Äthyl-, Methoxy-- oder Äthoxygruppen substituiert sein kann, bedeuten, sind bisher nicht bekanntgeworden. Wie gefunden wurde, stellen dieselben pharmakologisch, z. B. antiallergisch, wirk same Verbindungen dar. Vorliegende Erfin dung betrifft nun. die Herstellung dieser Verbindungen sowie ihrer Salze und quar- tären Ammoniumverbindungen.
Die netten Verbindungen lassen sieh nach verschiedenen Verfahren herstellen. So kann man ortho-substituierte Phenole der Formel

oder ihre Salze, insbesondere Alkali- und Erd- alkalimetallsalze, mit reaktionsfähigen Estern von Aminoalkanolen der allgemeinen Formel

insbesondere ihren Halogenwasserstoffsäure- estern, ferner z.
B. auch Alkyl-, oder Aryl- sulforsäureestern, Sehwefelsäureestern oder .Alkylschwefelsätzreestern, umsetzen. Bei Ver wendung der freien Phenole ist der Zusatz eines säurebindenden Mittels, wie z. B. Pottasche, von Vorteil.
Die als Ausgangsprodukte in Betracht kommenden ortho-substituierten Phenole der allgemeinen Formel II lassen sich z. B. durch Spaltung der zugehörigen Alkyl-, Aralkyl- oder Aryläther mit. üblichen Mitteln, wie z. B. Bromwasserstoff mit oder ohne Phos phor, oder Pyridinhydrochlorid herstellen.
Zu diesen Alkyl-, Aralkvl- oder Aryl- ä.thern gelang t man z. B. durch Kondensation von Alky 1-, Aralky 1- oder Aryläthern von in ortho-Stellung mit einer 0xymethylgruppe substituierten Phenolen mit einkernigen He- terocy elen mit l-'-)
ITeteroatomen. An Stelle der freien Oxymethylverbindungen können auch ihre Ester mit Chlor-, Brom- oder Jod- Wasserstoff verwendet werden. Geeignete Kondensationsmittel für diese Reaktion sind z. B. Zinkchlorid, Zinkgranalien, Eisenchlorid.
Eine weitere Methode zur Herstellung der vorgenannten Äther besteht in der Überfüh- rung von Alkyl-, Aralkyl- oder Aryläthern von in ortho-Stellung mit einer Halogen- methylgruppe substituierten Phenolen in ent sprechende metallorganische Verbindungen, z.
B. mit Magnesium, und Umsetzung der selben mit halogensubstituierten einkernigen Heterocvclen-mit 1-2 Heteroatomen nach Grignard.
Ferner kann man auch nach an sich be kannten Verfahren hergestellte Alkyl-, Aral- kyl- oder Aryläther von in ortho-Stellung mit einer Cyanmethylgruppe substituierten Phenolen mit Alkalimetallen, Alkaliamiden oder Alkalimetall-alkoholaten in entspre chende Metallverbindungen überführen, diese mit halogensubstituierten,
einkernigen He- terocyclen kondensieren und hierauf die Ni trilgruppe mit verdünnter Schwefelsäure ver seifen und die entstandene Säure gleichzeitig oder anschliessend decarboxylieren.
Als einkernige heterocyelische Reste mit <B>1-92</B> Fleteroatomen (Het--) seien beispiels- weise die von Pyrrol, Pyrrolidin, Pyridin, Piperidin, Pyrazolin, Imidazol, Pyridazin, Pyrimidin, Pyrazin, Furan, Tetrahydrofuran, Pyran, Thiophen,
Oxazol, Thiazol sowie ihren Methyl-, Äthyl-, Methoxy-, Äthoxy-, Chlor- oder Brom-Substitutionsprodukten sich ablei tenden Reste genannt.

Bromalkyl- oder Jodalkyläthern der ortho- substituierten Phenole der Formel II mit Tri- alkyl- oder Aralkyl-.dialkylaminen mit min destens 2 Alkylresten mit 1-4 C-Atomen oder durch Verwendung von quartären Ammoniumverbindungen von reaktionsfähi gen Estern von Dialkylaminoalkoholen der allgemeinen Formel III
in dem oben besehrie- benen Verätherungsverfahren an Stelle der tertiären basischen Verbindungen.
Mit anorganischen und organischen Säu ren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Essigsäure, Citronensäure Oxalsäure, Bernsteinsäure, Weinsäure, Benzoesäure, Phthalsäure bilden die tertiären Basisehen Äther gut kristallisie rende Salze, die in Wasser leicht löslich sind. Halogenhaltige basische Äther können auch durch nachträgliche Halogeniermg der halo genfreien Äther erhalten werden.
Gegenstand vorliegenden Patentes ist nun ein Verfahren zur Herstellung eines basischen Äthers eines ortho-substituierten Phenols. Das Verfahren ist dadurch gekennzeichnet, dass man einen reaktionsfähigen Ester des ss-Dimethylamino-äthanols mit einer den Rest

einführenden Verbindung veräthert.
Die erhaltene neue Verbindung, das 2-[o (ss-Dimethylamino-äthoxy) -benzyl] -thiophen, siedet unter 0,1 mm Druck bei 137-138 . Sie soll therapeutische Verwendung finden. keispiel: Unter gutem Rühren und Erwärmen am Rückfluss unter Feuchtigkeitsabschluss wird aus 20 Teilen 11ethylalkohol und 2,3 Teilen Natrium in 250 Teilen abs. Toluol das Alko- holat dargestellt.
Dann werden 19 Teile 2-(o- Oxy-benzyl)-thiophen zugegeben und der Me thylalkohol vollständig abdestilliert. Hierauf werden bei etwa 90-95 12 Teile ss-Dimethyl- amino-äthylehlorid zugetropft. Nach beendig tem Zutropfen wird noch 3 Stunden am Rück fluss erhitzt.
Nach dem Abkühlen und Ver setzen mit etwas Äther wird der basische Phenoläther mit total 300 Teilen 2n-Salzsäure in 2 Portionen aufgenommen, mit. konz. Na tronlauge unter guter Eiskühlung alkalisch gestellt und ausgeäthert. Die ätherische Lö sung wird mit Natriumsulfat getrocknet, ab filtriert, der Äther abdestilliert und der Rück stand der Vakuumdestillation unterworfen. Unter 0,1 mm Druck geht das 2-[O-(ss-Dl- methylamino - äthoxy) - benzyl] - thiophen bei 137-138" als leicht viskoses Öl über.
Das Chlorhydrat besitzt. den Schmelzpunkt. von 123-125 und ist in Wasser leicht löslich.
