worin R1 einen Arylrest, R2 einen Aralkylrest und R3 eine tertiäre Aminogruppe, die sich von einem nicht aromatischen Amin ableitet, bedeutet.
Arylreste sind vor allem Phenylreste. Aralkylreste sind vor allem Phenyl-niederalkyl-, z. B. Benzyl- oder 1- oder 2-Phenyläthylreste. Die genannten Aryl- und Aral- kylreste können im aromatischen Ring unsubstituiert oder ein-, zwei- oder mehrfach substituiert sein.
Sub- stituenten können beispielsweise niedere Alkylreste, wie Methyl-, Äthyl-, Propyl- oder Isopropylreste oder gerade oder verzweigte, in beliebiger Stellung verbundene Butyl- reste, niedere Alkoxygruppen, insbesondere Methoxy-, Äthoxy-, Propoxy- oder Butoxygruppen und Halogen atome, vor allem Fluor-, Chlor- oder Bromatome oder die Trifluormethylgruppe sein.
Als tertiäre Aminogruppe, die sich von einem nicht aromatischen Amin ableitet, kommt eine Aminogruppe in Betracht, die durch einen zweiwertigen oder zwei ein wertige Reste aliphatischen Charakters substituiert ist. Substituenten der Aminogruppe können vor allem Alkyl reste, wie niedere Alkylreste, z. B. die genannten, oder gerade oder verzweigte Alkylenreste, wie z. B. Butylen- (1,4)-, Pentylen-(1,5)-, 1,5-Dimethylpentylen-(1,5)-, Hexylen-(1,6)-, Hexylen-(1,5)- oder Oxa-, Aza- oder Thiaalkylenreste, insbesondere solche, die mit dem Stickstoffatom höchstens 8 Ringglieder ergeben, wie z. B.
3-Oxa- oder -Thiapentylen-(1,5)-, 3-Methyl-3-azahexy- len-(1,6), 3-Äthyl-1,5-dimethyl-3-azapentylen-(1,5) oder 3-Methyl-3-aza-pentylen-(1,5) sein. Die Gruppe R3 ist in erster Linie eine Pyrrolidino-, Piperidino-, Morpholino-, Thiamorpholino- oder N'- Niederalkylpiperazinogruppe, wie die N'-Methylpiper- azinogruppe, vor allem aber eine Di-Niederalkylamino- gruppe, wie die Diäthylamino- oder noch besser die Di- methylaminogruppe.
Die neuen Verbindungen besitzen wertvolle pharma kologische Eigenschaften, vor allem eine analgetische Wirkung, wie tierexperimentell z. B. an der Maus fest gestellt wurde. Ausserdem sind sie als Morphinantago nisten wirksam. Die Verbindungen können dementspre chend als Analgetika verwendet werden. Ferner wirken sie, z. B. an Ratten, Kaninchen, Katzen oder am Hund, diuretisch und besitzen weiterhin eine antitussive und spasmolytische Wirkung. Sie können daher auch in die ser Hinsicht entsprechende Anwendung als Heilmittel finden.
Daneben können die Verbindungen als Ausgangs= oder Zwischenprodukte für die Herstellung anderer wertvoller Verbindungen dienen. So kann man z. B. Verbindungen der Formel

