Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von neuen in 2-Stellung durch Chlor substituierten Steroiden der Pregnanreihe der allgemeinen Forrnel

worin Rl Wasserstoff, eine Methylgruppe oder ein Halogenatom, R Wasserstoff oder eine z- oder p-ständige Methylgruppe, R3 eine freie oder veresterte Hydroxylgruppe, insbesondere eine mit einer niederen Alkancarbonsäure veresterte Hydroxylgruppe und R4 eine freie, veresterte oder verätherte Hydroxylgruppe bedeuten und X und Y je für ein Halogenatom stehen.
Die genannten veresterten Hydroxylgruppen sind vor allem solche, die sich von organischen Carbonsäuren der aliphatischen, alicyclischen, aromatischen oder heterocyclischen Reihe ableiten, insbesondere von solchen mit 1-18 Kohlenstoffatomen, z.B. der Ameisensäure, Essigsäure, Propionsäure, der Buttersäuren, Valeriansäuren, wie n-Valeriansäure, oder Trimethylessigsäure, Trifluoressigsäure, der Capronsäuren, wie 5-Trimethyl-propionsäure oder Diäthylessigsäure, der Önanth-, Capryl-, Pelargon-, Caprin-, Undecylsäuren, z.B. der Undecylensäure, der Laurin-, Myristin-, Palmitin- oder Stearinsäuren, z.B.
der ÖIsäure, Cyclopropan-, -butan-, -pentan- und -hexancarbonsäure, Cyclopropylmethancarbonsäure, Cyclobutylmethancarbonsäure, Cyclopentyläthancarbonsäure, Cyclohexyläthancarbonsäure, der Cyclopentyl-, Cyclohexyl- oder Phenylessigsäuren oder -propionsäuren, der Benzoesäure, Phenoxyalkansäuren, wie Phenoxyessigsäure, Dicarbonsäuren, wie Bernsteinsäure, Phthalsäure, Chinolinsäure, der Furan-2carbonsäure, 5-tert.-Butyl-furan-2-carbonsäure, 5-Brom-furan -2-carbonsäure, der Nicotinsäure oder der Isonicotinsäure, oder von Sulfonsäuren, wie Benzolsulfonsäuren oder von anorganischen Säuren, wie z.B. Phosphor- oder Schwefelsäuren.
Die Estergruppen können sich aber auch von Orthocarbonsäuren wie Orthoameisensäure, Orthoessigsäure oder Orthopropionsäure ableiten, wobei diese Säuren sowie auch die oben genannten Dicarbonsäuren cyclische 17,21-Ester liefern können.
Als verätherte Hydroxygruppen sind besonders diejenigen zu nennen, welche sich von Alkoholen mit 1-8 Kohlenstoffatomen ableiten, wie niederaliphatischen Alkanolen, Äthylalkohol, Methylalkohol, Propylalkohol, iso-Propylalkohol, den Butyl- oder Amylalkoholen, von Cycloalkanolen, wie Cyclopentanol oder von araliphatischen Alkoholen, insbesondere von monocyclischen arylniederaliphatischen Alkoholen, wie Benzylalkohol, oder von heterocyclischen Alkoholen, wie x-Tetrahydropyranol oder -furanol.
Unter Halogenatomen sind Bromatome, in erster Linie aber Chlor- und Fluoratome zu verstehen.
Die erfindungsgemäss erhältlichen neuen Verbindungen der obigen Formel (1) besitzen wertvolle pharmakologische Eigenschaften. So weisen sie insbesondere eine lokale antiinflammatorische Wirkung auf, wie sich im Tierversuch, z.B. an der Ratte, in Dosen von 0,001 bis 0,1 mg lokal im Fremdkörpergranulom-Test zeigt. Die neuen Verbindungen können daher als lokale anti-inflammatorische Mittel Verwendung finden. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere von pharmakologisch wirksamen Verbindungen.
Besonders hervorzuheben sind Verbindungen obiger Formel, worin Rl Wasserstoff oder ein Fluoratom, R2 eine x-Methylgruppe, R3 Wasserstoff oder der Rest einer niederen Alkancarbonsäure, R; eine freie, veresterte oder mit einem niederen Alkanol, Tetrahydrofuranol oder Tetrahydropyranol verätherte Hydroxylgruppe bedeuten und X für ein Chloratom und Y für ein Chlor- oder Fluoratom stehen, und in allererster Linie das 6x-Fluor-2,9x,ll-trichlor-16;
;c-methyl-17.sc-hydroxy- -21 -acetoxy-3 ,20-dioxo-pregna- 1 ,4-dien, und das 2,9,l 1 -Tri- chlor- 1 6x-methyl- 1 7z-hydroxy-21 -acetoxy-3,20-dioxo-pregna- -1,4-dien, die beispielsweise an der Ratte bei lokaler Gabe in einer Dosis von 0,003 bis 0,01 mg eine deutliche antiinflammatorische Wirkung aufweisen.
Die Verbindungen der obigen Formel (I) werden erhalten, wenn man erfindungsgemäss in einer Verbindung der allgemeinen Formel

worin R1, R, R3, R4, X und Y die obgenannten Bedeutungen besitzen, Chlor an die 1,2-Doppelbindung addiert und aus der erhaltenen 1,2-Dichlorverbindung Chlorwasserstoffsäure abspaltet. Wenn erwünscht, wird in erhaltenen Verbindungen der Formel I eine vorhandene freie Hydroxygruppe verestert oder in 21-Stellung veräthert.
Die Addition von Chlor an die 1,2-Doppelbindung erfolgt in an sich bekannter Weise. Beispielsweise kann man sie in einem inerten Lösungsmittel, wie z.B. Propionsäure, bei tiefer Temperatur im Dunkeln ausführen. Die Abspaltung von Chlorwasserstoff aus den 1 .2-Dichlorverbindungen erfolgt durch Behandeln mit einer Base, vorzugsweise einer tertiären organischen Stickstoffbase wie z.B. Triäthylamin, Pyridin oder Kollidin.
