Verfahren zur Herstellung von neuen Derivaten des Sulfanilamids
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Derivate des Sulfanilamids.
Verbindungen der allgemeinen Formel I

in welcher R1 eine Alkylgruppe mit höchstens 5 Kohlenstoffatomen oder eine Cycloalkyl. oder Cycloalkenylgruppe von höchstens 7 Kohlenstoffatomen bedeutet, und ihre Additionssalze mit anorganischen oder organischen Säuren sind bisher nicht bekannt geworden.
Wie nun gefunden wurde, besitzen diese Verbindungen wertvolle pharmakologische Eigenschaften, insbesondere das l-Sulfanilyl-2-imino-3-butyl-, das 1-Sulfanilyl-2 -imino-3-tert.butyl- und das 1 -Sulfanilyl-2-imino-3-cyclo- hexyl-imidazolidin weisen bei peroraler oder parenteraler Verabreichung hypoglykämische Wirkung auf, die sich als geeignet zur Behandlung der Zuckerkrankheit charakterisieren. Die hypoglykämische Wirkung wird an Standardversuchen an Warmblütern, z.B. Kaninchen und Ratten, nachgewiesen.
In den Verbindungen der allgemeinen Formel I kann Rt als niedere Alkylgruppe z.B. die Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, sek.Butyl-, tert.Butyl-, Iso butyl-, Pentyl-, Isopentyl-, 2,2-Dimethyl-propyl-, l-Methyl-butyl-, l-Äthyl-propyl-, 1 ,2-Dimethyl-propylgruppe sein.
Ferner kann Rt beispielsweise sein: als Cycloalkylgruppe die Cyclopropyl-, Cyclopropylmethyl-, 2-Cyclopropyl-äthyl-, 3-Cyclopropyl-propyl-, Cyclobutyl-, Cyclobutyl-methyl-, 2-Cyclobutyl-äthyl-, 3-Cyclobutyl-propyl-, Cyclopentyl-, l-Methyl-cyclopentyl-, 2-Methyl-cyclopentyl-, 3-Methyl-cyclopentyl-, 1-Athyl-cyclopentyl-, 2-Äthyl cyclopentyl-, 3-Äthyl-cyclopentyl-, Cyclopentylmethyl-, 2- Methyl-cyclopentylmethyl-, 3- Methyl-cyclopentylmethyl-, Cycolhexyl-, l-Methyl-cyclohexyl-, 2-Methyl-cyclohexyl-, 3-Methyl-cyclohexyl-, 4-Methyl-cyclohexyl-,

in welcher R1 die unter Formel I angegebene Bedeutung hat, kondensiert und cyclisiert, nötigenfalls das Reak tionsprodukt zur Überführung des Restes X in die freie Aminogruppe hydrolysiert, reduziert oder reduktiv spaltet und gegebenenfalls die erhaltene Verbindung mit einer anorganischen oder organischen Säure in ein Additionssalz überführt.
Geeignete reaktionsfähige Ester von Hydroxyverbindungen der allgemeinen Formel III sind beispielsweise Halogenide. insbesondere Chloride oder Bromide, ferner Sulfonsäureester, z.B. der o- oder p-Toluolsulfonsäureester oder der Methansulfonsäureester. Die Kondensation erfolgt vorzugsweise in einem mit Wasser mischbaren oder nichtmischbaren Lösungsmittel in An- oder Abwesenheit von Wasser. Als Lösungsmittel können Alkanole, z.B. Butanol, ätherartige Flüssigkeiten, z.B. Dioxan, Diäthylenglykolmonomethyläther, Carbonsäureamide, wie N,N-Dimethylformamid, oder Sulfoxide, wie Dimethylsulfoxid. verwendet werden. Es ist vorteilhaft, die Kondensation in Gegenwart eines säurebindenden Mittels vorzunehmen.