worin R3 die eingangs gegebene Bedeutung besitzt und X für ein geeignetes Metallatom der IA-Gruppe des periodischen Systems, z. B. Lithium, oder für -Mg-Hal steht, wobei Hal Chlor, Brom oder Jod bedeutet, um setzt und den erhaltenen Komplex zersetzt.
Die Umsetzung der genannten Verbindungen kann in an sich bekannter Weise, vornehmlich in Anwesenheit eines Lösungs- oder Verdünnungsmittels, z. B. eines Äthers, wie Tetrahydrofuran, erfolgen. Die Zersetzung des erhaltenen Komplexes kann in üblicher Weise, z. B. durch Hydrolyse, vorgenommen werden.
Je nach den Verfahrensbedingungen und Ausgangs stoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Salze. Die Salze der Endstoffe können in an sich be kannter Weise, z. B. mit Alkalien oder Ionenaus- tauschern, in die freien Basen übergeführt werden. Von den letzteren lassen sich durch Umsetzung mit organi schen oder anorganischen Säuren, insbesondere solchen, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind, Salze gewinnen.
Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Per chlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Amei sen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxy- malein-, Brenztrauben- oder Lävulinsäure;
Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxy- benzoe-, Salicyl- oder p-Aminosalicylsäure, Methansul- fon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfon- säure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalin- sulfonsäure oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freimacht. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im vorausgegan genen und nachfolgend unter den freien Basen sinn- und zweckgemäss gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die Ausgangsstoffe können unter den Reaktions bedingungen gebildet werden.
Für die erfindungsgemässen Reaktionen werden vor nehmlich solche Ausgangsstoffe verwendet, die die oben erwähnten bevorzugten Verbindungen ergeben.
Die Ausgangsstoffe sind bekannt oder können nach bekannten Methoden hergestellt werden.
Erhaltene Racemate lassen sich nach bekannten Methoden, beispielsweise wie folgt in die optischen Anti poden zerlegen: Die racemischen Basen, gelöst in einem geeigneten inerten Lösungsmittel, setzt man mit einer optisch aktiven Säure um und trennt die erhaltenen Salze, z. B. auf Grund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, aus denen die Antipoden der neuen Basen durch Einwirkung alkalischer Mittel frei gesetzt werden können. Besonders gebräuchliche optisch aktive Säuren sind die D- und L-Formen von Wein säure, Di-o-Toluolweinsäure, Äpfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure.
Die Trennung kann beispielsweise auch durchgeführt werden, indem man das erhaltene reine Racemat aus einem optisch aktiven Lösungsmittel umkristallisiert. Vorteilhaft iso liert man den wirksameren der beiden Antipoden.
Die neuen Verbindungen können z. B. in Form phar mazeutischer Präparate Verwendung finden, welche sie in freier Form oder in Form ihrer Salze in Mischung mit einem für die enterale, parenterale oder topicale Appli kation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial ent halten.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
<I>Beispiel 1</I> In einem Rührkolben werden 7,2 g Magnesium (0,3 Mol) mit wenig Jod angeätzt und mit 20 ml ab solutem Tetrahydrofuran und 0,8 ml Äthylbromid ver setzt. Durch leichtes Erwärmen wird die Reaktion in Gang gesetzt und innert 15 bis 20 Minuten 49,2 g (0,3 Mol) N,N-Dimethyl-2-bromallylamin in 50 ml ab solutem Tetrahydrofuran so zugetropft, dass die Lösung dauernd im Sieden bleibt. Man erwärmt unter Rühren weitere 30 Minuten zum Sieden, bis das Magnesium bis auf kleine Reste gelöst ist.
Nun lässt man ohne Kühlung 39,3 g (0,2 Mol) Desoxybenzoin in 125 ml absolutem Tetrahydrofuran so zutropfen, dass die Reaktionslösung dauernd im Sieden bleibt. Anschliessend kocht man 6 Stunden unter Rückfluss. Man giesst auf 60 g Ammo- niumchiorid in 500 ml Wasser, extrahiert das ausge schiedene Öl mit Äther, wäscht den Äther mit Wasser und extrahiert ihn mehrmals mit 2-n.
Essigsäure (total mit 500 ml). Die essigsaure Lösung wird mit 10-n. Na- tronlauge (125 ml) alkalisch gestellt und das ausgeschie dene Öl in Äther aufgenommen. Die ätherische Lösung wird getrocknet und der Äther abgedampft. Man erhält so das 1,2-Diphenyl-3-dimethylaminomethyl-3-buten-2- ol der Formel

Durch Versetzen dieser Base in Essigester mit der berechneten Menge äthanolischer Salzsäure erhält man das 1,2-Diphenyl-3-dimethylaminomethyl-3-buten-2-ol- hydrochlorid, F. 236 bis 238 .

