Verfahren zur Hersteliung von Steroiden Gegenstand des Patentes Nr. 347<B>521</B> ist ein Ver fahren zur Herstellung von JO(ll)-Steroiden durch Dehydratisierung von 11-Oxysteroiden. Es wurde nun gefunden, dass sich auch Steroide der Androstan- und Pregnanreihe, die Hydroxylgruppen in anderen Stel lungen tragen, auf dieselbe Weise dehydratisieren las sen unter Bildung der entsprechenden ungesättigten Verbindungen.
Es ist so möglich, eine grosse Zahl un gesättigter Steroide herzustellen, die an sich bemer kenswerte physiologische Wirksamkeit besitzen oder wertvolle Zwischenprodukte zur Herstellung anderer physiologisch aktiver Steroidverbindung sind. Von spezieller Bedeutung sind infolge ihrer Wirksamkeit die i)(11)-Steroide, wie z.
B. 9(11)-Dehydro-desoxy- corticosteronacetat, 9(11)-Dehydro-progesteron, 9(11)- Dehydro-testosteronacetat, 9(11)-Dehydro-östradiol- acetat, 9(11)-Dehydro-17a-oxy-progesteron, 9(11)- Dehydro -17a - oxy - desoxycorticosteronacetat <B>USW.</B> Diese Verbindungen können nach dem Verfahren der vorliegenden Erfindung aus den entsprechenden 9a Oxysteroiden hergestellt werden.
Die d9(ll)-Steroide können auch in wertvolle Zwischenprodukte zur Her stellung physiologisch wirksamer Steroide übergeführt werden. So kann z. B. 9(11)-Dehydro-pregnenolon durch Oxydation nach Opperiauer zum 9(11)-Dehy- dro-progesteron oxydiert werden. Die d9(ll)-Steroide eignen sich ferner zur Herstellung der 9a-Chlor- und 9a-Fluor-11f-oxy-analoga der bekannten physiolo gisch aktiven Nebennierenrinden-und Sexualhormone, insbesondere von solchen, welche auch die d4-3,20 Diketofunktion enthalten.
(Siehe Fried und Sabo, J. Am. Chem. Soc. 75, 2273 [l953].) Hydriert man J9(11)-Steroide, so erhält man die entsprechenden 9(11)-Hydroverbindungen. Diese lassen sich nach be kannten Methoden in bekannte physiologisch aktive Steroide und/oder Zwischenprodukte überführen.
Von besonderer Bedeutung, entweder wegen ihrer physiologischen Wirksamkeit an sich oder als Zwi schenprodukte zur Herstellung physiologisch wirk samer Steroide, sind 4,9(11)-Androstadien-3,17-dion, 6-Methyl-4-pregnen-3,11,20-trion, 4,14-Pregnadien und 4,16-Androstadien-3-on.
Bisher verwendete man in der Steroidchemie Ha- logenamide und Halogenimide als Oxydations- oder Halogenisierungsmittel oder in gewissen Fällen für beide Zwecke gleichzeitig. Die Verwendung dieser Stoffe in einer Dehydratisierungsreaktion war hin- gegen nicht bekannt.
Die Reaktion von Hydrocorti- sonacetat mit einem N-Halogenimid unter Wasser ausschluss und in Gegenwart einer Base, wie Pyridin, zwecks Herstellung einer 11-Ketogruppe, ist Gegen stand des US-Patentes Nr. 2 751402. Das Produkt dieser Reaktion ist Cortisonacetat, d. h. es handelt sich um eine Oxydationsreaktion. Bei den früheren Verfahren wurde das bei der Oxydation mit N-Halo- genamiden oder N-Halogenimiden entstehende Pro dukt mit einem wässrigen Reduktionsmittel, z. B.
wässrigem Natriumsulfit versetzt, um die überschüs sige N-Halogenverbindung zu zerstören. Im Gegen satz dazu verwendet man gemäss vorliegender Erfin dung wasserfreies Schwefeldioxyd, um die Dehydrati- sierung und die Zersetzung der überschüssigen N- Halogenverbindung zu bewirken.
Gemäss dem Verfahren der vorliegenden Erfin dung wird ein kernhydroxyliertes Steroid der Andro- stan- oder Pregnanreihe unter wasserfreien Bedin gungen in einem basischen, nicht an der Reaktion teilnehmendem, organischen Medium mit einem N Halogenamid oder N-Halogenimid und wasserfreiem Schwefeldioxyd zusammengebracht und das gebildete dehydratisierte Steroid aus der Reaktionsmischung abgetrennt. Die Gegenwart von Wasser würde zur Re- generierung des hydroxylgruppenhaltigen Ausgangs materials führen.
Bei unzulässig langer Verzögerung des Schwefeldioxydzusatzes stabilisiert sich das mit dem Hypohalogenit gebildete Zwischenprodukt unter Ausbildung einer Ketogruppe, so dass man keine Dehydratisierung, sondern eine Oxydationsreaktion erhält.
Zur Erzielung einer hohen Ausbeute muss man deshalb das Schwefeldioxyd innert etwa 2 Stunden nach dem Vermischen des Steroids mit dem N-Halo- genamid oder N-Halogenimid zusetzen, wobei diese Zeit je nach dem speziellen verwendeten Ausgangs- steroid variieren kann.
