Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 18-Methylensteroiden der Formel

worin R Wasserstoff oder eine veresterte Hydroxygruppe bedeutet und in welcher in 6,7-Stellung eine weitere Doppelbindung vorhanden sein kann.
Eine veresterte Hydroxygruppe R leitet sich vorzugsweise von organischen Säuren der aliphatischen, aromatischen oder heterocyclischen Reihe ab, insbesondere von solchen mit 1-18 Kohlenstoffatomen, z.B. der Ameisensäure, Essigsäure, Propionsäure, der Buttersäuren, Valeriansäuren, wie n-Valeriansäure, oder Trimethylessigsäure, Trifluoressigsäure, der Capronsäuren, wie 8-Trimethylpropionsäure oder Diäthylessigsäure, der Oenanth-, Capryl-, Pelargon-, Caprin-, Undecylsäuren, der Undecylensäure, der Laurin-, Myristin-, Palmitinoder Stearinsäuren, der Ölsäure, oder einer Cyclopropan-, -butan-, -pentan- und -hexancarbonsäure, einer Cycloalkylalkancarbonsäure z.B.
Cyclopropylmethancarbonsäure, Cyclobutylmethancarbonsäure, Cyclopentyläthancarbonsäure, Cyclohexyläthancarbonsäure, der Benzoesäure, von Phenoxyalkansäuren, wie Phenoxyessigsäure, von Dicarbonsäuren, wie Bernsteinsäure, Phthalsäure, Chinolinsäure, der Furan-2 -carbonsäure, 5-tert.-Butyl-furan-2-carbonsäure, 5-Brom -furan-2-carbonsäure, der Nicotinsäure oder der Isonicotinsäure, oder von Sulfonsäuren, wie Benzolsulfonsäuren, oder von anorganischen Säuren, wie z.B. Phosphor- oder Schwefelsäuren.
Die neuen Verbindungen der obigen Formel I besitzen wertvolle pharmakologische Eigenschaften. So weisen sie neben einer gestagenen Wirkung insbesondere eine ovulationshemmende Wirkung auf, wie sich im Tierversuch z.B. an der Ratte zeigt. Die neuen Verbindungen können daher als Gestagene und Contraceptiva Verwendung finden. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere von pharmakologisch wirksamen Verbindungen.
Besonders hervorzuheben ist das 18-Methylen-19-nor-pro- gesteron, das 17a-Acetoxy-l 8-methylen- 1 9-nor-progesteron und ihre 6-Dehydro-Derivate.
Das Verfahren zur Darstellung der A4-3-Oxo-Verbindun- gen der obigen Formel ist dadurch gekennzeichnet, dass man Verbindungen der Formel

worin Y1 und Y je eine freie oder ketalisierte Oxogruppe und R Wasserstoff oder eine veresterte Hydroxygruppe bedeuten, und in welcher eine Doppelbindung ausgehend vom 5-Kohlenstoffatom vorhanden ist, wobei jedoch mindestens eine der Gruppen Yl, Y eine ketalisierte Oxogruppe ist, zur Spaltung der Ketalgruppen mit Säuren hydrolysiert.
Wenn erwünscht, können erhaltene Verbindungen in der 6,7-Stellung unter Einführung einer Doppelbindung dehydriert werden.
Die Ausgangsstoffe der Formel II können z.B. nach dem Verfahren des französischen Patentes 1 601140 hergestellt werden. Man geht z.B. von entsprechenden eventuell in 17 Stellung substituierten - 1 9-Nor-pregnen-20-hydroxy- 18- -säure(18 < 20)-laktonen aus, die in 3-Stellung eine freie oder geschützte Hydroxygruppe oder eine ketalisierte Oxogruppe aufweisen und gelangt durch Anwendung des Verfahrens lb) des genannten französischen Patentes zu Verbindungen der obigen Formel II, worin Y eine freie Oxogruppe darstellt.