Als solche können diejenigen Verbindungen verwendet werden, die anschliessend an das erste Verfahren genannt sind, ferner auch tertiäre organische Basen, wie z.B. N.N-Diisopropyl-äthylamin .
Die anschliessende Umwandlung der Gruppe X des Reaktionsproduktes in die freie Aminogruppe, welche dieses in eine Verbindung der allgemeinen Formel 1 überführt, wird je nach der Art der Gruppe X durch eine Hydrolyse, Reduktion oder reduktive Spaltung vorgenommen.
Durch Hydrolyse in die freie Aminogruppe überführbare Reste X sind beispielsweise Acylaminoreste, wie z.B. die Acetamidogruppe. Ferner sind solche Reste niedere Alkoxycarbonylaminoreste, wie z.B. die Äthoxycar bonylarninogruppe, Aryloxycarbonylaminoreste, wie der Phenoxycarbonylaminorest, oder Arylmethoxycarbonylaminoreste, wie der Benzyloxyearbonylaminorest, oder Reste von entsprechenden Thiokohlensäuredenvaten.
Weitere Beispiele sind substituierte Methylenaminoreste wie z.B. die Benzylidenamino- oder die p-Dimethylami no-benzylidenarninogruppe. Die Hydrolyse zur Freisetzung der Aminogruppe kann in saurem Medium, z.B. in methanolischer Salzsäure, oder in verdünnter wässriger Salzsäure oder Schwefelsäure erfolgen oder, falls X durch einen Alkoxycarbonylaminorest verkörpert ist, auch unter milden alkalischen Bedingungen, z.B. mittels 1-n oder 2-n Natronlauge, vorgenommen werden.
Ein Beispiel für einen durch Reduktion in die Aminogruppe überführbaren Rest X ist die Nitrogruppe und Beispiele für solche Reste. die durch reduktive Spaltung zur Aminogruppe führen, sind die Phenylazo- oder p -Dimethylamino-phenylazogruppen. Die Reduktion dieser Reste kann allgemein katalytisch, z.B. mittels Wasserstoff in Gegenwart von Raney-Nickel, Palladium- oder Platin-Kohle, in einem inerten Lösungsmittel, wie z.B.
Äthanol. erfolgen. Neben diesen kommen auch andere übliche Reduktionsverfahren in Betracht, beispielsweise die Reduktion von Nitrogruppen oder die reduktive Spaltung von Azogruppen mittels Eisen in Essigsäure oder Salzsäure.
Als Ausgangsprodukte des Verfahrens werden die reaktionsfähigen Ester einer Hydroxyverbindung der allgemeinen Formel III verwendet deren Symbol R1 mit den Gruppen übereinstimmt, die anschliessend an Formel 1 aufgezählt sind. Eine Gruppe von Bromiden der allgemeinen Formel III können z.B. erhalten werden, wenn man ein l-Alkyl- oder ein 1 -Cycloalkyl-aziridin [vgl. A.
Weissberger, Heterocyclic Compounds with Three and Four-Membered Rings, Part One, John Wiley Sons Inc., London (1964)] mit Bromcyan in Dioxan umsetzt.
Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden anschliessend gewünschtenfalls in ihre Salze mit anorganischen sowie organischen Säuren übergeführt. Die Herstellung dieser Salze erfolgt z.B. durch Umsetzung der Verbindungen der allgemeinen Formel I mit der äquivalenten Menge einer Säure in einem geeigneten wässrigorganischen oder organischen Lösungsmittel, wie z.B.
Methanol, Äthanol, Diäthyläther, Chloroform oder Methylenchlorid.