Das Steroidhypohalogenit erhält man gewöhnlich durch Umsetzung des Oxysteroids mit einem N-Halo- genamid oder N-Halogenimid, in welchem das Halo gen ein Atomgewicht von 35-127 hat und vorzugsweise Chlor oder Brom ist, oder mit einer anderen Verbin dung, die befähigt ist, eine Hydroxyverbindung in ein Hypohalogenit umzuwandeln.
Beispiele für solche Verbindungen sind N-Chlor- acetamid, N-Bromacetamid, N-Chlorsuccinimid, N- Bromsuccinimid, N-Jodsuccinimid, N-Bromphthali- mid, 3-Brom-5,5-dimethylhydantoin, 1,3-Dibrom-5,
5- dimethylhydantoin. Gewöhnlich verwendet man mehr als ein Moläquivalent bezogen auf das Oxysteroid. Falls das fdolekül andere Gruppen enthält, die oxy diert oder dehydratisiert werden können, sollte man entsprechend grössere Mengen der N-Halogenverbin- dung verwenden, um optimale Ausbeuten zu errei chen.
Die Reaktion des Oxysteroids mit der N-Halo- genverbindung erfolgt in Gegenwart einer basischen organischen Verbindung, die von den Reagenzien praktisch nicht angegriffen wird. Beispiele für solche Basen sind die tertiären Amine, deren Aminstickstoff ein Glied eines aromatischen Ringes ist, z.
B. die Pyridine wie Pyridin und Alkylpyridine, wie Picolin, Lutidin, Collidin, Conyrin, Parvulin usw., und die niedrigen Fettsäureamide, insbesondere Formamid, Methyl- und Dimethylformamid.
Wahrscheinlich entstehen Komplexe einiger der oben genannten Verbindungen mit dem Schwefel dioxyd, doch hat dies keinen nachteiligen Einfluss auf ihre Verwendbarkeit als Medium für die primäre Reaktion oder als Mittel zur wirksamen Entfernung von Halogenwasserstoff, wie er im Verlaufe der Reaktion entsteht.
Die Base wird vorzugsweise im molaren über schuss, bezogen auf das Oxysteroid, z. B. in zehnfach molarer Menge verwendet und stellt vorzugsweise das einzige Lösungsmittel für die Reaktion dar.
Die Umsetzung des Oxysteroids mit der N-Halo- genverbindung erfolgt unter wasserfreien Bedingun gen. Die Bezeichnung wasserfrei bedeutet, dass das Reaktionsgemisch, bezogen auf das Steroid, weniger als ein Moläquivalent Wasser enthalten soll. Wenn die Mischung ein Moläquivalent oder mehr Wasser enthält, so wird praktisch kein dehydriertes Steroid gebildet.
Je weniger Wasser die Mischung enthält, desto grösser ist die Ausbeute an dehydriertem Steroid. Vorzugsweise enthält die Mischung weniger als 0,1 Moläquivalent Wasser.
Die Reaktion zwischen N-Halogenamid oder N- Halogenimid und dem Oxysteroid lässt sich vorzugs weise zwischen -40 C und +70 C durchführen, wobei die untere Grenze durch die Löslichkeit der Reaktionsteilnehmer und eine angemessene Reak tionszeit, die obere Grenze durch die normalerweise bei Umsetzungen mit N-Halogenverbindungen bei höheren Temperaturen auftretenden Nebenreaktionen festgelegt werden. Die übliche Zimmertemperatur wird aus Zweckmässigkeitsgründen bevorzugt und lie fert stets hohe Ausbeuten an dem gewünschten Pro dukt.
Die Reaktionszeit wird vor allem durch die Ge schwindigkeit bestimmt, mit der sich das so erhaltene Hypohalogenit unter Bildung der Ketogruppe um lagert. Erhöht man die Temperatur über Zimmer temperatur, so wird die Reaktionszeit entsprechend verkürzt, während niedrigere Temperaturen dieselbe verlängern. In den meisten Fällen ist die Reaktion zwischen der Hydroxylgruppe und der Halogenver bindung bei Zimmertemperatur nach einer halben Stunde beendigt, und mindestens beim Hydrocorti- sonacetat bereits in weniger als 5 Minuten.
Das wasserfreie Schwefeldioxyd kann gasförmig oder flüssig sein bzw. in Form eines Materials, das wie z. B. ein Alkalimetallhyposulfit Schwefeldioxyd in situ bildet, vorliegen.
Die Reaktion zwischen der Hypohalogenitgruppe und dem Schwefeldioxyd bildet offenbar ein Halogen- sulfonat, das unter den wasserfreien Bedingungen und in Gegenwart einer Base in das Salz aus dehydrati siertem Steroid, Halogenwasserstoff, Base und Schwe- feltrioxyd umgewandelt wird. Der Bereich der Reak tionstemperatur ist praktisch der gleiche wie bei der Herstellung des Hypohalogenits. Die Reaktion scheint in den meisten Fällen momentan zu verlaufen und das Reaktionsprodukt kann fast sofort isoliert wer den.