Man giesst auf 60 g Ammoniumchlorid in 500 ml Wasser, nimmt das ausgeschiedene Öl in Äther auf und extrahiert die ätherische Lösung mit 2-n. Essigsäure. Die essigsaure Lösung wird mit 10-n. Natronlauge alkalisch gestellt und mit Äther ausgezogen. Den nach dem Verdampfen des Äthers erhaltenen Rückstand löst man in Essigester und versetzt mit äthanolischer Salzsäure. Man erhält so das 1,2-Diphenyl-3-diäthylaminomethyl-3-buten-2-ol- hydrochlorid der Formel

Salzsäure. Aus der salz sauren Lösung kristallisiert direkt das 1,2-Diphenyl-3- piperidinomethyl-3-buten-2-ol-hydrochlorid der Formel

Nach dem Umkristallisieren aus Iso- propanol schmilzt die Verbindung bei 202 bis 204 . <I>Beispiel 4</I> In einem Rührkolben werden 7,2 g Magnesium (0,3 Mol) mit wenig Jod angeätzt und mit 20 ml absolutem Tetrahydrofuran und 0,8 ml Äthylbromid versetzt. Natronlauge alkalisch gestellt, das ausgeschiedene Öl in Äther auf genommen und die ätherische Lösung mit Wasser ge waschen.
Der durch Verdampfen des Äthers erhaltene Rückstand wird in Essigester gelöst und ergibt beim Versetzen mit äthanolischer Salzsäure das 1,2-Di-(p- methoxyphenyl) -3-dimethylaminomethyl-3 -buten-2-ol- hydrochlorid der Formel

vom F. 199 . <I>Beispiel 5</I> 21,6 g Magnesium (0,9 Mol) werden mit wenig Jod angeätzt und mit 60 ml absolutem Tetrahydrofuran über gossen. Mit 1 ml Äthylbromid wird die Grignard-Reak- tion eingeleitet.
Dann werden unter Rühren 171,1 g (0,9 Mol) N-(2 Bromallyl)-pyrrolidin in 150 ml Tetra- hydrofuran zugetropft und 30 weitere Minuten unter Rückfluss gekocht, wobei unter lebhafter Reaktion das Magnesium in Lösung geht. Innert 20 Minuten werden 117,6 g (0,6 Mol) Desoxybenzoin in 375 ml Tetrahydro- furan portionenweise so zugegeben, dass die Reaktions lösung im Sieden bleibt. Man kocht 8 Stunden unter Rückfluss, destilliert die Hälfte des Tetrahydrofurans ab und giesst auf 180 g Ammoniumchlorid in 1500 ml Wasser. Dann nimmt man in Äther auf, wäscht mit Wasser, extrahiert den Äther mit total 1000 ml 2-n. Essigsäure und zum Schluss mit 100 ml 2-n.
Salzsäure. Die sauren Extrakte werden mit 250 ml 10-n. Natron lauge versetzt, das abgeschiedene Öl wiederum in Äther aufgenommen und mit Wasser gewaschen. Aus dem Äther gewinnt man durch Abdampfen das 1,2-Diphenyl- 3-(pyrrolidinomethyl)-3-buten-2-ol der Formel