Wie gesagt werden, wenn erwünscht, freie Hydroxygruppen in 17- und 21-Stellung verestert. Es können sowohl 17xoder 21-Monoester als auch 17x.21-Diester hergestellt werden.
Zur Herstellung der 21-Monoester behandelt man die 21-Hydroxyverbindungen in an sich bekannter Weise mit reaktiven funktionellen Carbonsäurederivaten, vorzugsweise mit solchen der oben genannten Säuren, wie z.B. mit einem Säureanhydrid oder einem Säurehalogenid, z.B. in einer tertiären Base wie Pyridin.
Eine freie Hydroxygruppe in 17x-Stellung kann nach an sich bekannten Methoden selektiv verestert werden. Man stellt z.B. durch Acylierung mit einem Carbonsäureanhydrid, wie z.B. Acetanhydrid, unter Zusatz einer starken Säure, besonders einer aromatischen Sulfonsäure, wie z.B. p-Toluolsulfonsäure. als Katalysator, die 17x.21-Diester her und verseift hernach die 21-Estergruppe unter milden Bedingungen. Man verwendet dazu beispielsweise Lösungen eines Alkalimetall carbonats oder -hydrogencarbonats in einem wässrigen, aliphatischen Alkohol wie Methanol oder Äthanol.
17x-Monoester können auch auf folgende, an sich bekannte Weise erhalten werden: Aus Verbindungen der Formel (1), die in 17.x- und 21-Stellung freie Hydroxygruppen aufweisen, erhält man beim Umsetzen mit einem Orthoester des Typs R'-C(OR"), wobei R' ein Wasserstoffatom oder einen Alkylrest und R" einen Alkylrest bedeuten, in Gegenwart einer starken Säure, wie z.B. p-Toluolsulfonsäure, als Katalysator, in einem inerten Lösungsmittel, wie z.B. Benzol, cyclische 17,x,21-Orthoester. Durch Hydrolyse mit einer schwachen-organischen Säure wie z.B. Oxalsäure, wird sodann die 21-Esterbindung selektiv hydrolysiert, wobei ein 17.cc-Mono- ester erhalten wird.
Wenn erwünscht, kann eine freie Hydroxygruppe in 21 Stellung in an sich bekannter Weise, vorzugsweise mit reaktiven Derivaten der oben genannten Alkohole, veräthert werden. Beispielsweise erhält man mit Dihydropyran in einem an der Reaktion sich nicht beteiligenden Lösungsmittel wie Tetrahydrofuran, Diäthyläther oder Chloroform in Gegenwart von Phosphoroxychlorid, die 21 -Tetrahydropyranyläther.
Die als Ausgangsstoffe zu verwendenden Verbindungen der obigen Formel II sind bekannt oder lassen sich nach an sich bekannten Methoden herstellen.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, bei denen Chlor unter den Reaktionsbedingungen gebildet wird.
Es können Präparate zur Anwendung in der Humanoder Veterinärmedizin, welche die neuen, oben beschriebenen pharmakologisch wirksamen Stoffe der Formel I als aktive Substanzen zusammen mit einem pharmazeutischen Trägermaterial enthalten, hergestellt werden. Als Träger verwendet man organische oder anorganische Stoffe, die für die enterale, z.B.
orale, parenterale oder topicale Gabe geeignet sind. Für die Bildung derselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren. Die pharmazeutischen Präparate können in fester Form, oder in flüssiger oder halbflüssiger Form vorliegen. Gegebenenfalls enthalten diese pharmazeutischen Präparate Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- od. Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die neuen Verbindungen können auch als Ausgangsprodukte für die Herstellung anderer wertvoller Verbindungen dienen.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
5,0 g 4-6-Fluor-9a,1 1 p.dichlor. 1 6-methyl-l 7x-hy- droxy-21-acetoxy-3,20-dioxo-pregnadien werden in 250 ml Dioxan gelöst. Nach Abkühlen bis zur beginnenden Kristallisation des Dioxans gibt man successive innerhalb von 8 Tagen bei 0-5" im Dunkeln 215 ml einer Lösung von 72 g Chlor in 1000 ml Propionsäure in Portionen von je 20-30 ml hinzu.
Anschliessend wird die Reaktionslösung auf 3 It Eiswasser ausgetragen. Man extrahiert mehrmals mit Methylenchlorid und wäscht die Extrakte nacheinander mit Wasser, gesättigter wässriger Natriumbicarbonatlösung und nochmals mit Wasser. Die gewaschenen, vereinigten Extrakte werden über Natriumsulfat getrocknet, filtriert und im Vakuum bei 30-35" Badtemperatur vollständig eingedampft. Man löst das erhaltene noch rohe #4-6α-Fluor-1,2,9α,11 -tetrachlor-16x-methyl- -17α-hydroxy-21-acetoxy-3,20-dioxo-pregnen (9,5 g) in 300 Pyridin und lässt 20 Stunden bei Raumtemperatur stehen, wobei ein allmählicher Farbumschlag von gelbbraun in dunkelbraun zu beobachten ist. Die Reaktionslösung wird darauf auf 6 lt normale, eisgekühlte Salzsäure ausgetragen.
Man extrahiert viermal mit je 1 It Methylenchlorid und wäscht die Extrakte nacheinander mit eisgekühlter, normaler Salzsäure, Wasser, gesättigter, wässriger Natriumbicarbonatlösung und nochmals mit Wasser. Die gewaschenen, vereinigten Extrakte werden über Natriumsulfat getrocknet, filtriert und im Vakuum vollständig eingedampft. Zur Reinigung löst man das erhaltene Rohprodukt (6,27 g) in Methylenchlorid, filtriert durch eine Säule von Silicagel und eluiert weiter mit Methylenchlorid. Die dünnschichtchromatographisch einheitliche Mittelfraktion wird aus Methylenchlorid-Äther umkristalli siert und liefert au 4-6a-Fluor-2,9a,11,3-trichlor-l 6a-methyl- -17α-hydroxy-21-acetoxy-3,20-dioxo-pregnadien vom F (2470) 248-249" (u.Z.).