Längere Reaktionszeiten scheinen auf die Aus beute keine oder nur geringe Wirkung zu haben.
Wie schon früher gesagt, kann das Verfahren der vorliegenden Erfindung zur Dehydratisierung von be liebig kernhydroxylierten Steroiden der Androstan- und Pregnanreihe Verwendung finden. Andere, in verschiedenen Stellungen am Steroidkern gebundene Substituenten scheinen keinen Einfluss auf die grund legende Operation des Verfahrens zu haben.
Es ist deshalb nach dem Verfahren möglich, aus geeigneten Ausgangsmaterialien Steroidverbindungen zu erhalten, die in 1-2-, 2-3-, 3-4-, 5-6-, 6-7-, 7-8-, 8-9-, 8-14-, 9-11-, 11-12-, 14-15-, 15-16- und 16-17- Stellung ungesättigt sind.
<I>Beispiel 1</I> 35 mg (0,25 Millimol) N-Bromacetamid in Pyri- din wurden so lange mit Schwefeldioxyd behandelt, bis eine Probe mit angesäuertem Kaliumjodidstärke- papier negativ ausfiel; die Mischung wurde sodann bei 25 C mit einer Lösung von 20 mg (0,06 Milli- mol) 3u-Oxy-pregnan-11,20-dion in über Barium oxyd destilliertem Pyridin versetzt.
Das resultierende Gemisch wurde etwa 45 Minuten gerührt, auf 12 C gekühlt und tropfenweise mit Wasser versetzt. Nach mehrstündigem Kühlen auf 0-5 C wurde vom Nie derschlag abfiltriert, mit Wasser gewaschen und ge trocknet. Man erhielt eine Rohausbeute von 24 mg 3-Pregnen-11,20-dion, das anhand seines Spektrums identifiziert wurde.
<I>Beispiel 2</I> Zu einer Lösung von 7,5 kg 5a-Oxy-6-methyl- pregnan-3,11,20-trion in 71,5 1 frisch destilliertem und getrocknetem Pyridin gab man bei Zimmer temperatur 6,950 kg N-Bromacetamid. Die Mischung wurde bei 25 C eine halbe Stunde gerührt und auf 0 C abgekühlt, dann während der Zugabe von 19,277 kg Schwefeldioxydgas bei 0-10 C gehalten. Die Mischung wurde sodann auf 25 C erwärmt, 11o Stunden gerührt und wieder auf<B>0-5'</B> C gekühlt.
Dann setzte man 200 1 10%ige Schwefelsäure und hernach weitere 36 1 Wasser zu und rührte noch etwa 3 Stunden. Man filtrierte vom entstandenen festen Produkt, wusch gründlich mit Wasser und trocknete im Vakuum bei 60 C bis zur Gewichtskonstanz. Man erhielt 6,484 kg (90,7 % der Theorie) 6ss-Methyl- 4-pregnen-3,11,20-trion. Schmelzpunkt 162,5 bis 165 C; [a]D: +207 C (Chloroform); Emat =16,067.
<I>Beispiel 3</I> Eine Lösung von 10,3 mg (0,03 Millimol) 6ss Oxy-11-keto-progesteron in über Bariumoxyd frisch destilliertem Pyridin wurde auf 25 C gebracht. Dann wurde eine Lösung von 8,6 mg (0,06 Millimol) N-Bromacetamid in Pyridin zugegeben und 30 Mi nuten bei 25 C gerührt. Nach dem Abkühlen, auf 12""C wurde so lange trockenes Schwefeldioxyd ein geleitet, bis die Reaktion mit angesäuertem Kalium- jodidstärkepapier negativ war.
Dann wurde unter Rühren und bei einer Temperatur der Mischung von unterhalb 30 C Wasser zugetropft. Die entstandene Suspension wurde auf 0-5 C gekühlt und einige Stunden bei dieser Temperatur gehalten. Der Nieder schlag wurde abfiltriert, mit Wasser gewaschen und getrocknet. Man erhielt 4,6-Pregnadien-3,11,20-trion in einer Rohausbeute von 9,5 mg.
Das Produkt ent hielt laut Papierchromatogramm kein 6ss-Oxy-11- keto-progesteron. Das IR-Spektrum zeigte Absorp tion bei 1698 cm-' (nicht konjugierte Ketogruppe), 1650 cm-' (konjugierte Ketogruppe) und 1620, 1586 cm-' (44.G C=C), was mit der zugeordneten Struktur übereinstimmt.
<I>Beispiel 4</I> Man arbeitete genau nach Beispiel 1, jedoch unter Verwendung von 6ss-Oxy-11-keto-progesteron als Ausgangsmaterial und erhielt 4,6-Pregnadien-3,11,20- trion. Die Infrarotanalyse stimmte mit der Struktur überein und war identisch mit der im Beispiel 3 für das gleiche Produkt angegebenen. <I>Beispiel 5</I> Man arbeitete gemäss Beispiel 3, jedoch mit 10,8 mg 6ss-Oxy-5ss-pregnan-3,11,20-trion als Aus gangsmaterial, und erhielt eine Rohausbeute von 8,1 mg 6-Pregnen-3,11,
20-trion. Das IR-Spektrum zeigte Spuren von restlichem Hydroxyl und starke Bande für nicht konjugierte Ketogruppen.
