Ces composés possèdent des propriétés antibac tériennes et parasiticides. Ils conviennent particuliè- rement bien dans la lutte contre le plasmodium de la malaria, les helminthes intestinaux, et Endamoeba histolytica, agent de l'amibiase.

On a intérêt à effectuer cette condensation avec des quantités pratiquement équivalentes des réactifs en présence d'un solvant qui peut être de l'eau, un phénol, un alcool aliphatique tel que l'éthanol, l'iso- propanol, le pentanol et l'octanol,
une cétone ali phatique telle que la méthyl-éthylcétone, la diisobu- tylcétone, un éther aliphatique tel que l'éther dibuty- lique, un éther monoalcoylique de l'éthylène glycol, un éther cyclique tel que le dioxane, un hydrocar bure tel quel le benzène, le toluène et le xylène. Le solvant préféré est le phénol. La température de réaction est avantageusement supérieure à 750 C et de préférence de 95 à 145o C.
On peut isoler le produit sous forme de base libre ou le transformer en sel d'addition d'acide non toxique tel que le chlorhydrate, bromhydrate, sulfate, phosphate, oxa- late, sulfamate, sulfonate, acétate, lactate, tartrate, gluconate, citrate, pénicillinate, arsanilate et arsonate. Parmi les sels préférés, on peut citer les suivants 5,5'-méthylène-di-salicylate ; 4,4'-méthylène-bis-(3- hydroxy-2-naphtoate) ;
méthylène-di-o-crésotinate ; 8- hydroxy-7-iodo-5-quinoléine-sulfonate ; N-acétyl-4- hydroxy-m-arsanilate ; N-glycolyl-p-arsanilate ; N,N'- éthylène-bis-(p-arsanilate) ; p-uréidobenzènearsonate ; benzylpénicillinate ; phénoxyméthylpénicillinate et fumagillinate. Certains des sels précités possèdent une action amibicide, et dans certains cas antibactérienne, renforcée.
<I>Exemple 1</I> Un mélange de 12,8g de 10-oxyde de 6,9-di- chloro-2-méthoxy-acridine, 11,7g de dichlorhydrate de 2-diéthylaminométhyl-4-amino-phénol et 25g de phénol est porté à une température de 130 à 140L, C pendant une heure et demie. On refroidit le mélange réactionnel, le verse dans une solution de 5 ml d'acide chlorhydrique concentré dans 200 ml d'acétone, puis le laisse reposer à température ambiante pendant dix-huit heures. On dilue alors le mélange avec de l'éther anhydre et le refroidit.
On recueille le produit solide par filtration, le dissout dans de l'eau, puis on filtre la solution aqueuse et ajoute au filtrat une solution d'hydroxyde d'ammonium en excès. Le pro duit séparé est récupéré par filtration et séché ; point de fusion de<B>1680C</B> avec décomposition, après re- cristallisation au sein d'un mélange chaud éthanol- eau. Ce produit est le 10'-oxyde de 4-(6'-chloro-2'- méthoxy-9'-acridinylamino)-a-diéthyl-amino-o-crésol.
De même, on peut préparer les composés sui vants à partir du 10-oxyde de 6,9-dichloro-2-mé- thoxy-acridine et d'une quantité équivalente de l'amino-phénol approprié 10'-oxyde de 4-(6'-chloro-2'-méthoxy-9'-acridinyl- amino)-a-dioctylamino-o-crésol ; 10'-oxyde de 4-(6'-chloro-2'-méthoxy-9'-acridinyl- amino)-a-pyrrolidino-o-créso1 ;
10'-oxyde de 4-(6'-chloro-2'-méthoxy-9'-acridinyl- amino)-a-pipéridino-o-crésol ; 10'-oxyde de 4-(6'-chloro-2'-méthoxy-9'-acridinyl- amino)-a-diméthylamino-o-crésol ; 10'-oxyde de 4-(6'-chloro-2'-méthoxy-9'-acridinyl- amino)-a-isobutylamino-o-crésol ; 10'-oxyde de 4-(6'-chloro-2'-méthoxy-9'-acridinyl- amino)-a-(4'-méthyl-pipérazino)-o-crésol ;
10'-oxyde de 2,2'-[5-(6'-chloro-2'-méthoxy-9'-acridi- nyl-amino)-2 hydroxy-benzylamino]-diéthanol ; 10'-oxyde de 4-(6'-chloro-2'-méthoxy-9'-acridinyl- amino)-a-morpholino-o-crésol. Le 10-oxyde de 6,9-dichloro-2-méthoxy-acridine de départ peut être préparé de la manière suivante une solution de 278g de 6,
9-dichloro-2-méthoxy- acridine dans 5 litres de chloroforme est traitée à température ambiante par une solution froide de 160g d'acide perbenzoïque dans 3 litres de chloro forme. Après l'avoir laissée reposer à température ambiante pendant trente-six heures, on ajoute une feuille de platine, on concentre le mélange sous vide jusqu'à un volume de 1 litre et demi et le refroidit dans un bain de glace. Le précipité formé est séparé par filtration et lavé à fond à l'éther ; le point de fusion est de 226 à 228 C.