Aus der Mutterlauge gewinnt man beim Aufarbeiten noch weitere Mengen desselben Produkts.
Beispiel 2
2,60 g A1 4-6α-Fluor-2,9α11 p-trichlor-l acc-methyl-17-hy- droxy-21 -acetoxy-3 ,20-dioxo-pregnadien werden in 200 ml Methanol unter Erwärmen zum Sieden und Durchleiten von Stickstoff gelöst. Anschliessend kühlt man die erhaltene Lösung unter Stickstoff auf 1-3" ab und lässt sodann innerhalb von 30-60 Minuten eine Auflösung von 0,50 g Natriumbicarbonat in 10 ml Wasser zutropfen. Die Reaktionslösung wird hernach über Nacht bei 1-3 unter Stickstoff weiter gerührt, wobei das Reaktionsprodukt successive kristallisiert. Man kontrolliert den Ablauf der Verseifung mittels Dünnschichtchromatographie auf Silicagel mit Toluol-Essigester 1:1 als Laufmittel.
Nach einer Verseifungsdauer von 10-15 Stunden lässt sich in der Reaktionslösung kein Ausgangsmaterial mehr feststellen. In diesem Moment wird das Reaktionsgemisch auf 1500 ml Wasser ausgetragen. Man extrahiert mehrmals mit Methylenchlorid, wäscht die Extrakte nacheinander wiederholt mit Wasser, trocknet diese über Natriumsulfat, filtriert und dampft sie vollständig ein. Die Ausbeute an kristallinem Rohprodukt beträgt 2,4 g, das zwischen 232-233 0 schmilzt; das Produkt ist dünnschichtchromatographisch rein (Silicagel, Toluol: Essigester 1:1). Zur weiteren Reinigung kristallisiert man das leicht gelbstichige Rohprodukt aus Methanol, wobei zunächst 1,0 g A1S4-6a-Fluor-2,9,a,11S-trichlor-16x-methyl- -17α,21-dihydroxy-3,20-dioxo-pregnadien vom F. (231 ) 2322330 resultieren.
Die Mutterlauge liefert nach Konzentrieren weitere Mengen desselben Produkts.
Beispiel 3
1,3 ml Trimethylessigsäurechlorid werden unter Rühren in Stickstoffatmosphäre in 25 ml reinem, trockenem Pyridin bei 0 gelöst. Zur erhaltenen klaren, farblosen Lösung lässt man innerhalb 5 Minuten eine Aulösung von 1,0 g A"4-6- -Fluor-2,9,l 1 p.trichlor. 1 6a-methyl- 17a,21 -dihydroxy-3,20- -dioxo-pregnadien in 15 ml reinem, trockenem Pyridin unter stetem Rühren bei 0 in Stickstoffatmosphäre zerfliessen. Die Reaktionslösung wird 8-12 Stunden bei 0 unter Stickstoff gerührt. Nach dieser Reaktionsdauer lässt sich im Reaktionsgemisch mittels Dünnschichtchromatographie [Silicagel, Toluol-Essigester 1:1] kein Ausgangsmaterial mehr nachweisen.
Die Reaktionslösung wird nun auf Eiswasser ausgetragen und mit verdünnter Salzsäure angesäuert. Man extrahiert die Reaktionsprodukte mit Essigester und wäscht die Essigesterextrakte der Reihe nach mit verdünnter Salzsäure, Wasser, verdünnter Sodalösung und Wasser, trocknet sie über Natriumsulfat, filtriert und dampft sie ein. Der weisse, feinkristalline Rückstand (1,3 g) wird aus Essigester umkristallisiert.
Es resultiert reines #1.4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl- -17α-hydroxy-21-trimethylacetoxy-3,20-dioxo-pregnadien vom F. 215.2170.
In analoger Weise gewinnt man aus #1,4-6α-Fluor-2,9α,- 11 -trichlor-16a-methyl- 17,z,21 -dihydroxy-3,20-dioxo-pregna- dien durch Umsetzen mit Undecylensäurechlorid das A1µ4-6a- -Fluor-2,9α,11ss-trchlor-16α-methyl-17α-hydroxy-21-unde.

Beispiel 4
4.8 g #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α,21-di- hydroxy-3,20-dioxo-pregnadien, 0,025 g p-Toluolsulfosäure, 6,0 ml Dimethylformamid und 6,0 ml Orthopropionsäure-tri- äthylester werden zusammen unter Rühren in Stickstoffatmosphäre während 4 Stunden auf 115 erwärmt. Man kühlt die klare Reaktionslösung ab, fügt 0,5 ml Pyridin hinzu und dampft im Vakuum vollständig ein. Der kristalline Rückstand wird mit 25 ml Methanol angerieben, abgenutscht, mit etwas gekühltem Methanol ausgewaschen und schliesslich bei 60 70 im Vakuum getrocknet.
Man erhält reines A1ç4-6a-Fluor- -2,9α11ss-trichlor-16α-methyl-17α,21-(1'-äthoxy)-propyliden- dioxy-3,20-dioxo-pregnadien in feinen Nadeln.
