Dans le brevet Nu 330478, est décrit un procédé d'obtention d'alcoylthiocolchicéines substituées ou non, de formule générale I :

La présente invention a pour objet un procédé de préparation de composés nouveaux de la série des alcoylthiocolchicéines substituées ou non précipitées, qui résultent du remplacement du groupement acétyle R'par l'hydro- gène, ces nouveaux composés répondant également à la formule générale 1 dans laquelle R représente de l'hydrogène ou un radical alcoyle,
R'représente de l'hydrogène et R"représente un radical alcoyle substitué ou non.
Le procédé suivant l'invention est caracté- risé par le fait qu'on fait réagir un agent hydrolysant acide sur une alcoylthiocolchicéine répondant à la formule générale :

dans laquelle R représente de l'hydrogène ou un radical alcoyle et R"représente un radical alcoyle substitué ou non, pour désacétyler le groupement amino.
La structure des nouvelles N-désacétyl- alcoylthiocolchicéines est démontrée par le fait qu'en les traitant par l'anhydride acétique en présence de pyridine, on obtient par acétylation de la fonction amine ou par acétylation simultanée des fonctions amine et phénol, les alcoyl thiocolchicéines ou les acétyl-déméthyl-alcoyl- thiocolchicéines correspondantes, ces dernières fournissant, par saponification en milieu alcalin de Facétoxyle phénolique, les déméthyl-alcoylthiocolchicéines d'où l'on peut repasser aux alcoylthiocolchicéines par méthylation suivant des procédés connus.
Les nouveaux composés obtenus trouvent leur application dans le domaine biologique et industriel de la colchicine, notamment dans les modifications de la mitose et la création de polyploides, soit par épandage de solutions aqueuses ou de suspensions des produits sur des sols cultivés, soit par traitement préalable des semences avec les produits purs ou dilués dans un solvant ou sur support. Du point de vue physiologique, ces nouveaux composés présentent, en général, une toxicité nettement in férieure à celle des dérivés colchiciques antérieurement utilisés. En effet, ainsi qu'on le voit dans le tableau A, l'indice thérapeutique de la N-désacétyl-thiocolchicine est de beaucoup supérieur à celui de la colchicine. L'indice thérapeutique résulte du rapport dose léthale sur dose antimitotique.

Le procédé selon la présente invention conduit aux dérivés N-désacétylés de structure thiocolchicique avec d'excellents rendements.
Les exemples suivants illustrent l'invention.
Les points de fusion sont instantanés ; ils ont été déterminés sur bloc.
Exemple 1 : N-désacétyl-thocolchicine (formule I, R=CH3 ; R'=H. ; R"=-CH3)
à partir de la thiocolchicine
A 4, 88 g de thiocolchicine, on ajoute 60 cm de méthanol et 60 cm3 d'acide chlorhydrique 2 N. On porte 18 heures à reflux, puis on distille 40 cm3 de méthanol aqueux et extrait à 3 reprises avec, chaque fois, 100 cm3 de chloroforme. Les extraits chloroformiques réunis sont lavés 4 fois avec, chaque fois, 100 cm3 d'eau. On sèche le chloroforme et le distille à sec, ce qui permet de récupérer 400 mg de thiocolchicine brute.
La couche aqueuse épuisée au chloroforme et les eaux de lavage réunies sont additionnées de soude ION jusqu'à pH'13, puis extraites à 5 reprises avec, chaque fois, 100 cm3 de chloroforme. Les extraits chloroformiques réunis sont lavés 2 fois avec 50 cm3 d'eau chaque fois, séchés et distillés à sec. On obtient un résidu pesant 5, 2 g à cause de la solvatation chloroformique. Ce résidu, repris par 20 cm3 de chloroforme et additionné de 120 cm3 d'éther, cristallise aussitôt et fournit 3, 52 g de désacétyl-thiocolchicine (formule I, R=-CH3 ; R'=H ; R"=CH3), soit un rendement de 80'o/o. Le produit fond à 195 C.
Après recristallisation dans le mélange dioxane-éther, le point de fusion s'élève à 2000 C ; [a] 2D0 = -207 (c = 0, 5'o/o dans le chloroforme).

