Gegenstand der Erfindung ist ein Verfahren zur Herstellung von substituierten p-Hydroxyphenylhydrazonen der Formel I in der

Weise umsetzt und gewünschtenfalls die Verfahrenspro dukte, sofern sie saure Gruppen enthalten, mittels nicht toxischer Basen in ihre physiologisch verträglichen Salze überführt.
Die nach dem erfindungsgemässen Verfahren als Aus gangsprodukte verwendeten p-Hydroxyphenylhydrazine der Formel II bzw. deren Säureadditionssalze können nach bekannten Methoden aus den entsprechend substi tuierten p-Hydroxyphenylaminen beispielsweise durch
Diazotieren, Umsetzung zu den Diazosulfonaten, Reduk tion zu den Hydrazosulfonaten und Spaltung zu den
Säureadditionssalzen mit wässriger oder alkoholischer
Säure hergestellt werden. Besonders bevorzugt sind als
Ausgangsstoffe solche p-Hydroxyphenylhydrazine, in denen R1 und R2 Wasserstoff bedeuten. Während die freien Basen unbeständige, leicht zersetzliche Flüssigkei ten oder Festkörper sind, sind die Säureadditionssalze gut zu handhabende, einigermassen beständige kristalline Substanzen.
Als zweite Reaktionskomponente kommen für das erfindungsgemässe Verfahren aromatische Aldehyde oder aliphatisch-aromatische Ketone der Formel III infrage.
Als Reste R3 seien genannt: Wasserstoff oder Methyl, Äthyl, n-Propyl oder i-Propyl. R4 kann ein gegebenenfalls ein- oder mehrfach substituierter Phenylkern sein, der in meta- oder para-Position eine Hydroxylgruppe tragen kann. Als weitere Substituenten kommen Hy droxy-, Alkoxy-, niedrigmolekulare Alkyl-, Trifluormethyl-, Halogen-, Carbalkoxy-, Carbamoyl- und/oder Carboxygruppen infrage. Anstelle der Carbonylverbindungen können auch deren funktionelle Derivate eingesetzt werden, beispielsweise die Oxime, Hydrazone, Phenylhydrazone, Semicarbazone, Imine, Azomethine, Aminale, Acetale, Halbacetale, Acetate, Ketale, Thioketale oder Enamine.
Enthalten die Carbonylverbindungen eine saure Funktion, so können auch basische Salze zur Anwendung kommen.
Die erfindungsgemässe Umsetzung der p-Hydroxyphenylhydrazine mit den Carbonylverbindungen führt man vorteilhaft in Wasser und/oder mit Wasser mischbaren Lösungsmitteln wie niedrigen aliphatischen Alkoholen, Dioxan, Tetrahydrofuran, Dimethylformamid oder Pyridin, vorzugsweise jedoch in Mischungen aus Wasser und Methanol oder Äthanol aus. Die Umsetzung kann bei niedriger oder mässig erhöhter Temperatur vorgenommen werden. Zweckmässig ist es jedoch, bei Temperaturen von 10 bis 150 C zu arbeiten. Es ist vorteilhaft, anstelle der freien p-Hydroxyphenylhydrazine die Säureadditionssalze anzuwenden, z.B. die Hydrochloride. Die Umsetzung kann dann in Gegenwart von schwach basischen Puffern, wie z.B. Natriumacetat, vorgenommen werden.
Vorteilhaft ist es ferner, unter Ausschluss von Luftsauerstoff unter einem Schutzgas zu arbeiten oder der Lösung des p-Hydroxyphenylhydrazins ein schwaches Reduktionsmittel, wie z.B. Ascorbinsäure, zuzusetzen. Man kann die erfindungsgemässe Umsetzung auch so ausführen, dass man ein p-Hydroxyphenylhydrazin in Anwesenheit einer Carbonylverbindung in der Reaktionslösung direkt aus einer Vorstufe, z.B. einer p-Hydroxyphenylhydrazo-acylverbindung, in Gegenwart von wässrigen oder alkoholischen Säuren erzeugt. Infrage kommt z.B. eine p-Hydroxyphenylhydrazosulfonsäure oder ein Salz derselben.