In analoger Weise gewinnt man aus #1,4-6α-Fluor-2,9α,- 11 p-trichlor- 16 -methyl-17a,21 dihydroxy-3,20-dioxo-pregna- dien durch Umsetzen mit Orthoessigsäure-triäthylester das #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α,21-(1'-äthoxy)- -äthylidendioxy-3,20-dioxo-pregnadien, durch Umsetzen mit Orthovaleriansäuretrimethylester das #1,4-6α-Fluor-2,9α,11ss- -trichlor-16α-methyl-17α,21-(1'-methoxy)-pentylidendioxy- -3,20-dioxo-pregnadien, durch Umsetzen mit Orthobenzoesäure-triäthylester das #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-me- thyl-17α
;,21-(1'-äthoxy)-benzylidendioxy-3,20-dioxo-pregna- dien, durch Umsetzen mit Cyclopentanon-diäthylacetal das #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α,21-cyclopen- tylidendioxy-3,20-dioxo-pregnadien, mit Benzaldehyddiäthylacetal das #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α,21- -benzylidendioxy-3,20-dioxo-pregnadien und mit ss-Carbo methoxyorthopropionsäuretrimethylester das \1 4-6a-Fluor -2,9α11ss-trichlor-16α-methyl-17α,21-(1'-methoxy-3'-carbo- methoxy)-propylidendioxy-3,20-dioxo-pregnadien.
Eine Mischung des #1,4-6α-Fluor-2,9α,11ss-trichlor-16α- -methyl-17α,21-(l'-methoxy-3'-carbomethoxy)-propyliden- dioxy-3,20-dioxo-pregnadien in Methanol mit einer wässrigen Lösung von Natriumbicarbonat wird unter Stickstoff verrührt bis mittels Dünnschichtchromatographie kein Ausgangsmaterial festzustellen ist. Die Lösung wird in Eiswasser gegossen, mit verdünnter Salzsäure angesäuert und mit Methylenchlorid ausgezogen. Der nach dem Eindampfen entstehende ölige Rückstand wird in Methanol aufgenommen, wobei das #1,4- -6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α,21-(1'-methoxy-3'- -carboxy)-propylidendioxy-3,20-dioxo-pregnadien auskristallisiert; die neue Verbindung löst sich in einer wässrigen Lösung von Natriumbicarbonat.
Beispiel 5
Eine Suspension von #1,4-6α-Fluor-2,9α,11ss-trichlor-16α- -methyl-17α,21-(1'-äthoxy)-propylidendioxy-3,20-dioxo-pre- gnadien in Methanol wird bei 45 mit einer 2-n. Lösung von Oxalsäure in Wasser behandelt bis eine klare Lösung entsteht.
Man giesst die Lösung hernach auf eine gesättigte wässrige Lösung von Natriumhydrogencarbonat und extrahiert mit Methylenchlorid. Die Extrakte werden mit Wasser gewaschen, getrocknet und eingedampft. Nach Umkristallisieren des Rückstandes aus Essigester erhält man #1,4-6α-Fluor-2,9α,11ss -trichlor-16α-methyl-21-hydroxy-17α-propionoxy-3,20-dioxo- -pregnadien.
In analoger Weise gewinnt man aus #1,4-6α-Fluor-2,9α,- 11ss-trichlor-16α-methyl-17α-21-(1'-methoxy-3'-carboxy)-pro- pylidendioxy-3.20-dioxo-pregnadien durch Spaltung mittels verdünnter wässriger Oxalsäure das #1,4-6α-Fluor-2,9α,11ss-tri- chlor-16α-methyl-21-hydroxy-17α-hernisuccinoyloxy-3,20- -dioxo-pregnadien, das in einer wässrigen Lösung von Natriumhydrogencarbonat löslich ist, aus #1,4-6α-Fluor-2,9α,- 11ss-trichlor-16α-methyl-17α,21-(1'-methoxy)-pentylidendioxy- -3,20-dioxo-pregnadien #1,4-6α-Fluor-2,9α,11ss-trichlor-16α- -methyl-21-hydroxy-17α

Beispiel 6 #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α,21-di- hydroxy-3,20-dioxo-pregnadien werden in Methyliodid unter Zusatz von Dimethylformamid gelöst und nach Zugabe von Silberoxyd bei Raumtemperatur gerührt, bis mittels Dünnschichtchromatographie kein Ausgangsmaterial mehr festzustellen ist. Die Silbersalze werden abgetrennt, die blanke Reaktionslösung vollständig eingedampft und der Rückstand an Silicagel chromatographisch gereinigt. In analoger Weise gewinnt man aus #1,4-6α-Fluor-2,9α- 11ss-trichlor-16α-methyl-17α,21-dihydroxy-3,20-dioxo-pregna- dien durch Umsetzen mit Äthyljodid #1,4-6α-Fluor-2,9α,11ss- -trichlor-16α-methyl-17α-hydroxy-21-äthoxy-3,20-dioxo-pre- gnadien, mit Allyljodid A1f4-6x-Fluor-2,9x,l l p-trichlor-16x- -methyl- 17a-hydroxy-21 -allyloxy-3,20-dioxo-pregnadien, mit Benzyljodid #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α- -hydroxy-21-benzyloxy-3,20-dioxo-pregnadien.
Beispiel 7 #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α,21-dihy- droxy-3,20-dioxo-pregnadien werden in 3,4-Dihydropyran unter Zusatz von p-Toluolsulfonsäure-Monohydrat bei Raumtemperatur gerührt bis im Dünnschichtchromatogramm einer Reaktionsprobe kein Ausgangsmaterial mehr festzustellen ist.
Man neutralisiert die umgesetzte p-Toluolsulfonsäure mit Pyridin und dampft die Reaktionslösung im Vakuum vollständig ein. Der Rückstand liefert nach Umkristallisieren aus Essigester reines #1,4-6α-Fluor-2,9α,11ss-trichlor-16α-methyl-17α- -hydroxy-21-(2'-tetrahydropyranyloxy)-3,20-dioxo-pregnadien.