<I>Beispiel 6</I> Man arbeitete wie im Beispiel 1 unter Verwen dung von 10,8 mg 6ss-Oxy-5ss-pregnan-3,11,20-trion und erhielt<B>11,0</B> mg rohes 6-Pregiien-3,11,20-trion. Das IR-Spektrum zeigte wie im vorangehenden Bei spiel Spuren restlicher Hydroxylgruppen und nicht konjugierte Ketogruppen an.
<I>Beispiel 7</I> Man arbeitete wie im Beispiel 3, aber unter Ver wendung von 9,7 mg 7ss-Oxy-4-androsten-3,17-dion als Ausgangsmaterial. Man erhielt 6,8 mg Rohpro dukt, das zur Hauptsache aus 4,6-Androstadien-3,17- dion bestand. Das IR-Spektrum bestätigte die Struk tur und zeigte Absorption bei 1730 cm-' (17-Keto- gruppe), 1657 cm-' (konjugierte Ketogruppe), 1615, 1580 cm-' (konjugierte C=C). UV-Absorption bei 283 m,u (44,s C=C).
<I>Beispiel 8</I> Man arbeitete wie im Beispiel 1, jedoch mit 7ss-Oxy-4-androsten-3,17-dion als Ausgangsstoff, und erhielt 4,3 mg rohes 4,6-Androstadien-3,17-dion. Die Infrarotanalyse bestätigte die Struktur und Identität mit dem Produkt des Beispiels 7.
<I>Beispiel 9</I> Man arbeitete wie im Beispiel 3, jedoch unter Verwendung von 9,9 mg 9a-Oxy-progesteron als Ausgangsmaterial. Man erhielt 3,0 mg rohes 9(I1)- Dehydroprogesteron. Die papierchromatographische und Infrarotanalyse bestätigten die Struktur. Fol gende Absorptionen wurden gemessen: 1695 cm-' (20-Ketogruppe); 1668 cm' (konjugierte Ketogruppe) 1611 cm-1 (44 C=C).
<I>Beispiel 10</I> Man arbeitete wie im Beispiel 1, jedoch unter Verwendung von 19,8 mg 9a-Oxy-progesteron als Ausgangsmaterial, und erhielt eine Rohausbeute von 14,9 mg 9(11)-Dehydro-progesteron. Papierchromato- gramm und Infrarotanalyse stimmten mit der Struk tur überein; das Produkt war mit dem Produkt nach Beispiel 9 identisch.
<I>Beispiel 11</I> Arbeitet man genau nach Beispiel 3, jedoch un ter Verwendung von 9,9 mg 14a-Oxy-progesteron als Ausgangsmaterial, so erhält man 8,6 mg rohes 14(15)-Dehydro-progesteron, dessen Struktur durch IR-Analyse bestätigt wurde. Das Produkt zeigte kein restliches Hydroxyl und folgende Absorptionsbanden: 1695 cm-' (20-Ketogruppe) 1668 cm-1 (konjugierte Ketogruppe), 1611 cm-1 (d4 C=C).
<I>Beispiel 12</I> Arbeitet man genau wie im Beispiel 1, jedoch unter Verwendung von 9,9 mg 14a-Oxy-progesteron als Ausgangsmaterial, so erhält man 7,5 mg rohes 14(15)-Dehydro-progesteron. Die Infrarotanalyse stimmt mit der zugeordneten Struktur überein. Das IR-Spektrum ist identisch mit demjenigen des Pro duktes von Beispiel 11.
<I>Beispiel 13</I> Arbeitet man wie im Beispiel 3, aber unter Ver wendung von 19,6 mg 14a-Oxy-l-dehydro-progeste- ron als Ausgangsmaterial, so erhält man 15,5 mg rohes 1,14(16)-Dehydro-progesteron. Die papierchro- matographische Analyse zeigt kein Ausgangsmaterial im Endprodukt.
Die Infrarotanalyse bestätigt die Struktur mit folgenden Absorptionsmaxima: 1690, 1688 cm-' (nichtkonjugierte Ketogruppe), 1652 cm-1 (konjugierte Ketongruppe), <B>1618,</B> 1597 cm-1 (d1,14_ C=C).
<I>Beispiel 14</I> Arbeitet man genau wie im Beispiel 1, jedoch unter Verwendung von 9,8 mg 14a-Oxy-l-dehydro- progesteron, so erhält man 8,1 mg rohes 1,14(15)- Dehydro-progesteron. Die Infrarotanalyse bestätigte die Struktur der mit dem Produkt von Beispiel 13 identischen Verbindung.
<I>Beispiel 15</I> Arbeitet man wie im Beispiel 3, jedoch unter Verwendung<I>von 9,9 mg</I> 15a-Oxy-progesteron als Ausgangsmaterial, so erhält man 7,3 mg Produkt. Die papierchromatographische Untersuchung zeigt das Vorhandensein einer Spur von unverändertem 15a-Oxy-progesteron. Die Infrarotanalyse bestätigt die Struktur für 14(15)-Dehydro-progesteron.
