Verfahren zur Herstellung neuer pharmazeutisch wertvoller Thiophenverbindungen.
Gegenstand vorliegender Erfindung ist ein Verfahren zur Herstellung von Verbindungen der Formel

worin Alkl eine gerade oder verzweigte Alkylidengruppe mit bis zu 4 C-Atomen, Alk2 eine gerade oder verzweigte gesättigte niedere Alkylengruppe mit mindestens 2 C-Atomen als Kette zwischen der Gruppe NR1 und dem Phenylrest, deren zunächst am Phenylrest stehendes C-Atom auch eine Hydroxylgruppe tragen kann und R Wasserstoff oder eine niedere Alkylgruppe ist. R2 und R3 sind gleich oder verschieden und bedeuten Wasserstoff, Halogen, eine Hydroxylgruppe oder eine Alkoxygruppe.
Das Verfahren ist dadurch gekennzeichnet, dass man aus einer Verbindung der Formel

worin Alkl eine der Alkylidengruppe Alkl entsprechende Alkylengruppe ist, Wasser abspaltet.
Es empfiehlt sich, die Abspaltung des Wassers in einem sauren, nicht oxydierenden Medium durchzufüh ren, vorzugsweise bei pH-Werten von 3 oder weniger.
Hierbei ist es in vielen Fällen zweckmässig, bei erhöhter Temperatur zu arbeiten.
Die so erhaltenen Basen können mit Hilfe von anorganischen oder organischen Säuren in die entsprechenden Salze übergeführt werden. Man kann auch mit Hilfe von Alkylhalogeniden oder anderen Alkylderivaten die entsprechenden quartären Salze gewinnen.
Die neuen Verbindungen und ihre Salze haben wertvolle zentralstimulierende und coronarerweiterende Eigenschaften.
Beispiel 1
Aus 4,8 g Mg-Spänen (0,2 Mol) und 31,4 g Brombenzol (0,2 Mol) wird in üblicher Weise die Grignard Verbindung bereitet. Zu dieser Lösung lässt man 31 g (0,1 Mol) 2-{N-[3'-Thienyl-3'-oxo-propyl-(1')]}amino-3phenyl-propan-HCl in Äther suspendiert eintropfen. Es wird 4 Stunden am leichten Sieden gehalten und anschliessend in der Kälte mit NH4Cl und Wasser zersetzt. Das 2-{N-[3'-Phenyl-3'-thienyl-3'-hydroxy-propyl (1)]}-amino-3-phenylpropan wird wie üblich isoliert.
In eine Lösung von 40 g 2-{N-[3'-Phenyl-3'-thienyl 3'-hydroxy-propyl-(1)] }-amino-3 -phenylpropan in 400ml Eisessig wird 20 Minuten trockenes HCl-Gas eingeleitet. Anschliessend wird 1 Stunde am Rückfluss gekocht, der Eisessig abdestilliert, der Rückstand in Wasser gelöst und mit Ammoniak alkalisch gestellt.
Das 2-tN-[3'-Phenyl-3'-thienyl-propan-1'-yl-(1')]}-ami- no-3-phenyl-propan wird als Öl isoliert. Die Base wird mit isopropanolischer HCl neutralisiert und das HCl Salz aus Isopropanol umkristallisiert ; Fp. 1740 C.
Beispiel 2
In eine Suspension von 9 g 2-{[3-Phenyl-3-thienyl (2)-3-hydroxypropyl-(1)]-amino}-3-(p-chloro-phenyl)propan-HCl in 100 ml Eisessig wird so lange Chlorwasserstoff eingeleitet, bis alles gelöst ist. Nach Abdestillieren des Eisessigs wird mit konzentriertem Ammoniak der Rückstand alkalisch gemacht und mit insgesamt 500 ml Äther extrahiert. Aus der getrockneten Ätherlösung wird mit isopropanolischer HCl-Lösung das 2 {(3-Phenyl-3-thienyl-(2)-propan-(1)-yl-(1)]-amino}-3 (p-chloro-phenyl)-propen-hydrochlorid erhalten (7 g, 81 lo Ausbeute). Aus Isopropanol umkristallisiert liegt der Schmelzpunkt bei 193 bis 194 C.
Beispiel 3
