-1970.
Das als Ausgangsmaterial verwendete 1-(2-Methylthio-5 -pyrimidinyloxy)-2,3-epoxypropan kann wie folgt hergestellt werden:
1,42 g 5-Hydroxy-2-methylthiopyrimidin und 2,1 g Kaliumcarbonat werden in 30 ml Aceton unter Rühren mit 0,9 ml Epichlorhydrin versetzt und anschliessend 40 Stunden unter Rückfluss zum Sieden erhitzt. Dann wird abgenutscht, der Filterrückstand mit Aceton nachgewaschen und das Filtrat unter Vakuum eingedampft. Man erhält als Eindampfrückstand ein braunes Öl, bestehend aus rohem l-(2-Methyl thio-5-pyrimidinyloxy)-2,3-epoxypropan, welches ohne Reinigung weiter verwendet werden kann.
Beispiel 15
Analog Beispiel 1 werden hergestellt: a) aus 20,2 g 3-(2',3'-Epoxy-propoxy)-2-(n-butylamino- -carbonyl)-pyridin das 3-(3'-Isopropylamino-2'-hydroxy- -propoxy)-2- (n-butylaminocarbonyl)-pyridin, welches bei 65-67" schmilzt; b) aus 15,0 g 6-Methoxy-3-(2',3'-epoxy-propoxy)-pyridazin das 3-(3'-Isopropylamino-2'-hydroxy-propoxy)-6-methoxy -pyridazin, welches bei 115-116 schmilzt; c) aus 15,0 g 6-Chlor.3-(2',3'-epoxy-propoxy)-pyridazin das 3-(3'-Isopropylamino-2'-hydroxy-propoxy-6-chlor-pyridazin, welches bei 98-99" schmilzt; d) aus 13,5 g 3-(2',3'-Epoxy-propoxy)-pyridazin das 3-(3' -Isopropylamino-2'-hydroxy-propoxy)-pyridazin, welches bei 91-930 schmilzt.
e) aus 14,4 g 2-(2',3'-Epoxy-propoxy)-5-acetylamino-py din das 2-(3'-Isopropylamino.2'-hydroxy-propoxy)-pyridin, welches bei 139-141 schmilzt.

The compounds mentioned either belong to the class of the non-cardioselective p-receptor blockers, i.e. They block the p-receptors on the vessels or in the trachea in similar or even smaller doses or concentrations than the 53 receptors in the heart, or they belong to the class of so-called cardioselective receptor blockers, i.e. they block the p-receptors of the heart even in a dose or concentration range which does not yet cause a blockage of the p-receptors in the vessels or in the trachea.

3.2 g of crude 1- (5-pyrimidinyloxy) -2,3-epoxypropane are dissolved in 50 ml of isopropanol and, after addition of 15 ml of isopropylamine, heated to boiling under reflux for 18 hours.
The reaction mixture is then evaporated in vacuo, the residue is taken up in 25 ml of acetone, heated to boiling and mixed with 25 ml of ethyl acetate. Then the solution is decanted off from undissolved resins and then evaporated in vacuo. The evaporation residue is dissolved in acetone and a solution of 1.8 g of oxalic acid in 10 ml of acetone is added.
