Verfahren zur Herstellung von 17a-Oxy-20-keto-21-acyloxy-steroiden Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 17a-Oxy-20-keto-21- acyloxy-steroiden durch Sauerstoffeinführung in J i7 (ao)-21-acyloxy-steroide mit Osmiumtetroxyd und Wasserstoffperoxyd, das dadurch gekennzeichnet ist, dass man in Gegenwart eines aromatischen tertiären N-heterocyclischen Amins arbeitet.
Mit Hilfe dieses Verfahrens können Cortison, Hydrocortison, 11-Epihydrocortison und 17a-Oxy-- 11-desoxy-corticosteron und deren Ester leichter als bisher hergestellt werden. Insbesondere lassen sich auch 17a-Oxy-20-keto-21 acetoxy-steroide nach dem neuen Verfahren erhalten.
Ferner erhält man eine wesentliche höhere Aus beute an 17u-Oxy-20-keto-21-acyloxy-steroid, als wenn man in Abwesenheit des Amins arbeitet. Diese höheren Ausbeuten erscheinen als höhere Ausbeute pro Einheit der Reaktionszeit und des Osmium- tetroxyds sowie als höhere Ausbeute an isolierbarem Produkt. Demzufolge bietet das Verfahren Wirtschaft lichkeit hinsichtlich des Ausgangsmaterials, der Reagenzien und der Zeit und liefert zudem noch ein Produkt, das weniger Osmiumtetroxyd enthält oder aus dem letzteres leichter entfernbar ist, sowie von verbesserter Farbe.
Die Verwendung von Osmiumtetroxyd zur I7ber- führung von Verbindungen mit Doppelbindung in Glycole ist eine bekannte Reaktion. Dabei lagert sich das Osmiumtetroxyd unter Bildung eines cyclischen Esters der Osmiumsäure an die Doppelbindung an, der nach Hydrolyse, die gewöhnlich mit wässrigem Natriumsulfit erfolgt, das Glycol liefert.
Diese Reak tion wurde in den USA-Patenten Nrn. <B>2265143,</B> 2275790, 2492194 und 2493780 beschrieben, wo (z0)-21-Acyloxy-pregnansteroide gewöhnlich mit äquimolekularen Mengen Osmiumtetroxyd behandelt und dann mit wässrigem Natriumsulfit hydrolysiert werden, um ein 17,20,21-Trioxy-pregnansteroid her zustellen.
Das USA-Patent Nr. 2493780 beschreibt auch, dass man Wasserstoffperoxyd zusammen mit einer katalytischen Menge Osmiumtetroxyd verwen den kann. Ähnliche Hydroxylierungs(Glycolysie- rungs)reaktionen mit eine Doppelbindung enthalten den Verbindungen, H202 und katalytischen Mengen Metalloxyd finden sich in den USA-Patenten Nm. 2373942, 2402566, 2414385 und 2437648.
Prins und Reichstein, Helv. Chim. Acta, 25, 300 (1942) teilen mit, dass die Oxydation des cyclischen Osmiumsäureesters eines in der Seitenkette unge sättigten Steroids mit Chlorsäure zum Oxy-ketosteroid führt, statt nur zum Glycolsteroid bei der Hydrolyse des Osmiumsäureesters mit wässrigem Natriumsulfit. Miescher und Schmidlin,
Helv. Chim. Acta, 33, 1840 (1950) ersetzten die Chlorsäure durch H202 als Oxy dationsmittel für den Osmiumsäureester des Steroids und sagen im USA-Patent Nr. 2668816, dass man auch Alkylperoxyde und Persäuren verwenden- kann. Diese Reaktion ergibt aber pro Zeiteinheit ausser ordentlich schlechte Ausbeuten, da sie 48-96 Stun den oder mehr zur Erreichung einer maximalen Aus beute von etwa 4804 erfordert.
Seither teilten Miescher und Schmidlin mit, dass die Reaktion durch Licht katalysiert wird (USA-Patent Nr. 2662854), doch ist die Zunahme der Reaktionsgeschwindigkeit selbst dann nur gering. So zeigen ihre Beispiele Reak tionszeiten von 20 Stunden oder mehr bis zu 80 Stun den.
Es zeigte sich nun, dass gemäss vorliegender Er findung die Reaktionsgeschwindigkeit stark erhöht werden kann, wenn man in Anwesenheit eines aro matischen tert. N-heterocyclischen Amins arbeitet. Das erfindungsgemässe Verfahren ermöglicht eine 10- bis 600fache oder noch höhere Steigerung der Reak tionsgeschwindigkeit ohne Einbusse der Gesamtaus- beute an 17a-Oxy-20-keto-21-acyloxy-steroid. Ausser dem ist diese Beschleunigung der Reaktion in der Regel von einer merklichen Erhöhung der Gesamt ausbeute an 17a-Oxy-20-keto-21-acyloxy-steroid be gleitet.
Ausserdem werden diese Ergebnisse mit klei neren Mengen Osmiumtetroxyd als beim Verfahren an Miescher und Schmidlin erhalten. So zeigt es sich, dass man im erfindungsgemässen Verfahren nur 0,002 Moläquivalente Osmiumtetroxyd, bezogen auf das Ausgangssteroid, zu verwenden braucht, um die Reaktion wirksam, ohne Einbusse an der Ausbeute des 17a-Oxy-20-keto-21-acyloxy-steroids durchzufüh ren.
Mit nur 0,05 Moläquivalenten Osmiumtetroxyd, was etwa der Hälfte der von Miescher und Schmidlin verwendeten Menge entspricht, erhält man wesentlich verbesserte Ausbeuten des gewünschten 17a-Oxy-20- keto-21-acyloxy-steroids. Ausserdem braucht man kein Licht wie beim Verfahren von Miescher und Schmidlin, wodurch der Prozess auf industrieller Basis leichter durchzuführen ist.
Die im Verfahren verwendeten tert. N-hetero- cyclischen Amine der aromatischen Reihe sind da durch gekennzeichnet, dass ihr Aminstickstoff ein Glied einer >C-N = C < -Gruppe in einem aroma tischen Ring darstellt, wie z.
B. in den Pyridinen, wie Pyridin, Picolin, a- und f-Lutidin, <I>a-,</I> P- und y- Collidin, Propylpyridin, die Parvoline und Chinolin, Isochinolin, Pyridopyridin, Naphtyridin usw.
Obschon man irgendeines dieser aromatischen tert.-N-hetero- cyclischen Amine im erfindungsgemässen Verfahren verwenden kann, bevorzugt man die Anwendung der Pyridine, das heisst des Pyridins und seiner Homo logen, z. B. der Picoline, fl-Lutidine, a-Lutidine, Collidine usw., wobei Pyridin am meisten bevor zugt ist.
Die Menge des verwendeten Amins ist nicht kri tisch und kann in weiten Grenzen variieren, ohne die Reaktionsgeschwindigkeit oder die Reaktionsprodukte wesentlich zu schädigen, obschon übermässige Men gen an Amin die Reaktionsgeschwindigkeit unter die optimale herabsetzen können. Zur Erzielung bester Ergebnisse verwendet.man vorzugsweise einen deut lichen molaren überschuss, bezogen auf das Steroid.