Les sels d'addition peuvent être obtenus de la manière suivante Une solution chaude de 9,0 g de 10'-oxyde de 4- (6'-chloro-2'-méthoxy-9'-acridinyl-amino) - u - diéthyl- amino-o-crésol dans 25 ml d'éthanol est ajoutée à une solution chaude de 12,6 g d'acide N-glycolyl-p- arsanilique dans 150 ml d'éthanol à 80%. On chauffe la solution au bain-marie pendant dix minutes, on la filtre puis on chasse le solvant sous vide.
Par tritura tion du résidu avec plusieurs portions d'un mélange éther-acétone, on sépare le bis-(N-glycolyl-p-arsani- late du 10'-oxyde de 4-(6'-chloro-2'-méthoxy-9'-acri- dinyl-amino)-a-diéthylamino-o-crésol.
D'autres sels du 10'-oxyde de 4-(6'-chloro-2'- méthoxy-9'-acridinyl-amino) - u -diéthylamino-o-crésol peuvent être préparés à partir de la base libre et d'un ou deux équivalents des acides suivants( en présence des solvants suivants) fumagillinique (acétone) acide sulfurique (éther) acide bromhydrique (éthanol) acide acétique (acétone) acide phosphorique (éthanol Des solutions froides, filtrées, de 22,7 g de di- chlorhydrate sesquihydraté de 10'-oxyde de 4-(3',
6'- dichloro-9'- acridiny 1-amino) - a - pyrrolidino - o -crésol dans 75 ml d'eau et de 31 g de benzylpénicilline po tassique dans 200 ml d'eau sont mélangées dans un bain de glace ; on refroidit l'huile qui se sépare et provoque sa solidification en grattant les parois du récipient avec une baguette. On purifie le produit ainsi obtenu par dissolution dans de l'éthanol et re- précipitation avec de l'éther.
Ce produit est le bis- benzylpénicillinate de 10'-oxyde de 4-(3',6'-dichloro- 9'-acridinyl-amino)-a-pyrrolidino-o-crésol.
De même, on peut préparer des sels de 10'-oxyde de 4-(3',6'-dichloro-9'-acridinyl-amino)-a-pyrrolidino- o-crésol à partir du mono- ou du dichlorhydrate de la base et d'un ou de deux équivalents, respective ment, des sels suivants Sel de sodium d'acide 8-hydroxy-7-iodo-5-quinoléine- sulfonique ; sel de potassium de phénoxyméthylpénicilline ; sel monosodique d'acide n-uréidobenzènearsonique ;
sel monosodique d'acide N-acétyl-4-hydroxy-m-arsa- nilique ; sel disodique d'acide N,N'-éthylène-bis-(p-arsanili- que). <I>Exemple 2</I> Un mélange de 14,6 g de dichlorhydrate de 4- amino-2-pyrrolidinométhyl-phénol et de 25 g de phé nol est chauffé sous vide au bain-marie pendant une heure, on ajoute 14,9 g de 10-oxyde de 3,6,
9-tri- chloro-acridine et on chauffe le mélange sous agita tion entre 125 à 135o C pendant deux heures. On refroidit le mélange réactionnel puis le verse dans un mélange de 5 ml d'acide chlorhydrique con centré dans 300 ml d'acétone. Après une autre dilu tion avec de l'acétone jusqu'à un litre, on recueille la matière précipitée rouge par filtration, on la lave à l'acétone. On la redissout dans de l'eau et on alca linise la solution aqueuse par addition d'une solution d'hydroxyde d'ammonium. On extrait le mélange au chloroforme, lave les extraits à l'eau, les sèche sur du carbonate de potassium anhydre puis on les filtre.
On évapore le filtrat à sec sous vide, on reprend le résidu dans de l'éthanol bouillant, on décolore la solution, l'évapore à faible volume et lui ajoute un excès d'acide chlorhydrique concentré. On recueille le précipité et le cristallise au sein d'un mélange mé- thanol-acétone, point de fusion 2301, C (déc.). Le produit cristallisé est le dichlorhydrate de 10'-oxyde de 4-(3',6'-dichloro-9'-acridinyl-amino)-a-pyrrolidino- o-crésol.
De même, les composés suivants peuvent être préparés à partir du 10-oxyde de 3,6,9-trichloro-acri- dine et d'une quantité correspondante de l'amino- phénol approprié 10'-oxyde de 4-(3',6'-dichloro-9'-acridinyl-amino)-a- diéthylamino-o-crésol ; 10'-oxyde de 4-(3',6'-dichloro-9'-acridinyl-amino)-a- pipéridino-o-crésol ;
10'-oxyde de 4-(3',6'-dichloro-9'-acridinyl-amino)-a- décylamino-o-crésol ; 10'-oxyde de a-[5-(3',6'-dichloro-9'-acridinyl-amino)- 2-hydroxy-benzylamino]-éthanol ; 10'-oxyde de 4-(3',6'-dichloro-9'-acridinyl-amino)-a- pipérazino-o-crésol ; 10'-oxyde de a-fc-[5-(3',6'-dichloro-9'-acridinyl- amino)-2-hydroxy-benzyl]-éthylaminoÎ-éthanol ;
10'-oxyde de 4-(3',6'-dichloro-9'-acridinyl-amino)-a- cyclohexylimino-o-crésol.
Le 10-oxyde de 3,6,9-trichloro-acridine, point de fusion 236 à 238- C, utilisé comme matière première dans le procédé ci-dessus, peut être préparé à partir de la 3,6,9-trichloro-acridine et de l'acide perbenzoï- que selon le procédé indiqué à l'exemple 1 ci-dessus, pour la préparation du 10-oxyde de 6,9-dichloro-2- méthoxy-acridine.