<I>Beispiel 16</I> Arbeitet man genau wie im Beispiel 1, jedoch unter Verwendung von 9,9 mg 15a-Oxy-progesteron als Ausgangsmaterial, so erhält man 9,8 mg Produkt. Die papierchromatographische Untersuchung zeigt das Vorhandensein geringer Mengen eines Pyridin- Schwefeldioxyd-Komplexes an; die Infrarotanalyse bestätigt die Struktur des 14(15)-Dehydro-progeste- rons.
<I>Beispiel 17</I> Arbeitet man wie im Beispiel 1, jedoch unter Ver wendung von 15a-Oxy-androsten-3,17-dion als Aus gangsmaterial, so erhält man 10,8 mg rohes 4,14(15)- Androstadien-3,17-dion. Die Infrarotanalyse zeigt eine Spur restliches Hydroxyl und bestätigt ansonsten die zugeordnete Struktur. Absorption: 1730 cm-' (nicht konjugierte Ketogruppe), 1660 cm-' (44-3 Ketogruppe), 1615 cm-1 (44 C=C).
<I>Beispiel 18</I> Arbeitet man wie im Beispiel 3, jedoch unter Verwendung von 8,7 mg Testosteron als Ausgangs material, so erhält man 7,2 mg 4,16-Androstadien- 3-on. Die Infrarotanalyse zeigt kein restliches Hy- droxyl und bestätigt die Struktur mit folgenden Ab sorptionsbanden: 1670 cm-' (J--3-Ketogruppe), 1615 cm-' (44 C=C).
<I>Beispiel 19</I> Arbeitet man genau wie im Beispiel 1, jedoch unter Verwendung von 8,7 mg Testosteron als Aus gangsmaterial, so erhält man 6,4 mg Produkt. Das Produkt ist identisch mit der nach Beispiel 18 her gestellten Verbindung.
Aus den vorstehenden Beispielen wird ersichtlich, dass die vorliegende Erfindung zur Dehydratisierung von beliebig kernhydroxylierten Steroiden der Andro- stan- und Pregnanreihe dienen kann.
Die Konstitu tion des restlichen Moleküls des Ausgangssteroids ist von geringem Einfluss auf die Durchführbarkeit des Verfahrens. Es ist notwendig, dass die Reaktion unter wasserfreien Bedingungen und in Gegenwart eines basischen nicht reagierenden organischen Mediums erfolgt; dagegen sind Änderungen der Reaktionszei ten, Temperatur und Konzentration der Reaktions teilnehmer zulässig, ohne den normalen Verlauf des Verfahrens zu verändern.
Wie in den Beispielen 1 und 3 sowie den. darauf folgenden gezeigt wurde, ist es gleichgültig, ob Ausgangssteroid und N-Halogenver- bindung zuerst vermischt und dann das Schwefel dioxyd zugegeben wird, oder ob man die N-Halogen- verbindung zuerst mit dem Schwefeldioxyd zusam menbringt und diese Mischung dem Ausgangssteroid zusetzt.
Die Bezeichnung < basisches nicht reagierendes organisches Medium umschreibt ein Medium, das mit den Ausgangssteroiden sowie mit den N-Halogen- amiden nicht reagiert. Das Medium reagiert jedoch mit dem Halogenwasserstoff in dem Masse, wie er bei der Dehydratisierung entsteht, und verhindert so eine Halogenierung des Steroidkerns. Zudem ist es wahr- scheinlich, dass zwischen Schwefeldioxyd und einigen der beschriebenen Medien Komplexe gebildet werden.
Process for the production of steroids The subject of patent no. 347 <B> 521 </B> is a process for the production of JO (II) steroids by dehydrating 11-oxysteroids. It has now been found that steroids of the androstane and pregnane series, which carry hydroxyl groups in other positions, can also be dehydrated in the same way with formation of the corresponding unsaturated compounds.
It is thus possible to produce a large number of unsaturated steroids which in themselves have remarkable physiological effectiveness or are valuable intermediates for the production of other physiologically active steroid compounds. Due to their effectiveness, the i) (11) steroids, such as z.
B. 9 (11) -Dehydro-deoxycorticosterone acetate, 9 (11) -Dehydro-progesterone, 9 (11) -Dehydro-testosterone acetate, 9 (11) -Dehydro-estradiol-acetate, 9 (11) -Dehydro-17a -oxy-progesterone, 9 (11) - dehydro -17a - oxy - deoxycorticosterone acetate <B> ETC. </B> These compounds can be prepared from the corresponding 9a oxysteroids by the process of the present invention.
The d9 (II) steroids can also be converted into valuable intermediate products for the manufacture of physiologically effective steroids. So z. B. 9 (11) -Dehydro-pregnenolone can be oxidized to 9 (11) -Dehydro-progesterone by oxidation according to Opperiauer. The d9 (II) steroids are also suitable for producing the 9a-chlorine and 9a-fluoro-11f-oxy analogs of the known physiologically active adrenal cortex and sex hormones, in particular those which also have the d4-3,20 diketo function contain.