Die gemäss dem Verfahren gebildeten p-Hydroxyphenylhydrazone kristallisieren meist aus dem Reaktionsgemisch aus und können abfiltriert werden. Andernfalls kann man sie entweder durch schonendes Einengen der Reaktionslösung bei möglichst tiefer Temperatur oder durch vorsichtiges Versetzen mit Wasser kristallin aus dem Reaktionsgemisch abscheiden. Die Verfahrensprodukte sind schwach gelb bis orangerot gefärbte kristalline Substanzen, die meist keinen definierten Schmelzpunkt haben, sondern einen Zersetzungspunkt zeigen. Je nach der Art der Substituenten lösen sie sich in Wasser mehr oder weniger gut.
Enthalten die Verfahrensprodukte saure Gruppen, so können sie in Salze mit nichttoxischen Basen übergeführt werden. Geeignete Basen sind z.B. Alkali- und Erdalkalihydroxyde, Alkalicarbonate und -bicarbonate, ferner tertiäre Amine wie Triäthylamin.
Die nach dem Verfahren gemäss der Erfindung hergestellten p-Hydroxyphenylhydrazone können als solche oder in Form ihrer Salze mit nichttoxischen Basen als Heilmittel verwendet werden. Sie besitzen eine sehr starke antibakterielle Aktivität, die sich insbesondere gegen gramnegative Keime wie z.B. Escherichia Coli, Bacterium proteus oder Pseudomonas aeruginosa richtet und sind bekannten Bakteriostatika in ihrer Wirkung überlegen.
Die folgende Tabelle zeigt die Resultate von Vergleichsprüfungen zwischen einerAnzahl von erfindungsgemäss erhaltenen Substanzen und zwei bekannten Bakteriostatika.
In der Tabelle sind die minimalen bakteriostatischen Hemmkonzentrationen (MHK) aufgeführt. Die bakteriostatische Wirkung wurde mit Hilfe des Reihenverdünnungstestes bestimmt. Als Nährmedium diente Merck Standard Bouillon I. Die Keim-Einsaat betrug ca.
105 Keime/ml. Die angegebenen Werte sind an jeweils drei Reihen mit differenten Verdünnungsfaktoren ermittelt worden. Die Werte der MHK stellen diejenigen Verdünnungen dar, bei denen in der flüssigen Primärkultur noch kein Wachstum auftrat. Sie sind umgekehrt proportional der bakteriostatischen Aktivität einer Substanz: je kleiner der MHK-Wert ist um so grösser ist die bakteriostatische Aktivität. Die Werte der Tabelle wurden nach der Methode von J.C. Gould (Brit. med. Bull. 16, 29 (1960)) bestimmt. Die akuten Toxizitäten der Verbindungen bei peroraler Verabreichung an der Ratte liegen in der Grössenordnung von ca. 500-1000 mg/kg.
Die erfindungsgemäss hergestellten Substanzen können als antibakterielle Zusätze zu pharmazeutischen, kosmetischen oder technischen Produkten Verwendung finden. Sie können als Oberflächenantiseptika oder zur Desinfektion von Wunden lokal angewendet werden. Sie lassen sich jedoch auch zur Behandlung von Infektionen des Mund- und Rachenraumes verwenden. Bei peroraler Verabreichung werden sie im Harn in unveränderter Form ausgeschieden und verleihen dem Harn eine hohe bakteriostatische Aktivität. Sie eignen sich daher besonders zur Bekämpfung von Infektionen der Harnwege.
Die Verfahrensprodukte können - soweit sie für die medizinische Anwendung infrage kommen - als solche oder in Form entsprechender Salze, gegebenenfalls unter Beimengung pharmazeutisch üblicher inerter Hilfs- oder Trägerstoffe angewendet werden.
Mögliche Anwendungsformen sind z.B. Tabletten, Dragees, Salben, Cremes, Lotions oder Puder. Die Herstellung der galenischen Präparate erfolgt nach den üblichen Methoden. Feste Verabreichungsformen enthalten vorzugsweise 50-100 mg an Wirkstoff pro Verabreichungseinheit.