Le dichlorhydrate de 4-amino-2-pyrrolidinomé- thyl-phénol de départ peut être préparé de la manière suivante<B>:</B> 71 g de pyrrolidine, 30 g de paraformal- déhyde et 100 ml d'éthanol sont chauffés au bain- marie pendant trente minutes. On ajoute la solution résultante à une solution de 151 g de 4-acétylamino- phénol dans 500 ml d'éthanol et on chauffe le mé lange à reflux pendant une heure et quart.
Après traitement au noir, le solvant est éliminé sous vide et on chauffe le résidu au bain-marie avec 660 ml d'acide chlorhydrique 1 : 1, pendant trois heures. On évapore le mélange sous vide jusqu'à obtention d'une huile, on ajoute plusieurs portions d'éthanol et de benzène et les élimine sous vide, on dissout le résidu dans de l'éthanol et le reprécipite par de l'éther an hydre. Après de nombreuses triturations avec de l'éther et/ou de l'acétone, le résidu huileux cristal lise ; point de fusion 210 à 2150 C (avec décomposi tion).
<I>Exemple 3</I> Un mélange de 16,7 g de dichlorhydrate de 2-n- décylaminométhyl-4-amino-phénol et de 30 g de phé nol est chauffé au bain-marie sous vide pendant une heure, puis on ajoute 14 g de 10-oxyde de 6,9-di- chloro-2-méthoxy-acridine et agite le mélange entre 130 et 1400 C pendant deux heures. On refroidit le mélange réactionnel, le verse dans 1 litre de solution normale d'hydroxyde de sodium, et l'extrait au chlo roforme. Les extraits chloroformés réunis sont lavés à fond à l'eau, séchés sur du carbonate de potassium anhydre, filtrés et évaporés sous vide.
On reprend le résidu dans de l'éthanol et ajoute un excès de gaz chlorhydrique alcoolique. On ajoute de l'acétone et de l'éther anhydre, recueille le précipité sur un filtre, le lave à l'éther et le sèche sous-vide ; point de fusion 175,1 C avec décomposition. Le produit est le dichlorhydrate de 10'-oxyde de 4-(6'-chloro-2'- méthoxy-9'-acridinyl-amino) - a #- décylamino-o-crésol.
Le dichlorhydrate de 2-n-décylaminométhyl-4- amino-phénol, de point de fusion de 100 à 1100 C, utilisé comme matière première pour la préparation ci-dessus, peut être obtenu, en faisant réagir 51,8 g de n-décylamine, 9 g de paraformaldéhyde et 45,3 g de 4-acétylamino-phénol selon le procédé indiqué ci- dessus à l'exemple 2 pour la préparation du dichlor- hydrate de 2-pyrrolidinométhyl-4-amino-phénol. <I>Exemple 4</I> Un mélange de 25 g de phénol, 7,
0 g de dichlor- hydrate de 4-amino-a-diéthylamino-o-crésol et 6,8 g de 10-oxyde de 6,9-dichloro-acridine hémihydrate est agité et chauffé sur un bain de métal entre 125 et 135 C pendant deux heures. Après refroidisse ment, on dilue le mélange réactionnel avec plusieurs volumes d'éther et d'acétone. On recueille le produit brut par filtration, le dissout dans de l'eau tiède, alca linise la solution avec de l'hydroxyde d'ammonium, l'extrait au chloroforme, puis on lave les extraits chloroformés à fond à l'eau et les sèche sur du car bonate de potassium anhydre.
On évapore le chloro forme sous vide, puis on traite le résidu goudronneux par un excès de gaz chlorhydrique alcoolique. Plu sieurs cristallisations de ce résidu au sein d'un mé lange éthanol-acétone-éther donnent le dichlorhy- drate de 10-oxyde de 4-(6'-chloro-9'-acridinyl-ami- no)-a-diéthylamino-o-crésol ; point de fusion entre 135 et 140o C avec décomposition.
Le 10-oxyde hémihydrate de 6,9-dichloro-acri- dine, point de fusion 164 à 166 C, pris comme ma tière première, peut être préparé à partir de la. 6,9- dichloro-acridine et de l'acide perbenzoïque selon le procédé indiqué à l'exemple 1 ci-dessus pour la pré paration du 10-oxyde de 6,9-dichloro-2-méthoxy- acridine. <I>Exemple 5</I> Un mélange de 4,
8 g de dichlorhydrate de 2-di- éthylaminométhyl-4-amino-phénol, de 20g de phé nol, de 5 g de 10-oxyde de 6,9-dichloro-3-méthyl- acridine, est chauffé et agité entre 120 et 125o C pendant deux heures. Après refroidissement, on verse le mélange dans une solution de 5 ml d'acide chlor hydrique concentré dans 500 ml d'acétone. On dilue le mélange avec de l'éther, recueille le produit brut par filtration, et le lave à l'éther.
On dissout dans de l'eau le sel ainsi obtenu, filtre la solution et précipite la base par l'addition d'un excès d'une solution d'hy droxyde d'ammonium. On recueille le précipité par filtration et le sèche. Une recristallisation au sein d'un mélange chaud éthanol-eau donne le 10'-oxyde de 4-(6'-chloro-3'-méthyl-9'-acridinyl-amino)-u-di- éthylamino-o-crésol.