Exemple 2 :
Préparation
de la N-désacétyl-déméthylthiocolchicine (formule I, R=H, R'=H, R"=-CH3)
à partir de la démethylthiocolchicine
On chauffe pendant vingt-quatre heures au reflux 4, 2 g de déméthylthiocolchicine,
F. 308 C, [a] =-249 (c= 0, 5 /0, chloroforme), dans 60 cm3 de méthanol et 60 cm3 d'acide chlorhydrique 2N. Après élimination du méthanol, on neutralise par le bicarbonate de sodium et extrait au chloroforme qu'on évapore à sec.
Le résidu recristallisé en éthanol fournit 3, 1 g (80 o/o) de substance pure,
F. 185 C, [a] D =-213 (c=0, 5 /o, éthanol).
La N-désacétyl-déméthyl-thiocolchicine cristallise en aiguilles prismatiques, colorées en jaune orangé. Elle est soluble dans 15 volumes d'éthanol chaud et 100 volumes d'éthanol froid, soluble dans le chloroforme, insoluble dans l'éther, l'éther de pétrole ainsi que dans l'eau. Elle est par contre soluble dans les alcalis et les acides grâce à son caractère amphotère dû à la présence d'une fonction amine et d'un hydroxyle phénolique libres.

La N-désacétyl-déméthyl-thiocolchicine fournit un chlorhydrate soluble dans l'eau qu'on prépare en traitant une solution éthanolique de base par un léger excès d'acide chlorhydrique 2 N et en précipitant par l'éther.
Exemple 3 :
Préparation
de la N-dÚsacÚtyl-ÚthylthiocolchicÚine
Formule I (R=-CH3; R'=H; R"=-C2H5)
On dissout 700 mg d'ÚthylthiocolchicÚine dans 10 cm3 de méthanol et ajoute 10 cm3 d'acide chlorhydrique 2 N. On chauffe au reflux pendant seize heures, laisse refroidir et verse dans l'eau. La solution aqueuse est lavée au chloroforme, alcalinisée par la soude, puis extraite au chloroforme. L'extrait est lavé, séché et évaporé à sec. Le résidu est cristallisé dans un mélange d'acétate d'éthyle et d'éther et fournit 520 mg (83 0/0) de N-désacétyl-ethyl thiocolchicéine qui, après recristallisation, fond à 163 C, [a] D =-219 (c = 0, 5 /0, chloroforme).
Ce produit, qui n'a pas encore été décrit, se présente sous forme de cristaux jaunes, insolubles dans l'eau, l'éther, l'éther de pétrole, solubles dans les acides aqueux, le chloroforme, le diméthylformamide, l'acétone, solubles dans 9 volumes d'alcool à chaud.

On dissout 700 mg d'éthylthiocolchicéine dans 10 cm3 de méthanol et ajoute 10 cm3 d'acide bromhydrique 2 N. On chauffe au reflux pendant seize heures et poursuit le traitement comme il est indiqué dans l'exemple 3.
On obtient ainsi la N-désacétyl-éthylthiocolchi- céine, F. 163 C, [a] 20=-219o (c=0, 5, D/o, chloroforme) avec un rendement brut de 60 /o.
Ce produit est identique à celui décrit dans l'exemple précédent.

The present invention relates to a process for preparing new compounds of the series of substituted or non-precipitated alkylthiocolchiceins, which result from the replacement of the acetyl group R ′ by hydrogen, these new compounds also corresponding to general formula 1 in which R represents hydrogen or an alkyl radical,
R ′ represents hydrogen and R ″ represents a substituted or unsubstituted alkyl radical.
The process according to the invention is characterized by the fact that an acid hydrolyzing agent is reacted with an alkylthiocolchicein corresponding to the general formula:

in which R represents hydrogen or an alkyl radical and R "represents a substituted or unsubstituted alkyl radical, to deacetylate the amino group.
The structure of the new N-deacetyl-alkylthiocolchiceins is demonstrated by the fact that, by treating them with acetic anhydride in the presence of pyridine, the alkyl thiocolchiceins are obtained by acetylation of the amine function or by simultaneous acetylation of the amine and phenol functions. or the corresponding acetyl-demethyl-alkyl-thiocolchiceins, the latter providing, by saponification in an alkaline medium of the phenolic acetoxyl, the demethyl-alkylthiocolchiceins from which it is possible to switch back to the alkylthiocolchiceins by methylation according to known methods.
The new compounds obtained find their application in the biological and industrial field of colchicine, in particular in the modifications of mitosis and the creation of polyploids, either by spreading aqueous solutions or suspensions of the products on cultivated soils, or by preliminary treatment. seeds with the pure products or diluted in a solvent or on a support. From a physiological point of view, these new compounds show, in general, a toxicity markedly lower than that of the colchicium derivatives previously used. Indeed, as can be seen in Table A, the therapeutic index of N-desacetyl-thiocolchicine is much higher than that of colchicine. The therapeutic index results from the lethal dose to antimitotic dose ratio.

