Verfahren zur Herstellung von neuen N-heterocyclischen Verbindungen Die vorliegende Erfindung betrifft ein Ver fahren zur Herstellung von neuen N-heterocychschen Verbindungen mit wertvollen pharmakologischen Eigenschaften.
In einem aromatischen Ring monohalogenierte N-substituierte 10,11-Dihydro-5-dibenzo(b,f)azepine und 5-Dibenzo(b,f)azepine sind bisher nicht bekannt geworden.

Quaternäre Ammoniumsalze, die sich von den vorstehend definierten tertiären Basen ableiten, wirken als Ganglioplegica. Das erfindungsgemässe Verfahren zur Herstel lung der neuen Verbindungen ist nun dadurch ge kennzeichnet, dass man ein 3-Halogen-iminodibenzyl oder 3-Halogen-iminostilben der Formel

in Gegenwart eines Kondensationsmittels mit einem reaktionsfähigen Ester eines Aminoalkohols der Formel HO-Z-Am III umsetzt.
Als Kondensationsmittel eignen sich hierfür insbesondere Natriumamid, Lithiumamid, Kalium- amid, Natrium oder Kalium, Butyllithium, Phenyl- lithium oder Lithiumhydrid. Die Umsetzung kann in An- oder Abwesenheit eines inerten organischen Lösungsmittels, wovon als Beispiele Benzol, Toluol und Xylole genannt seien,
durchgeführt werden.
Als reaktionsfähige Ester von Aminoalkoholen der Formel 111 kommen insbesondere die Halo genide in Frage, im einzelnen seien genannt:
Dimethylaminoäthylchlorid, Diäthylaminoäthylchlorid, Methyläthylaminoäthylchlorid, f-Dimethylamino-propylchlorid, ,B-Dimethylamino-isopropylchlorid, y-Dimethylamino-propylchlorid, @-Dimethylamino-butylchlorid, a-Methyl-y-dimethylamino-n-amylchlorid, ss-(Di-n-propylamino)-äthylchlorid, ss-(Methyl-isopropyl-amino)-äthylchlorid, /3-(Di-n-butylamino)-äthylchlorid, ss-(Di-isobutylamino)-äthylchlorid,
Pyrrolidinoäthylchlorid, Piperidinoäthylchlorid, y-Piperidino-propylchlorid, Morpholinoäthylchlorid, ,8-(4-Methyl-piperazino)-äthylchlorid, P-(4-Acetoxyäthyl-piperazino)-äthylchlorid, y-(4-Acetoxyäthyl-piperazino)-propylchlorid und 1-Methyl-piperidyl-(3)-methylchlorid, sowie die entsprechenden Bromide und Jodide.
Bei den Ausgangsstoffen der Formel II handelt es sich um das 3-Chlor- und 3-Brom-iminodibenzyl und -iminostilben. Die vorgenannten Iminodibenzyl- derivate kann man aus dem unsubstituierten Imino- dibenzyl durch N-Acylierung, Umsetzung mit Acetylchlorid nach Friedel-Crafts, Behandlung mit Stickstoffwasserstoffsäure nach Schmidt,
partielle Hydrolyse des erhaltenen 3-Acetamido-5-acyl-imino- dibenzyls, Diazotierung der 3-Amino-5-acylverbin- dungen, Umsetzung der Diazoniumhalogenide mit, Cuprihalogeniden nach Sandmeyer und hydrolytische Abspaltung des 5-Acylrestes erhalten.
Die Imino- stilbenderivate können aus den entsprechenden 3-Halogen-5-acyl-iminodibenzylen, Halogenierung in 10-Stellung, z. B. mit N-Brom-succinimid, Halogen wasserstoffabspaltung und Hydrolyse, z. B. mit Ätzalkalien in der Kälte, erhalten werden.
Durch Anlagerung von reaktionsfähigen Estern, insbesondere Halogeniden oder Sulfaten, aliphatischer oder araliphatischer Alkohole, z. B. von Methyljodid, Dimethylsulfat, Äthylbromid, Äthyljodid oder Benzyl- chlorid, können aus den erfindungsgemäss erhaltenen tertiären Aminen der Formel I in üblicher Weise monoquatemäre Ammoniumverbindungen hergestellt werden, wobei die Gruppe Am reagiert.