Le 10-oxyde de 6,9-dichloro-3-méthyl-acridine, point de fusion 172 à 1740 C, utilisé comme matière première, peut être préparé à partir de la 6,9-di- chloro-3-méthyl-acridine et de l'acide perbenzoïque selon le procédé indiqué à l'exemple 1 ci-dessus pour la préparation du 10-oxyde de 6,9-dichloro-2-mé- thoxy-acridine. La 6,9-dichloro-3-méthyl-acridine peut elle-même être préparée de la manière suivante 191 g d'acide 2,4-dichlorobenzoïque,
134 g de m- toluidine, 138 g de carbonate de potassium anhydre, 5 g de poudre de cuivre et 750 ml d'alcool n-amyli- que sec sont chauffés au reflux sous agitation pen dant cinq heures. On refroidit le mélange, lui ajoute 70 g d'hydroxyde de potassium dans 100 ml d'eau, et l'entraîne à la vapeur d'eau. Le résidu aqueux est filtré à chaud, légèrement acidifié avec de l'acide chlorhydrique concentré et refroidi. On recueille par filtration l'acide précipité, le lave à l'eau, puis à l'éther de pétrole (point d'ébullition 30 à 600 C).
On fait bouillir le précipité avec 300 ml d'éthanol, le refroidit, le recueille sur un filtre et le lave à l'éther de pétrole (point d'ébullition de 30 à 60o C).
Une recristallisation au sein d'un mélange monochloro- benzène-ligroïne (point d'ébullition 60 à 800 C) après traitement au noir, donne l'acide 4-chloro-N-(m-to- luyl)-anthranilique, de point de fusion 197 à 1990 C, sous forme d'aiguilles presque incolores. Un mélange de 64 g de cet acide et de 200 ml d'oxychlorure de phosphore est tiédi modérément jusqu'à la fin de la réaction exothermique, puis chauffé au reflux pen dant deux heures.
On élimine sous vide une partie de l'oxychlorure de phosphore en excès, on dissout le résidu dans du chloroforme sec, et on verse lente ment la solution chloroformée sous agitation dans un excès de solution d'hydroxyde d'ammonium et de glace. On ajoute une seconde portion de chloroforme et sèche les solutions chloroformées réunies, sur du carbonate de potassium anhydre. On distille sous vide le chloroforme, on fractionne le résidu par cris tallisation au sein de benzène et d'acétone et on obtient la 6,9-dichloro-3-méthyl-acridine de point de fusion de 206o C.
<I>Exemple 6</I> Un mélange de 11,7g de dichlorhydrate de 2- diéthylaminométhyl-4-amino-phénol, de 30 g de phé nol et de 13,6 g de 10-oxyde de 6,9-dichloro-3-nitro- acridine est chauffé entre 130 et 140 C pendant deux heures sous agitation mécanique. Après refroi dissement, on verse le mélange sous agitation dans un mélange de 5 ml d'acide chlorhydrique concentré et de 500 ml d'acétone. On dilue ensuite le mélange à l'éther, on recueille le produit par filtration et le lave à l'éther.
Le chlorhydrate ainsi obtenu est dis sous dans de l'eau, la solution est filtrée et la base libre précipitée par addition d'un excès de solution d'hydroxyde d'ammonium. On recueille le précipité par filtration, le sèche sous vide, le cristallise au sein d'un mélange méthanol-eau pour obtenir le 10'-oxyde de 4-(6'-chloro-3'-nitro-9'-acridinyl-amino)-a-diéthyl- amino-o-créso1.
Le 10-oxyde de 6,9-dichloro-3-nitro-acridine de départ peut s'obtenir à partir d'acide 2,4-dichloro- benzoïque et de m-nitro-aniline selon le procédé indi qué.
à l'exemple 5 ci-dessus pour la préparation du 10-oxyde de 6,9-dichloro-3-méthyl-acridine. <I>Exemple 7</I> Un mélange de 11,7g de dichlorhydrate de 2- diéthylaminométhyl-4-amino-phénol, de 30 g de phé nol et de 15 g de 10-oxyde de 6,9-dichloro-2-phényl- acridine, est chauffé entre 125 et 1350 C pendant deux heures sous agitation mécanique. Après refroi dissement, on verse le mélange dans une solution de 5 ml d'acide chlorhydrique concentré dans 500 ml d'acétone. On dilue le mélange avec 500 ml d'éther et l'on recueille le produit par filtration.
Le chlorhy- drate ainsi obtenu est dissous dans de l'eau, et la base précipitée avec un excès de solution d'hydro- oxyde d'ammonium. On recueille le précipité par fil tration, et le sèche sous vide. Une recristallisation au sein d'un mélange chaud éthanol-eau donne le 10'- oxyde de 4-(6'-chloro-2'-phényl-9'-acridinyl-amino)- a-diéthylamino-o-créso1.