Beispiel 8
25,4 g #1,4-6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl- -17α-hydroxy-21-acetoxy-3,20-dioxo-pregnadien werden in 200 ml Methanol unter Erwärmen zum Sieden und Durchleiten von Stickstoff gelöst. Anschliessend kühlt man die erhaltene Lösung unter Stickstoff auf 0 und lässt hernach innerhalb von 30-60 Minuten eine Auflösung von 0,50 g Natriumbicarbonat in 10 ml Wasser zutropfen. Die Reaktionslösung wird hernach über Nacht bei 0 unter Stickstoff weiter gerührt. Man kontrolliert den Ablauf der Verseifung mittels Dünnschichtchromatographie. Nach einer Verseifungsdauer von 10-15 Stunden lässt sich in der Reaktionslösung kein Ausgangsmaterial mehr feststellen und in diesem Moment wird das Reaktionsgemisch auf 1520 ml Wasser ausgetragen.
Man extrahiert mehrmals mit Methylenchlorid, wäscht die Extrakte nacheinander wiederholt mit Wasser, trocknet sie über Natriumsulfat und dampft sie vollständig ein. Die Ausbeute an Rohprodukt beträgt 2,5 g #1,4-6α,11ss-Difluor-2-chlor-9α- -brom-16α-methyl-17α,21-dihydroxy-3,20-dioxo-pregnadien, das durch Umkristallisieren aus Methanol reines #1,4-6α,11ss- -Difluor-2-chlor-9a-brom- 1 6a-methyl- 1 7z,21 -dihydroxy-3 ,20 -dioxo-pregnadien liefert.
Zur Herstellung des #1,4-6α,1 liP-Difluor-2-chlor-9.a-brom- -16α-methyl-17α-hydroxy-21-trimethylacetoxy-3,20-dioxo-pre- gnadiens werden 1,3 ml Trimethylessigsäurechlorid bei 0 in 25 ml Pyridin gelöst. Man lässt sodann eine Auflösung von 0,9 g #1,4-6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl-17α,21- -dihydroxy-3,20-dioxo-pregnadien in 15 ml Pyridin bei 0 zufliessen und rührt 10-15 Stunden bei 00. Zur Aufarbeitung giesst man die Reaktionslösung auf Eiswasser, extrahiert die ausgefällten Reaktionsprodukte mit Essigester, wäscht die Extrakte nacheinander mit verdünnter Salzsäure, Wasser, verdünnter Sodalösung und Wasser, trocknet sie und dampft sie vollständig ein.
Der weisse, fein kristalline Rückstand wird aus Essigester umgelöst und liefert reines #1,4-6α,11 1ss-Difluor- -2-chlor-9a-brom- 1 6a-methyl- 1 7α-hydroxy-21 -trimethylacetoxy-3 ,20-dioxo-pregnadien.
In analoger Weise erhält man aus #1,4-6α,1 19-Difluor-2 -chlor-9α-brom-16α-methyl-17α,21-dihydroxy-3,20-dioxo-pre- gnadien durch Umsetzen mit n-Valeriansäurechlorid #1,4-6α,- 1 ,B-Difluor-2-chlor-9a-brom-1 6a-methyl- 1 7,a-hydroxy-21 -n- -valeroyloxy-3,20-dioxo-pregnadien, mit Undecylensäurechlorid #1,4-6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl-17α-hy- droxy-21-undecylonoyloxy-3,20-dioxo-pregnadien, mit Benzolychlorid #1,4-6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl- -17α-hydroxy-21 -benzoyloxy-3 ,20-dioxo-pregnadien, mit Phenylpropionsäurechlorid #1,4-6α,11ss-Difluor-2-chlor-9α

1,0 g #1,4-6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl-17α,- 21-dihydroxy-3,20-dioxo-pregnadien werden zusammen mit 1,0 g Bernsteinsäureanhydrid in 25 ml Pyridin mehrere Stunden auf dem siedenden Wasserbad erwärmt. Die Reaktionslösung wird gekühlt, im Vakuum weitgehend eingedampft, und die Essigesterlösung des Rückstandes nach Waschen mit verdünnter Salzsäure und Wasser bis zur beginnenden Kristallisation konzentriert. Man erhält #1,4-6α,11ss-Difluor-2- -chlor-9α-brom-16α-methyl-17α-hydroxy-21-hemisuccinoyloxy- -3,20-dioxo-pregnadien.
Nach derselben Arbeitsweise gewinnt man aus #1,4-6α,- 11 p-Difluor-2-chlor-9x-brom- 16α-methyl-17α,21 -dihydroxy -3,20-dioxo-pregnadien durch Umsetzen mit Glykolsäureanhydrid A1 4-6x,11 -Difluor-2-chlor-9a-brom- 16a-methyl-17a- -hydroxy-21-hemidiglykoloyloxy-3,20-dioxo-pregnadien, mit Phthalsäureanhydrid #1,4-6α,11ss-Difluor-2-chlor-9α-brom- -1 6 -methyl- 1 7 -hydroxy-2 1 -hemiphthaloyloxy-3.20-dioxo- -pregnadien, mit Chinolinsäureanhydrid #1,4-6α
;,11ss-Difluor- -2-chlor-9 -brom- 1 6 -methyl- 1 7 -hydroxy-21 -hemichinolin- oyloxy-3,20-dioxo-pregnadien, mit Propionsäureanhydrid #1,4- -6α,11ss-Difluor-2-chlor-9α-brm-16α-methyl-17-hydroxy-21- -propionyloxy-3 ,20-dioxo-pregnadien.
5,0 #1,4-6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl- -17α,21-dihydroxy-3,20-dioxo-pregnadien, 0,02 g p-Toluolsulfosäure-Monohydrat, 6,0 ml Dimethylformamid und 6,0 ml Orthopropionsäure-triäthylester werden zusammen unter Rühren in Stickstoffatmosphäre während 4 Stunden auf 110 erwärmt. Man kühlt die klare Reaktionslösung ab, fügt 0,5 ml Pyridin hinzu und dampft im Vakuum vollständig ein. Der kristalline Rückstand wird mit 25 ml Methanol angerieben, abgenutscht, mit etwas gekühltem Methanol ausgewaschen und schliesslich bei 60-70 im Vakuum getrocknet. Man erhält reines #1,4-6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl- -17α,21-(1'-äthoxy)-propylidendioxy-3,20-dioxo-pregnadien in feinen Nadeln.