Zur Durchführung des erfindungsgemässen Ver fahrens wird das Ausgangssteroid vorzugsweise in einem inerten organischen Lösungsmittel gelöst, wie zum Beispiel in tert. Butylalkohol, Diäthyläther, Tetrahydrofuran usw., und das Osmiumtetroxyd, das aromatische tert. N-heterocyclische Amin und das Wasserstoffperoxyd zugemischt. Es ist vorteilhaft, jedoch nicht notwendig, das Osmiumtetroxyd und das Amin vor dem Wasserstoffperoxyd zuzugeben.
Ferner empfiehlt es sich, das Osmiumtetroxyd und das Was serstoffperoxyd im gleichen Lösungsmittel, das als Reaktionsmedium verwendet wird, gelöst zuzugeben. Das aromatische tert. N-heterocyclische Amin kann man als solches oder in der gleichen Lösung zu setzen.
Die Menge des Osmiumtetroxyds kann in weiten Grenzen variieren, z. B. von 0,2 Moläquivalenten bis zu 0,002 Moläquivalenten. Vorzugsweise ver wendet man jedoch nicht mehr als 0,05 Moläquiva- lente. Hält man die Menge des Osmiumtetroxyds zwischen 0,002 und 0,05 Moläquivalenten, so ist es möglich, hohe Ausbeuten des gewünschten Produktes zu erzielen und das restliche Osmiumtetroxyd wirk sam zu entfernen, indem man die Reaktionsmischung mit einem Fällungsmittel für Osmiumtetroxyd,
wie Natriumsulfit, ohne Anwendung von Wärme oder langen Kontaktzeiten behandelt. Man erhält so hohe Ausbeuten an Produkten von verbesserter Farbe und Zusammensetzung, ohne dass die 21-Acyloxygruppe wesentlich entacyliert würde.
Theoretisch sind für jedes Mol in der Reaktion sich bildenden Osmiumsäureester 2 Mol Wasserstoff peroxyd erforderlich. Es wurde jedoch gefunden, dass beim Verfahren der vorliegenden Erfindung mehr als die theoretische Menge H_0.., benutzt wer den muss, um eine vollständige Reaktion zu erzielen. Zur Erreichung bester Resultate muss man deshalb einen überschuss an Wasserstoffperoxyd anwenden. Beste Ergebnisse erhält man in der Regel bei An wendung von zwischen 2,2 und 2,7 Äquivalenten Wasserstoffperoxyd.
Verwendet man weniger H20". so wird die Ausbeute zufolge unvollständiger Reak tion herabgesetzt. Man kann mehr als 2,75 Mol- äquivalente H20" verwenden, doch sind diese un erwünscht, speziell wenn man ein 3-Keto-d 4.i7 (zo)- 21-acyloxy-steroid behandelt.
Es wurde bei solchen Steroiden beobachtet, dass übermässige H20.-Kon- zentrationen, dazu neigen, den A-Ring abzubauen; diese Nebenreaktion verschlechtert jedoch die Ge samtausbeute an dem gewünschten d 4-3,20-Diketo- 17a-oxy-21-acyloxy-steroid.
Gewünschtenfalls kann man das H202 langsam zur Mischung aus Steroid, Osmiumtetroxyd und Amin geben, z. B. während einer Periode von meh reren Minuten bis einigen Stunden. Manchmal ver läuft die Reaktion unter diesen Bedingungen vorteil hafter, besonders wenn das Ausgangssteroid eine "11-3-Ketogruppe enthält, die intakt bleiben soll. Normalerweise kann jedoch das H.=0., praktisch aui einmal zugegeben werden, da die Reaktionsgeschwin digkeit sehr hoch ist.
In diesem Falle kann es er wünscht sein, zu kühlen, da die Reaktion etwas exo- therm verläuft. Vorteilhafterweise wird die Geschwin digkeit des H102 Zusatzes so bemessen, dass die Reaktion beendet ist, wenn der letzte Anteil zuge setzt wird. Dies kann leicht ermittelt werden, indem man auf restliche H.0., titriert.
Zweckmässigerweise verwendet man das Wasser stoffperoxyd in einem organischen Lösungsmittel ge löst, z. B. in tert. Butylalkohol oder dergleichen, wobei man zu Beginn für gute Wasserfreiheit sorgt. Da bei der Reaktion Wasser entsteht, bleibt die Reaktionsmischung normalerweise nicht wasserfrei; doch kann man manchmal mit Vorteil ein wirksames inertes Trocknungsmittel oder andere Massnahmen zur Entfernung des Reaktionswassers verwenden.
Arbeitet man im Anfang unter wasserfreien Bedin- gungen, so erhöht sich die Ausbeute am erwünsch ten 17-Oxy-20-keto-steroid, während die Ausbeute an unerwünschtem 17,20-Dioxy-steroid entsprechend zurückgeht.
Eine Reaktionstemperatur zwischen etwa 15 und 30 C wird bevorzugt; doch kann man auch bei etwas höheren oder niedrigeren Temperaturen arbei ten, z. B. zwischen etwa - 10 C und etwa -I- 70 C. Man arbeitet nicht nur aus Zweckmässigkeitsgründen, sondern auch zwecks Erzielung optimaler Ausbeuten an 17-Oxy-20-keto-steroid vorzugsweise bei Zimmer temperatur.
Anstelle der obenerwähnten Lösungsmittel kann man irgendein Lösungsmittel verwenden, das die Reaktionsteilnehmer löst. Der Ausdruck inertes Lösungsmittel entspricht hier der zweiten Definition von solvent in Hackh's Chemical Dictionary, 3. Auflage und bedeutet Eine Flüssigkeit, welche eine andere Substanz ohne Änderung der chemischen Zusammensetzung auflöst im Gegensatz zu den Lösungsmitteln, die durch chemische Reaktion lösen.
Geeignete andere Lösungsmittel sind Cyclohexanon, Dioxan, tert: Amylalkohol, Äthanol und Methanol. Vorzugsweise siedet das Lösungsmittel niedrig genug, um durch Destillation unter vermindertem Druck bei Zimmertemperatur ohne Erhitzen aus der Reaktions mischung entfernt werden zu können.
Während die Reaktion des 417(20)-21-Acyloxy- steroids mit Osmiumtetroxyd und H.,02 träge ist und bis zur Beendigung mindestens 20 Stunden oder mehr erfordert, ist die Reaktion eines 417(20)-21-Acyloxy- steroids mit Osmiumtetroxyd und H,02 in Gegenwart eines aromatischen, tertiären N-heterocyclischen Amins gewöhnlich in weniger als einer Stunde und manchmal in weniger als 5 Minuten beendet.
Manch mal braucht man auch eine längere Zeit; doch selbst dann wird die Gesamtdauer, wenn überhaupt, nur selten 8 Stunden übersteigen. Die Ausbeute pro Zeit einheit wird deshalb im untersten Bereich etwa 0,15 /u pro Minute und manchmal bis zu 10 % pro Minute betragen.
Erwägt man, dass die früher be kannten Verfahren im besten Falle Ausbeuten von nicht wesentlich mehr als 0,015%. pro Minute ergaben, so sieht man, dass die Gegenwart eines aromatischen tertiären-N-heterocyclischen Amins die Reaktion stark beschleunigt. So entsteht nach dem Verfahren der vorliegenden Erfindung das gewünschte 17-Oxy-20-keto-steroid mit einer Geschwindigkeit, die einige 10-600 Mal grösser ist als bei den be kannten Verfahren.