Le 10-oxyde de 6,9-dichloro ? lphényl-acridine, point de fusion 209 à 21l C, servant de matière première, peut être obtenu à partir de la 6,9-dichloro- 2-phényl-acridine et d'acide perbenzoïque selon le procédé indiqué à l'exemple 1 ci-dessus pour la pré paration du 10-oxyde de 6,9-dichloro-2-méthoxy- acridine. La 6,
9-dichloro-2-phényl-acridine peut elle- même être préparée de la manière suivante: un mé lange de 111,5 g de 4-amino-diphényle, de 114,6 g d'acide 2,4-dichlorobenzoïque, de 82,8 g de carbo nate de potassium anhydre, de 3 g de poudre de cuivre et de 800 ml d'alcool n-amylique sec est chauffé au reflux pendant six heures avec agitation mécanique. On retire de l'eau par un collecteur d'eau Dean-Stark au cours de la réaction. On ajoute une solution de 70 g d'hydroxyde de potassium dans 100 ml d'eau, puis on entrame à la vapeur d'eau le mélange, le dilue avec de l'eau et le filtre.
On préci- pite l'acide brut au moyen d'acide chlorhydrique dilué, le recueille par filtration et le lave à fond à l'eau. On fait digérer le précipité humide avec 1,5 li tre d'éthanol à l'ébullition, on refroidit le mélange, le filtre, on lave le précipité avec de l'éther de pé trole (point d'ébullition 30 à 60 C) et le sèche. Le produit, qui est l'acide 4-chloro-N-(4-diphényl)-an- thranilique, est recristallisé au sein de monochloro- benzène ; point de fusion 246o C.
Un mélange de 130 g d'acide 4-chloro-N-(4-diphényl)-anthranilique et de 400 ml d'oxychlorure de phosphore est chauffé modérément jusqu'à la fin de la réaction exothermi que ; on chauffe ultérieurement le mélange au reflux pendant deux heures et l'on chasse sous vide l'excès d'oxychlorure de phosphore. On dissout le résidu dans du chloroforme sec et verse la solution lente ment, sous agitation, dans un excès de solution d7hy- droxyde d'ammonium et de glace. On ajoute une seconde portion de chloroforme, et sèche les solu tions chloroformées réunies sur du carbonate de po tassium anhydre.
On chasse le chloroforme sous vide, et on cristallise au sein du benzène le produit rési duel, à savoir la 6,9-dichloro-2-phényl-acridine ; point de fusion 2400C.

It is advantageous to carry out this condensation with substantially equivalent amounts of the reactants in the presence of a solvent which can be water, a phenol, an aliphatic alcohol such as ethanol, isopropanol, pentanol and l. 'octanol,
an aliphatic ketone such as methyl ethyl ketone, diisobutyl ketone, an aliphatic ether such as dibutyl ether, a monoalkyl ether of ethylene glycol, a cyclic ether such as dioxane, a hydrocarbon as is benzene, toluene and xylene. The preferred solvent is phenol. The reaction temperature is preferably above 750 ° C and preferably 95-145 ° C.
The product can be isolated in the form of a free base or transformed into a non-toxic acid addition salt such as hydrochloride, hydrobromide, sulfate, phosphate, oxalate, sulfamate, sulfonate, acetate, lactate, tartrate, gluconate, citrate, penicillinate, arsanilate and arsonate. Among the preferred salts, mention may be made of the following 5,5'-methylene-di-salicylate; 4,4'-methylene-bis- (3-hydroxy-2-naphthoate);
methylene-di-o-cresotinate; 8-hydroxy-7-iodo-5-quinoline-sulfonate; N-acetyl-4-hydroxy-m-arsanilate; N-glycolyl-p-arsanilate; N, N'-ethylene-bis- (p-arsanilate); p-ureidobenzenearsonate; benzylpenicillinate; phenoxymethylpenicillinate and fumagillinate. Some of the aforementioned salts have an amoebicidal action, and in certain cases antibacterial, reinforced.
<I> Example 1 </I> A mixture of 12.8 g of 6,9-dichloro-2-methoxy-acridine 10-oxide, 11.7 g of 2-diethylaminomethyl-4-amino-phenol dihydrochloride and 25g of phenol is brought to a temperature of 130 to 140L, C for one and a half hours. The reaction mixture is cooled, poured into a solution of 5 ml of concentrated hydrochloric acid in 200 ml of acetone, then left to stand at room temperature for eighteen hours. The mixture is then diluted with anhydrous ether and cooled.
The solid product was collected by filtration, dissolved in water, then the aqueous solution was filtered and to the filtrate was added an excess ammonium hydroxide solution. The separated product is recovered by filtration and dried; melting point of <B> 1680C </B> with decomposition, after recrystallization in a hot ethanol-water mixture. This product is 4- (6'-Chloro-2'-methoxy-9'-acridinylamino) -a-diethyl-amino-o-cresol 10'-oxide.
Likewise, the following compounds can be prepared from 6,9-dichloro-2-methoxy-acridine 10-oxide and an equivalent amount of the appropriate amino-phenol 10'-4- oxide. (6'-Chloro-2'-methoxy-9'-acridinyl-amino) -a-dioctylamino-o-cresol; 4- (6'-Chloro-2'-methoxy-9'-acridinyl-amino) -a-pyrrolidino-o-creso1 10'-oxide;
4- (6'-Chloro-2'-methoxy-9'-acridinyl-amino) -a-piperidino-o-cresol 10'-oxide; 4- (6'-Chloro-2'-methoxy-9'-acridinyl-amino) -a-dimethylamino-o-cresol 10'-oxide; 4- (6'-Chloro-2'-methoxy-9'-acridinyl-amino) -α-isobutylamino-o-cresol 10'-oxide; 4- (6'-Chloro-2'-methoxy-9'-acridinyl-amino) -a- (4'-methyl-piperazino) -o-cresol 10'-oxide;
2,2 '- [5- (6'-Chloro-2'-methoxy-9'-acridinyl-amino) -2 hydroxy-benzylamino] -diethanol 10'-oxide; 4- (6'-Chloro-2'-methoxy-9'-acridinyl-amino) -a-morpholino-o-cresol 10'-oxide. The starting 6,9-dichloro-2-methoxy-acridine 10-oxide can be prepared in the following manner, a solution of 278g of 6,
9-dichloro-2-methoxy-acridine in 5 liters of chloroform is treated at room temperature with a cold solution of 160 g of perbenzoic acid in 3 liters of chloroform. After allowing it to stand at room temperature for thirty-six hours, a platinum foil is added, the mixture is concentrated in vacuo to a volume of 1.5 liters and cooled in an ice bath. The precipitate formed is separated by filtration and washed thoroughly with ether; the melting point is 226-228 C.