Beispiel 1 4-Hydroxy-benzaldehyd-p4iydrnxyphenylhydrazon
3,38 Mol p-Hydroxyphenylhydrazin HCl (680 g eines Produktes von 80 0/obiger Reinheit) werden unter Rühren in einer Mischung aus 3,41 Wasser, 2,01 Methanol und 1,5 1 gesättigter Natriumacetatlösung, der 50g Ascorbinsäure zugesetzt wurden, gelöst und auf 10 abgekühlt. Dazu lässt man unter Rühren eine Lösung von 3,38 Mol (412,7 g) 4-Hydroxy-benzaldehyd in 2,01 Methanol zutropfen. Es scheidet sich schnell ein gelber kristalliner Niederschlag ab. Man lässt noch 15 Minuten nachrühren, saugt ab und wäscht den Niederschlag nacheinander mit 21 Wasser und 1,5 1 Wasser/Methanol (4: 1).
Der Niederschlag wird in 3,5 1 heissem Methanol gelöst, die Lösung langsam mit 0,51 Wasser versetzt und abkühlen gelassen. Nach dem Absaugen und Trocknen erhält man derbe, gelbe Kristallnadeln, die einen Schmelzpunkt von 180-181 (unter Zersetzung) zeigen.
Beispiel 2 3-Methyl-4-hydroxy-5-carboxy-benzaldehyd hydroxyphenylhydrazon

Produktes von 90 < ?/oiger Reinheit), kühlt auf 150 ab und ]ässt eine Lösung von 0,1 Mol (18,0g) 3-Methyl-4-hydroxy-5-carboxy-benzaldehyd in 300mI Methanol zutropfen.
Man rührt 15 Minuten nach, gibt dann tropfenweise 2n HCl hinzu, bis ein pH von 3 erreicht ist, rührt weitere 30 Minuten, saugt ab, wäscht den Niederschlag mit Wasser und kristallisiert ihn aus wenig Methanol oder Äthanol/Wasser um. Man erhält grüngelbe Kristallblättchen vom Schmelzpunkt 1860 (unter Zersetzung).
Beispiel 3 3-Methoxy-4-hydroxy-benzaldehyd-p-hydroxy- phenylhydrazon
Zu einer Lösung von 0,51 Wasser, 0,5 1 gesättigter Natriumacetatlösung und 0,51 Methanol gibt man unter Rühren 0,5 Mol p-Hydroxyphenylhydrazin. HC1 (119,0 g eines Produktes von 67 O/oiger Reinheit) zu, kühlt auf 100 ab und lässt eine Lösung von 0,5 Mol (76,0g) Vanillin in 500 ml Methanol langsam zutropfen. Man lässt eine Stunde nachrühren, saugt den Niederschlag ab, wäscht ihn mit Wasser und kristallisiert den Rückstand aus Methanol/Wasser um. Man erhält gelbe Kristalle vom Schmelzpunkt 171-1720 (unter Zersetzung).
In analoger Weise erhält man b) 3 -Hydroxy-benzaldehyd-p-hydroxyphenylhydrazon,

4-Hydroxy-benzaldehyd-3'-chlor-4'-hydroxy-5'-methyl phenylhydrazon
Man löst in einem Gemisch von 150 ml Wasser und 200ml Methanol unter Rühren 67,5mMol 3-Chlor-4hydroxy-5-methylphenylhydrazin HC1 (Fp. 2620 unter Zers.) (21 g eines Produktes von 67 obiger Reinheit) und kühlt auf 150 ab. Dazu lässt man eine Lösung von 67,2 mMol (8,2 g) p-Hydroxybenzaldehyd in 100 ml Methanol zutropfen. Nach 15 Min Nachrühren wird der Niederschlag abgesaugt, mit Wasser gewaschen und aus Methanol/Wasser umkristallisiert. Man erhält gelbe Kriställchen vom Schmp. 1570 (unter Zers.).
In analoger Weise erhält man aus 3-Chlor-4-hydroxyphenylhydrazin HCl (Fp. 1120 unter Zers.) das 4-Hydroxy-benzaldehyd-3'-chlor-4'-hydroxy-phenylhydrazon, Schmp. 153-1540 (unter Zers.).

4-Hydroxy-benzaldehyde-3'-chloro-4'-hydroxy-5'-methyl phenylhydrazone
67.5 mmol of 3-chloro-4hydroxy-5-methylphenylhydrazine HC1 (melting point 2620 with decomposition) (21 g of a product of 67 of the above purity) are dissolved in a mixture of 150 ml of water and 200 ml of methanol, and the mixture is cooled to 150 .