Zur Verwendung als Arzneistoffe können anstelle der freien Verbindungen der allgemeinen Formel I deren pharmazeutisch annehmbare Salze mit Säuren eingesetzt werden. Geeignete Additionssalze sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, -Hydroxyäthansulfonsäure, Essigsäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keineswegs die einzige Ausführungsform desselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I a) 9,9 g 1-tert.Butyl-aziridin [vgl. A. Weissberger, Heterocyclic Compounds with Three and Four-Membered Rings, John Wiley & Sons Inc., London (1964), Seite 530] werden in 60 ml Dioxan gelöst und mit 10,6 g Bromcyan versetzt. In einer exothermen Reaktion wird eine Lösung von N-(2-Brom-äthyl)-N-tert.butyl-cyanamid erhalten, die man unter Rühren zu einer Lösung von 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml 1-n Natronlauge tropft.
Man kocht das Reaktionsgemisch eine Stunde unter Rückfluss und dampft es anschliessend im Vakuum auf das halbe Volumen ein. Die ausgefallenen Kristalle werden abfiltriert, mit Wasser gewaschen, bei 600 im Vakuum getrocknet und aus Essigsäureäthylester umkristallisiert. Das erhaltene 4'-(2-Imino-3-tert.butyl-imidazo- lidin-1-ylsulfonyl)-acetanilid schmilzt bei 245 - 245-2470.
b) 33,8 g des erhaltenen Anilids werden mit 100 ml 2-n Salzsäure zum Endprodukt l-Sulfanilyl-2-imino-3- -tert.butyl-imidazolidin vom Smp. 187 - 1890 (aus Methanol/Wasser) hydrolysiert.
Beispiel 2
Analog Beispiel 1 erhält man: a) aus 5,7g 1-Methyl-aziridin und 10.6g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-methyl-cyan- amid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml 1 -n Natronlauge das 4'-(2-Imino-3-methyl-imidazoli- din-l-ylsulfonyl)-acetanilid vom Smn. 266 - 2670 liefert; dieses wird analog Beispiel 1 b) zum 1 -Sulfanilyl-2-imino- -3-methyl-imidazolidin vom Smp. 209 - 2110 hydrolysiert;
b) aus 7,1 g 1-Äthyl-aziridin und 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyD-N-äthyl-cyanamid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml l-n Natronlauge das 4'-(2-Imino-3 -äthyl-imidazolidin- 1 -ylsul- fonyl)-acetanilid vom Smp. 267 - 2690 liefert; dieses wird analog Beispiel 1 b) zum 1-Sulfanilyl-2-imino-3-äthyl -imidazolidin vom Smp. 171 - 1720 hydrolysiert; c) aus 8,5 g 1-Propyl-aziridin und 10,6g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-propyl-cyan amid, welches mit 21,4g 4'-Sulfamoyl-acetanilid in 100 ml 1-n Natronlauge das 4'-(2-Imino-3-propyl-imidazolidin-1-ylsulfonyl)-acetanilid vom Smp. 253 - 2550 liefert;
dieses wird analog Beispiel 1 b) zum 1 -Sulfanilyl-2-imino- -3-propyl-imidazolidin vom Smp. 164 - 1660 hydrolysiert; d) aus 8,5 g 1-Isopropyl-aziridin und 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-isopropyl-cyan- amid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml 1-n Natronlauge das 4'-(2-Imino-3-isopropyl-imidazo lidin-1-ylsulfonyl)-acetanilid vom Smp. 253 - 254 liefert; dieses wird analog Beispiel 1 b) zum 1-Sulfanilyl-2-imino -3-isopropyl-imidazolidin vom Smp. 183 - 1840 hydrolysiert;
e) aus 9,9 g 1-Isobutyl-aziridin und 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-isobutyl-cyanamid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml 1-n Natronlauge das 4'-(2-Imino-3-isobutyl-imidazo- lidin-1-ylsulfonyl)-acetanilid vom Smp. 264-265 liefert; dieses wird analog Beispiel 1 b) zuh 1 -Sulfanilyl-2- -imino-3-isobutyl-imidazolidin vom Smp. 146- 1470 hydrolysiert; f) aus 9,9 g 1-sek.Butyl-aziridin und 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-sek.butyl-cyanamid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml 1-n Natronlauge das 4'-(2-Imino-3-sek.butyl-imidazolidin-1-ylsulfonyl)-acetanilid vom Smp. 265 - 2660 liefert;
dieses wird analog Beispiel 1 b) zum 1 -Sulfanilyl-2-imi- no-3-sek.butyl-imidazolidin vom Smp. 173 - 173,50 hy.