The process according to the present invention results in N-deacetylated derivatives with a thiocolchic structure in excellent yields.
The following examples illustrate the invention.
Melting points are instantaneous; they were determined on the block.
Example 1: N-deacetyl-thocolchicine (formula I, R = CH3; R '= H.; R "= - CH3)
from thiocolchicine
To 4.88 g of thiocolchicine, 60 cm3 of methanol and 60 cm3 of 2N hydrochloric acid are added. The mixture is refluxed for 18 hours, then 40 cm3 of aqueous methanol are distilled and extracted 3 times with, each time, 100 cm3 of chloroform. The combined chloroform extracts are washed 4 times with 100 cm3 of water each time. The chloroform is dried and distilled to dryness, which makes it possible to recover 400 mg of crude thiocolchicine.
The aqueous layer exhausted with chloroform and the combined washing waters are added with ION sodium hydroxide to pH ′ 13, then extracted 5 times with, each time, 100 cm3 of chloroform. The combined chloroform extracts are washed twice with 50 cm3 of water each time, dried and distilled to dryness. A residue weighing 5.2 g is obtained due to chloroform solvation. This residue, taken up in 20 cm3 of chloroform and added with 120 cm3 of ether, immediately crystallizes and provides 3.52 g of deacetyl-thiocolchicine (formula I, R = -CH3; R '= H; R "= CH3), i.e. a yield of 80%. The product melts at 195 C.
After recrystallization from a dioxane-ether mixture, the melting point rises to 2000 C; [a] 2D0 = -207 (c = 0.5% in chloroform).

N-desacetyl-demethylthiocolchicine (formula I, R = H, R '= H, R "= - CH3)
from demethylthiocolchicine
4.2 g of demethylthiocolchicine are heated for twenty-four hours at reflux,
F. 308 C, [a] = -249 (c = 0.5 / 0, chloroform), in 60 cm3 of methanol and 60 cm3 of 2N hydrochloric acid. After removing the methanol, neutralized with sodium bicarbonate and extracted with chloroform which is evaporated to dryness.
The residue recrystallized from ethanol gives 3.1 g (80 o / o) of pure substance,
F. 185 C, [a] D = -213 (c = 0.5 / o, ethanol).
N-deacetyl-demethyl-thiocolchicine crystallizes in prismatic needles, colored orange yellow. It is soluble in 15 volumes of hot ethanol and 100 volumes of cold ethanol, soluble in chloroform, insoluble in ether, petroleum ether as well as in water. On the other hand, it is soluble in alkalis and acids thanks to its amphoteric character due to the presence of an amine function and a free phenolic hydroxyl.

N-deacetyl-demethyl-thiocolchicine provides a water-soluble hydrochloride which is prepared by treating an ethanolic solution of base with a slight excess of 2N hydrochloric acid and precipitating with ether.
Example 3:
Preparation
N-dÚsacÚtyl-ÚthylthiocolchicÚine
Formula I (R = -CH3; R '= H; R "= - C2H5)
700 mg of ÚthylthiocolchicÚine are dissolved in 10 cm3 of methanol and 10 cm3 of 2N hydrochloric acid are added. The mixture is heated under reflux for sixteen hours, allowed to cool and poured into water. The aqueous solution is washed with chloroform, basified with sodium hydroxide, then extracted with chloroform. The extract is washed, dried and evaporated to dryness. The residue is crystallized from a mixture of ethyl acetate and ether and gives 520 mg (83 0/0) of N-deacetyl-ethyl thiocolchiceine which, after recrystallization, melts at 163 C, [a] D = - 219 (c = 0.5 / 0, chloroform).
This product, which has not yet been described, is in the form of yellow crystals, insoluble in water, ether, petroleum ether, soluble in aqueous acids, chloroform, dimethylformamide, acetone, soluble in 9 volumes of hot alcohol.

700 mg of ethylthiocolchiceine are dissolved in 10 cm3 of methanol and 10 cm3 of 2N hydrobromic acid is added. The mixture is heated under reflux for sixteen hours and the treatment is continued as indicated in Example 3.
There is thus obtained N-deacetyl-ethylthiocolchiein, mp 163 C, [a] 20 = -219o (c = 0.5, D / o, chloroform) with a crude yield of 60 / o.
This product is identical to that described in the previous example.