The addition salts can be obtained as follows A hot solution of 9.0 g of 10'-4- (6'-chloro-2'-methoxy-9'-acridinyl-amino) - u - diethyl oxide - amino-o-cresol in 25 ml of ethanol is added to a hot solution of 12.6 g of N-glycolyl-p-arsanilic acid in 150 ml of 80% ethanol. The solution is heated in a water bath for ten minutes, filtered and then the solvent is removed in vacuo.
By triturating the residue with several portions of an ether-acetone mixture, the bis- (N-glycolyl-p-arsanilate from 4- (6'-chloro-2'-methoxy- 10'-oxide is separated. 9'-acridinyl-amino) -a-diethylamino-o-cresol.
Other salts of 4- (6'-chloro-2'-methoxy-9'-acridinyl-amino) - u -diethylamino-o-cresol 10'-oxide can be prepared from the free base and one or two equivalents of the following acids (in the presence of the following solvents) fumagillinic (acetone) sulfuric acid (ether) hydrobromic acid (ethanol) acetic acid (acetone) phosphoric acid (ethanol Cold, filtered solutions of 22.7 g of di - hydrochloride sesquihydrate of 10'-4- (3 'oxide,
6'- dichloro-9'- acridiny 1-amino) - a - pyrrolidino - o -cresol in 75 ml of water and 31 g of tasic benzylpenicillin in 200 ml of water are mixed in an ice bath; the oil which separates and causes its solidification is cooled by scraping the walls of the container with a rod. The product thus obtained is purified by dissolving in ethanol and reprecipitating with ether.
This product is 4- (3 ', 6'-dichloro-9'-acridinyl-amino) -a-pyrrolidino-o-cresol 10'-oxide bis-benzylpenicillinate.
Likewise, 10'-oxide salts of 4- (3 ', 6'-dichloro-9'-acridinyl-amino) -a-pyrrolidino-o-cresol can be prepared from the mono- or dihydrochloride of base and one or two equivalents, respectively, of the following salts 8-hydroxy-7-iodo-5-quinoline-sulfonic acid sodium salt; potassium salt of phenoxymethylpenicillin; monosodium salt of n-ureidobenzenearsonic acid;
N-acetyl-4-hydroxy-m-arsanilic acid monosodium salt; N, N'-ethylene-bis- (p-arsanil) disodium salt. <I> Example 2 </I> A mixture of 14.6 g of 4-amino-2-pyrrolidinomethyl-phenol dihydrochloride and 25 g of phe nol is heated under vacuum in a water bath for one hour, 14 , 9 g of 10-oxide of 3.6,
9-Trichloro-acridine and the mixture is heated with stirring at 125-135 ° C for two hours. The reaction mixture is cooled and then poured into a mixture of 5 ml of hydrochloric acid concentrated in 300 ml of acetone. After further dilution with acetone to one liter, the red precipitated material is collected by filtration, washed with acetone. It is redissolved in water and the aqueous solution is alkalinized by the addition of an ammonium hydroxide solution. The mixture is extracted with chloroform, the extracts washed with water, dried over anhydrous potassium carbonate and then filtered.
The filtrate is evaporated to dryness in vacuo, the residue is taken up in boiling ethanol, the solution is decolorized, evaporated to low volume and an excess of concentrated hydrochloric acid is added thereto. The precipitate is collected and crystallized in a methanol-acetone mixture, melting point 2301, C (dec.). The crystallized product is 4- (3 ', 6'-dichloro-9'-acridinyl-amino) -a-pyrrolidino-o-cresol 10'-oxide dihydrochloride.
Likewise, the following compounds can be prepared from 3,6,9-trichloro-acridine 10-oxide and a corresponding amount of the appropriate amino-phenol 10'-4- (3 'oxide. , 6'-Dichloro-9'-acridinyl-amino) -α-diethylamino-o-cresol; 4- (3 ', 6'-dichloro-9'-acridinyl-amino) -a-piperidino-o-cresol 10'-oxide;
4- (3 ', 6'-dichloro-9'-acridinyl-amino) -a-decylamino-o-cresol 10'-oxide; 10'-alpha- [5- (3 ', 6'-dichloro-9'-acridinyl-amino) - 2-hydroxy-benzylamino] -ethanol oxide; 4- (3 ', 6'-dichloro-9'-acridinyl-amino) -a-piperazino-o-cresol 10'-oxide; 10'-alpha-fc- [5- (3 ', 6'-dichloro-9'-acridinyl-amino) -2-hydroxy-benzyl] -ethylamino-ethanol;
4- (3 ', 6'-Dichloro-9'-acridinyl-amino) -a-cyclohexylimino-o-cresol 10'-oxide.