drolysiert; g) aus 11,3 g 1-Pentyl-aziridin und 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-pentyl-cyanamid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml 1-n Natronlauge das 4'-(2-Imino-3-pentyl-imidazoli din-1-ylsulfonyl)acetanilid vom Smp. 248 - 2500 liefert; dieses wird analog Beispiel 1 b) zum 1-Sulfanilyl-2-imino-3-pentyl-imidazolidin vom Smp. 167 - 1680 hydrolysiert; h) aus 12,7 g 1 -Hexyl-aziridin und 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-hexyl-cyanamid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml 1-n Natronlauge das 4'(2-Imino-3-hexyl-imidazoli- din-1-ylsulfonyl)-acetanilid vom Smp. 236 - 2380 liefert;
dieses wird nach Beispiel 1 b) zum 1-Sulfanilyl-2-imino -3-hexyl-imidazolidin vom Smp. 182 - 1830 hydrolysiert; i) aus 11,1 g 1-Cyclopentyl-aziridin und 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-cyclopentylcyanamid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100 ml 1-n Natronlauge das 4'-(2-Imino-3-cyclopentyl-imidazolidin- 1 -ylsulfonyl)-acetanilid vom Smp.
261 - 2630 liefert; dieses wird nach Beispiel lb) zum 1-Sulfanilyl-2-imino-3-cyclopentyl-imidazolidin vom Smp.
192 - 1930 hydrolysiert, und j) aus 12,5 g 1-Cyclohexyl-aziridin und 10,6g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-cyclo hexylcyanamid, welches mit 21,4 g 4'-Sulfamoyl-acetanilid in 100ml 1-n Natronlauge das 4'-(2-Imino-3-cyclo hexyl-imidazolidin- 1 -ylsulfonyl)-acetanilid vom Smp. 283 bis 2840 liefert; dieses wird nach Beispiel 1 b) zum 1 -Sulfanilyl-2-imino-3-cyclohexyl-imidazolidin vom Smp.
178 - 1790 hydrolysiert.
Beispiel 3
12,5 g 1-Cyclohexyl-aziridin werden in 60 ml Dioxan gelöst und mit 10,6g Bromcyan versetzt. In einer exothermen Reaktion entsteht das N-(2-Brom-äthyl)-N-cyclohexyl-cyanamid, welches in der Reaktionslösung unter Rühren zu einer Lösung von 17,2 g Sulfanilamid in 100 ml 1-n Natronlauge gegeben wird. Man kocht das Reaktionsgemisch eine Stunde unter Rückfluss und dampft es im Vakuum ein. Der Rückstand wird mit Wasser versetzt, der entstandene Niederschlag abfiltriert und aus Essigsäureäthylester umkristallisiert. Das erhaltene 1 Sulfanilyl-2-imino-3-cyclohexyl-imidazolidin schmilzt bei 178- 1790.
Beispiel 4
Analog Beispiel 3 erhält man: a) aus 5,7 g 1-Methyl-aziridin mit 10,6g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-methyl-cyanamid, welches mit 17,2 g Sulfanilamid in 100 ml 1-n Natronlauge das 1 -Sulfanilyl-2-imino-3 -methyl-imidazolidin vom Smp. 209 - 2110 liefert; b) aus 7,1 g 1-Athyl-aziridin mit 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-äthyl-cyanamid, welches mit 17,2g Sulfanilamid in 100ml 1-n Natronlauge das 1 -Sulfanilyl-2-imino-3-äthyl-imidazolidin vom Smp. 171 - 1720 ergibt; c) aus 8,5 g 1-Propyl-aziridin mit 10,6g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-cyanamid, welches mit 17,2g Sulfanilamid in 100 ml 1-n Natronlauge das 1-Sulfanilyl-2-imino-3-propyl-imidazolidin vom Smp.