(See Fried and Sabo, J. Am. Chem. Soc. 75, 2273 [1953].) If J9 (11) -steroids are hydrogenated, the corresponding 9 (11) -hydrocompounds are obtained. These can be converted into known physiologically active steroids and / or intermediates by known methods.
Of particular importance, either because of their physiological effectiveness per se or as intermediates for the production of physiologically active steroids, are 4,9 (11) -androstadien-3,17-dione, 6-methyl-4-pregnen-3,11, 20-trione, 4,14-pregnadiene and 4,16-androstadien-3-one.
Up to now, halogenamides and halogenimides have been used in steroid chemistry as oxidizing or halogenating agents or, in certain cases, for both purposes at the same time. The use of these substances in a dehydration reaction was not known.
The reaction of hydrocortisone acetate with an N-haloimide under exclusion of water and in the presence of a base such as pyridine to produce an 11-keto group is the subject of US Pat. No. 2,751,402. The product of this reaction is cortisone acetate; . H. it is an oxidation reaction. In the earlier process, the product resulting from the oxidation with N-halo-amides or N-haloimides was treated with an aqueous reducing agent, e.g. B.
added aqueous sodium sulfite to destroy the excess N-halogen compound. In contrast, according to the present invention, anhydrous sulfur dioxide is used to effect the dehydration and the decomposition of the excess N-halogen compound.
According to the process of the present invention, a nucleus-hydroxylated steroid of the androstane or pregnane series is brought together under anhydrous conditions in a basic organic medium that does not participate in the reaction with an N haloamide or N-haloimide and anhydrous sulfur dioxide and the dehydrated one formed Separated steroid from the reaction mixture. The presence of water would lead to the regeneration of the hydroxyl-containing starting material.
If the addition of sulfur dioxide is delayed for an inadmissibly long time, the intermediate product formed with the hypohalite stabilizes with the formation of a keto group, so that instead of dehydration, an oxidation reaction is obtained.
To achieve a high yield, the sulfur dioxide must therefore be added within about 2 hours after the steroid has been mixed with the N-halo-amide or N-halo-imide, although this time may vary depending on the particular starting steroid used.
The steroid hypohalite is usually obtained by reacting the oxysteroids with an N-halo-amide or N-halo-imide, in which the halogen has an atomic weight of 35-127 and is preferably chlorine or bromine, or with another compound which is capable to convert a hydroxy compound into a hypohalite.
Examples of such compounds are N-chloroacetamide, N-bromoacetamide, N-chlorosuccinimide, N-bromosuccinimide, N-iodosuccinimide, N-bromophthalimide, 3-bromo-5,5-dimethylhydantoin, 1,3-dibromo-5 ,
5-dimethylhydantoin. Usually more than one molar equivalent is used based on the oxysteroid. If the molecule contains other groups that can be oxidized or dehydrated, correspondingly larger amounts of the N-halogen compound should be used in order to achieve optimal yields.
The reaction of the oxysteroid with the N-halogen compound takes place in the presence of a basic organic compound which is practically not attacked by the reagents. Examples of such bases are the tertiary amines whose amine nitrogen is a member of an aromatic ring, e.g.
B. the pyridines such as pyridine and alkyl pyridines such as picoline, lutidine, collidine, conyrin, parvulin, etc., and the lower fatty acid amides, especially formamide, methyl and dimethylformamide.
Complexes of some of the above compounds with the sulfur dioxide are likely to form, but this does not adversely affect their utility as a medium for the primary reaction or as a means of effectively removing hydrogen halide produced in the course of the reaction.
The base is preferably in molar excess, based on the oxysteroid, for. B. used in ten times the molar amount and is preferably the only solvent for the reaction.
The reaction of the oxysteroid with the N-halogen compound takes place under anhydrous conditions. The designation anhydrous means that the reaction mixture, based on the steroid, should contain less than one molar equivalent of water. If the mixture contains one molar equivalent or more of water, virtually no dehydrated steroid will be formed.
The less water the mixture contains, the greater the yield of dehydrated steroid. Preferably the mixture contains less than 0.1 molar equivalent of water.
The reaction between N-haloamide or N-haloimide and the oxysteroid can preferably be carried out between -40 ° C. and +70 ° C., the lower limit being due to the solubility of the reactants and an appropriate reaction time, the upper limit being due to the normal reaction times side reactions occurring with N-halogen compounds at higher temperatures are determined. The usual room temperature is preferred for reasons of convenience and always delivers high yields of the desired product.
The reaction time is mainly determined by the speed with which the hypohalite obtained in this way is rearranged to form the keto group. If the temperature is increased above room temperature, the reaction time is shortened accordingly, while lower temperatures lengthen the same. In most cases, the reaction between the hydroxyl group and the halogen compound is completed after half an hour at room temperature, and at least in the case of hydrocortisone acetate in less than 5 minutes.
The anhydrous sulfur dioxide can be gaseous or liquid or in the form of a material such. B. an alkali metal hyposulfite forms sulfur dioxide in situ.
