EMI0003.0047
Beispiel 2: 4 g d'-3-Acetoxy-21-bromacetoxy-pregnen- 20-onund 6 g Zinkflitter werden im Hochvakuum getrocknet. Dann fügt man 40 cm3 Dioxan hinzu.
Nun fügt man 2 cm' abs.

where Ri, R2, Yi to Y3 and n have the meaning already mentioned and Me represents a metal atom.
Compounds with a 5- or 6-membered saturated and hydroxylated lactone ring or unsaturated lactone ring are thus obtained as process products. In addition to compounds with the 13, r-unsaturated lactone ring recorded above, compounds with a r, δ or α, 13 double bond can also be obtained from the compounds with a 6-membered hydroxylated latone ring.
The condensation is in a known manner on the type of Reformatzky reaction by means of metals such as zinc or magnesium, in a suitable inert Lö solvent such as. B. benzene, toluene, dioxane or ether, and subsequent treatment with dilute acid. Partial elimination of water generally takes place even during the condensation reaction. Mixtures are then obtained which can either be separated or processed further as such.
If necessary, the condensation product is then subjected to one of the known methods for splitting off water, of which the distillation or. Sublima tion under reduced pressure has proven to be particularly suitable, if necessary after mixing with surface-active Stof fen such as Aluininitttnoxyd, silica gel, activated carbon, fuller's earth, or with anhydrous salts such.
B. copper sulfate. To isolate pure compounds, the resulting reaction product can be subjected to the usual separation processes, such as, for example, fractional crystallization or cliromographic adsorption analysis.
The present patent relates to a process for the preparation of a lactone of the cyclopentanopolyhydrophenanthrene series, which is characterized in that a compound of the general formula

3 where X is a group that can be converted into a hydroxyl group by hydrolysis and Y is a halogen atom, treated with metals in the presence of an inert solvent, the resulting organometallic compound is mixed with dilute acid and the reaction product is subjected to dehydration and hydrolysis.
The new lactone obtained in this way, 45,6i20,22-3,21-dioxy-norcholadien-acid lactone, of F. 258-260, is identical to the compound prepared according to Patent No. 234780. It should find therapeutic use or serve as an intermediate for the preparation of therapeutically usable compounds.

(prepared for example from d-5 6-3-acetoxy-21-diazo-pregnen-20-one by reaction with bromoacetic acid) are dissolved in 20 cm 'absolute benzene and the solution is refluxed with 1.56 g of zinc flakes until the zinc is almost completely dissolved.
The benzene solution is carefully concentrated, the residue is mixed with absolute ethyl alcohol and the solution is filtered off from the zinc sludge. The alcoholic solution is poured into ice-cold dilute hydrochloric acid and the precipitated reaction product is taken up in ether. The ethereal layer containing alcohol is washed with plenty of water; the main amount of the reaction product precipitates as a white powder and is separated off by filtration. This product, which is relatively difficult to dissolve in ether, can be purified by crystallization from glacial acetic acid or absolute alcohol.
It then melts at 239 and represents the d'-3-acetoxy-20,21-dioxy-norcholenic acid lactone of the formula
EMI0003.0030
The elimination of water to give the corresponding unsaturated lactone takes place, for example, as was easily shown, by heating to 240 in vacuo. The unsaturated lactone, namely 45, ss2o, 22-3-acetoxy-21-oxy-norcholadienoic acid-lactone, from 173 to 174 m.
Acid saponification is used to obtain the / 5,6,20,22-3 21-dioxy-norcholadien-acid lactone from F.258-260. The following formula applies to him:
EMI0003.0047
Example 2: 4 g d'-3-acetoxy-21-bromoacetoxy-pregnen-20-one and 6 g zinc flakes are dried in a high vacuum. Then 40 cm3 of dioxane are added.
The reaction mixture is carefully heated and refluxed for a short time. Now add 2 cm 'abs. Add alcohol and cook on the water bath for another hour. Then 30 cms of the solution are distilled off, the contents of the flask in 80 cm 'abs. Dissolved alcohol and filtered off the zinc residue. The clear filtrate is concentrated in vacuo to about 40 cm3 and ether and dilute hydrochloric acid are added in a separating funnel.
After a short shake, a yellowish precipitate separates out of the ethereal solution in the form of rather coarse grains. It is filtered off, washed with ether until the yellow color disappears, then with dilute hydrochloric acid and water and finally dried in vacuo. This reaction product is then heated to 60 for 2 hours with acetic anhydride in pyridine.
The solution is then evaporated and the residue is chromatographed using a column of 60 g of aluminum prepared with benzene. Elution with ether gives the lactone of d5.B; 2n, 22-3_Acetoxy-21-oxy-norcholadienoic acid, which melts at 173-174, which is then converted to the lactone of 258-260 as described in Example 1 is hydrolyzed.