Mit anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthandisulfon- säure, Essigsäure, Citronensäure, Äpfelsäure, Bern steinsäure, Fumarsäure, Maleinsäure, Weinsäure, Benzoesäure und Phthalsäure bilden die tertiären Basen Salze, welche zum Teil wasserlöslich sind.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsius graden angegeben.
<I>Beispiel 1</I> 22,9 Teile 3-Chlor-irninodibenzyl werden in 300 Volumteilen Xylol gelöst und unter Rühren in einer Stickstoffatmosphäre 4 Teile Natriumamid in Toluol pulverisiert und suspendiert hinzugefügt. Die Xylol- lösung färbt sich sofort dunkel, hellt aber beim Auskristallisieren des Natriumsalzes wieder auf.
Das Reaktionsgemisch wird während etwa 2 Stunden bei 80 gerührt, bis die Ammoniakentwicklung be endet ist. Zum Natriumsalz wird eine Lösung von y-Dimethylamino-propylchlorid in Toluol (mit Na tronlauge aus 17,4 Teilen des entsprechenden Hy- drochlorids, das heisst etwa 10 /o Überschuss, frei gesetzt, in Toluol aufgenommen und mit wasser freiem Natriumsulfat 2 Stunden getrocknet) gegeben und 15 Stunden unter Kochen am Rückfluss gerührt.
Das ausgeschiedene Natriumchlorid wird abfiltriert und das Filtrat eingeengt. Der Rückstand wird mit Äther verdünnt und durch Einleiten von trockener Salzsäure das Hydrochlorid von 5-(y-Dimethylamino- propyl) - 3 - chlor - iminodibenzyl ausgefällt. Es wird abgenutscht und durch mehrmaliges Umkristallisieren aus Aceton gereinigt, worauf es bei 191,5-192 schmilzt.
In analoger Weise erhält man unter Verwen dung der aus 17,4 Teilen ;,-Diäthylamino-propyl- chlorid-hydrochlorid freigesetzten Base das 5-(;,-Di- äthylaminopropyl)-3-chlor-iminodibenzyl, und unter Verwendung der aus 20,5 Teilen /3-Piperidino-äthyl- chlorid-hydrochlorid freigesetzter.
Base das 5-(P-Pi- peridino-äthyl)-3-chlor-iminodibenzyl. Anderseits er hält man ausgehend von 27,4 Teilen 3-Brom-imino- dibenzyl analog obigem Beispiel das 5-(y -Dimethyl- amino-propyl)-3-brom-iminodibenzyl.
<I>Beispiel 2</I> 23 Teile 3-Chlor-iminodibenzyl werden in 190 Volumteilen Xylol gelöst, 4,3 Teile Natriumamid (in Toluol pulverisiert) hinzugefügt und 3 Stunden bei 80 in einer Stickstoffatmosphäre gerührt. Die Lösung färbt sich zuerst dunkel, hellt sich aber auf, sobald das Natriumsalz zu kristallisieren beginnt. Eine Morpholinoäthylchloridlösung (aus 20,5 Teilen Hydrochlorid mit verdünnter Natronlauge freigesetzt und in Benzol aufgenommen) wird zum Natrium salz gegeben.
Hierauf wird das Reaktionsgemisch unter ständigem Rühren 16 Stunden am Rückfluss erhitzt. Nach dem Einengen und Abkühlen wird das ausgeschiedenen Natriumchlorid abfiltriert und mit abs. Äther gewaschen. In die Lösung wird in der Kälte trockenes Salzsäuregas eingeleitet.
Das Hydro- chlorid des 3 - Chlor - 5 - p) -morpholino-äthyl-imino- dibenzyls wird abgenutscht und aus Alkohol um- kristallisiert. Es schmilzt bei 242 .