In analoger Weise gewinnt man aus #1,4-6α,1 1ss-Difluor-2- -chlor-9α-brom-16α-methyl-17α,21-dihydroxy-3,20-dioxo-pre- gnadien durch Umsetzen mit Orthoessigsäuretriäthylester #1,4- -6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl-17α,21-(1'- -äthoxy)-äthylidendioxy-3,20-dioxo-pregnadien, mit Orthobenzoesäure-triäthylester #1,4-6α,11ss-Difluor-2-chlor-9α-brom- -16x-methyl-17a,21 -(1 '-äthoxy)-benzylidendioxy-3,20-dioxo- pregnadien, mit Ortho-n-valeriansäure-trimethylester #1,4-6α,- 11ss-Difluor-2-chlor-9α-brom-16α-methyl-17α

Eine Mischung des #1,4-6α,1 -Difluor-2-chlor-9a-brom- -16α-methyl-17α,21-(1'-methoxy-3'-carbomethoxy)-propyliden- dioxy-3,20-dioxo-pregnadien in Methanol mit einer wässrigen Lösung von Natriumhydrogencarbonatlwird unter Stickstoff verrührt, bis im Dünnschichtchromatogramm einer Reaktionsprobe kein Ausgangsmaterial mehr festzustellen ist. Die Reaktionslösung wird in Eiswasser gegossen, mit verdünnter Salzsäure angesäuert und mit Methylenchlorid ausgezogen.
Der nach dem Eindampfen des mehrmals mit Wasser gewaschenen Extrakts verbleibende Rückstand wird in Methanol-Essigester aufgenommen, wobei reines #1,4-6α,11ss-Difluor-2-chlor-9α- -brom-16α-methyl-17α,21-(1'-methoxy-3'-carboxy)-propyliden- dioxy-3,20-dioxo-pregnadien auskristallisiert; die neue Verbindung löst sich in einer wässrigen Lösung von Natriumhydrogencarbonat.
Eine Aufschwemmung von #1,4-6α,11ss-Difluor-2-chlor.
-9a-brom- 1 6a-methyl- 17a,21 -(1 '-äthoxy)-propylidendioxy -3,20-dioxo-pregnadien in Methanol wird bei 45 mit einer 2-n. Lösung von Oxalsäure in Wasser behandelt bis eine klare Lösung entsteht. Man giesst die Lösung sodann auf eine wässrige Lösung von Natriumhydrogencarbonat und extrahiert mit Methylenchlorid. Die Extrakte werden mit Wasser gewaschen, getrocknet und eingedampft. Der Rückstand lie- fert nach Umkristallisieren aus Essigester reines #1,4-6α,11ss- -Difluor-2-chlor-9α-brom-16α-methyl-21-hydroxy-17α-propionoxy-3,20-dioxo-pregnadien.
Analog gewinnt man aus #1,4-6α,11ss-Difluor-2-chlor-9α- -brom-16α-methyl-17α,21-(1'-methoxy)-pentylidendioxy-3,20- -dioxo-pregnadien durch partielle Spaltung mittels verdünnter wässriger Oxalsäure #1,4-6α,11ss-Difluor-2-chlor-9α-brom- methyl-21-hydroxy-17α-n-valeroyloxy-3,20-dioxo-pregnadien, aus #1,4-6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl-17α,21- -(1'-äthoxy)-benzylidendioxy-3,20-dioxo-pregnadien das #1,4- -6α,11ss-Difluor-2-chlor-9α-brom-16α-methyl-21-hydroxy-17α -benzoyloxy-3,20-dioxo-pregnadien, aus #1,4-6α,11ss-Difluor- -2-chlor-9α-brom-16α-methyl-17α

Halogen atoms are bromine atoms, but primarily chlorine and fluorine atoms.
The new compounds of the above formula (1) obtainable according to the invention have valuable pharmacological properties. In particular, they have a local anti-inflammatory effect, as shown in animal experiments, e.g. in the rat, in doses of 0.001 to 0.1 mg locally in the foreign body granuloma test. The new compounds can therefore be used as local anti-inflammatory agents. However, the new compounds are also valuable intermediates for the preparation of other useful substances, in particular pharmacologically active compounds.
Particularly noteworthy are compounds of the above formula in which R1 is hydrogen or a fluorine atom, R2 is an x-methyl group, R3 is hydrogen or the residue of a lower alkanecarboxylic acid, R; a free, esterified or etherified with a lower alkanol, tetrahydrofuranol or tetrahydropyranol hydroxyl group and X stands for a chlorine atom and Y for a chlorine or fluorine atom, and first and foremost 6x-fluoro-2,9x, ll-trichloro-16;
; c-methyl-17.sc-hydroxy- -21-acetoxy-3, 20-dioxo-pregna-1, 4-diene, and the 2.9, l 1 -tri-chloro-16x-methyl-17z -hydroxy-21 -acetoxy-3,20-dioxo-pregna-1,4-diene, which, for example, have a marked anti-inflammatory effect on rats when given locally in a dose of 0.003 to 0.01 mg.

in which R1, R, R3, R4, X and Y have the abovementioned meanings, chlorine is added to the 1,2-double bond and hydrochloric acid is split off from the 1,2-dichloro compound obtained. If desired, any free hydroxyl group present in the compounds of the formula I obtained is esterified or etherified in the 21-position.
The addition of chlorine to the 1,2 double bond takes place in a manner known per se. For example, they can be placed in an inert solvent such as e.g. Propionic acid, run at low temperature in the dark. Hydrogen chloride is split off from the 1,2-dichloro compounds by treatment with a base, preferably a tertiary organic nitrogen base such as e.g. Triethylamine, pyridine or collidine.