Beim Verfahren la) des genannten französischen Patentes geht man vorteilhaft von in 17-Stellung gemäss der Formel II substituierten Verbindungen aus, in denen die 20-Oxogruppe ketalisiert ist, wobei auch eine 3-Oxogruppe mit Vorteil solchermassen geschützt ist. Man gelangt so zu 20-Monoketalen und 3,20-Diketalen der obigen Formel II, in welchen dann verfahrensgemäss die ketalisierten Oxogruppen freigesetzt werden. Man kann auch von Verbindungen mit einer freien oder geschützten 3-Hydroxygruppe ausgehen und durch Anwendung der Verfahren des genannten Patentes zu 20-Ketalen entsprechend der obigen Formel II gelangen, in denen jedoch eine 3-Hydroxygruppe oder eine geschützte 3-Hydroxygruppe vorhanden ist: diese wird dann in an sich bekannter Weise zur Oxogruppe oxydiert, gegebenenfalls unter Einführung der 4,5-Doppelbindung.
Die verfahrensgemässe Spaltung der ketalisierten Oxogruppen erfolgt in an sich bekannter Weise mit einer Säure, z.B. 60-80S0iger wässeriger Essigsäure. wenn erwünscht, in Gegenwart von Perchlorsäure.
Die gegebenenfalls vorzunehmende Einführung einer zusätzlichen Doppelbindung in 6.7-Stellung kann in an sich bekannter Weise ausgeführt werden, z.B. mit einem Chinon, z.B. Chloranil. oder Dicyanchlorbenzochinon. z.B. in Toluollösung.
Die gemäss der vorliegenden Erfindung herzustellenden Verbindungen der obigen Formel I können zur Herstellung von pharmazeutischen Präparaten für die Human- oder Veterinärmedizin verwendet werden. Als pharmazeutische Träger verwendet man organische oder anorganische Stoffe, die für die enterale, z.B. orale, parenterale oder topische Gabe geeignet sind. Für die Bildung derselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z.B. Wasser. Gelatine. Milchzucker, Stärke. Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole. Gummi, Polyalkylenglykole, Vaseline, Cholesterin und andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können in fester Form, z.B. als Tabletten. Dragees oder Kapseln, oder in flüssiger oder halbflüssiger Form als Lösungen, Suspensionen, Emulsionen, Salben oder Cremen vorliegen.
Gegebenenfalls sind diese pharmazeutischen Präparate sterilisiert und bwz. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die neuen Verbindungen können auch als Ausgangsprodukte für die Herstellung anderer wertvoller Verbindungen dienen.
Die erfindungsgemäss hergestellten Verbindungen können auch als Futterzusatzmittel verwendet werden.
Die Erfindung wird im folgenden Beispiel näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
Zu einer unter Stickstoff gerührten Suspension von 2,0 g Triphenylmethylphosphoniumbromid in 35 ml Äther werden 1,0 g einer 32%igen n-Butyllithium-Paraffin-Suspension zugefügt. Man rührt das Reaktionsgemisch 2 Stunden bei Raumtemperatur, tropft dann eine Lösung von 500 mg A5(10) 3,3 ,20,20-Bisäthylendioxy- 1 8-oxo- 1 9-nor-pregnen in 15 ml Tetrahydrofuran zu und kocht anschliessend 18 Stunden unter Rückfluss. Die abgekühlte Lösung wird mit Äther verdünnt, von unlöslichen Anteilen abfiltriert, das Filtrat mit Eis-Wasser gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft. Den amorphen Rückstand löst man in 15 ml Aceton, setzt 60 mg p-Toluolsulfonsäure zu und lässt 18 Stunden bei Raumtemperatur stehen.
Das Reaktionsgemisch wird anschliessend auf Eis-Wasser gegossen und die wässerige Suspension dreimal mit Methylenchlorid extrahiert. Die mit gesättigter Natriumbicarbonatlösung und mit Wasser neutralgewaschenen und getrockneten Methylenchlorid-Auszüge liefern nach Eindampfen im Wasserstrahlvakuum, -Chromatographie des anfallenden Rohproduktes an Silicagel und anschliessender Kristallisation aus Methylenchlorid/Petroläther, reines 18 -Methylen-19-nor-progesteron. [D20 = +2300 (c = 1,00).
Das als Ausgangsstoff verwendete 510 > -3,3,20,20-Bis- methylendioxy-18,oxo-19-nor-pregnen kann auf folgendem Wege erhalten werden:
Zu einer kurz auf 80" erwärmten Suspension von 76 g Bleitetraacetat und 26 g Calciumcarbonat in 5 1 Tetrachlorkohlenstoff werden 14,0 g des 18-20 Laktons der 3B-Acetoxy- -Sa-chlor-6p,2Q,-dihydroxy-pregnan-18-säure (hergestellt aus dem entsprechenden bekannten As-Lakton durch Anlagerung unterchloriger Säure) und 17 g Jod zugegeben. Das Reaktionsgemisch wird anschliessend unter Rühren und Bestrahlen mit einer 1000-Watt-Lampe 90 Minuten am Rückflusskühler gekocht.
