Abstract

"VETOR DE áCIDO NUCLéICO, CéLULAS DE MAMìFEROS, PROCESSOS PARA A PREPARAçãO DE CéLULAS E DE UMA PROTEìNA DE INTERESSE, USO DE UM GENE, <225>-INTERFERON HUMANO, ERITROPOIETINA D HUMANA, E, COMPOSIçãO FARMACêUTICA". Proteínas como <225> interferon humano ou eritropoietina humana são preparados por cultivo de células de mamíferos que abrigam uma sequência de ácido nucleico compreendendo (i) uma sequência de codificação que codifica a proteína desejada e que é ligada de modo operativo a um promotor capaz de dirigir expressão da sequência de codificação em uma célula de mamíferos na presença de um íon de metal pesado; e (ii) uma primeira sequência de marcador selecionável compreende um gene de metalotioneína e que é ligada de modo operativo a um promotor capaz de dirigir expressão do gene de metalotioneína em uma célula de mamíferos na presença de um íon de metal pesado; e opcionalmente (iii) uma segunda sequência de marcador selecionável que compreende um gene neo e, que é ligada de modo operativo a um promotor capaz de dirigir expressão do gene neo em uma célula de mamíferos.