As stated, free hydroxyl groups in the 17- and 21-positions are esterified if desired. Both 17x or 21 monoesters and 17x.21 diesters can be produced.
To prepare the 21-monoesters, the 21-hydroxy compounds are treated in a manner known per se with reactive functional carboxylic acid derivatives, preferably with those of the acids mentioned above, such as e.g. with an acid anhydride or an acid halide, e.g. in a tertiary base such as pyridine.
A free hydroxyl group in the 17x position can be selectively esterified by methods known per se. E.g. by acylation with a carboxylic acid anhydride, e.g. Acetic anhydride, with the addition of a strong acid, especially an aromatic sulfonic acid, such as e.g. p-toluenesulfonic acid. as a catalyst, produces the 17x.21-diester and then saponifies the 21-ester group under mild conditions. For this purpose, for example, solutions of an alkali metal carbonate or hydrogen carbonate in an aqueous, aliphatic alcohol such as methanol or ethanol are used.
17x-monoesters can also be obtained in the following manner known per se: Compounds of the formula (1) which have free hydroxyl groups in the 17.x- and 21-positions are obtained on reaction with an orthoester of the type R'-C (OR "), where R 'is a hydrogen atom or an alkyl radical and R" is an alkyl radical, in the presence of a strong acid, such as p-toluenesulfonic acid, as a catalyst, in an inert solvent, e.g. Benzene, 17, x, 21 cyclic orthoesters. By hydrolysis with a weak organic acid such as e.g. Oxalic acid, the 21-ester linkage is then selectively hydrolyzed to give a 17.cc monoester.
If desired, a free hydroxyl group in the 21 position can be etherified in a manner known per se, preferably with reactive derivatives of the alcohols mentioned above. For example, with dihydropyran in a solvent that does not participate in the reaction, such as tetrahydrofuran, diethyl ether or chloroform in the presence of phosphorus oxychloride, the 21-tetrahydropyranyl ether is obtained.
The compounds of the above formula II to be used as starting materials are known or can be prepared by methods known per se.
The invention also relates to those embodiments of the process in which chlorine is formed under the reaction conditions.
Preparations for use in human or veterinary medicine which contain the new pharmacologically active substances of the formula I described above as active substances together with a pharmaceutical carrier material can be produced. The carrier used is organic or inorganic substances which are suitable for enteral, e.g.
oral, parenteral or topical administration are suitable. Substances that do not react with the new compounds come into question for the formation of the same. The pharmaceutical preparations can be in solid form, or in liquid or semi-liquid form. If necessary, these pharmaceutical preparations contain auxiliaries, such as preservatives, stabilizers, wetting agents or emulsifiers, salts to change the osmotic pressure or buffers. They can also contain other therapeutically valuable substances. The new compounds can also serve as starting materials for the production of other valuable compounds.
In the following examples the temperatures are given in degrees Celsius.
example 1
5.0 g 4-6-fluoro-9a, 11 p-dichlor. 1 6-methyl-1 7x-hydroxy-21-acetoxy-3,20-dioxo-pregnadiene are dissolved in 250 ml of dioxane. After cooling until the dioxane starts to crystallize, 215 ml of a solution of 72 g of chlorine in 1000 ml of propionic acid are added successively within 8 days at 0-5 "in the dark in portions of 20-30 ml each.
The reaction solution is then poured onto 3 l of ice water. It is extracted several times with methylene chloride and the extracts are washed successively with water, saturated aqueous sodium bicarbonate solution and again with water. The washed, combined extracts are dried over sodium sulphate, filtered and completely evaporated in vacuo at a bath temperature of 30-35 ". The resulting still crude # 4-6? -Fluoro-1,2,9?, 11-tetrachloro-16x- is dissolved. methyl- 17α-hydroxy-21-acetoxy-3,20-dioxo-pregnene (9.5 g) in 300% pyridine and left to stand for 20 hours at room temperature, a gradual color change from yellow-brown to dark brown being observed is then poured onto 6 liters of normal, ice-cold hydrochloric acid.
It is extracted four times with 1 It methylene chloride each time and the extracts are washed successively with ice-cold normal hydrochloric acid, water, saturated aqueous sodium bicarbonate solution and again with water. The washed, combined extracts are dried over sodium sulfate, filtered and evaporated completely in vacuo. For purification, the crude product obtained (6.27 g) is dissolved in methylene chloride, filtered through a column of silica gel and eluted further with methylene chloride. The middle fraction, which is uniform according to thin-layer chromatography, is recrystallized from methylene chloride-ether and gives 4-6a-fluoro-2,9a, 11,3-trichloro-l 6a-methyl-17α-hydroxy-21-acetoxy-3,20-dioxo -pregnadien vom F (2470) 248-249 "(oc).
Further amounts of the same product are obtained from the mother liquor during work-up.
Example 2
2.60 g of A1 4-6? -Fluoro-2.9? 11 p-trichloro-1 acc-methyl-17-hydroxy-21-acetoxy-3, 20-dioxo-pregnadiene are dissolved in 200 ml of methanol with warming dissolved for boiling and passing nitrogen through. The resulting solution is then cooled to 1-3 "under nitrogen and a solution of 0.50 g of sodium bicarbonate in 10 ml of water is then added dropwise over the course of 30-60 minutes. The reaction solution is then continued overnight at 1-3" under nitrogen The progress of the saponification is checked by means of thin-layer chromatography on silica gel with toluene-ethyl acetate 1: 1 as the mobile phase.
