Caryophyllen ist in ätherischen Ölen in der Natur weitverbreitet und kommt vor allem im Nelkenöl (Eugenia caryophyllata) vor, aus dem es im Gemisch mit etwa 10% Humulen isoliert werden kann. Aus dem handelsüblichen Caryophyllen lässt sich reines Caryophyllen leicht durch Säulenchromatographie an Kieselgel gewinnen.

Die reduktive Eliminierung der 6,7-Epoxygruppe einer Verbindung der Formel II gemäss Verfahrensvariante (a) kann in an sich bekannter Weise erfolgen, beispielsweise durch Behandlung mit einem Chrom(II)-Äthylendiaminkomplex [I. K. Kochi et al., Tetrahedron 24, 3503 (1968)] oder mittels schwach verkupfertem Zink in kochendem Äthanol [S. M. Kupchan und M. Marnyama, J. Org. Chem. 36, 1187 (1971)].
Die Reduktion eines 12-Norcaryophyll-6-en-3-ons der Formel Ia zum entsprechenden sekundären Alkohol der Formel Ib gemäss Verfahrensvariante (b) kann nach an sich bekannten Methoden mit einem geeigneten Reduktionsmittel wie einem komplexen Alkalimetallhydrid, z. B. LiAIH4, NaAIH4 oder NaBH4, einem komplexen Trialkoxy-alkalimetallhydrid, z. B. LiAIH(OCH3)3 oder LiAIH[OC(CH3)3]3, einem substituierten Aluminiumhydrid, wie Diäthylaluminiumhydrid, Diisobutylaluminiumhydrid, oder einer Trialkylaluminiumverbindung wie Triisopropylaluminium, erfolgen. Die Verwendung komplexer Alkalimetallhydride, insbesondere von Lithiumaluminiumhydrid, ist bevorzugt.
Die Reaktion wird zweckmässig in einem organischen Lösungsmittel wie einem Äther, z. B. in Diäthyläther, Dioxan oder Tetrahydrofuran, ausgeführt. Es ist empfehlenswert, bei niedrigen Temperaturen, d. h. unterhalb der Raumtemperatur, z. B. bei 0 , zu arbeiten.
Der sekundäre Alkohol kann aber auch direkt aus dem Epoxyketon II hergestellt werden, wenn man die oben angegebene Methode der Expoxidreduktion so modifiziert, dass ein hoher Kupferanteil im Metall vorhanden ist.
Die Überführung eines 12-Norcaryophyll-6-en-3-ons der Formel Ia mit einer Grignardverbindung der Formel H3C-Mg-Hal (Hal = Cl, Br, J) in ein Caryophyll-6-en-3-ol der Formel Id gemäss Verfahrensvariante (c) kann ebenfalls in für den Fachmann bekannter Weise erfolgen.
Schliesslich kann die Verätherung der 3-Hydroxyfunktion eines Caryophyll-6-en-3-ols oder eines 12-Norcaryophyll6-en-3-ols der Formel Ie gemäss Verfahrensvariante (d) nach einer der üblichen Methoden, beispielsweise mit einem Alkylhalogenid oder einem Alkylsulfat, vorgenommen werden.
Die Herstellung des 6,7-Epoxy-12 -norcaryophyllan3-ons der Formel II aus Caryophyllen (III) kann in an sich bekannter Weise über 6,7-Epoxycaryophyllen der Formel