Die Verwendung des aromatischen tert.-N-hetero- cyclischen Amins bringt aber nicht nur die erwähnte wesentliche Verkürzung der Reaktionszeit mit sich, sondern bewirkt auch eine wesentliche Ausbeute steigerung an 17-Oxy-20-keto-steroid. So erforderte die Reaktion von 11,;
-Oxy-21-acetoxy-4,17(20)- pregnadien-3-on mit H20, und Osmiumtetroxyd bis zur Beendigung 48 Stunden bei einer Ausbeute von nur 47 % Hydrocortisonacetat, während beim Arbei- ten gemäss vorliegendem Verfahren nach 2 bzw. 6 Stunden Reaktionszeit die Ausbeuten 53 bzw.
60 % betragen (siehe Beispiel 1).
Die ausgesprochene Erhöhung der Reaktionsge schwindigkeit macht das erfindungsgemässe Verfah ren technisch günstiger und wirtschaftlicher als das bekannte Verfahren. Auch die überraschende und merkliche Steigerung der Ausbeute an Hydrocortison- acetat macht das Verfahren wirtschaftlich wertvoll. Das Verfahren kann mit gleichem Erfolg auch auf andere A17(20)_21-Acyloxy-steroide Anwendung fin den.
Die aromatisch heterocyclischen tert. Amine, die erfindungsgemäss verwendet werden, sind alle dadurch gekennzeichnet, dass sie von Wasserstoffperoxyd unter den Reaktionsbedingungen praktisch nicht angegriffen werden. Primäre, sekundäre und die meisten nichtaromatischen tertiären Amine werden durch H202 leicht oxydiert und reagieren mit dem selben, wodurch der Reaktionsverlauf gestört wird.
Ausserdem scheint die Anwesenheit eines andern primären, sekundären oder tertiären Amins als der aromatischen tertiären-N-heterocyclischen Amine die Reaktionsgeschwindigkeit nachteilig zu beeinflussen.
Die aromatischen tertiären-N-heterocyclischen Amine jedoch wirken in überraschender und vorteil hafter Weise als Beschleuniger der Reaktion und zur Erhöhung der Ausbeute. Diese Beschleunigung der Reaktionsgeschwindigkeit ermöglicht es auch, prak tisch mit katalytischen Mengen Osmiumtetroxyd zu arbeiten, wodurch 1. der Osmiumgehalt des Produktes ausgesprochen verringert, 2. eine grössere Wirt schaftlichkeit erreicht und 3. die Reaktion sicherer gesteuert werden kann.
Die d17(20)-21-Acyloxy-steroide, vorzugsweise der Pregnenreihe, die sich nach dem Verfahren der Erfindung in die entsprechenden 17a-Oxy-20- keto-21-acyloxy-steroide überführen lassen, können durch folgende Formel

dargestellt werden, in der Ac den Acylrest einer organischen Carbonsäure, der vorzugsweise 1 bis 8 Kohlenstoffatome enthält, bedeutet, vorzugsweise den Rest einer niedrigaliphatischen Carbonsäure, ins besondere der Essigsäure,
während St einen Steroiden Cyclopentanpolyhydrophenanthrenkern bedeutet, der durch eine Doppelbindung am Kohlenstoffatom 17 an die oben dargestellte Seitenkette gebunden ist. Vorteilhafterweise ist sowohl der Cyclopentanpoly- hydrophenanthrenrest als auch der Acylrest frei von Substituenten oder Gruppen, die unter den Reaktions bedingungen von Osmiumtetroxyd angegriffen wer den.
Ein bevorzugter Typ des Ausgangssteroids ist entweder gesättigt oder enthält nur eine 44-Doppel- bindung, die mit einer 3-Ketogruppe gekuppelt ist.
Es ist ferner von Vorteil, wenn die bevorzugten Ausgangssteroide nur gesättigte Acylgruppen ent halten, da Doppelbindungen, seien sie im Kern oder im Acylrest, manchmal von Osmiumtetroxyd und H202 ebenfalls angegriffen werden. Ausserdem kann der Kern des Ausgangssteroids vorteilhafterweise eine Hydroxyl-, Keto-, Acyloxy-, Hydrocarbonoxy-, Benzyloxygruppe,
Halogen- oder Oxydogruppe oder Gruppen in 3, 6, 11 und 12 sowie andern Stellungen des Kernes aufweisen. Das erfindungsgemässe Ver fahren ist jedoch als Verbesserung früherer Prozesse zur Einführung von Sauerstoff in d17(ao)_21-Acyloxy- steroide unter Bildung von 17-Oxy-20-keto-21- acyloxy-steroiden in seinem weitesten Umfang auf die Erzeugung von 17-Oxy-20-keto-steroiden aus irgendwelchem A17(20)-21-Acyloxy-steroid anwendbar.
Im speziellen jedoch eignet sich das erfindungs gemässe Verfahren zur Oxydation von in 3-Stellung Sauerstoff enthaltenden A17(20)-21-Acyloxy-steroiden der Pregnan- und Allopregnanreihe, wie z. B.
3a- und 3ss-Oxy-21-acetoxy-17(20)-pregnen, 3a,21- und 3ss,21-diacetoxy-17(20)-pregnen, 3a,l 1ss- und 3ss,11f-dioxy-21-acetoxy-17(20)- pregnen;
3a,llss- und 3ss,llss dioxy-21-acetoxy-17(20)- allopregnen, 21-Acetoxy-17(20)-pregnen-3-on, 21-Acetoxy-17(20)-pregnen-3,11-dion, l lss-Oxy-21-acetoxy-4,17(20)-pregnadien-3-on, 11 a-Oxy-21-acetoxy-4,17(20)-pregnadien-3-on, 21-Acetoxy-4,17(20)-pregnadien-3,1 1-dion, 21-Acetoxy-4,9(11), 17(20)

in der Ac der Acylrest einer organischen Carbon- säure, vorzugsweise einer niedrig-aliphatischen Säure, insbesondere Essigsäure, und R Wasserstoff, eine a-Acyloxygruppe, vorzugsweise eine niedrige, z. B.
Acetoxygruppe, eine ss-Oxygruppe oder ein Keton- sauerstoff (= 0) ist, da sich diese Steroide unter Verwendung des vorliegenden Verfahrens leicht in die physiologisch aktiven Nebennierenrindenhormone Cortison (Kendall's Substanz E) und Hydrocortison (Kendall's Substanz F) überführen lassen.
Ebenfalls bevorzugt sind die di-Analoga der in Formel 1I dar- gestellten Verbindungen, in denen R eine ss-Oxy- gruppe ist. Diese Verbindungen lassen sich in dl Hydrocortison bzw. dl-Cortison überführen.
So ergibt beispielsweise die Umsetzung von 21-Acetoxy- 4,17(20)-pregnadien-3,11-dion mit 0s 04 und H.20.., in Gegenwart von Pyridin nach dem Verfahren der vorliegenden Erfindung Cortisonacetat. In glei cher Weise entsteht aus l lss-Oxy-21-acetoxy-4,17(20)- pregnadien-3-on das llss,17a-Dioxy-21-acetoxy-4- pregnen-3,20-dion (Hydrocortisonacetat);
aus lla- Oxy-21-acetoxy-4,17(20)-pregnadien-3-on das 11a,17a- Dioxy-21-acetoxy-4-pregnen-3,20-dion, das mit Chromsäure leicht zum Cortisonacetat oxydiert wer den kann (USA-Patent Nr. 2602769 von Murray und Peterson, veröffentlicht am B. Juli 1952).