164 - 1660 liefert; d) aus 8,5 g l-Isopropyl-aziridin mit 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-isopropyl-cyanamid, welches mit 17,2 g Sulfanilamid in 100 ml 1-n Natronlauge das 1 -Sulfanilyl-2-imino-3-isopropyl-imid- azolidin vom Smp. 183 - 1840 ergibt; e) aus 9,9 g 1-Butyl-aziridin mit 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-butyl-cyanamid, welches mit 17,2g Sulfanilamid in 100ml 1-n Natronlauge das 1 -Sulfanilyl-2-imino-3-butyl-imidazolidin vom Smp. 179 - 1810 liefert;
f) aus 9,9 g 1-Isobutyl-aziridin mit 10,6g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-isobutyl-cyan- amid, welches mit 17,2 g Sulfanilamid in 100 ml 1-n Na tronlauge das l-Sulfanilyl-Zimino 1 -Sulfanilyl-2-imino-3-isobutyl-imidazolidin vom Smp. 146 - 1470 ergibt:
g) aus 9,9 g 1-sek.Butyl-aziridin mit 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-sek.butyl-cyanamid, welches mit 17,2 g Sulfanilamid in 100 ml 1-n Natronlauge das 1-Sulfanilyl-2-imino-3-sek.butyl-imidazoli- din vom Smp. 173 - 173,50 liefert; h) aus 11,3 g 1-Pentyl-aziridin mit 10,6g Bromcyan in 60 ml Dioxan das N-(2-Bromäthyl)-N-pentyl-cyanamid, welches mit 17,2g Sulfanilamid in 100ml 1-n Natronlauge das 1-Sulfanilyl-2-imino-3-pentyl-imidazolidin vom Smp. 167 - 1680 ergibt; i) aus 12,7 g 1-Hexyl-aziridin mit 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-hexyl-cyanamid, welches mit 17,2g Sulfanilamid in 100ml 1-n Natronlauge das 1 -Sulfanilyl-2-imino-3-hexyl-imidazolidin vom Smp. 182- 1830 liefert;
j) aus 11,1 g 1-Cyclopentyl-aziridin mit 10,6 g Bromcyan in 60 ml Dioxan das N-(2-Brom-äthyl)-N-cyclopentyl-cyanamid, welches mit 17,2 g Sulfanilamid in 100 ml 1 -n Natronlauge das 1-Sulfanilyl-2-imino-3-cyclopentyl- -imidazolidin vom Smp. 192 - 1930 liefert.
Beispiel 5
Ein Gemisch von 16,0 g N-(2-Chlor-äthyl)-N-tert.butyl-cyanamid, 5,6 g Kaliumhydroxid und 17,2 g Sulfanilamid in 100 ml Diäthylenglykolmonomethyläther wird 30 Minuten in einem Bad von 160- 1700 erhitzt. Anschliessend giesst man das Reaktionsgemisch auf Eis und filtriert die Suspension. Der Filterrückstand wird in 2-n Salzsäure gelöst, die Lösung mit Aktivkohle entfärbt, filtriert und mit konz. Natronlauge alkalisch gestellt. Die ausgefallenen Kristalle werden abfiltriert, mit Wasser gewaschen und aus Äthanol umkristallisiert. Man erhält das l-Sulfanilyl-2- imino-3-tert.butyl-imidazolidin vom Smp.188-1900.

Suitable reactive esters of hydroxy compounds of the general formula III are, for example, halides. in particular chlorides or bromides, also sulfonic acid esters, e.g. the o- or p-toluenesulfonic acid ester or the methanesulfonic acid ester. The condensation is preferably carried out in a water-miscible or immiscible solvent in the presence or absence of water. Alkanols, e.g. Butanol, ethereal liquids, e.g. Dioxane, diethylene glycol monomethyl ether, carboxamides, such as N, N-dimethylformamide, or sulfoxides, such as dimethyl sulfoxide. be used. It is advantageous to carry out the condensation in the presence of an acid-binding agent.