The reaction between the hypohalite group and the sulfur dioxide evidently forms a halosulfonate which, under the anhydrous conditions and in the presence of a base, is converted into the salt of dehydrated steroid, hydrogen halide, base and sulfur trioxide. The range of the reaction temperature is practically the same as in the preparation of the hypohalite. In most cases, the reaction appears to be instantaneous and the reaction product can be isolated almost immediately.
Longer reaction times seem to have little or no effect on the yield.
As stated earlier, the process of the present invention can be used for the dehydration of any nuclear hydroxylated steroids of the androstane and pregnane series. Other substituents bound in various positions on the steroid nucleus appear to have no effect on the basic operation of the method.
It is therefore possible according to the method to obtain steroid compounds from suitable starting materials, which are divided into 1-2-, 2-3-, 3-4-, 5-6-, 6-7-, 7-8-, 8-9 -, 8-14-, 9-11-, 11-12-, 14-15-, 15-16- and 16-17- positions are unsaturated.
<I> Example 1 </I> 35 mg (0.25 millimoles) of N-bromoacetamide in pyridine were treated with sulfur dioxide until a sample with acidified potassium iodide starch paper turned out negative; the mixture was then treated at 25 C with a solution of 20 mg (0.06 millimoles) of 3u-oxy-pregnane-11,20-dione in pyridine distilled over barium oxide.
The resulting mixture was stirred for about 45 minutes, cooled to 12 C, and water was added dropwise. After cooling for several hours to 0-5 ° C., the precipitate was filtered off, washed with water and dried. A crude yield of 24 mg of 3-pregnene-11,20-dione, which was identified on the basis of its spectrum, was obtained.
<I> Example 2 </I> To a solution of 7.5 kg of 5a-oxy-6-methyl-pregnane-3,11,20-trione in 71.5 l of freshly distilled and dried pyridine was added 6.950 at room temperature kg of N-bromoacetamide. The mixture was stirred at 25 C for half an hour and cooled to 0 C, then kept at 0-10 C while 19.277 kg of sulfur dioxide gas were added. The mixture was then warmed to 25 ° C., stirred for 110 hours and cooled again to 0-5 ° C.
200 liters of 10% strength sulfuric acid were then added, followed by a further 36 liters of water, and the mixture was stirred for a further 3 hours. The solid product formed was filtered off, washed thoroughly with water and dried in vacuo at 60 ° C. to constant weight. 6.484 kg (90.7% of theory) 6ss-methyl-4-pregnen-3,11,20-trione were obtained. Melting point 162.5 to 165 ° C; [a] D: +207 C (chloroform); Emat = 16.067.
<I> Example 3 </I> A solution of 10.3 mg (0.03 millimoles) 6ss oxy-11-keto-progesterone in pyridine freshly distilled over barium oxide was brought to 25 ° C. Then a solution of 8.6 mg (0.06 millimoles) of N-bromoacetamide in pyridine was added and the mixture was stirred at 25 ° C. for 30 minutes. After cooling to 12 "" C, dry sulfur dioxide was introduced until the reaction with acidified potassium iodide starch paper was negative.
Water was then added dropwise to the mixture at a temperature of below 30 ° C. with stirring. The resulting suspension was cooled to 0-5 ° C. and kept at this temperature for a few hours. The precipitate was filtered off, washed with water and dried. 4,6-Pregnadiene-3,11,20-trione was obtained in a crude yield of 9.5 mg.
According to the paper chromatogram, the product contained no 6ss-oxy-11-keto-progesterone. The IR spectrum showed absorption at 1698 cm- '(non-conjugated keto group), 1650 cm-' (conjugated keto group) and 1620, 1586 cm- '(44th G C = C), which is consistent with the assigned structure.
<I> Example 4 </I> The procedure was exactly as in Example 1, but using 6ss-oxy-11-keto-progesterone as the starting material, and 4,6-pregnadiene-3,11,20-trione was obtained. The infrared analysis was consistent with the structure and was identical to that given in Example 3 for the same product. <I> Example 5 </I> The procedure was as in Example 3, but with 10.8 mg of 6ss-oxy-5ss-pregnane-3,11,20-trione as starting material, and a crude yield of 8.1 mg of 6 was obtained -Pregnen-3.11,
20-trion. The IR spectrum showed traces of residual hydroxyl and strong bands for unconjugated keto groups.
<I> Example 6 </I> The procedure was as in Example 1 using 10.8 mg of 6ss-oxy-5ss-pregnan-3,11,20-trione and obtained 11.0 mg of crude 6-Pregiien-3,11,20-trione. As in the previous example, the IR spectrum showed traces of residual hydroxyl groups and non-conjugated keto groups.
<I> Example 7 </I> The procedure was as in Example 3, but using 9.7 mg of 7ss-oxy-4-androstened-3,17-dione as starting material. 6.8 mg of Rohpro product were obtained, which mainly consisted of 4,6-androstadiene-3,17-dione. The IR spectrum confirmed the structure and showed absorption at 1730 cm- '(17-keto group), 1657 cm-' (conjugated keto group), 1615, 1580 cm- '(conjugated C = C). UV absorption at 283 m, u (44, s C = C).