After a saponification time of 10-15 hours, no more starting material can be found in the reaction solution. At this point the reaction mixture is poured into 1500 ml of water. It is extracted several times with methylene chloride, the extracts are washed successively repeatedly with water, dried over sodium sulphate, filtered and evaporated completely. The yield of crystalline crude product is 2.4 g, which melts between 232-233 °; the product is pure by thin-layer chromatography (silica gel, toluene: ethyl acetate 1: 1). For further purification, the slightly yellowish crude product is crystallized from methanol, initially 1.0 g of A1S4-6a-fluoro-2,9, a, 11S-trichloro-16x-methyl- -17α, 21-dihydroxy-3,20- dioxo-pregnadiene from F. (231) 2322330 result.
After concentration, the mother liquor yields further quantities of the same product.
Example 3
1.3 ml of trimethyl acetic acid chloride are dissolved in 25 ml of pure, dry pyridine at 0 while stirring in a nitrogen atmosphere. A solution of 1.0 g of A "4-6- -fluoro-2,9, l 1 p.trichlor. 1 6a-methyl-17a, 21-dihydroxy-3, is left to the resulting clear, colorless solution within 5 minutes. Dissolve 20-dioxo-pregnadiene in 15 ml of pure, dry pyridine with constant stirring in a nitrogen atmosphere at 0. The reaction solution is stirred under nitrogen for 8-12 hours at 0. After this reaction time, the reaction mixture can be purified by thin layer chromatography [silica gel, toluene-ethyl acetate 1: 1] no longer detect any starting material.
The reaction solution is then poured onto ice water and acidified with dilute hydrochloric acid. The reaction products are extracted with ethyl acetate and the ethyl acetate extracts are washed in sequence with dilute hydrochloric acid, water, dilute soda solution and water, dried over sodium sulfate, filtered and evaporated. The white, finely crystalline residue (1.3 g) is recrystallized from ethyl acetate.
The result is pure # 1.4-6? -Fluoro-2,9?, 11ss-trichloro-16? -Methyl-17? -Hydroxy-21-trimethylacetoxy-3,20-dioxo-pregnadiene with a melting point of 215.2170.

4.8 g # 1,4-6α-fluoro-2,9α, 11ss-trichloro-16α-methyl-17α, 21-di-hydroxy-3,20-dioxo-pregnadiene, 0.025 g p-toluenesulfonic acid, 6 , 0 ml of dimethylformamide and 6.0 ml of tri-ethyl orthopropionate are heated together to 115 for 4 hours while stirring in a nitrogen atmosphere. A suspension of # 1,4-6α-fluorine-2,9α, 11ss-trichloro-16α-methyl-17α, 21- (1'-ethoxy) -propylidenedioxy-3,20-dioxo-pre- grace in methanol is given at 45 with a 2-n. Treated solution of oxalic acid in water until a clear solution is obtained.

25.4 g of # 1,4-6α, 11ss-difluoro-2-chloro-9α-bromo-16α-methyl-17α-hydroxy-21-acetoxy-3,20-dioxo-pregnadiene are added in 200 ml of methanol dissolved while heating to boiling and passing nitrogen through. The resulting solution is then cooled to 0 under nitrogen and a solution of 0.50 g of sodium bicarbonate in 10 ml of water is then added dropwise within 30-60 minutes. The reaction solution is then stirred further overnight at 0 under nitrogen. The progress of the saponification is checked by means of thin-layer chromatography. After a saponification time of 10-15 hours, no more starting material can be found in the reaction solution and at this point the reaction mixture is poured into 1520 ml of water.
It is extracted several times with methylene chloride, the extracts are washed successively repeatedly with water, dried over sodium sulphate and evaporated completely. The yield of crude product is 2.5 g of # 1,4-6α, 11ss-difluoro-2-chloro-9α-bromo-16α-methyl-17α, 21-dihydroxy-3,20-dioxo-pregnadiene which, by recrystallization from methanol, yields pure # 1,4-6α, 11ss- -difluoro-2-chloro-9a-bromo-1 6a-methyl-1 7z, 21-dihydroxy-3, 20 -dioxo-pregnadiene.
For the preparation of the # 1,4-6α, 1liP-difluoro-2-chloro-9.a-bromo-16α-methyl-17α-hydroxy-21-trimethylacetoxy-3,20-dioxo-pre-gnadiene 1.3 ml of trimethyl acetic acid chloride are dissolved in 25 ml of pyridine at 0. A solution of 0.9 g of # 1,4-6α, 11ss-difluoro-2-chloro-9α-bromo-16α-methyl-17α, 21- -dihydroxy-3,20-dioxo- Inflow pregnadiene in 15 ml of pyridine at 0 and stir for 10-15 hours at 00. For work-up, the reaction solution is poured into ice water, the precipitated reaction products are extracted with ethyl acetate, the extracts are washed successively with dilute hydrochloric acid, water, dilute soda solution and water, and they are dried and completely evaporates them.
The white, finely crystalline residue is redissolved from ethyl acetate and yields pure # 1,4-6α, 11 lss-difluoro--2-chloro-9a-bromo-1 6a-methyl-1 7α-hydroxy-21 -trimethylacetoxy- 3, 20-dioxo-pregnadiene.

A mixture of the # 1,4-6α, 1-difluoro-2-chloro-9a-bromo-16α-methyl-17α, 21- (1'-methoxy-3'-carbomethoxy) -propylidene-dioxy- 3,20-dioxo-pregnadiene in methanol with an aqueous solution of sodium hydrogen carbonate is stirred under nitrogen until no more starting material can be found in the thin-layer chromatogram of a reaction sample. The reaction solution is poured into ice water, acidified with dilute hydrochloric acid and extracted with methylene chloride.
The residue remaining after the extract, which has been washed several times with water has been evaporated, is taken up in methanol-ethyl acetate, where pure # 1,4-6α, 11ss-difluoro-2-chloro-9α-bromo-16α-methyl-17α is obtained. , 21- (1'-methoxy-3'-carboxy) -propylidene-dioxy-3,20-dioxo-pregnadiene crystallized out; the new compound dissolves in an aqueous solution of sodium hydrogen carbonate.