The subsequent conversion of group X of the reaction product into the free amino group, which converts it into a compound of general formula 1, is carried out, depending on the type of group X, by hydrolysis, reduction or reductive cleavage.
X radicals which can be converted into the free amino group by hydrolysis are, for example, acylamino radicals, e.g. the acetamido group. Furthermore, such groups are lower alkoxycarbonylamino groups, e.g. the ethoxycarbonylamino group, aryloxycarbonylamino groups, such as the phenoxycarbonylamino group, or aryl methoxycarbonylamino groups such as the benzyloxyearbonylamino group, or groups of corresponding thiocarbonic acid derivatives.
Further examples are substituted methylene amino radicals such as e.g. the benzylideneamino or the p-Dimethylami no-benzylideneamino group. The hydrolysis to liberate the amino group can be carried out in an acidic medium, e.g. in methanolic hydrochloric acid, or in dilute aqueous hydrochloric acid or sulfuric acid or, if X is represented by an alkoxycarbonylamino radical, also under mild alkaline conditions, e.g. using 1-n or 2-n sodium hydroxide solution.
An example of a radical X which can be converted into the amino group by reduction is the nitro group and examples of such radicals. which lead to the amino group through reductive cleavage are the phenylazo or p -dimethylamino-phenylazo groups. The reduction of these residues can generally be catalytic, e.g. by means of hydrogen in the presence of Raney nickel, palladium or platinum carbon, in an inert solvent such as e.g.
Ethanol. respectively. In addition to these, other customary reduction processes can also be used, for example the reduction of nitro groups or the reductive cleavage of azo groups using iron in acetic acid or hydrochloric acid.
The reactive esters of a hydroxyl compound of the general formula III, the symbol R1 of which corresponds to the groups listed below in formula 1, are used as starting products for the process. A group of bromides of general formula III can e.g. can be obtained if a 1-alkyl- or a 1-cycloalkyl-aziridine [cf. A.
Weissberger, Heterocyclic Compounds with Three and Four-Membered Rings, Part One, John Wiley Sons Inc., London (1964)] with cyanogen bromide in dioxane.
The compounds of general formula I obtained by the process according to the invention are then, if desired, converted into their salts with inorganic and organic acids. These salts are produced e.g. by reacting the compounds of general formula I with the equivalent amount of an acid in a suitable aqueous, organic or organic solvent, such as e.g.
Methanol, ethanol, diethyl ether, chloroform or methylene chloride.
For use as medicinal substances, their pharmaceutically acceptable salts with acids can be used instead of the free compounds of the general formula I. Suitable addition salts are e.g. Salts with hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulfonic acid, ethanesulfonic acid, -hydroxyethanesulfonic acid, acetic acid, lactic acid, oxalic acid, succinic acid, fumaric acid, maleic acid, malic acid, tartaric acid, citric acid, benzoic acid, salicylic acid, phenyl acetic acid.
The following examples explain the preparation of the new compounds of general formula I and of hitherto not described intermediates in more detail, but by no means represent the only embodiment of the same. The temperatures are given in degrees Celsius.
Example I a) 9.9 g of 1-tert-butyl-aziridine [cf. A. Weissberger, Heterocyclic Compounds with Three and Four-Membered Rings, John Wiley & Sons Inc., London (1964), page 530] are dissolved in 60 ml of dioxane and mixed with 10.6 g of cyanogen bromide. In an exothermic reaction, a solution of N- (2-bromo-ethyl) -N-tert-butyl-cyanamide is obtained, which is mixed with stirring to a solution of 21.4 g of 4'-sulfamoyl-acetanilide in 100 ml of 1- n Sodium hydroxide solution drips.