<I> Example 8 </I> The procedure was as in Example 1, but with 7ss-oxy-4-androstene-3,17-dione as the starting material, and 4.3 mg of crude 4,6-androstadiene-3,17 were obtained -dion. Infrared analysis confirmed the structure and identity with the product of Example 7.
<I> Example 9 </I> The procedure was as in Example 3, but using 9.9 mg of 9a-oxy-progesterone as starting material. 3.0 mg of crude 9 (I1) -dehydroprogesterone were obtained. Paper chromatographic and infrared analysis confirmed the structure. The following absorptions were measured: 1695 cm- '(20-keto group); 1668 cm '(conjugated keto group) 1611 cm-1 (44 C = C).
<I> Example 10 </I> The procedure was as in Example 1, but using 19.8 mg of 9a-oxy-progesterone as starting material, and a crude yield of 14.9 mg of 9 (11) -dehydro-progesterone was obtained. Paper chromatogram and infrared analysis agreed with the structure; the product was identical to the product according to example 9.
<I> Example 11 </I> If you work exactly as in Example 3, but using 9.9 mg of 14a-oxy-progesterone as starting material, you get 8.6 mg of crude 14 (15) -dehydro-progesterone, the structure of which was confirmed by IR analysis. The product showed no residual hydroxyl and the following absorption bands: 1695 cm- '(20-keto group) 1668 cm-1 (conjugated keto group), 1611 cm-1 (d4 C = C).
<I> Example 12 </I> If you work exactly as in Example 1, but using 9.9 mg of 14a-oxy-progesterone as starting material, 7.5 mg of crude 14 (15) -dehydro-progesterone are obtained. The infrared analysis is consistent with the assigned structure. The IR spectrum is identical to that of the product from Example 11.
<I> Example 13 </I> If you work as in Example 3, but using 19.6 mg of 14a-oxy-l-dehydro-progesterone as starting material, 15.5 mg of crude 1.14 are obtained (16) -Dehydro-progesterone. The paper chromatographic analysis shows no starting material in the end product.
The infrared analysis confirms the structure with the following absorption maxima: 1690, 1688 cm- '(non-conjugated ketone group), 1652 cm-1 (conjugated ketone group), <B> 1618, </B> 1597 cm-1 (d1,14_ C = C) .
<I> Example 14 </I> If you work exactly as in Example 1, but using 9.8 mg of 14a-oxy-l-dehydro-progesterone, you get 8.1 mg of raw 1.14 (15) - Dehydro-progesterone. Infrared analysis confirmed the structure of the compound identical to the product of Example 13.
<I> Example 15 </I> If one works as in Example 3, but using <I> 9.9 mg </I> 15a-oxy-progesterone as starting material, 7.3 mg of product are obtained. The paper chromatographic examination shows the presence of a trace of unchanged 15a-oxy-progesterone. The infrared analysis confirms the structure for 14 (15) -dehydro-progesterone.
<I> Example 16 </I> If you work exactly as in Example 1, but using 9.9 mg of 15a-oxy-progesterone as starting material, 9.8 mg of product are obtained. The paper chromatographic examination shows the presence of small amounts of a pyridine-sulfur dioxide complex; the infrared analysis confirms the structure of the 14 (15) -dehydro-progesterone.
<I> Example 17 </I> If you work as in Example 1, but using 15a-oxy-androstene-3,17-dione as the starting material, you get 10.8 mg of crude 4.14 (15) - Androstadiene-3,17-dione. Infrared analysis shows a trace of residual hydroxyl and otherwise confirms the assigned structure. Absorption: 1730 cm- '(non-conjugated keto group), 1660 cm-' (44-3 keto group), 1615 cm-1 (44 C = C).
<I> Example 18 </I> If one works as in Example 3, but using 8.7 mg testosterone as starting material, 7.2 mg of 4,16-androstadien-3-one are obtained. The infrared analysis shows no residual hydroxyl and confirms the structure with the following absorption bands: 1670 cm- '(J - 3-keto group), 1615 cm-' (44 C = C).
<I> Example 19 </I> If you work exactly as in Example 1, but using 8.7 mg of testosterone as the starting material, 6.4 mg of product are obtained. The product is identical to the compound produced according to Example 18.
It can be seen from the above examples that the present invention can serve for the dehydration of any nuclear hydroxylated steroids of the androstane and pregnane series.
The constitution of the remaining molecule of the parent steroid has little impact on the feasibility of the method. It is necessary that the reaction take place under anhydrous conditions and in the presence of a basic non-reacting organic medium; however, changes to the reaction times, temperature and concentration of the reaction participants are permitted without changing the normal course of the procedure.
As in Examples 1 and 3 and the. was subsequently shown, it does not matter whether the starting steroid and N-halogen compound are first mixed and then the sulfur dioxide is added, or whether the N-halogen compound is first brought together with the sulfur dioxide and this mixture is added to the starting steroid.
The term <basic non-reacting organic medium describes a medium that does not react with the parent steroids or with the N-haloamides. However, the medium reacts with the hydrogen halide to the extent that it is produced during dehydration and thus prevents halogenation of the steroid nucleus. It is also likely that complexes are formed between sulfur dioxide and some of the media described.
