-reste, oder 3-Oxa-heptyl-(6)-reste, und als Halo genalkylreste Halogenniederalkylreste, wie Chlor- äthylreste, zu nennen. Als Cycloalkylreste bzw. Cycloalkylalkylreste kommen z.
B. Cyclopentyl-, oder Cyclohexylreste, Cyclopentyl-oder Cyclohexyl-methyl-, -äthyl- oder -propylreste in Frage.
Aralkylreste sind speziell Phenalalkyl-, wie 1- oder 2-Phenyläthyl- oder Phenylmethylreste, in denen die aromatischen Kerne Substituenten tragen können, wie niedere Alkylreste oder freie oder substituierte Oxy-, Amino- oder Mercaptogruppen, Halogenatome, Trifluor- methyl- oder Nitrogruppen. Heterocyclylalkyl-reste sind z.
B. Pyridylmethyl- Thenyl- oder Furfurylreste, die auch substituiert sein können, z. B. wie für die Aralkylreste angegeben. In den genannten substituierten Oxy-, Mercapto- oder Aminogruppen sind die Substituenten insbeson dere solche der oben genannten Art, vor allem niedere Alkylreste, so dass es sich z.
B. um Methoxy-, Äthoxy-, Propoxy- oder Butoxygruppen, entspre chende Alkylmercaptogruppen, Alkylendioxy-, wie Methylendioxygruppen, Mono- oder Dialkylamino- gruppen, wie Mono- oder Dimethyl-, -äthyl-, -propyl-, -butyl- oder pentylaminogruppen handelt. Als Halogenatome sind vor allem Fluor, Chlor oder Brom zu nennen.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. Insbesondere sind sie coronarerweiternd, wirksam. Die neuen Verbin dungen können somit als Heilmittel, vor allem bei Durchblutungsstörungen des Herzmuskels, aber auch als Zwischenprodukte zur Herstellung solcher dienen.

Vor allem betrifft die Erfindung die Herstellung des ausgezeichnete coronarerweitemde Wirkung auf weisenden 1-Isopropyl-4-mereapto-6-methyl-pyr- azolo[3,4-d]pyrimidins, sowie des ebenfalls ausge zeichnete coronarerweiternde Wirkung aufweisenden 1-Isopropyl-4-mercapto- 6-benzyl-pyrazolo[3,4 - d]py- rimidins und ihrer Salze.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man ein 1-Ri 3-R3 6-Rs 4-hydroxy-pyrazolo[3, 4-d]pyrimidin, worin R1, R3 und R, die oben an gegebenen Bedeutungen haben, mit einem Sulfurie- rungsmittel behandelt. Als Sulfurierungsmittel kommt vor allem Phosphorpentasulfid in Frage.
Diese Umsetzung kann in üblicher Weise, vor zugsweise bei erhöhter Temperatur, in Abwesenheit oder Anwesenheit eines Lösungsmittels, im geschlos senen oder offenen Gefäss, durchgeführt werden.
Je nach der Arbeitsweise und den Ausgangs stoffen erhält man die neuen Verbindungen in freier Form oder in Form ihrer Salze. Erhaltene freie Ver bindungen können in ihre Metallsalze übergeführt werden, z. B. durch Lösen in Alkalilaugen. Erhal tene Salze lassen sich in die freien Verbindungen überführen. Die neuen pharmakologisch wertvollen Verbin dungen, ihre Salze oder entsprechende Gemische können z.
B. in Form pharmazeutischer Präparate Verwendung finden, welche die genannten Verbin dungen in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial enthalten.
Die für die Herstellung der 4-Mercapto-Verbin- dungen verwendeten 4-Hydroxy-Verbindungen kön nen erhalten werden, wenn man 2-R1-3-Amino-py- razol-4-.carbonsäure-alkylester mit Nitrilen der For mel RCCN kondensiert.
Als Ausgangsstoffe werden gemäss der vorliegen den Erfindung vorzugsweise diejenigen verwendet, die zu den eingangs als besonders wertvoll geschil derten Endstoffen führen.
In den nachfolgenden Beispielen sind die Tem peraturen in Celsiusgraden angegeben. <I>Beispiel 1</I> 1.8,2 g 1-Isopropyl-4-hydroxy-6-methyl-pyrazolo- [3,4-d]pyrimidin werden in 200 cm?, Pyridin gelöst. 30 g Phosphorpentasulfid werden zu der Lösung ge geben und es wird während 8 Stunden zum Sieden erhitzt. Man giesst dann die Reaktionslösung auf 3 Liter Eiswasser, lässt über Nacht stehen und nutscht am Morgen von dem gelb ausgefallenen Niederschlag ab.
Durch Umkristallisation aus Äthanol erhält man 1-Isopropyl-4-mercapto-6-methyl-pyrazolo[3,4-d]-py- rimidin der Formel

Das Ausgangsmaterial wird z. B. wie folgt er halten: Zu einer Lösung von 9,9 g 2-Isopropyl-3-amino- 4-.carbäthaxy-pyrazol in 50 cma Acetonitril gibt man 2,3g Natrium unter Eiskühlung. Die Temperatur darf während der Reaktion nicht über 30 steigen. Sobald die exotherme Reaktion beendet ist, erhitzt man während 4 Stunden auf 90-95 . Das Reak tionsgemisch wird dann abgekühlt und mit 100 cm3 Äthanol versetzt. Man dampft im Vakuum das Äthanol ab, gibt zum Rückstand 150 em3 2-n.
Natronlauge und -extrahiert das überschüssige Ace- tonitril mit Chloroform. Die wässerige Phase wird durch Zugabe von 5-n. Salzsäure auf pH 3-4 ge stellt, wobei nach längerem Stehen ein fester Nieder schlag ausfällt, den man abfiltriert und aus Äthanol umkristallisiert.
Man erhält das 1-Isopropyl-4 hy- droxy-6-methyl-pyrazolo[3,4-d]pyrimidin der Formel

Die wäss- rige Phase wird durch Zugabe von 5-n. Salzsäure auf pH 5-6 gestellt, wobei ein festes Produkt aus fällt. Letzteres wird mehrmals aus Äthanol umkri- stallisiert. 1,6-Di-isopropyl-4-hydroxy-pyrazolo[3,4-d] pyrimidin wird so in weissen Kristallen vom F. 175 bis 177 erhalten.