Aus der Base lässt sich in Essigesterlösung mit alko holischer Salzsäure das 1,2-Diphenyl-3-(pyrrolidino- methyl)-3-buten-2-ol-hydrochlorid, F. 223-225 , gewin nen.
Das als Ausgangsmaterial verwendete N-(2-Brom- allyl)-pyrrolidin wird analog zu Beispiel 1 aus Pyrrolidin und 2,3-Dibrompropen in Benzol gewonnen, Kp. 84 bis 88 l23 mm.
<I>Beispiel 6</I> 7,2 g Magnesium (0,2 Mol) werden mit Jod angeätzt und in 20 ml absolutem Tetrahydrofuran suspendiert. Mit 0,8 ml Äthylbromid wird die Reaktion in Gang ge bracht und dann 49,2 g (0,3 Mol) N,N-Dimethyl-2- bromallylamin in 50 ml absolutem Tetrahydrofuran so zugetropft, dass dauernde Rückflusstemperatur erhalten bleibt und anschliessend 1 Stunde im Wasserbad unter Rückfluss erwärmt.
Anschliessend gibt man 46,0 g (0,2 Mol) Benzyl-(p-chlorphenyl)-keton, gelöst in 125 ml absolutem Tetrahydrofuran, so zu, dass die exotherme Reaktion erhalten bleibt und kocht anschliessend 8 Stun den unter Rückfluss. Man giesst auf 60 g Ammonium chlorid in 500 ml Wasser, nimmt in Äther auf und trennt vom noch vorhandenen Keton durch Extraktion mit total 500 ml 2-n. Salzsäure. Der salzsaure Auszug, in welchem sich Kristalle abscheiden, wird mit 10-n. Natronlauge (etwa 120 ml) alkalisch gestellt und die freigesetzte Base in Äther aufgenommen.
Die ätherische Lösung wird mit Wasser gewaschen, getrocknet und der Äther abge dampft. Man erhält so als Rückstand das 1-Phenyl-2-(p- chlorphenyl)-3-(dimethylaminomethyl)-3-buten-2-ol der Formel

Durch Lösen in Essigester und Versetzen mit 2,5-n. äthanolischer Salzsäure bis zur sauren Reaktion erhält man das 1-Phenyl-2-(p-chlorphenyl)-3-(dimethylamino- methyl)-3-buten-2-ol-hydrochlorid, F. 241-243'.

Anschliessend gibt man 45,0 g (0,2 Mol) Benzyl-(p-methoxyphenyl)-keton, ge löst in 125 ml absolutem Tetrahydrofuran, so zu, dass die exotherme Reaktion erhalten bleibt und kocht an- schliessend 8 Stunden unter Rückfluss. Man giesst auf 60 g Ammoniumchlorid in 500 ml Wasser, nimmt in Äther auf und trennt vom noch vorhandenen Keton durch Extraktion mit verdünnter Essigsäure. Durch Ver setzen mit Natronlauge wird alkalisch gestellt und die freigesetzte Base in Äther aufgenommen. Die ätherische Lösung wird mit Wasser gewaschen, getrocknet und der Äther abgedampft.
Man erhält so als Rückstand das 1 Phenyl-2-(p-methoxyphenyl)-3-(dimethylaminomethyl) 3-buten-2-ol der Formel

Man giesst auf 60g Ammo niumchlorid in 500 ml Wasser, nimmt in Äther auf und trennt vom noch vorhandenen Keton durch Extraktion mit 2 mal 100 ml 2-n. Essigsäure. Der ausfallende Nie derschlag wird isoliert und in warmem Benzol gelöst. Die ätherische und die benzolische Lösung werden nun mit total 350 ml 2-n. Salzsäure ausgezogen. Die Essig säure- und Salzsäureauszüge werden vereinigt, wobei sich Kristalle bilden. Die vereinigten Auszüge werden mit 150 ml 10-n. Natronlauge alkalisch gestellt und die freigesetzte Base in Äther aufgenommen. Die ätherische Lösung wird mit Wasser gewaschen, getrocknet und der Äther abgedampft.
Man erhält so als Rückstand das 1-(p-Chlorphenyl)-2-phenyl-3-(dimethylaminomethyl)-3- buten-2-ol der Formel

Anschliessend gibt man 49,0 g (0,25 Mol) Desoxybenzoin, gelöst in 125 ml absolutem Tetrahydrofuran, so zu, dass die exotherme Reaktion erhalten bleibt und kocht anschliessend 8 Stunden unter Rückfluss. Man giesst auf 60 g Ammoniumchlorid in 500 ml Wasser, nimmt in Äther auf und trennt vom noch vorhandenen Keton durch Extraktion mit total 600 ml 2-n. Essigsäure. Aus dem essigsauren Auszug wird mit 150 ml 10-n.

