Abstract

MÉTODOS E COMPOSIÇÕES PARA A MODIFICAÇÃO E DISTRIBUIÇÃO DE LINFÓCITOS. A presente divulgação fornece métodos e composições para modificação genética de linfócitos, por exemplo, células T e/ou células NK. Em algumas modalidades, os métodos incluem misturas de reação e formulações de células resultantes, que são criadas usando sangue total ou um componente do mesmo que não é uma PBMC e, adicionalmente, compreendem células T e partículas retrovirais recombinantes com polinucleotídeos que codificam um CAR. Em algumas modalidades, os linfócitos modificados são reintroduzidos em um sujeito por via subcutânea. Em algumas modalidades, são fornecidos polinucleotídeos que fornecem às células T a capacidade de regular a sobrevivência e proliferação celular em resposta à ligação a um CAR.