Da die meisten physiologisch aktiven Steroide eine 17a-Oxy-20-keto-21-oxy- oder Acyloxygruppierung besitzen, ermöglicht das erfindungsgemässe Verfah ren die Umwandlung von d17('o)-21-Acyloxy-steroiden in Steroide, die mindestens einen Teil der zur Bil dung eines physiologisch aktiven Steroids erforder lichen Struktur aufweisen.
Da man viele synthetische Methoden zur Einführung einer J4-3-Ketogruppe und eines 11-Sauerstoffatoms in den Steroidkern kennt, ferner solche zur Entfernung inaktivierender Gruppen aus demselben, sind auch andere Steroide als die durch Formel 1I dargestellten über das erfin dungsgemässe Verfahren leicht in physiologisch aktive Rindenhormone überführbar.
Die im Verfahren verwendeten J17(2 -21-Acyl- oxysteroide können auf verschiedene Weise erhalten werden, z. B. nach der Reaktion von Dimroth [Dim- roth, Berichte, 71B, 1343 (1938)], die auf 17ss-Oxy- 20-pregnen-steroide angewendet 21-Oxy-, 21-Acet- oxy- oder 21-Brom-17,20-pregnen-steroide liefert,
die nach bekannten Verfahren in andere 21-Acyl- oxy-steroide übergeführt werden können [siehe Serini, Brit. Pat. Nr. 213630, Berichte<B>7111,</B> 1313, 1362 (1938), USA-Patente Nrn. 2267258, 2305727, Brit. Pat. Nr. 467790, Miescher und Mitarb. Helv. Chim. Acta 22, 120, 894 (1939, Ruzicka und Müller, ibid., 22, 416 755 (1939)].
Gewünschtenfalls können diese Verbindungen auch nach der nachfolgend beschriebenen #Methode oder nach der USA-Patent- schrift Nr. 2 774 776 erhalten werden.
Da es gewöhnlich vorteilhaft ist, das Osmium vom Reaktionsprodukt zu trennen, wird das Reak tionsgemisch meist mit einem Mittel behandelt, das, wie beispielsweise Natriumsulfit, Schwefelwasserstoff oder dergleichen, befähigt ist, das Osmium entweder als freies Osmium oder als Kation eines unlöslichen Salzes auszufällen. Die übliche Arbeitsweise bestand bisher darin, das Reaktionsgemisch mit wässrigem Natriumsulfit auf höhere Temperaturen, z. B. auf die Rückflusstemperatur der Mischung, zu erhitzen.
Unter diesen Bedingungen wird eine im Reaktions produkt vorhandene Acetatgruppe mindestens teil weise hydrolysiert, und man muss wieder acetylieren, um ein vollkommen acetyliertes Reaktionsprodukt zu erhalten. Nach einer besonderen Ausführungsform der vor liegenden Erfindung vermischt man das Reaktions produkt bei etwa Zimmertemperatur oder niedriger mit wässrigem Natriumsulfit, um das Osmium aus der Mischung zu entfernen.
Unter diesen Bedin gungen bleibt die Acetat- oder eine andere leicht hydrolysierbare Estergruppe im Molekül intakt, und die beim Arbeiten nach den früheren Verfahren erfor derliche Wiederveresterung kann wegfallen, wodurch man eine verbesserte Ausbeute an Steroidprodukt erhält.
Die Anwendung geringer Os 04 Konzentrationen im Reaktionsgemisch, z. B. weniger als etwa 0,05 und vorzugsweise so wenig wie 0,002 Moläquiva- lente, berechnet auf das Ausgangssteroid, ergibt einen unbedeutenden Verlust an Reaktionsprodukt, der am Ende der Reaktion als Ester der Osmiumsäure auf gefunden werden kann, wenn die Reaktionsmischung nicht hydrolysiert wird.
Falls Spuren von Osmium- tetroxyd nicht unerwünscht sind, kann man die Hydrolyse weglassen, wenn man die bevorzugten niedrigen Konzentrationen anwendet, wodurch das Verfahren weiter vereinfacht wird. Die ausserordent lich langsame Reaktion bei den früher angewendeten Prozessen verbietet bei diesen in der Regel die An wendung so geringer Osmiumtetroxydkonzentratio- nen. Da die Liefermöglichkeiten für Osmiumtetroxyd etwas beschränkt sind und das Produkt toxisch und gefährlich ist, erleichtert die Anwendung dieser sehr schwachen Konzentrationen die Anpassung des Ver fahrens an grosstechnische Anforderungen.
Präparation 11ss3 Oxy-21-acetoxy-4,17(20)-pregnadien-3-on Eine Lösung von 0,518 g 11ss',21-Dioxy-4,17(20)- pregnadien-3-on, hergestellt gemäss USA-Patent Nr. 2705621, in 5 cm3 Pyridin wird mit 2 cm- Essigsäureanhydrid vermischt und das Ganze 17 Stunden bei Zimmertemperatur aufbewahrt und dann mit zerstossenem Eis versetzt.
Das ausgefällte llss-Oxy-21-acetoxy-4,17(20)-pregnadien-3-on wird abfiltriert, in Benzol gelöst und dann über einer Säule von 75 g synthetischem Magnesiumsüikat < Florisil chromatographiert. Die Säule wird mit 75-cm?-Portionen Lösungsmittel folgender Zusam mensetzung und Reihenfolge eluiert:
Benzol, drei mal Hexankohlenwasserstoffe Skellysolve B plus 1% Aceton, Skellysolve B plus 5 % Aceton, Skelly- solve B plus 10 % Aceton,
Skellysolve B plus 15 0/0 Aceton, Skellysolve B plus 20 % Aceton und schliess- lich zweimal mit Aceton.
Die Fraktionen mit 10 0/0 und 15 % Aceton werden vereinigt, das Lösungs- mittel enfernt und der feste Rückstand aus einer Mischung von Äthylacetat und Skellysolve B um kristallisiert, wobei man als erste Fraktion 0,253 g (45% Ausbeute) 11f3-Oxy-21-acetoxy-4,17(20)
- pregnadien-3-on vom Smp. 183-186 C erhält.
Analyse: Ber. für C23H3204: C 74,16; H 8,66 Gef. : C 74,18; H 8,45 C 73,95; H 8,74 In gleicher Weise erhält man durch Veresterung von 21-Oxy-4,17(20)-pregnadien-3-on in der oben beschriebenen Weise mit Essigsäureanhydrid 21- Acetoxy-4,17(20) -pregnadien-3 - on. 11 a-Oxy-21- acetoxy-4,17(20)-pregnadien-3-on und lla,
21-Di- acetoxy-4,17(20)-pregnadien-3-on erhält man in gleicher Weise, indem man auf ein Moläquivalent einen grossen molaren überschuss Essigsäureanhydrid einwirken lässt.
Andere Ester des lla,21-Dioxy-4,17(20)- pregnadien-3-ons, 11ss,21-Dioxy-4,17(20)-pregnadien- 3-ons und 21-Oxy-4,17(20)-pregnadien-3-ons wer den erhalten, wenn man in der oben beschriebenen Reaktion andere Säureanhydride oder Säurehaloge- nide einsetzt, wie Propionsäureanhydrid, Benzoyl- chlorid, Phthalsäureanhydrid, Bernsteinsäureanhydrid, Oxalylchlorid,
Trimethylacetylchlorid, Cyclopentano- propionylchlorid, Diäthylacetylchlorid, Phenylpro- pionylchlorid und dergleichen. Die Ameisensäureester erhält man in der Regel durch Verwendung von etwa 98 o/o.iger Ameisensäure und eines Veresterungskata- lysators.
