In the following examples the temperatures are given in degrees Celsius.
Example 1 To a solution of 15 g of 2-chloro-9- (diethylaminomethyl) anthracene in 300 cm3 of benzene, ethylene is passed in the autoclave until a pressure of 45 atmospheres is reached. This is followed by heating to 170 for 17 hours. After cooling, it is extracted with 2N hydrochloric acid. The hydrochloric acid extract is mixed with 2N sodium hydroxide solution until an alkaline reaction occurs, and the base which has separated out is then extracted with ether.
After drying and evaporation of the solvent, the 2-chloro-9- (diethylamino- methyl) -9,10-dihydro-9,10-ethano- (1,2) -anthracene of the formula remains
EMI0002.0016
its hydrochloride melts at 231.
The 2-chloro-9- (diethylaminomethyl) -anthracene used as starting material can be Herge as follows: 23 g of 2-chloro-9-anthracenaldehyde are added with stirring to a suspension of 6 g of sodium borohydride in 250 ml of ethanol. After one hour, 500 ml of water are added and the precipitate which has separated out is extracted with chloroform. After evaporation of the solvent, the 2-chloro-9- (hydroxymethyl) -anthracene remains, which melts at 140-142 after recrystallization from ethanol.
15 g of 2-chloro-9- (hydroxymethyl) -anthracene are boiled with 7 g of thionyl chloride in 50 ml of dioxane and then evaporated to dryness at 12 mm pressure. What remains is 2-chloro-9- (chloromethyl) anthracene, which melts at 140-143. This is heated to 90 in an autoclave with 15 g of diethylamine in 200 ml of ethanol for 4 hours. It is evaporated to dryness, the residue is dissolved in methylene chloride and extracted with water.
After evaporation of the solvent, the crude 2-chloro-9- (diethyl aminomethyl) anthracene remains, which is used for the Diels-Alder reaction.
Example 2 In a manner analogous to that described in Example 1, 2-chloro-9- (dimethylaminomethyl) -9,10-dihydro can be obtained from 2-chloro-9- (dimethylaminomethyn- anthracene and ethylene -9,10-ethano- (1,2) -anthracene received th, the hydrochloride of which melts at 244-245.