erfolgen.
Die Überführung des 6,7-Epoxycaryophyllens in das entsprechende 6,7-Epoxy-12-norcaryophyllan-3 -on (II) kann z. B. durch Ozonolyse der exocyclischen Doppelbindung oder durch Oxydation mit Permanganat erfolgen, während die selektive Epoxydierung der endocyclischen Doppelbindung des Caryophyllens mittels einer Persäure, beispielsweise Peressigsäure, in einem geeigneten Lösungsmittel, z. B. in Methylenchlorid, und in Gegenwart von Natriumacetat erfolgen kann.
Die Verbindungen der Formel I zeichnen sich durch besondere geruchliche Qualitäten aus. Sie sind durch holzige, erdige, teilweise an Sandel oder Vetiver erinnernde Geruchsnoten charakterisiert. Der Zusatz von Verbindungen der Formel I zu Riechstoffkompositionen macht diese in geruchlicher Hinsicht wärmer und voller. Sie vereinigen sich besonders gut mit holzig riechenden Kompositionen und ätherischen Ölen. Die Verbindungen können deshalb als Riechstoffe in der Parfümerie verwendet werden, beispielsweise zur Herstellung bzw. zur geruchlichen Modifizierung von Riechstoffkompositionen, wie Parfüms, Parfümbasen usw. durch Zusatz olfaktorisch wahrnehmbarer Mengen, z. B. 0,1 bis 10 Gew.%, zu Gemischen von bekannten Riechstoffen. Ein weites Anwendungsgebiet stellt z. B. die Herstellung chypreartiger Riechstoffkompositionen dar.
Die Verbindungen können auch zur Parfümierung von technischen und kosmetischen Produkten aller Art, z. B. von festen und flüssigen Detergentien, synthetischen Waschmitteln, Aerosolen, Seifen, Cremes, Lotions usw., z. B. in Konzentrationen von etwa 0,02 bis 0,1 Gel. %, verwendet werden.
Herstellungsvorschriften
A. Ein Gemisch aus 2,22 g 6,7-Epoxy-12-norcaryophyllan-3-on, 88 g schwach verkupfertem Zink [R. D. Smith und H. E. Simmens, Org. Syn. 41, 72 (1961)] und 400 ml Äthanol wurde 35 Stunden unter Rückfluss erhitzt. Nach Abkühlung des Reaktionsgemisches wurde der Niederschlag abfiltriert und das Filtrat unter vermindertem Druck eingeengt. Chromatographie des Rohproduktes an Kieselgel lieferte 1,7 g reines 12-Norcaryophyll-6 -en-3 -on.
IR: Vmax = 2930, 2858, 1695, 1450, 1381, 1369, 1351, 1331, 1279, 1225, 1160, 1113, 1090, 1059, 1018, 990 und 811 cm.
Die Verbindung hat einen holzigen, leicht camphrigen, an Sandel, Vetiver, Ceder erinnernden Geruch.
B. Ein Gemisch aus 2,22 g 6,7-Epoxy-12-norcaryophyllan-3-on, 88 g verkupfertem Zink [hergestellt wie von R. D. Smith und H. E. Simmens in Org. Syn. 41, 72 (1961) beschrieben, Zinkpulver jedoch mit der doppelten Menge Kupfersulfat behandelt) und 400 ml Äthanol wurde 60 Stunden unter Rückfluss erhitzt und dann in gleicher Weise wie in A beschrieben aufgearbeitet. Neben 0,4 g 12-Norcaryophyll6-en-3-on wurden 1,2 g 12-Norcaryophyll-6-en-3-ol erhalten mit holzigem, leicht camphrigem, erdigem, an Vetiver erinnerndem Geruch.
IR: Vmax = 3450, 2920, 2856, 1670, 1448, 1381, 1366, 1274, 1093, 1058, 990, 939, 890, 840 und 810 cm-t.
C. Zu einem Grignardreagenz, hergestellt aus 0,48 g Magnesium und 2,8 g Methyljodid, wurde im Verlauf von 5 Minuten eine Lösung von 3 g 12-Norcaryophyll-6-en-3-on in 15 ml Äther getropft. Nach 15 Minuten wurde das Reaktionsgemisch unter Eiskühlung vorsichtig mit konzentrierter Ammoniumchloridlösung versetzt und dann mit Äther extrahiert. Die ätherische Phase wurde mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingeengt. Kugelrohrdestillation des Rückstandes lieferte 3,05 g (94% d. Th.) reines Caryophyll-6-en-3 -ol.