Die Oxydation von 11-Oxy-21-acyloxy-4,17(20)- pregnadien-3-on mit Chromsäure liefert das entspre chende 21-Acyloxy-4,17(20)-pregnadien,3,11-dion. <I>Beispiel 1</I> a) Zu einer Lösung von 744 mg (2,0 Millimol) 1 lss-Oxy-21-acetoxy-4,17 (20) - pregnadien - 3 - an in 30 cm3 tert. Butylalkohol gibt man 1 cm3 Pyridin und 25 mg (0,1 Millimol)
0s 04 in 8 cm3 tert. Butylalkohol. Zu dieser Lösung gibt man 8,36 cm3 (5,0 Millimol) einer 0,6 molaren Lösung von 11,02 in tert. Butylalkohol. Der Zusatz dauert eine Stunde, und die Mischung wird dann eine weitere Stunde ge rührt.
Der verwendete tert. Butylalkohol war mit Natrium getrocknet und wies einen Feuchtigkeits- gehalt von etwa 0,04,% auf.
Das so erhaltene Reaktionsgemisch wurde sofort wie folgt aufgearbeitet: Man gibt 600 mg Natrium sulfit in 25 cm3 Wasser gelöst zu, rührt 5 Minuten und konzentriert durch Destillation bei etwa 50 mm Hg (abs.) auf etwa 15 cm3. Das Konzentrat wird mit Methylenchlorid extrahiert.
Der Extrakt wird getrocknet und über 80 g synthetischem Magne- siumsilikat ( Florisü ) chromatographiert. Die Säule wird mit Äthylenchlorid, das zunehmende Mengen Aceton enthält, entwickelt.
Das Ausgangssteroid be- findet sich in der Fraktion, die 2,5%, Aceton ent- hält. Ihre Menge ist 1600 cm3 mit einem Gehalt von 140 mg Ausgangssteroid (20%). Das gewünschte Produkt,
Hydrocortisonacetat, befindet sich in den 711/o Aceton enthaltenden Fraktionen, mit einem Gesamtvolumen von 2000 cm3 und einem Gehalt von 428 mg (53%,) Hydrocortisonacetat. Die Aus- beute, bezogen auf das in Reaktion getretene Aus- gangssteroid beträgt 67 o/oa der Theorie.
Das Glycol- Nebenprodukt, 11ss,17a,20 - Trioxy - 21- acetoxy-4- RTI ID="0005.0217" WI="20" HE="4" LX="1128" LY="2629"> pregnen-3-on befindet sich in den 15 % Aceton ent- haltenden Fraktionen, deren Gesamtvolumen 800 cm3 mit einem Gehalt an 101 mg (12,5 0/0) des Neben produktes beträgt.
b) Zu Vergleichszwecken wird 11ss-Oxy-21- acetoxy-4,17(20)-pregnadien-3-on nach dem Verfah ren von Miescher und Schmidlin unter Anwendung von im wesentlichen gleichen Mengen wie unter a) in Hydrocortisonacetat übergeführt.
Zu einer Lösung von 744 mg (2,0 Millimol) 1 lss-Oxy-21-acetoxy-4,17 (20) - pregnadien - 3 - an in 30 cm3 tert. Butylalkohol gibt man bei Zimmer temperatur 6,18 cm3 (4,0 Millimol) einer 0,684 mola- ren Lösung von H20, in mit Natrium getrocknetem tert. Butylalkohol und dann tropfenweise eine Lö- sung von 52 mg (0,2 Millimol)
Os 04 in 8 cm3 über Natrium getrocknetem tert. Butylalkohol. Die Zu gabe erfolgt im Verlauf von 8 Stunden und die Mischung wird dann weitere 48 Stunden bei Zim mertemperatur gehalten. Der getrocknete tert. Butyl- alkohol hatte einen Feuchtigkeitsgehalt von 0,04 0/0.
Die Reaktionsmischung wird dann genau wie unter a) aufgearbeitet. Man erhält 53 mg (7 %) Aus- gangsmaterial, 397 mg (47 %) Hydrocortisonacetat und 162 mg (20")/o) des Glycol-Nebenproduktes 11 ss,17a,
20-Trioxy-21-acetoxy-4-pregnen-3-on.

Weitere Reaktionen wurden durchgeführt, bei denen die unter a) beschriebenen Bedingungen abge ändert wurden. Zum Beispiel zeigte es sich, dass unter optimalen Bedingungen Reaktionszeiten von nur 5 Minuten genügen, um die Reaktion zu Ende zu führen. Weiter ergab sich, dass nur 0,002 Moläqui- valente Os 04, berechnet auf das Ausgangssteroid er forderlich sind, um beste Ausbeuten an gewünschtem Produkt zu erhalten. Obschon bei diesen niedrigen Konzentrationen an Os 04 die Reaktionsgeschwindig keit etwas abfällt, kann man sie durch Erhöhung der Reaktionstemperatur leicht wieder erhöhen.
Es zeigte sich auch, dass manchmal der Zusatz von 0s 04 und Amin vor der Zugabe des H202 die Ausbeute an gewünschtem Produkt erhöht. Obschon die Reak- tion selbst in Anwesenheit von merklichen Mengen Wasser in einer Weise verläuft, die derjenigen der bekannten Verfahren überlegen ist, wurde gefunden, dass beim Arbeiten unter möglichst wasserfreien Be dingungen die Ausbeute an 17-Oxy-20-ketoprodukt verbessert wird.
<I>Beispiel 2</I> Verwendet man im Verfahren des Beispiels la als Ausgangsmaterial 21-Acetoxy-4,17(20)-pregnadien- 3,11-dion, so erhält man 17a-Oxy-21-acetoxy-4- pregnen-3,11,20-trion (Cortisonacetat) in gleicher Ausbeute wie das Hydrocortison im Beispiel la.
<I>Beispiel 3</I> In gleicher Weise wie im Beispiel la erhält man 17a-Oxy-21-acetoxy-4-pregnen-3,20-dion (Acetat der Substanz S) bei Verwendung von 21-Acetoxy- 4,17(20)-pregnadien-3-on als Ausgangssteroid in der dort beschriebenen Reaktion.
<I>Beispiel 4</I> 11 a,17a-Dioxy-21-acetoxy-4-pregnen - 3,20 - dion erhält man nach der im Beispiel la angegebenen Arbeitsweise, wenn man l la-Oxy-21-acetoxy- 4,17(20)-pregnadien-3-on als Ausgangssteroid ver wendet.
In analoger Weise kann man andere Acyloxyester von Cortison, Substanz S und 1 l a,17a,21-Trioxy-4- pregnen-3,20-dion erhalten, indem man die in den Beispielen 1-4 verwendeten 21-Acetoxy-4,17(20)- pregnadienone durch die geeigneten 21-Acyloxy- 4,17(20)-pregnadienone ersetzt, in denen die Acyloxygruppe Formyloxy, Propionyloxy, Butyryl- oxy, Dimethylacetoxy,
Valeryloxy, Trimethyl- acetoxy, Phenylacetoxy, Benzoyloxy, Heptanoyloxy, Hexanoyloxy, Cyclopentanopropionyloxy, Phenyl- propionyloxy oder dergleichen ist.
llf=Oxy-21-acet- oxy-1,4,17(20)-pregnatrien-3-on und 21-Acetoxy- 1,4,17(20)-pregnatrien-3,11-dion werden in analoger Weise in 11fl',17a-Dioxy-21-acetoxy-1,4-pregnadien- 3,20-dion bzw. in 17a-Oxy-21-acetoxy-1,4-preg- nadien-3,11,20-trion übergeführt.
