Verfahren zur Herstellung von in 17a-Stellung substituierten 1-Dehydro-testosteronen Gegenstand der vorliegenden Erfindung ist ein neues Verfahren zur Herstellung von 1-Dehydro- testosteronen, die in 17a-Stellung einen Alkyl- oder Alkenylrest von 2-4. Kohlenstoffatomen enthalten, und ihren Estern.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man 41.4-3,17-Dioxo-androstadien unter vorübergehendem Schutz der 3-Oxogruppe mit einer organischen Metallverbindung umsetzt, die einen Alkyl- oder Alkenylrest von 2-4 Kohlenstoff atomen enthält.
Die 1-Dehydro-testosterone mit dem erwähnten aliphatischen Kohlenwasserstoffrest in 17a-Stel- lung, z. B. 1-Dehydro-17a-äthyl-, 17a-Propyl-, 17a- Butyl-, 17a-Allyl- oder -Methallyl-, 17a-Isopropenyl- oder 17a-Vinyl-testosteron, und Ester besitzen auf Grund ihrer hohen anabolen Wirkung therapeu tisches Interesse und können als Heilmittel bei ent sprechenden Krankheiten Verwendung finden.
Die anabole Wirkung ist dabei überraschenderweise nicht mit hervorstechenden sexualspezifischen Wirkungen, verknüpft. Diese Eigenschaften machen diese Ver bindungen besonders wertvoll für die Behandlung solcher Affektionen, die einen vermehrten Protein aufbau verlangen und bei welchen die Sexualwirkung unerwünscht ist, wie Untergewichtigkeit, postopera tive oder postinfektiöse Zustände, Asthenie nach Schwangerschaft und in der Geriatrie.
Die verfahrensgemässe Einführung des erwähnten aliphatischen Kohlenwasserstoffrestes in 17a-Stel- lung, z. B. des Äthylrestes, erfolgt durch Umsetzung, mit der entsprechenden aliphatischen Metallverbin dung, wie Grignard- oder Alkalimetallverbindung, z.
B. Äthyl-magnesium-chlorid, -bromid oder -jodid oder Lithiumäthyl. Ein Äthylenrest lässt sich an schliessend durch geeignete Wahl der Reduktions bedingungen in einen Äthylrest, ein Allyl- in den n-Propylrest und ein Methallylrest in den i-Butylrest umwandeln.
Vor der Umsetzung mit der metallorga nischen Verbindung wird die 3-Oxogruppe geschützt, beispielsweise durch Überführung in einen 3-Enol- äther, wie z. B. 3-Enoläthyläther, in ein 3-Ketal, wie z. B. eine 3-Äthylendioxyverbindung oder in ein 3-Enamin, abgeleitet z. B. von Pyrrolidin oder Mor- pholin. Die Herstellung dieser Derivate erfolgt nach bekannten Methoden.
Nach der Umsetzung mit der metallorganischen Verbindung wird die geschützte 3-Oxogruppe in Freiheit gesetzt, z. B. mittels Salz säure, Schwefelsäure oder p-Toluolsulfonsäure.
Die erhaltenen Verfahrensprodukte lassen sich in bekannter Weise in ihre funktionellen Derivate, wie z. B. Ester und/oder Enolester, verwandeln. Zur Esterbildung können beliebige organische oder an organische Säuren, wie aliphatische, alicyclische, araliphatische, aromatische oder heterocyclische Carbon-, Thioncarbon-, Thiolcarbon- oder Sulfon- säuren, z. B.
Essigsäure, Propionsäure, Buttersäure, Capronsäure, Önanthsäure, Undecylensäure, ss Cyclo- pentylpropionsäure, Phenylpropionsäure, Schwefel säuren oder Phosphorsäuren, verwendet werden. <I>Beispiel</I> 28,4 g d1,4-Androstadien-3,17-dion, 15 g Pyrrol- idin und 0,15 g p-Toluolsulfosäure werden zusam men in 500 cm?, Benzol 2 Stunden zum Sieden er hitzt.
Die mit Wasser gewaschene und getrocknete Benzollösung lässt man innerhalb einer Stunde unter Rühren zu einer Lösung von 10,8g Äthyllithium in 500 cm3 Äther fliessen und erwärmt anschliessend noch 1 Stunde zum Sieden. Das Reaktionsgemisch wird sodann gekühlt und in 2 Liter Wasser einge tragen. Man extrahiert mit Äther, wäscht den Atherextrakt mit Wasser aus und dampft das Lö sungsmittel im Vakuum ab.
Der Rückstand wird in 400 cms Methanol aufgenommen und mit 40g Na triumacetat, 100 em3 Wasser und 40 cm3 Eisessig 6 Stunden zum Sieden erhitzt. Hernach konzentriert man im Vakuum, verrührt den Rückstand mit 1000 cms verdünnter Salzsäure und extrahiert mit Methylenchlorid. Die Lösung wird mit verdünnter Sodalösung und Wasser gewaschen, über Natrium sulfat getrocknet und filtriert. Man dampft bis zur beginnenden Kristallisation ein und lässt zu ihrer Vervollständigung über Nacht bei 0 stehen.
Das er haltene 1-Dehydro-17a-äthyl-testosteron wird abge- nutscht und mit wenig eisgekühltem Äther gewaschen. Ausbeute: 21,9 g.
