Procédé de préparation d'un dicarbamate
La présente invention a pour objet un procédé de préparation du dicarbamate de 2-phényl-1, 3-pro pane-diol, qui est un composé organique nouveau, possédant des propriétés antispasmodiques exception nellement intenses préventives des attaques survenant au cours des. électrochocs.
Ce composé est un solide cristallisé blanc soluble dans la plupart des solvants organiques, mais peu soluble dans l'eau. Il donne des solutions stables avec 1'eau et les solvants organiques. Par chauffage ou ébullition avec un acide ou une base, ce composé s'hydrolyse avec formation de 1,3-propane-diol substitué en 2 correspondant, d'ammoniaque et d'an- hydride carbonique.
Ce composé est préparé, selon l'invention, par réaction d'échange d'ester entre le 2-phényl-1, 3-propane-dol et un uréthane de faible poids moléculaire.
Dans cette réaction, l'alcool de faible poids moléculaire contenu dans 2 équivalents d'uréthane est remplacé par le 2-phényl-1, 3-propane-diol avec formation du dicarbamate désiré.
Le 2-phényl-1, 3-propane-diol utilisé dans la fabrication de ce composé nouveau peut tre préparé par un procédé quelconque connu, tel que, par exemple, la réduction de l'ester malonique correspondant substitué en position 2.
On dissout 20 g de 2-phényl-1, 3-propane-diol et 25 g d'éthyl-uréthane dans 3 20 ce de toluène anhydre.
On ajoute 3 g d'isopropylate d'aluminium et on chasse par distillation l'éthanol formé dans la con densation de l'éthyl-uréthane avec le diol. L'alcool distille sous forme d'azéotrope avec le toluène d'un point d'ébullition d'environ 770 C.
On poursuit la distillation jusqu'à élimination de sensiblement la quantité théorique d'éthanol. On chasse par distillation le toluène du mélange sous pression réduite et on extrait le produit solide au moyen d'une solution aqueuse d'isopropanol. On obtieJlt de cette solution, par refroidissement, 16,5 g de produit punfié, ce qui représente un rendement d'environ 52 /0 du rende- ment théorique. Le produit purifié possède un point de fusion de 151-152 C et n'est que légèrement soluble dans 1'eau à la température ambiante ordinaire.

L'essai de ce composé sur des animaux expéri- mentaux, tels que la souris, montre qu'il possède des propriétés antispasmodiques remarquables propres à prévenir les attaques au cours de l'électrochoc. On constate toutefois qu'il est relativement inefficace quant à la protection des animaux contre les convulsions provoquées par le pentylène-tétrazol. Ce composé nouveau ne possède pas de propriétés hypnotiques et ne provoque pas la paralysie des muscles volontaires sauf aux très fortes doses.
Les doses provoquant le sommeil ou la paralysie chez la souris sont supérieures à 2000 mg/kg, alors que les dbses protectrices contre les attaques survenant au cours de e l'électrochoc sont de l'ordre de 60 à 80 mg/kg. Le composé est remarquable en raison de sa très faible toxicité. La dose moyenne léthale chez la souris par administration intrapéritonéale est d'environ 4000 mg/kg et la dose moyenne léthale chez la souris par administration orale est d'environ 8000 mg/kg.
Le composé obtenu selon le procédé de l'inven- tion est principalement destiné aux usages par voie buccale et mis sous forme de pilules, de comprimés ou de capsules par les procédés bien connus. I1 peut tre dissous dans un solvant approprié tel qu'un n mélange d'eau et de polyéthylène-glycol, de manière à donner une solution propre aux injections à l'administration par voie rectale.
Le produit de départ peut tre préparé comme suit :
On réduit 50 g de phényl-malonate de diéthyle de la manière usuelle à l'aide de 12 g d'hydrure de lithium et d'aluminium, la réduction étant effectuée dans 500 cc d'éther anhydre. On utilise l'excès d'agent réducteur avec un petit volume d'acétate d'éthyle. On traite le mélange réactionnel de la manière usuelle à l'aide d'eau et d'acide sulfurique étendu, les composants organiques étant extraits au moyen d'éther. On obtient 16 g (soit environ 50 ID/o du rendement théorique) de produit purifié fondant à 49-520C.

3 g of aluminum isopropylate are added and the ethanol formed in the condensation of ethyl urethane with the diol is distilled off. The alcohol distils as an azeotrope with toluene with a boiling point of about 770 C.
The distillation is continued until substantially the theoretical amount of ethanol has been removed. The toluene was distilled off from the mixture under reduced pressure and the solid product was extracted with an aqueous solution of isopropanol.