Process for the preparation of 17a-oxy-20-keto-21-acyloxy-steroids The present invention relates to a process for the preparation of 17a-oxy-20-keto-21-acyloxy-steroids by introducing oxygen into J i7 (ao) -21 -acyloxy-steroids with osmium tetroxide and hydrogen peroxide, which is characterized in that one works in the presence of an aromatic tertiary N-heterocyclic amine.

Furthermore, you get a much higher yield of 17u-oxy-20-keto-21-acyloxy-steroid than if you work in the absence of the amine. These higher yields appear as a higher yield per unit of reaction time and of osmium tetroxide and as a higher yield of isolable product. Accordingly, the process offers economy in terms of starting material, reagents and time and also provides a product that contains less osmium tetroxide or is easier to remove from the latter, and of improved color.
The use of osmium tetroxide to convert compounds with double bonds into glycols is a well-known reaction. The osmium tetroxide is attached to the double bond to form a cyclic ester of osmic acid which, after hydrolysis, which usually takes place with aqueous sodium sulfite, supplies the glycol.
This reaction has been described in U.S. Patent Nos. 2265143, 2275790, 2492194 and 2493780, where (z0) -21-acyloxy-pregnancy steroids are usually treated with equimolecular amounts of osmium tetroxide and then hydrolyzed with aqueous sodium sulfite to produce a 17,20,21-trioxy-pregnan steroid.
U.S. Patent No. 2493780 also describes that hydrogen peroxide can be used in conjunction with a catalytic amount of osmium tetroxide. Similar hydroxylation (glycolysis) reactions with a double bond contain the compounds H2O2 and catalytic amounts of metal oxide can be found in the USA patents Nm. 2373942, 2402566, 2414385 and 2437648.
Prins and Reichstein, Helv. Chim. Acta, 25, 300 (1942) report that the oxidation of the cyclic osmic acid ester of a steroid unsaturated in the side chain with chloric acid leads to the oxy-ketosteroid, instead of only to the glycol steroid during hydrolysis of the osmic acid ester with aqueous sodium sulfite. Miescher and Schmidlin,
Helv. Chim. Acta, 33, 1840 (1950) replaced the chloric acid with H 2 O 2 as an oxidizing agent for the osmic acid ester of the steroid and say in US Pat. No. 2668816 that alkyl peroxides and peracids can also be used. However, this reaction gives exceptionally poor yields per unit of time, since it requires 48-96 hours or more to achieve a maximum yield of about 4804.
Since then, Miescher and Schmidlin have reported that the reaction is catalyzed by light (U.S. Patent No. 2662854), but even then the rate of increase is only slight. Their examples show reaction times of 20 hours or more up to 80 hours.
It has now been found that according to the present invention, the reaction rate can be greatly increased if you tert in the presence of an aromatic. N-heterocyclic amine works. The process according to the invention enables a 10- to 600-fold or even higher increase in the reaction rate without loss of the overall yield of 17α-oxy-20-keto-21-acyloxy-steroid. In addition, this acceleration of the reaction is usually accompanied by a noticeable increase in the overall yield of 17a-oxy-20-keto-21-acyloxy-steroid.
In addition, these results are obtained with smaller amounts of osmium tetroxide than in the Miescher and Schmidlin method. It turns out that in the process according to the invention only 0.002 molar equivalents of osmium tetroxide, based on the starting steroid, need to be used in order to carry out the reaction effectively without loss of the yield of the 17a-oxy-20-keto-21-acyloxy-steroid .
With only 0.05 molar equivalents of osmium tetroxide, which corresponds to about half the amount used by Miescher and Schmidlin, one obtains significantly improved yields of the desired 17a-oxy-20-keto-21-acyloxy-steroid. In addition, there is no need for light as in the Miescher and Schmidlin process, which makes the process easier to carry out on an industrial basis.
The tert used in the process. N-heterocyclic amines of the aromatic series are characterized in that their amine nitrogen is a member of a> C-N = C <group in an aromatic ring, such as.
B. in the pyridines, such as pyridine, picoline, a- and f-lutidine, <I> a-, </I> P- and y- collidine, propylpyridine, the parvoline and quinoline, isoquinoline, pyridopyridine, naphtyridine, etc.
Although any of these aromatic tert-N-heterocyclic amines can be used in the process according to the invention, preference is given to using the pyridines, that is to say the pyridine and its homologues, e.g. B. the picolines, fl-lutidines, a-lutidines, collidines, etc., with pyridine being given most before.
The amount of amine used is not critical and can vary within wide limits without significantly damaging the reaction rate or the reaction products, although excessive amounts of amine can reduce the reaction rate below the optimum. To achieve the best results, it is preferable to use a significant molar excess, based on the steroid.
To carry out the inventive method, the starting steroid is preferably dissolved in an inert organic solvent, such as in tert. Butyl alcohol, diethyl ether, tetrahydrofuran, etc., and the osmium tetroxide, the aromatic tert. N-heterocyclic amine and the hydrogen peroxide mixed in. It is advantageous, but not necessary, to add the osmium tetroxide and the amine before the hydrogen peroxide.
It is also advisable to add the osmium tetroxide and the hydrogen peroxide dissolved in the same solvent that is used as the reaction medium. The aromatic tert. N-heterocyclic amine can be put as such or in the same solution.
The amount of osmium tetroxide can vary within wide limits, e.g. B. from 0.2 molar equivalents to 0.002 molar equivalents. Preferably, however, no more than 0.05 molar equivalents are used. If the amount of osmium tetroxide is kept between 0.002 and 0.05 molar equivalents, it is possible to achieve high yields of the desired product and effectively remove the remaining osmium tetroxide by treating the reaction mixture with a precipitant for osmium tetroxide,
treated like sodium sulfite, without the application of heat or long contact times. This gives high yields of products of improved color and composition without the 21-acyloxy group being substantially deacylated.
Theoretically, 2 moles of hydrogen peroxide are required for each mole of osmic acid ester formed in the reaction. However, it has been found that in the process of the present invention, more than the theoretical amount H_0 .. must be used in order to achieve a complete reaction. Therefore, to achieve the best results, you have to use an excess of hydrogen peroxide. The best results are usually obtained when using between 2.2 and 2.7 equivalents of hydrogen peroxide.
If less H2O "is used, the yield is reduced as a result of incomplete reaction. More than 2.75 molar equivalents of H2O" can be used, but these are undesirable, especially if one uses a 3-keto-d 4.i7 (zo ) - 21-acyloxy-steroid treated.
It has been observed with such steroids that excessive H2O concentrations tend to degrade the A-ring; However, this side reaction worsens the overall yield of the desired d 4-3,20-diketo-17a-oxy-21-acyloxy-steroid.
If desired, the H202 can be added slowly to the mixture of steroid, osmium tetroxide and amine, e.g. B. during a period of several minutes to a few hours. Sometimes the reaction proceeds more favorably under these conditions, especially if the starting steroid contains a 11-3-keto group which is supposed to remain intact. Normally, however, the H. = 0. can be added practically all at once, since the reaction rate is very rapid is high.