Die übliche Aufarbeitung und Chromatographie liefert das bei 220-2200 schmelzende 1 8,20-Lacton der 3z-Acetoxy- -Scc-chlor-6p,19-oxido-20p-hydroxy-pregnan- 18-säure. Dieses wird in an sich bekannter Weise in Stellung 3 hydrolysiert und das anfallende 18,20-Lacton der 3l,20fi-Dihydroxy-Sa- -chlor-6lS,19-oxido-pregnan-18-säure (F. 302-303 0) in Aceton -Methylenchlorid-Gemisch mit 8n.-Chromsäure in verdünnter Schwefelsäure zum entsprechenden 3-Keton oxydiert.
Durch Erwärmen einer methanolischen Lösung der letzteren Verbindung mit Kaliumacetat wird das Chloratom in Stellung 5 in Form von Salzsäure eliminiert, wobei das Lacton der 84-3-0xo-6P,1 9-oxido-2qp-hydroxy-pregnan- 18-säure (F. 260 261") anfällt. 6,6 g dieser Verbindung liefern bei der Reduktion mit 130 g Zinkpulver in 130 ml Eisessig und 7 ml Wasser das bei 256-258" schmelzende 1 8,20-Lacton der \4-3-Oxo- - 19,20P-dihydroxy-pregnen- 18-säure. Durch Oxydation mit 8n.-Chromsäure in Aceton-Methylenchlorid und anschliessende Decarboxylierung der gebildeten 19-Säure durch einstündiges Erwärmen auf 50 in Pyridin wird das 1 8,20-Lacton der (10) <RTI
ID=2.26> -3-Oxo-20p-hydroxy- l 9-nor-pregnen- 18-säure erhalten.
3,0 g dieser Verbindung werden unter Rühren zu einer Lösung von 1,0 g Oxalsäure in 40 ml Äthylenglykol zugegeben und das Reaktionsgemisch 30 Minuten bei Raumtemperatur stehen gelassen. Die Aufarbeitung liefert das 1 8,20-Lacton der Xs (10) 3,3-Athylendioxyd-20 S-hydroxy- l 9-nor-pregnen- 18 -säure.
Durch Erwärmen mit 20%iger Kaliumhydroxydlösung, anschliessende Neutralisation der Lauge mit fester Kohlensäure, Lyophilisation der Lösung, Methylierung des isolierten Kaliumsalzes des 18-Säure mit Methyljodid und Oxydation der nun freien Hydroxygruppe in Stellung 20 erhält man den Methylester der 5 (10) -3,3 -Äthylendioxy-20-oxo- 19- -nor-pregnen-18-säure, der nach Ketalisierung, Lithiumaluminiumhydrid-Reduktion und darauffolgender Oxydation der gebildeten 18-Hydroxygruppe mit Chromsäure in Pyridin das gewünschte ,5(l )-3,3,20,20-Bisäthylendioxy-1 8-oxo-19-nor- -pregnen liefert.

The new compounds of the above formula I have valuable pharmacological properties. The compounds of the above formula I to be prepared according to the present invention can be used for the preparation of pharmaceutical preparations for human or veterinary medicine. The pharmaceutical carriers used are organic or inorganic substances which are suitable for enteral, e.g. oral, parenteral or topical administration are suitable. Substances that do not react with the new compounds, e.g. Water. Gelatin. Milk sugar, starch. Magnesium stearate, talc, vegetable oils, benzyl alcohols. They can also contain other therapeutically valuable substances. The new compounds can also serve as starting materials for the production of other valuable compounds.
The compounds prepared according to the invention can also be used as feed additives.

1.0 g of a 32% strength n-butyllithium-paraffin suspension is added to a suspension, stirred under nitrogen, of 2.0 g of triphenylmethylphosphonium bromide in 35 ml of ether. The 510> -3,3,20,20-bis-methylenedioxy-18, oxo-19-nor-pregnen used as starting material can be obtained in the following way:
14.0 g of the 18-20 lactone of 3B-acetoxy- -Sa-chloro-6p, 2Q, -dihydroxy-pregnan-18.