<I>Beispiel 3</I> 23 Teile 3-Chlor-iminodibenzyl werden in 190 Volumteilen Xylol gelöst, 4,3 Teile Natriumamid (in Toluol pulverisiert) hinzugefügt und 3 Stunden in einer Stickstoffatmosphäre bei 80 gerührt. Hier auf wird Dimethylaminoisopropylchlorid in Benzol (aus 18 Teilen Hydrochlorid des Dimethylamino-iso- propylchlorids mit verdünnter Natronlauge freigesetzt und in Benzol aufgenommen, die Benzollösung 2 Stunden mit wasserfreiem Natriumsulfat getrocknet)
hinzugefügt und das Reaktionsgemisch 16 Stunden unter Rückfluss gekocht. Das ausgeschiedene Na triumchlorid wird abfiltriert, mit absolutem Äther nachgewaschen und aus dem Filtrat durch Einleiten von trockenem Chlorwasserstoff das Hydrochlorid des so entstandenen 3-Chlor-5-(ss-dimethylamino-a- methyl-äthyl)-iminodibenzyl gefällt. Es wird aus viel Aceton umkristallisiert und schmilzt rein bei 247 .
<I>Beispiel 4</I> 23 Teile 3-Chlor-iminodibenzyl werden in 150 Volumteilen absolutem Xylol gelöst, worauf eine Suspension von 4,3 Teilen in Toluol pulverisiertes Natriumamid hinzugefügt wird. Das Gemisch wird gerührt und auf 90 erhitzt, wobei Ammoniak ent weicht und sich das Natriumsalz bildet.
Die ent standene Suspension desselben wird anschliessend mit a-[N-methyl-piperidyl-(2)]-äthylchlorid (aus 22 Teilen des Hydrochlorids freigesetzt und in Toluol aufgenommen) versetzt und 16 Stunden unter Rück fluss gekocht. Nach dem Abkühlen wird das ausge schiedene Natriumchlorid abgetrennt, die Lösung eingeengt und das Öl bei 0,1 mm Druck destilliert. Das 3 - Chlor - 5 - [i3-N'-Methyl-piperidyl-(2')-äthyl]- iminodibenzyl geht bei 195 über.
<I>Beispiel 5</I> 23 Teile 3-Chlor-iminostilben in 250 Volum- teilen abs. Xylol werden mit 4,3 Teilen Natrium- amid, in Toluol pulverisiert und suspendiert, 3 Stun den unter Rückfluss gekocht. Hierauf wird y-Di- methylamino-propylchlorid (freigesetzt aus 18 Teilen des Hydrochlorids und in Xylol aufgenommen) hinzugefügt und das Ganze 20 Stunden unter Rühren weiter zum Sieden erhitzt.
Das erhaltene Reaktionsgemisch wird zur Trockne eingedampft, und aus dem mit Äther versetzten Rückstand werden die basischen Anteile mit verdünnter Salzsäure extrahiert. Die sauren Auszüge werden alkalisch gestellt, und die freie Base wird in Äther aufgenom men. Nach dem Trocknen und Einengen der Äther lösung erhält man das gewünschte 3-Chlor-5-(y-di- methylamino-propyl))-iminostilben, das aus Petrol- äther umkristallisiert bei 53 schmilzt.

The resulting suspension is then treated with a- [N-methyl-piperidyl- (2)] ethyl chloride (released from 22 parts of the hydrochloride and taken up in toluene) and refluxed for 16 hours. After cooling, the sodium chloride which has separated out is separated off, the solution is concentrated and the oil is distilled at 0.1 mm pressure. The 3 - chloro - 5 - [i3-N'-methylpiperidyl- (2 ') - ethyl] - iminodibenzyl passes over at 195.
<I> Example 5 </I> 23 parts of 3-chloro-iminostilbene in 250 parts by volume of abs. Xylene is pulverized and suspended in toluene with 4.3 parts of sodium amide and refluxed for 3 hours. The reaction mixture obtained is evaporated to dryness, and the basic components are extracted from the residue mixed with ether with dilute hydrochloric acid. The acidic extracts are made alkaline and the free base is taken up in ether. After drying and concentrating the ether solution, the desired 3-chloro-5- (γ-dimethylamino-propyl)) - iminostilbene is obtained, which melts at 53 when recrystallized from petroleum ether.