If you work under anhydrous conditions at the beginning, the yield of the desired 17-oxy-20-keto-steroid increases, while the yield of undesired 17,20-dioxy-steroid decreases accordingly.
A reaction temperature between about 15 and 30 C is preferred; but you can also work at slightly higher or lower temperatures, z. B. between about -10 C and about -I- 70 C. You work not only for reasons of expediency, but also to achieve optimal yields of 17-oxy-20-keto-steroid preferably at room temperature.
Any solvent which dissolves the reactants can be used in place of the solvents mentioned above. The term inert solvent corresponds to the second definition of solvent in Hackh's Chemical Dictionary, 3rd Edition and means a liquid that dissolves another substance without changing its chemical composition, in contrast to solvents that dissolve through chemical reaction.
Suitable other solvents are cyclohexanone, dioxane, tert: amyl alcohol, ethanol and methanol. Preferably, the solvent boils low enough to be removed from the reaction mixture by distillation under reduced pressure at room temperature without heating.
While the reaction of the 417 (20) -21-acyloxstereroids with osmium tetroxide and H., 02 is sluggish and requires at least 20 hours or more to complete, the reaction of a 417 (20) -21-acyloxysteroids with osmium tetroxide is and H, 02 in the presence of an aromatic tertiary N-heterocyclic amine usually terminated in less than an hour and sometimes in less than 5 minutes.
Sometimes you need a longer time; but even then, the total time will rarely, if at all, exceed 8 hours. The yield per unit time will therefore be in the lowest range about 0.15 / u per minute and sometimes up to 10% per minute.
If one considers that the previously known processes, at best, yields not significantly more than 0.015%. per minute, it can be seen that the presence of an aromatic tertiary-N-heterocyclic amine greatly accelerates the reaction. Thus, according to the process of the present invention, the desired 17-oxy-20-keto-steroid is produced at a rate that is some 10-600 times greater than with the known processes.
The use of the aromatic tert-N-heterocyclic amine not only brings about the abovementioned substantial shortening of the reaction time, but also brings about a substantial increase in the yield of 17-oxy-20-keto-steroid. So 11's response required;
-Oxy-21-acetoxy-4,17 (20) - pregnadien-3-one with H2O, and osmium tetroxide until the end of 48 hours with a yield of only 47% hydrocortisone acetate, while when working according to the present process after 2 or 6 hours reaction time the yields 53 resp.
60% (see example 1).
The pronounced increase in the reaction rate makes the process according to the invention technically more favorable and more economical than the known process. The surprising and noticeable increase in the yield of hydrocortisone acetate also makes the process economically valuable. The method can also be applied to other A17 (20) _21-acyloxy steroids with the same success.
The aromatic heterocyclic tert. Amines which are used according to the invention are all characterized in that they are practically not attacked by hydrogen peroxide under the reaction conditions. Primary, secondary and most of the non-aromatic tertiary amines are easily oxidized by H2O2 and react with the same, whereby the course of the reaction is disturbed.
In addition, the presence of a primary, secondary or tertiary amine other than the aromatic tertiary-N-heterocyclic amines appears to adversely affect the rate of the reaction.
The aromatic tertiary-N-heterocyclic amines, however, act in a surprising and advantageous manner as accelerators of the reaction and to increase the yield. This acceleration of the reaction rate also makes it possible to work practically table with catalytic amounts of osmium tetroxide, whereby 1. the osmium content of the product is significantly reduced, 2. greater economic efficiency is achieved and 3. the reaction can be controlled more reliably.
The d17 (20) -21-acyloxy-steroids, preferably of the Pregnen series, which can be converted into the corresponding 17a-oxy-20-keto-21-acyloxy-steroids by the process of the invention, can have the following formula

A preferred type of parent steroid is either saturated or contains only one 44 double bond which is coupled to a 3-keto group.
It is also advantageous if the preferred starting steroids only contain saturated acyl groups, since double bonds, whether they are in the nucleus or in the acyl radical, are sometimes also attacked by osmium tetroxide and H 2 O 2. Acetoxy group, an β-oxy group or a ketone oxygen (= 0), since these steroids can easily be converted into the physiologically active adrenal cortex hormones cortisone (Kendall's substance E) and hydrocortisone (Kendall's substance F) using the present method.
Likewise preferred are the di-analogs of the compounds shown in formula 1I in which R is an β-oxy group. These compounds can be converted into dl hydrocortisone or dl cortisone.
For example, the reaction of 21-acetoxy-4,17 (20) -pregnadiene-3,11-dione with 0s 04 and H.20 .., in the presence of pyridine by the process of the present invention gives cortisone acetate. In the same way, lss-oxy-21-acetoxy-4,17 (20)-pregnadien-3-one results in llss, 17a-dioxy-21-acetoxy-4-pregnen-3,20-dione (hydrocortisone acetate) ;
from 11a-oxy-21-acetoxy-4,17 (20) -pregnadien-3-one the 11a, 17a-Dioxy-21-acetoxy-4-pregnen-3,20-dione, which is easily oxidized with chromic acid to cortisone acetate (U.S. Patent No. 2602769 to Murray and Peterson, published July 8, 1952).
Since most physiologically active steroids have a 17a-oxy-20-keto-21-oxy or acyloxy grouping, the process according to the invention enables the conversion of d17 ('o) -21-acyloxy steroids into steroids which contain at least part of the have the structure required for the formation of a physiologically active steroid.
Since there are many synthetic methods for introducing a J4-3 keto group and an 11-oxygen atom into the steroid nucleus, as well as those for removing inactivating groups therefrom, steroids other than those represented by Formula 1I are also easily physiological via the method according to the invention active bark hormones can be transferred.
The J17 (2-21-acyl oxysteroids used in the process can be obtained in various ways, e.g. according to the reaction of Dimroth [Dimroth, reports, 71B, 1343 (1938)], who based on 17ss-oxy- 20-pregnen-steroids applied 21-oxy-, 21-acetoxy- or 21-bromo-17,20-pregnen-steroids delivers,
which can be converted into other 21-acyl oxy steroids by known processes [see Serini, Brit. Pat. No. 213630, Reports <B> 7111, </B> 1313, 1362 (1938), U.S. Pat. Nos. 2267258, 2305727, Brit. Pat. No. 467790, Miescher et al. Helv. Chim. Acta 22, 120, 894 (1939, Ruzicka and Müller, ibid., 22, 416 755 (1939)].
If desired, these compounds can also be obtained by the #method described below or by US Pat. No. 2,774,776.
Since it is usually advantageous to separate the osmium from the reaction product, the reaction mixture is usually treated with an agent which, such as sodium sulfite, hydrogen sulfide or the like, is capable of precipitating the osmium either as free osmium or as a cation of an insoluble salt. The usual procedure so far has been to increase the reaction mixture with aqueous sodium sulfite to higher temperatures, e.g. B. to the reflux temperature of the mixture to heat.
Under these conditions, an acetate group present in the reaction product is at least partially hydrolyzed, and you have to acetylate again in order to obtain a completely acetylated reaction product. According to a particular embodiment of the present invention, the reaction product is mixed with aqueous sodium sulfite at about room temperature or lower in order to remove the osmium from the mixture.
Under these conditions, the acetate or other readily hydrolyzable ester group in the molecule remains intact and the re-esterification required when working according to the earlier methods can be omitted, resulting in an improved yield of steroid product.