<I>Beispiel 14</I> 56,28 g (0;2 Mol) 1,2-Diphenyl-3-(dimethylamino- methyl)-3-buten-2-ol werden in 200 ml Aceton gelöst und mit einer Lösung von 15,0 g (0,1 Mol) D(+)Wein- säure in 400 ml Aceton versetzt. Man reibt an und lässt 4 Stunden bei Raumtemperatur stehen. Die ausgeschie denen Kristalle werden abgenutscht und mit Aceton nachgewaschen.
(Aus dieser Mutterlauge kann, wie im folgenden Beispiel beschrieben, die l-Form gewonnen werden.) Durch Umkristallisation der erhaltenen Kri stalle aus einem Gemisch von 250 ml Aceton und 285 ml absolutem Äthanol erhält man das d-1,2-Di- phenyl-3-(dimethylaminomethyl)-3-buten-2-ol-D-tartrat, F.142-145 , [a]20 = + 86 1 (in Wasser, c =1%).
58 g d-1,2-Diphenyl-3-(dimethylaminomethyl)-3- buten-2-ol-D-tartrat werden in 300 ml Wasser gelöst und mit 200 ml 2-n. Natronlauge versetzt und mit Äther extrahiert. Die ätherische Lösung wird zweimal mit Was ser gewaschen, getrocknet und am Vakuum zur Trockne eingedampft. Man erhält so als Rückstand das d-1,2- Diphenyl-3-(dimethylaminomethyl)-3-buten-2-ol vom F. 79-84 , [a120 = +195 0,5 (in Chloroform, c = 2 0/0).
<I>Beispiel 15</I> Die Aceton-Mutterlauge aus dem vorhergehenden Beispiel wird mit einer Lösung von 15,0 g (0,1 Mol) D(+)Weinsäure in 400 ml Aceton versetzt. Man reibt an und lässt 4 Stunden stehen. Die erhaltenen Kristalle werden abgenutscht und mit Aceton gewaschen. Man erhält so ein Tartrat mit einem [a]20D von -26 bis -36 (in Wasser).
Aus verschiedenen Versuchen erhaltene Tartrate mit [a] D von -26 bis -36 werden vereinigt, in Wasser gelöst, mit 2-n. Natronlauge die Base gefällt und diese mit Äther isoliert. 79,4 g (0,282 Mol) des so erhaltenen 1,2-Diphenyl-3-(dimethylaminomethyl)-3 -buten-2-ols, das ein [a] j 0 von ungefähr -92 (in Chloroform) be sitzt, werden in 300 ml Aceton gelöst und mit einer Lösung von 21,0 g (0,141 Mol) D-Weinsäure in 400 ml Aceton warm versetzt. Nach 5 Stunden wird abgenutscht.
Man erhält 54 g eines Produktes vom F. 138-141 und [a]20' = -42 (in Wasser) (die Mutterlauge, erneut mit 21,0g D-Weinsäure in 400 ml Aceton versetzt, gibt wei tere 57 g vom F. 130-137 und [a120' = -14 ).
Das D-Tartrat mit [a]20D = -42 wird aus einem Gemisch von 250 ml Aceton und 285 ml absolutem Äthanol umkristallisiert und liefert ein Kristallisat vom F. 139-145' und [a]20' = -49 (in Wasser).
Erneutes Umkristallisieren aus einem Gemisch von 250 ml Aceton und 200 ml Äthanol ergibt ein Produkt vom F. 147-149 und [a] D = -67 (in Wasser). Dieses Tartrat wird in Wasser gelöst, mit 2-n. Natronlauge die Base gefällt und diese mittels Äther isoliert. Die äthe rische Lösung wird zweimal mit Wasser gewaschen, ge trocknet und der Äther verdampft. Man erhält so das l-1,2-Diphenyl-3-(dimethylaminomethyl)-3-buten-2-ol vom F. 79-80', [a]201 = -201' <B>0,5'</B> (in Chloroform, c = 2 %).