The reaction mixture is refluxed for one hour and then evaporated to half its volume in vacuo. The precipitated crystals are filtered off, washed with water, dried at 600 in a vacuum and recrystallized from ethyl acetate. The 4 '- (2-imino-3-tert-butyl-imidazolidin-1-ylsulfonyl) acetanilide obtained melts at 245-245-2470.
b) 33.8 g of the anilide obtained are hydrolyzed with 100 ml of 2N hydrochloric acid to give the end product l-sulfanilyl-2-imino-3-tert-butyl-imidazolidine with a melting point of 187-1890 (from methanol / water).
Example 2
Analogously to Example 1 one obtains: a) from 5.7 g of 1-methyl-aziridine and 10.6 g of cyanogen bromide in 60 ml of dioxane, the N- (2-bromo-ethyl) -N-methyl-cyanamide, which with 21.4 g 4'-sulfamoyl acetanilide in 100 ml 1 -N sodium hydroxide solution the 4 '- (2-imino-3-methyl-imidazoli- din-1-ylsulfonyl) -acetanilide from Smn. 266-2670 provides; this is hydrolyzed analogously to Example 1 b) to give 1-sulfanilyl-2-imino- -3-methyl-imidazolidine of melting point 209-2110;
b) from 7.1 g of 1-ethyl aziridine and 10.6 g of cyanogen bromide in 60 ml of dioxane, the N- (2-bromo-ethyD-N-ethyl-cyanamide, which with 21.4 g of 4'-sulfamoyl-acetanilide in 100 ml of sodium hydroxide solution gives 4 '- (2-imino-3-ethyl-imidazolidin-1 -ylsulfonyl) -acetanilide with a melting point of 267-2690; this is converted into 1-sulfanilyl-2- analogously to Example 1 b) hydrolyzed imino-3-ethylimidazolidine of melting point 171-1720; c) from 8.5 g of 1-propyl-aziridine and 10.6 g of cyanogen bromide in 60 ml of dioxane, the N- (2-bromo-ethyl) -N-propyl-cyanamide, which with 21.4 g of 4'-sulfamoyl-acetanilide in 100 ml of 1N sodium hydroxide solution, the 4 '- (2-imino-3-propyl-imidazolidin-1-ylsulfonyl) acetanilide with a melting point of 253-2550 is obtained;
this is hydrolyzed analogously to Example 1 b) to give 1-sulfanilyl-2-imino- -3-propyl-imidazolidine of melting point 164-1660; d) from 8.5 g of 1-isopropyl-aziridine and 10.6 g of cyanogen bromide in 60 ml of dioxane, the N- (2-bromo-ethyl) -N-isopropyl-cyanamide, which with 21.4 g of 4'- Sulfamoyl acetanilide in 100 ml of 1N sodium hydroxide solution gives 4 '- (2-imino-3-isopropyl-imidazo lidin-1-ylsulfonyl) acetanilide with a melting point of 253-254; this is hydrolyzed analogously to Example 1 b) to 1-sulfanilyl-2-imino -3-isopropyl-imidazolidine of melting point 183-1840;
e) from 9.9 g of 1-isobutyl-aziridine and 10.6 g of cyanogen bromide in 60 ml of dioxane, N- (2-bromo-ethyl) -N-isobutyl-cyanamide, which with 21.4 g of 4'-sulfamoyl acetanilide in 100 ml of 1N sodium hydroxide solution gives the 4 '- (2-imino-3-isobutyl-imidazolidin-1-ylsulfonyl) acetanilide of melting point 264-265; this is hydrolyzed analogously to Example 1 b) to give 1-sulfanilyl-2-imino-3-isobutyl-imidazolidine with a melting point of 146-1470; f) from 9.9 g of 1-sec-butyl-aziridine and 10.6 g of cyanogen bromide in 60 ml of dioxane, N- (2-bromo-ethyl) -N-sec.butyl-cyanamide, which with 21.4 g of 4 '-Sulfamoyl-acetanilide in 100 ml of 1N sodium hydroxide solution gives the 4' - (2-imino-3-sec.butyl-imidazolidin-1-ylsulfonyl) -acetanilide of melting point 265-2660;

this is hydrolyzed according to Example 1 b) to 1-sulfanilyl-2-imino -3-hexyl-imidazolidine of melting point 182-1830; i) from 11.1 g of 1-cyclopentyl-aziridine and 10.6 g of cyanogen bromide in 60 ml of dioxane, the N- (2-bromo-ethyl) -N-cyclopentylcyanamide, which with 21.4 g of 4'-sulfamoyl-acetanilide in 100 ml of 1N sodium hydroxide solution of the 4 '- (2-imino-3-cyclopentyl-imidazolidin-1 -ylsulfonyl) -acetanilide of melting point.