The use of low Os 04 concentrations in the reaction mixture, e.g. B. less than about 0.05 and preferably as little as 0.002 molar equivalents, calculated on the starting steroid, results in an insignificant loss of reaction product which can be found as an ester of osmic acid at the end of the reaction if the reaction mixture is not hydrolyzed .
If traces of osmium tetroxide are not undesirable, hydrolysis can be omitted using the preferred low concentrations, which further simplifies the process. The extraordinarily slow reaction in the processes used earlier generally prohibits the use of such low osmium tetroxide concentrations. Since the supply options for osmium tetroxide are somewhat limited and the product is toxic and dangerous, the use of these very weak concentrations makes it easier to adapt the process to industrial-scale requirements.
Preparation 11ss3 Oxy-21-acetoxy-4,17 (20) -pregnadien-3-one A solution of 0.518 g of 11ss', 21-dioxy-4,17 (20)-pregnadien-3-one, manufactured according to the USA patent No. 2705621, in 5 cm3 pyridine is mixed with 2 cm acetic anhydride and the whole thing is stored for 17 hours at room temperature and then crushed ice is added.
The precipitated 11ss-oxy-21-acetoxy-4,17 (20) -pregnadien-3-one is filtered off, dissolved in benzene and then chromatographed over a column of 75 g of synthetic magnesium sulfate (Florisil). The column is eluted with 75 cm? Portions of solvent of the following composition and sequence:
Benzene, three times hexane hydrocarbons Skellysolve B plus 1% acetone, Skellysolve B plus 5% acetone, Skellysolve B plus 10% acetone,
Skellysolve B plus 15% acetone, Skellysolve B plus 20% acetone and finally twice with acetone.
The fractions with 10 0/0 and 15% acetone are combined, the solvent is removed and the solid residue is crystallized from a mixture of ethyl acetate and Skellysolve B, the first fraction being 0.253 g (45% yield) of 11f3-Oxy- 21-acetoxy-4.17 (20)

21-Di-acetoxy-4,17 (20) -pregnadien-3-one is obtained in the same way by allowing a large molar excess of acetic anhydride to act on one molar equivalent.
Other esters of lla, 21-dioxy-4,17 (20)-pregnadien-3-one, 11ss, 21-dioxy-4,17 (20) -pregnadien-3-one and 21-oxy-4,17 (20 ) -pregnadien-3-ones are obtained if other acid anhydrides or acid halides are used in the reaction described above, such as propionic anhydride, benzoyl chloride, phthalic anhydride, succinic anhydride, oxalyl chloride,
Trimethylacetyl chloride, cyclopentanopropionyl chloride, diethylacetyl chloride, phenylpropionyl chloride and the like. The formic acid esters are generally obtained by using about 98% formic acid and an esterification catalyst.
The oxidation of 11-oxy-21-acyloxy-4,17 (20) - pregnadien-3-one with chromic acid provides the corresponding 21-acyloxy-4,17 (20) -pregnadiene, 3,11-dione. <I> Example 1 </I> a) To a solution of 744 mg (2.0 millimoles) of 1 lss-Oxy-21-acetoxy-4.17 (20) - pregnadiene - 3 - an in 30 cm3 of tert. Butyl alcohol is added 1 cm3 of pyridine and 25 mg (0.1 millimole)
0s 04 in 8 cm3 tert. Butyl alcohol. 8.36 cm3 (5.0 millimoles) of a 0.6 molar solution of 11.02 in tert. Butyl alcohol. The addition takes one hour and the mixture is then stirred for an additional hour.
The used tert. Butyl alcohol was dried with sodium and had a moisture content of about 0.04%.
The reaction mixture thus obtained was worked up immediately as follows: 600 mg of sodium sulfite dissolved in 25 cm3 of water are added, the mixture is stirred for 5 minutes and concentrated by distillation at about 50 mm Hg (absolute) to about 15 cm3. The concentrate is extracted with methylene chloride.
The extract is dried and chromatographed over 80 g of synthetic magnesium silicate (Florisu). The column is developed with ethylene chloride containing increasing amounts of acetone.
The starting steroid is found in the fraction that contains 2.5% acetone. Their amount is 1600 cm3 with a content of 140 mg of starting steroid (20%). The desired product,
Hydrocortisone acetate is in the fractions containing 711 / o acetone, with a total volume of 2000 cm3 and a content of 428 mg (53%) of hydrocortisone acetate. The yield, based on the starting steroid which has reacted, is 67% of theory.
The glycol by-product, 11ss, 17a, 20 - Trioxy - 21- acetoxy-4- RTI ID = "0005.0217" WI = "20" HE = "4" LX = "1128" LY = "2629"> pregnen-3- One is in the fractions containing 15% acetone, the total volume of which is 800 cm3 with a content of 101 mg (12.5%) of the by-product.
b) For comparison purposes, 11ss-oxy-21-acetoxy-4,17 (20) -pregnadien-3-one is converted into hydrocortisone acetate by the method of Miescher and Schmidlin using essentially the same amounts as under a).
To a solution of 744 mg (2.0 millimoles) 1 lss-Oxy-21-acetoxy-4,17 (20) - pregnadiene - 3 - an in 30 cm3 tert. Butyl alcohol is added at room temperature 6.18 cm3 (4.0 millimoles) of a 0.684 molar solution of H20, in sodium-dried tert. Butyl alcohol and then a solution of 52 mg (0.2 millimoles) dropwise
Os 04 in 8 cm3 dried over sodium tert. Butyl alcohol. The addition takes place over the course of 8 hours and the mixture is then kept at room temperature for a further 48 hours. The dried tert. Butyl alcohol had a moisture content of 0.04%.
The reaction mixture is then worked up exactly as under a). 53 mg (7%) of starting material, 397 mg (47%) of hydrocortisone acetate and 162 mg (20 ") / o) of the glycol by-product 11 ss, 17a, are obtained
20-Trioxy-21-acetoxy-4-pregnen-3-one.

Further reactions were carried out in which the conditions described under a) were changed abge. For example, it was found that, under optimal conditions, reaction times of just 5 minutes are sufficient to complete the reaction. It was also found that only 0.002 molar equivalents of Os 04, calculated on the starting steroid, are required in order to obtain the best yields of the desired product. Although the reaction speed drops somewhat at these low concentrations of Os 04, it can easily be increased again by increasing the reaction temperature.
It was also found that the addition of 0s 04 and amine before the addition of the H 2 O 2 sometimes increases the yield of the desired product. Although the reaction proceeds in a manner which is superior to that of the known processes even in the presence of significant amounts of water, it has been found that the yield of 17-oxy-20-keto product is improved when working under as anhydrous conditions as possible.
<I> Example 2 </I> If 21-acetoxy-4,17 (20) -pregnadiene-3,11-dione is used as starting material in the process of Example 1a, 17a-oxy-21-acetoxy-4- pregnen-3,11,20-trione (cortisone acetate) in the same yield as the hydrocortisone in example la.
<I> Example 3 </I> In the same way as in Example la, 17a-oxy-21-acetoxy-4-pregnen-3,20-dione (acetate of substance S) is obtained when using 21-acetoxy-4, 17 (20) -pregnadien-3-one as the starting steroid in the reaction described there.
<I> Example 4 </I> 11a, 17a-Dioxy-21-acetoxy-4-pregnen - 3.20 - dione is obtained according to the procedure given in Example la, if one la-Oxy-21-acetoxy- 4,17 (20) -pregnadien-3-one was used as the starting steroid.