3,6,9-trichloro-acridine 10-oxide, melting point 236-238-C, used as a raw material in the above process, can be prepared from 3,6,9-trichloro. acridine and perbenzoic acid according to the process indicated in Example 1 above, for the preparation of 6,9-dichloro-2-methoxy-acridine 10-oxide.
The starting 4-amino-2-pyrrolidinomethyl-phenol dihydrochloride can be prepared as follows <B>: </B> 71 g of pyrrolidine, 30 g of paraformaldehyde and 100 ml of ethanol are heated in a water bath for thirty minutes. The resulting solution is added to a solution of 151 g of 4-acetylaminophenol in 500 ml of ethanol and the mixture is heated under reflux for one and a quarter hours.
After charcoal treatment, the solvent is removed in vacuo and the residue is heated in a water bath with 660 ml of 1: 1 hydrochloric acid for three hours. The mixture is evaporated in vacuo until an oil is obtained, several portions of ethanol and benzene are added and removed in vacuo, the residue is dissolved in ethanol and reprecipitated with anhydrous ether. . After numerous triturations with ether and / or acetone, the oily residue crystallizes; melting point 210 to 2150 C (with decomposition).
<I> Example 3 </I> A mixture of 16.7 g of 2-n-decylaminomethyl-4-amino-phenol dihydrochloride and 30 g of phe nol is heated in a water bath under vacuum for one hour, then 14 g of 6,9-dichloro-2-methoxy-acridine 10-oxide are added and the mixture is stirred at 130 to 1400 C for two hours. The reaction mixture is cooled, poured into 1 liter of normal sodium hydroxide solution, and extracted with chloroform. The combined chloroform extracts are washed thoroughly with water, dried over anhydrous potassium carbonate, filtered and evaporated in vacuo.
The residue is taken up in ethanol and an excess of alcoholic hydrochloric gas is added. Acetone and anhydrous ether are added, the precipitate is collected on a filter, washed with ether and dried in vacuo; melting point 175.1 C with decomposition. The product is 4- (6'-Chloro-2'-methoxy-9'-acridinyl-amino) - α # - decylamino-o-cresol 10'-oxide dihydrochloride.
2-n-Decylaminomethyl-4-amino-phenol dihydrochloride, melting point 100 to 1100 C, used as a raw material for the above preparation, can be obtained by reacting 51.8 g of n- decylamine, 9 g of paraformaldehyde and 45.3 g of 4-acetylamino-phenol according to the process indicated above in Example 2 for the preparation of 2-pyrrolidinomethyl-4-amino-phenol dichlorhydrate. <I> Example 4 </I> A mixture of 25 g of phenol, 7,
0 g of 4-amino-a-diethylamino-o-cresol dihydrochloride and 6.8 g of 6,9-dichloro-acridine 10-oxide hemihydrate is stirred and heated on a metal bath between 125 and 135 C during two hours. After cooling, the reaction mixture is diluted with several volumes of ether and acetone. The crude product is collected by filtration, dissolved in lukewarm water, alkalinized the solution with ammonium hydroxide, extracted with chloroform, then the chloroform extracts are washed thoroughly with water and dried over anhydrous potassium carbonate.
The chloro form is evaporated off in vacuo, then the tarry residue is treated with an excess of alcoholic hydrochloric gas. Several crystallizations of this residue from an ethanol-acetone-ether mixture give 4- (6'-chloro-9'-acridinyl-amino) -a-diethylamino- 10-oxide dichloride. o-cresol; melting point between 135 and 140o C with decomposition.
6,9-Dichloroacridine 10-oxide hemihydrate, melting point 164-166C, taken as raw material, can be prepared from there. 6,9-dichloro-acridine and perbenzoic acid according to the process indicated in Example 1 above for the preparation of 6,9-dichloro-2-methoxy-acridine 10-oxide. <I> Example 5 </I> A mixture of 4,
8 g of 2-di-ethylaminomethyl-4-amino-phenol dihydrochloride, 20 g of phe nol, 5 g of 6,9-dichloro-3-methyl-acridine 10-oxide, is heated and stirred between 120 and 125o C for two hours. After cooling, the mixture is poured into a solution of 5 ml of concentrated hydrochloric acid in 500 ml of acetone. The mixture is diluted with ether, the crude product is collected by filtration, and washed with ether.
The salt thus obtained is dissolved in water, the solution filtered and the base precipitated by the addition of an excess of an ammonium hydroxide solution. The precipitate is collected by filtration and dried. Recrystallization from a hot ethanol-water mixture gives 4- (6'-chloro-3'-methyl-9'-acridinyl-amino) -u-di-ethylamino-o-cresol 10'-oxide.
6,9-dichloro-3-methyl-acridine 10-oxide, melting point 172-1740 C, used as a raw material, can be prepared from 6,9-dichloro-3-methyl-acridine and perbenzoic acid according to the process indicated in Example 1 above for the preparation of 6,9-dichloro-2-methoxy-acridine 10-oxide. 6,9-dichloro-3-methyl-acridine can itself be prepared as follows: 191 g of 2,4-dichlorobenzoic acid,
134 g of m-toluidine, 138 g of anhydrous potassium carbonate, 5 g of copper powder and 750 ml of dry n-amyl alcohol are refluxed with stirring for five hours. The mixture is cooled, 70 g of potassium hydroxide in 100 ml of water are added to it, and it is carried off with steam. The aqueous residue is filtered while hot, slightly acidified with concentrated hydrochloric acid and cooled. The precipitated acid is collected by filtration, washed with water and then with petroleum ether (boiling point 30 to 600 ° C.).