Das als Ausgangsprodukt für die erfindungsgemässe Wasserabspaltung dienende 2-(N-[3'-Phenyl-3'-thienyl3'-hydroxy-propyl-{1'}])-amino-3-phenyl-3-hydroxypropan, einer Base, die ein Hydrochlorid mit dem Smp.
2030 C (umkristallisiert aus Isopropanol) bildet, kann nach folgenden beiden Varianten hergestellt werden: a) Eine Lösung von 77 g Butyllithium (1,17 Mol) in 1030 ml Isopentan wird auf 15 C gekühlt. Dann lässt man 49 g Thiophen (0,58 Mol) bei dieser Temperatur zutropfen. Nach einer halben Stunde wird auf 5 C gekühlt und 177 g (0,58 Mol) 2-'N-[3'-Phenyl-3'-oxo-pro pyl-(1')]}-amino-3-phenyl-3-hydroxy-propan in Äther zugegeben. Danach wird noch eine Stunde gerührt und schliesslich unter Kühlung mit wässriger NH4Cl-Lösung zersetzt. Die jaitherschicht wird getrennt und in üblicher Weise aufgearbeitet.
Die Fraktion zwischen 200 und 2450 C bei 0,02 Torr enthielt das 2-(N-[3'-Phenyl-3'- thienyl-3'-hydroxy-propyl-(1')]}-amino-3-phenyl-3-hydroxy-propan. b) Aus 24,3 g Mg-Spänen (1 Mol) und 163 g 2 Brom-thiophen (1 Mol) wird in üblicher Weise die Grignardverbindung bereitet. Zu dieser Lösung lässt man 159,7 g (0,5 Mol) 2-{N-[3'-Phenyl-3'-oxo-propyl (1')] }-amino-3-phenyl-3-hydroxy-propan-HCl in 1 Liter Äther suspendiert eintropfen. Die Reaktion ist sofort beendet. Es wird in der Kälte mit NH4Cl und H2O zersetzt und wie üblich aufgearbeitet. Das 2-{N-[3'-Phenyl3'-thienyl-3'-hydroxy-propyl-(1')]}-amino-3-phenyl-3hydroxy-propan kann mit Benzin und wenig Benzol zur Kristallisation gebracht werden. Fp. 72 bis 730 C.
Das Hydrochlorid hat einen Schmelzpunkt von 203 C und ist identisch mit der nach der vorstehend beschriebenen Variante a) hergestellten Verbindung.
36,7 g (0,1 Mol) der nach den vorstehend beschriebenen Weisen hergestellten Hydroxylverbindung werden in 63 mi Eisessig gelöst und durch zweistündiges Kochen unter Rückfluss mit Jodwasserstoff in statu nascendi dehydratisiert. Die Lösung wird mit Natriumbisulfit entfärbt, die Essigsäure abdestilliert und die Base mit Ammoniak in Freiheit gesetzt. Das rohe 2-{N-[3'-Phenyl 3' - thienyl - propen-1'- yl-(l')] }-aniino-3-phenyl-3-hydro- xy-propan wird in das HCl-Salz übergeführt und aus Isopropanol umkristallisiert. Fp. 2040 C.
Die als Ausgangsstoffe des erfindungsgemässen Verfahrens dienenden Verbindungen der Formel II können beispielsweise so hergestellt werden, dass man eine Verbindung der allgemeinen Formel

The Grignard compound is prepared in the usual manner from 4.8 g of Mg turnings (0.2 mol) and 31.4 g of bromobenzene (0.2 mol). 31 g (0.1 mol) of 2- {N- [3'-thienyl-3'-oxo-propyl- (1 ')]} amino-3phenyl-propane-HCl suspended in ether are added dropwise to this solution. It is kept boiling gently for 4 hours and then decomposed in the cold with NH4Cl and water. The 2- {N- [3'-phenyl-3'-thienyl-3'-hydroxypropyl (1)]} -amino-3-phenylpropane is isolated as usual.
Dry HCl gas is passed into a solution of 40 g of 2- {N- [3'-phenyl-3'-thienyl 3'-hydroxypropyl- (1)]} -amino-3-phenylpropane in 400 ml of glacial acetic acid for 20 minutes . In a suspension of 9 g of 2 - {[3-phenyl-3-thienyl (2) -3-hydroxypropyl- (1)] -amino} -3- (p-chloro-phenyl) propane HCl in 100 ml of glacial acetic acid so long hydrogen chloride passed in until everything is dissolved.