Example 3
12.5 g of 1-cyclohexyl aziridine are dissolved in 60 ml of dioxane, and 10.6 g of cyanogen bromide are added. In an exothermic reaction, N- (2-bromo-ethyl) -N-cyclohexyl-cyanamide is formed, which is added to a solution of 17.2 g of sulfanilamide in 100 ml of 1N sodium hydroxide solution in the reaction solution with stirring. The reaction mixture is refluxed for one hour and evaporated in vacuo. The residue is mixed with water, the resulting precipitate is filtered off and recrystallized from ethyl acetate. The 1 sulfanilyl-2-imino-3-cyclohexyl-imidazolidine obtained melts at 178-1790.
Example 4
Analogously to example 3 one obtains: a) from 5.7 g of 1-methyl-aziridine with 10.6 g of cyanogen bromide in 60 ml of dioxane the N- (2-bromo-ethyl) -N-methyl-cyanamide, which with 17.2 g Sulfanilamide in 100 ml of 1N sodium hydroxide solution gives the 1-sulfanilyl-2-imino-3-methyl-imidazolidine with a melting point of 209-2110; b) from 7.1 g of 1-ethyl aziridine with 10.6 g of cyanogen bromide in 60 ml of dioxane, the N- (2-bromo-ethyl) -N-ethyl-cyanamide, which with 17.2 g of sulfanilamide in 100 ml of 1-n Sodium hydroxide solution gives 1-sulfanilyl-2-imino-3-ethyl-imidazolidine of melting point 171-1720; c) from 8.5 g of 1-propyl-aziridine with 10.6 g of cyanogen bromide in 60 ml of dioxane, the N- (2-bromo-ethyl) -N-cyanamide, which with 17.2 g of sulfanilamide in 100 ml of 1 N sodium hydroxide solution 1-sulfanilyl-2-imino-3-propyl-imidazolidine of mp.

j) from 11.1 g of 1-cyclopentyl-aziridine with 10.6 g of cyanogen bromide in 60 ml of dioxane, the N- (2-bromo-ethyl) -N-cyclopentyl-cyanamide, which with 17.2 g of sulfanilamide in 100 ml of 1 -n sodium hydroxide solution which supplies 1-sulfanilyl-2-imino-3-cyclopentyl- imidazolidine with a melting point of 192-1930.
Example 5
A mixture of 16.0 g of N- (2-chloro-ethyl) -N-tert-butyl-cyanamide, 5.6 g of potassium hydroxide and 17.2 g of sulfanilamide in 100 ml of diethylene glycol monomethyl ether is placed in a bath of 160-1700 for 30 minutes heated. The reaction mixture is then poured onto ice and the suspension is filtered. The filter residue is dissolved in 2N hydrochloric acid, the solution decolorized with activated charcoal, filtered and treated with conc. Sodium hydroxide solution made alkaline. The precipitated crystals are filtered off, washed with water and recrystallized from ethanol. The l-sulfanilyl-2-imino-3-tert-butyl-imidazolidine with a melting point of 188-1900 is obtained.