The precipitate is boiled with 300 ml of ethanol, cooled, collected on a filter and washed with petroleum ether (boiling point 30 to 60 ° C.).
Recrystallization from a monochloro-benzene-ligroin mixture (boiling point 60 to 800 C) after black treatment, gives 4-chloro-N- (m-to-luyl) -anthranilic acid, of point 197 to 1990 C, as almost colorless needles. A mixture of 64 g of this acid and 200 ml of phosphorus oxychloride is moderately warmed until the end of the exothermic reaction, then heated to reflux for two hours.
Part of the excess phosphorus oxychloride is removed in vacuo, the residue is dissolved in dry chloroform, and the chloroform solution is slowly poured with stirring into an excess of ice ammonium hydroxide solution. A second portion of chloroform is added and the combined chloroform solutions are dried over anhydrous potassium carbonate. The chloroform is distilled off under vacuum, the residue is fractionated by crystallization in benzene and acetone and the 6,9-dichloro-3-methyl-acridine with a melting point of 206o C.
<I> Example 6 </I> A mixture of 11.7 g of 2-diethylaminomethyl-4-amino-phenol dihydrochloride, 30 g of phe nol and 13.6 g of 6,9-dichloro 10-oxide -3-nitro-acridine is heated between 130 and 140 C for two hours with mechanical stirring. After cooling, the mixture is poured with stirring into a mixture of 5 ml of concentrated hydrochloric acid and 500 ml of acetone. The mixture is then diluted with ether, the product is collected by filtration and washed with ether.
The hydrochloride thus obtained is dissolved in water, the solution is filtered and the free base precipitated by adding an excess of ammonium hydroxide solution. The precipitate is collected by filtration, dried under vacuum and crystallized in a methanol-water mixture to obtain the 10'-oxide of 4- (6'-chloro-3'-nitro-9'-acridinyl-amino ) -a-diethyl-amino-o-creso1.
The starting 6,9-dichloro-3-nitro-acridine 10-oxide can be obtained from 2,4-dichloro-benzoic acid and m-nitro-aniline according to the method indicated.
in Example 5 above for the preparation of 6,9-dichloro-3-methyl-acridine 10-oxide. <I> Example 7 </I> A mixture of 11.7 g of 2-diethylaminomethyl-4-amino-phenol dihydrochloride, 30 g of phe nol and 15 g of 6,9-dichloro-2 10-oxide -phenyl-acridine is heated between 125 and 1350 C for two hours with mechanical stirring. After cooling, the mixture is poured into a solution of 5 ml of concentrated hydrochloric acid in 500 ml of acetone. The mixture is diluted with 500 ml of ether and the product collected by filtration.
The hydrochloride thus obtained is dissolved in water, and the base precipitated with an excess of ammonium hydroxide solution. The precipitate is collected by filtration, and dried in vacuo. Recrystallization from a hot ethanol-water mixture gives 4- (6'-chloro-2'-phenyl-9'-acridinyl-amino) - a-diethylamino-o-creso1 oxide.
6,9-dichloro 10-oxide? Phenyl-acridine, melting point 209-21 C, serving as a raw material, can be obtained from 6,9-dichloro-2-phenyl-acridine and perbenzoic acid according to the process given in Example 1 below above for the preparation of 6,9-dichloro-2-methoxy-acridine 10-oxide. The 6,
9-dichloro-2-phenyl-acridine can itself be prepared as follows: a mixture of 111.5 g of 4-amino-diphenyl, 114.6 g of 2,4-dichlorobenzoic acid, 82.8 g of anhydrous potassium carbonate, 3 g of copper powder and 800 ml of dry n-amyl alcohol is heated under reflux for six hours with mechanical stirring. Water is removed by a Dean-Stark water collector during the reaction. A solution of 70 g of potassium hydroxide in 100 ml of water is added, then the mixture is steamed with water, diluted with water and filtered.
The crude acid is precipitated with dilute hydrochloric acid, collected by filtration and washed thoroughly with water. The wet precipitate is digested with 1.5 liters of boiling ethanol, the mixture is cooled, filtered, the precipitate is washed with petroleum ether (boiling point 30 to 60 ° C.) and dry it. The product, which is 4-chloro-N- (4-diphenyl) -anthranilic acid, is recrystallized from monochlorobenzene; melting point 246o C.
A mixture of 130 g of 4-chloro-N- (4-diphenyl) -anthranilic acid and 400 ml of phosphorus oxychloride is heated moderately until the end of the exothermic reaction; the mixture is subsequently heated under reflux for two hours and the excess phosphorus oxychloride is removed in vacuo. The residue was dissolved in dry chloroform and the solution poured slowly, with stirring, into an excess of ammonium hydroxide solution and ice. A second portion of chloroform is added, and the combined chloroform solutions are dried over anhydrous potassium carbonate.
The chloroform is removed in vacuo, and the residual product, namely 6,9-dichloro-2-phenyl-acridine, is crystallized from benzene; melting point 2400C.