The reductive elimination of the 6,7-epoxy group of a compound of the formula II according to process variant (a) can be carried out in a manner known per se, for example by treatment with a chromium (II) ethylenediamine complex [I. K. Kochi et al., Tetrahedron 24, 3503 (1968)] or by means of weakly copper-plated zinc in boiling ethanol [S. M. Kupchan and M. Marnyama, J. Org. Chem. 36, 1187 (1971)].
The reduction of a 12-norcaryophyll-6-en-3-one of the formula Ia to the corresponding secondary alcohol of the formula Ib according to process variant (b) can be carried out by methods known per se with a suitable reducing agent such as a complex alkali metal hydride, e.g. B. LiAIH4, NaAIH4 or NaBH4, a complex trialkoxy-alkali metal hydride, e.g. B. LiAIH (OCH3) 3 or LiAIH [OC (CH3) 3] 3, a substituted aluminum hydride such as diethyl aluminum hydride, diisobutyl aluminum hydride, or a trialkyl aluminum compound such as triisopropyl aluminum. The use of complex alkali metal hydrides, particularly lithium aluminum hydride, is preferred.
The reaction is conveniently carried out in an organic solvent such as an ether, e.g. B. in diethyl ether, dioxane or tetrahydrofuran executed. It is recommended that at low temperatures, i. H. below room temperature, e.g. B. at 0 to work.

B. by ozonolysis of the exocyclic double bond or by oxidation with permanganate, while the selective epoxidation of the endocyclic double bond of the caryophyllene by means of a peracid, for example peracetic acid, in a suitable solvent, e.g. B. in methylene chloride, and in the presence of sodium acetate.
The compounds of the formula I are distinguished by particular odorous qualities. They are characterized by woody, earthy odor notes that are partly reminiscent of sandal or vetiver. The addition of compounds of the formula I to fragrance compositions makes them warmer and fuller in terms of odor. They combine particularly well with woody-smelling compositions and essential oils. The compounds can therefore be used as odoriferous substances in perfumery, for example for the production or odor modification of odoriferous compositions, such as perfumes, perfume bases, etc. by adding olfactory perceptible amounts, e.g. B. 0.1 to 10 wt.% To mixtures of known fragrances. A wide field of application is z. B. the production of chypre-like fragrance compositions.
The compounds can also be used for perfuming technical and cosmetic products of all kinds, e.g. B. of solid and liquid detergents, synthetic detergents, aerosols, soaps, creams, lotions, etc., z. B. in concentrations of about 0.02 to 0.1 gel. %, be used.
Manufacturing regulations
A. A mixture of 2.22 g of 6,7-epoxy-12-norcaryophyllan-3-one, 88 g of slightly copper-plated zinc [R. D. Smith and H. E. Simmens, Org. Syn. 41, 72 (1961)] and 400 ml of ethanol were refluxed for 35 hours. After the reaction mixture had cooled, the precipitate was filtered off and the filtrate was concentrated under reduced pressure. Chromatography of the crude product on silica gel yielded 1.7 g of pure 12-norcaryophyll-6-en-3 -one.

The compound has a woody, slightly camphor-like odor reminiscent of sandal, vetiver, cedar.
B. A mixture of 2.22 g of 6,7-epoxy-12-norcaryophyllan-3-one, 88 g of copper-plated zinc [prepared as described by RD Smith and HE Simmens in Org. Syn. 41, 72 (1961), zinc powder but treated with twice the amount of copper sulfate) and 400 ml of ethanol was refluxed for 60 hours and then worked up in the same way as described in A. In addition to 0.4 g of 12-norcaryophyll-6-en-3-one, 1.2 g of 12-norcaryophyll-6-en-3-ol were obtained with a woody, slightly camphoric, earthy odor reminiscent of vetiver.

C. A solution of 3 g of 12-norcaryophyll-6-en-3-one in 15 ml of ether was added dropwise to a Grignard reagent, prepared from 0.48 g of magnesium and 2.8 g of methyl iodide, over the course of 5 minutes. After 15 minutes, the reaction mixture was carefully mixed with concentrated ammonium chloride solution while cooling with ice and then extracted with ether. The ethereal phase was washed with water, dried with sodium sulfate and concentrated. Kugelrohr distillation of the residue yielded 3.05 g (94% of theory) of pure caryophyll-6-en-3-ol.
